At high...To design an accelerated method to evaluate thymopentin release from PLGA microspheres in vitro. Microspheres were prepared by double emulsion technique, using poly(lactide-co-glycolide) (PLGA) as carrier. At higher medium temperature (45℃, 50℃ and 55℃), an accelerated release testing in short time was studied and correlated with the conventional release (37℃) in vitro. The release in vitro of thymopentin from PLGA microspheres at 45 ℃, 50℃ and 55℃ was significantly accelerated (P 〈 0.05). In particular, at 50℃, an accelerated release (30 h) of the hydrophilic peptide from the PLGA matrix was achieved and correlated well with the conventional release (30 d). An accelerated release testing in vitro at higher temperature could be used to monitor thymopentin release from PLGA microspheres.展开更多

Results The plate insert has a stability of at least 15 d underculture condition. TEER increased significantly under co-culture condition from (66.1 +- 13.3)Ωcm^2 to (182.2 +- 6.7) Ωcm^2. Conclusion This micropore membrane culture plate insert can beeasily made, on which BCEC culture can be successfully performed. Moreover, it is adjustable andrecyclable. The spatiotemporal subcel...Using laser scanning confocal microscopy, we have found that the in cells loaded with fluo-3/AM, highest intracellular Ca(2+) in the perinuclear region is associated with the Golgi apparatus. The spatiotemporal subcellu lar distribution of Ca(2+) in living human fibroblasts exposing to calcium-free medium in response to agonists has been investigated. PDGF, which releases Ca(2+) from intracellular stores by inositol(1, 4, 5)-trisphosphate pathway,produced a biphasic transient rise in intracellular calcium.The initial rise was resulted from a direct release of calcium from the Golgi apparatus. Calcium could be also released from and reaccumulated into the Golgi apparatus by the stimulation of thapsigargin, an inhibitor of the Ca(2+) transport ATPase of intracellular calcium store. Permeablizing the plasma membrane by 10 μM digitonin resulted in the calcium release from the Golgi apparatus and depletion of the internal calcium store. These results suggest that the Golgi apparatus plays a role in Ca(2+) regulation in signal transduction.展开更多

The gain fac...The enhancement effect of the phase-conjugate wave produced by a third order nonlinear optical medium in a (AB)^N (D)^2 (BA)^N Bragg microcavity made of one-dimensional photonic crystals is studied. The gain factor of the phase-conjugate wave is derived when the resonance condition is satisfied by the incident optical wave and the Bragg microcavity mode. Because of stronger resonance enhancement to incident pump beam given by the Bragg microcavity, the enhancement effect of the phase-conjugate wave produced by a third order nonlinear optical medium inside Bragg microcavity is more outstanding than that caused by the naked third order nonlinear optical medium. The theoretical analysis and the numerical calculation show the Bragg microcavity with larger number N of periods is identical with a Fabry-Perot cavity when the resonance condition is satisfied by the incident optical wave and the Bragg microcavity mode.展开更多

METHODS: A novel rigid confocal probe (diameter 7 mm) was designed wit...AIM: To evaluate a newly developed hand-held confocal probe for in vivo microscopic imaging of the complete gastrointestinal tract in rodents. METHODS: A novel rigid confocal probe (diameter 7 mm) was designed with optical features similar to the flexible endomicroscopy system for use in humans using a 488 nm single line laser for fluorophore excitation, Light emission was detected at 505 to 750 nm. The field of view was 475 μm × 475 μm. Optical slice thickness was 7 μm with a lateral resolution of 0.7 μm. Subsurface serial images at different depths (surface to 250 μm) were generated in real time at 1024 × 1024 pixels (0.8 frames/s) by placing the probe onto the tissue in gentle, stable contact. Tissue specimens were sampled for histopathological correlation.RESULTS: The esophagus, stomach, small and large intestine and meso, liver, pancreas and gall bladder were visualised in vivo at high resolution in n = 48 mice. Real time microscopic imaging with the confocal minimicroscopy probe was easy to achieve. The different staining protocols (fluorescein, acriflavine, FITC-labelled dextran and L. esculentum lectin) each highlighted specific aspects of the tissue, and in vivo imaging correlated excellently with conventional histology. In vivo blood flow monitoring added a functional quality to morphologic imaging.CONCLUSION: Confocal microscopy is feasible in vivo allowing the visualisation of the complete GI tract at high resolution even of subsurface tissue structures. The new confocal probe design evaluated in this study is compatible with laparoscopy and significantly expands the field of possible applications to intra-abdominal organs. It allows immediate testing of new in vivo staining and application options and therefore permits rapid transfer from animal studies to clinical use in patients.展开更多

This microstructural coarsening is related to the thermoelectric current on the l/s interface, which is induced by difference of temperature on the inter- face. Within a magnetic field Lorentz force may be generated in front of the interface, causing a thermoelectric-magneto convection that makes the microstructure coarsened. It is inferred from the researches that the symbiot...Recent researches have shown that some compounds isolated from marine organisms have striking structural similarities with the metabolites from known microorganisms. It is inferred from the researches that the symbiotic or associated marine microorganisms may be the true sources of those compounds or at least involved in the biosynthesizing process. This view has been further evidenced by the researches for many sponges and sponge-associated microorganisms. Importantly, growing evidence has highlighted that the symbiotic or associated marine microorganisms live in the microenvironment within the hosts, and they also produce secondary metabolites which are new and original in structure and unique in activity. All these suggest that the microorganisms associated with marine organisms are the sources with very high potential to be new natural bioactive agents. This article reviews briefly the research advances in the study of new bioactive metabolites from marine organisms-associated microorganisms since 2000.展开更多

Methods: Fluoridated LnS+-doped HAP (Ln3+-HAP) NPs and iron oxides (lOs) can be encapsulated with biocompatible polymers via a modified solvent exaction/evaporation technique to prepare polymeric nanocomposites with fluoridated Ln3+-HAP/iron oxide. The nanocomposites were characterized for surface morphology, fluorescence spectra, magnetic properties and in vitro cytotoxicity. Magnetic targeted cellular imaging of such nanocomposites was also evaluated with confocal laser scanning microscope using A549 cells with or without magnetic field. Results: The fabricated nanocomposites showed good stability and excellent luminescent properties, as well as low in vitro cytotoxicity, indicating that the nanocomposites are suitable for biological applications. Nanocomposites under magnetic field achieved much higher cellular uptake via an energy-dependent pathway than those without magnetic field. Conclusion: 1tie nanocomposites fabricated in this study will be a promising tool for magnetic targeted cellular imaging with improved specificity and enhanced selection.展开更多

The effect of cholinergic agonists on [Ca2+]i was examined using laser scanning confocal microscopy and the Ca2+ sensitive dye Fluo-3.Results The results showed that the addition of acetylcholine (ACh) and carbachol (CCh), muscarnic and nicotinic agonists, induced [Ca2+]i increases in all the VHCs, whereas acetylcholine bromide (ACh-Br), a nicotinic agonist, induced the [Ca2+]i increase in only a small percentage of VHCs. The ACh or CCh-induced Ca2+ response could be partially suppressed by atropine. In the presence of 0.1?mmol/L atropine, the amplitudes of ACh or CCh-induced [Ca2+]i responses became significantly smaller than those in atropine free medium (P<0.01). Conclusions The results suggest the existence of cholinergic receptors in guinea pig VHCs. It is the muscarnic agonists rather than nicontic receptors that dominate [Ca2+]i variation. Fast Fourier transformation of high-resolution transmission elect...A novel and facile seed-mediated method for the preparation of monodispersed gold nanorods(GNRs) is presented by introducing pyrogallol as a reductant. Fast Fourier transformation of high-resolution transmission electron microscopy reveals that the synthesized GNRs are single crystalline. The longitudinal surface plasmon resonance of GNRs can be finely tuned by varying silver ion concentrations or seed amounts. Also, both thick(diameter >30 nm) and thin(diameter <10 nm) GNRs with exceptional monodispersity can be well prepared by this method. These findings indicate that this method has a greater performance in controlling the morphology of GNRs than that of traditional approach with ascorbic acid as a reductant.展开更多

Their compositions were determined by C, H elemental analyzer and EDTA titration. The infrared spectra, ultraviolet absorption spectra, and fluorescence spectra were also measured to identify the complexes. Elemental analysis showed that the compositions of these complexes were Tb(p-BrBA)3- H20, Tb(p-CIBA)3- 2H20, Tb(p-FBA)3- H20, Tb(o-FBA)3·2H20, Tb(o-CIBA)3· H20, and Tb(o-BrBA)3. H20, respectively. The monodispersed Ag@SiO2 core-shell nanoparticles with silica thicknesses of 10, 15, and 25 nm were success- fully prepared and characterized by transmission-electron microscopy. Fluorescence intensities of the complexes were detected before and after Ag@SiO2core-shell nanoparticles were added; the enhancement times were related to the silica-shell thick- ness. The fluorescence enhancement times were largest when the thickness of the silica shell was 25 nm. The mechanism may be attributed to the localized surface-plasmon resonance. Furthermore, the enhancement effect of terbium fluoro-benzoate complexes was the strongest in these complexes. This result may be attributed to the hydrogen bond between the hydroxyl on the surface of the silica shell and the fluorine atom.展开更多

Elemental analysis showed that the compositions of these complexes were Tb(p-BrBA)3- H20, Tb(p-CIBA)3- 2H20, Tb(p-FBA)3- H20, Tb(o-FBA)3·2H20, Tb(o-CIBA)3· H20, and Tb(o-BrBA)3. H20, respectively. The monodispersed Ag@SiO2 core-shell nanoparticles with silica thicknesses of 10, 15, and 25 nm were success- fully prepared and characterized by transmission-electron microscopy. Fluorescence intensities of the complexes were detected before and after Ag@SiO2core-shell nanoparticles were added; the enhancement times were related to the silica-shell thick- ness. The fluorescence enhancement times were largest when the thickness of the silica shell was 25 nm. The mechanism may be attributed to the localized surface-plasmon resonance. Furthermore, the enhancement effect of terbium fluoro-benzoate complexes was the strongest in these complexes. This result may be attributed to the hydrogen bond between the hydroxyl on the surface of the silica shell and the fluorine atom.
