[Method]The movemen...[Objective]The aim of this study was to identify transient expression of movement protein (MP) gene in Nicotinana benthaminana rapidly and further investigate the function of this exogenous gene. [Method]The movement protein gene of barley yellow dwarf virus (BYDV) was cloned into potato virus X (PVX) viral vector of pGR107,and PVX-recombinant vector was obtained. After electroporation of Agrobacterium tumefaciens,PVX was inoculated into the lower leaves of tobacco by Agrobacterium infiltration assay to observe the infection of virus on tobacco. [Result]After infection for 7 days,upper non-inoculated leaves of tobacco infected by the PVX-recombinant vector showed the virus infection symptoms,while the control group had no viral infection phenomenon. Daily follow-up observations for two groups revealed that tobacco infected by PVX-recombinant vector had severe symptoms of virus infection and curling leaves,or even led to necrosis both in infiltrated and systemic leaves in late period. However,tobacco infected by PVX vector had only slight symptoms of virus infection and could recover from infection. RT-PCR of the infected tobacco indicated that exogenous gene BYDV-MP had a normal transcription and expression in tobacco. [Conclusion]As a determinant factor for viral disease,BYDV-MP promotes the systemic infection rate of PVX and its symptom. In addition,it is feasible to express exogenous MP gene in Nicotiana benthaminan via PVX expression vector.展开更多

Genomic in situ hybridizat...The wheat_ Thinopyrum intermedium addition lines Z1,Z2 contain a pair of Th. intermedium chromosomes 2Ai_2 carrying the gene with resistance to barley yellow dwarf virus (BYDV). Genomic in situ hybridization (GISH) was used to analyze the chromosome constitution of Z1,Z2 by using genomic DNA probes from Th. intermedium and Pseudoroegneria strigosa . The results showed that the chromosome constitution of either Z1 or Z2 composes of 42 wheat chromosomes and two Th. intermedium chromosomes (2Ai_2). The 2Ai_2 chromosome is St_E intercalary translocation, in which the E genomic chromosome segment translocated into the middle region of the long arm of chromosome belonging to St genome. With the genomic DNA probe of Ps. strigosa , the GISH pattern specific to the 2Ai_2 chromosome may be used as a molecular cytogenetic marker. A detailed RFLP analysis on Z1, Z2 and their parents was carried out by using 12 probes on the wheat group 2 chromosomes. Twenty RFLP markers specific to the 2Ai_2 chromosome were identified. Two RAPD markers of OPR16 -350 and OPH09 -1580 , specific to the 2Ai_2 chromosome, were identified from 280 RAPD primers. These molecular markers could be used to assisted_select translocation lines with small segment of the 2Ai_2 chromosome and provide tools to localize the BYDV resistance.展开更多
Objective To investigate the effect of M5 muscarinic receptor subtype on the locomotor sensitization induced by heroin priming, and it's effect on the FosB expression in the nucleus accumbens (NAc) and the hippocam...Objective To investigate the effect of M5 muscarinic receptor subtype on the locomotor sensitization induced by heroin priming, and it's effect on the FosB expression in the nucleus accumbens (NAc) and the hippocampus in the heroin sensitized rats. Methods Locomotor activity was measured every 10 min for 1 h after subcutaneous injection of heroin. FosB expression was assayed by immunohistochemistry, and the antisense oligonucleotides (AS-ONs) targeting M5 muscarinic receptor was transferred with the lipofectin. Results Microinjection of AS-ONs targeting M5 muscarinic receptor in the ventral tegmental area (VTA) blocked the expression of behavioral sensitization induced by heroin priming in rats. Meanwhile, the expression of FosB-positive neurons in either the NAc or the dentate gyrus (DG) of the hippocam- pus increased in heroin-induced locomotor sensitized rats. The enhancement of FosB-positive neurons in the NAc or DG could be inhibited by microinjection of M5 muscarinic receptor AS-ONs into the VTA before the heroin-induced locomotor sensitization was performed. In contrast, microinjection of M5 muscarinic receptor sense oligonucleotide (S-ONs) into the VTA did not block the expression of behavioral sensitization or the expression of FosB in the NAc or DG in the heroin sensitized rats. Conclusion Blocking M5 muscarinic receptor in the VTA inhibits the expression of heroin-induced locomotor sensitization, which is associated with the regulation of FosB expression in the NAc and hippocampus neurons. M5 muscarinic receptor may be a useful pharmacological target for the treatment of heroin addiction.展开更多

Gexin III induced by palmin were studied at molecular and cellular level. The burst of superoxide, intercellular diffusion of hydrogen peroxide and process of cell death induced by palmin were investigated in tobacco plants by biochemical methods and Confocal microscopy. The results showed that a large amount of O-2(.-) was rapidly generated in tobacco cell elicited by palmin as a result of activation of NADPH oxidase, and the O-2(.-) was dismutated into H2O2 immediately by superoxide dismutase (SOD). Accumulation and intercellular diffusion of H2O2 were shown to be a trigger for hypersensitive cell death; and Ca2+ and some specific protein kinase were also shown to be involved in the activation of oxidative burst in tobacco cell induced by palmin.展开更多

The toxicities of sever-al germicides to Col etotrichum fragariae were determined with indoor mycelial growth inhibition assay. Field control test was preformed to further verify the control efficacy of germicides. [Result] The isolated strawberry anthracnose pathogenic strains were identified as C. gloeosporioide from genus Col etotrichum of subphylum Deuteromycotina. Prochloraz, pyrazole Kresoxim and hexaconazole al exhibited good indoor control effects; 25% pyrazole Kresoxim EC2000-fold solution, 24% azoxys-trobin·hexaconazole (azoxystrobin∶hexaconazole=1∶2) SC1500-fold solution and 25%prochloraz EC1000-fold solution al exhibited significant field control efficacy. [Result] Exogenous salicylic acid derivative increased the activities of PAL and POX, while did not influence the resistance to TMV. The crucian serum was treated at the levels, 0 μg L-1, 0.114 μg L-1, 0.57 μg L-1, 1.14 μg L-1 for 0 d 5 d 10 d 15 d and 20 d, respectively. The results showed that beta-cypermethrin could make great damage to crucian biochemical characteristics, and suggested that the effects of toxicant on serum GPT and GOT could be used as an index of toxicological assessment.展开更多

[Method] Chironomus sinicus larvae were exposed to different concentrations of clethodim, cypermethrin ...[Objective] The aim of this study was to detect the acute toxicity of clethodim, cypermethrin and 2,4-D-butylate. [Method] Chironomus sinicus larvae were exposed to different concentrations of clethodim, cypermethrin and 2,4-D-butylate. Survival rate, lethal concentration 50 (LC50) and superoxide dismutase (SOD) activity in the homogenate of the larvae were monitored to detect the toxicity of the pesti- cides. [Result] The survival rates of C. sinicus larvae exposed to cypermethrin and 2,4-D-butylate for 12 h were almost unchanged at different concentrations, but de- creased with concentration increasing 48 h later. For each pesticide, the differences in survival rates of C. sinicus larvae at 48 h were significant. The 48 h LC50 of clethodim, cypermethrin and 2,4-D-butylate to C. sinicus larvae were 1.842, 0.150 and 1.999 mg/L, respectively. The acute toxicity of cypermethrin was the highest a- mong the three pesticides and that of 2,4-D-butylate was the lowest. Clethodim sig- nificantly reduced the SOD content in a dose-dependent manner. 2,4-D-butylate also reduced SOD content in C. sinicus larvae, but the reduction was not significantly re- lated to its dosage. However, cypermethrin showed no significant effect on SOD ac- tivity in C. sinicus larvae.展开更多

The hemolytic substances produced by K. mikimotoi are thought...Karenia mikimotoi (Miyake & Kominami ex Oda) Hansen & Moestrup is associated with harmful algal blooms in temperate and subtropical zones of the world. The hemolytic substances produced by K. mikimotoi are thought to cause mortality in fishes and invertebrates. We evaluated the composition of the hemolytic toxin produced by K. mikimotoi cultured in the laboratory using thin-layer chromatography. In addition, we evaluated the effect of co-occuring algae (Prorocentrum donghaiense and Alexandrium tamarense) and the cladoceran grazer Moina mongoliea on hemolytic toxin production in K. mikimotoi. The hemolytic toxins from K. mikimotoi were a mixture of 2 liposaccharides and 1 lipid. Waterborne clues from P. donghaiense and A. tamarense inhibited the growth of K. mikimotoi but increased the production of hemolytic toxins. Conversely, K. mikimotoi strongly inhibited the growth of caged P. donghaiense and A. tamarense. In addition, the ingestion of K. mikimotoi by M. mongolica induced the production of hemolytic toxins in K. mikimotoi. Taken together, our results suggest that the presence of other microalgae and grazers may be as important as environmental factors for controlling the production of hemolytic substances. K. mikimotoi secreted allelochemicals other than unstable fatty acids with hemolytic activity. The production of hemolytic toxins in dinofiagellates was not only dependent on resource availability, but also on the risk of predation. Hemolytic toxins likely play an important role as chemical deterrents secreted by K. mikimotoi.展开更多

AIM:Ribavirin (RBV) shows a strong antiviral effect on hepatitis C virus when used in combination with interferon. However, RBV-induced anemia is a major problem in this therapy. It would be of great clinical importan...AIM:Ribavirin (RBV) shows a strong antiviral effect on hepatitis C virus when used in combination with interferon. However, RBV-induced anemia is a major problem in this therapy. It would be of great clinical importance to ameliorate the anemia without reducing the RBV dose. We report here that, Ninjinyoeito (NYT), a herbal medicine can reduce the RBV-induced anemia. METHODS: Twenty-three patients with chronic hepatitis C were treated with interferon alpha 2b plus RBV with (NYT group) or without (control group) NYT by a randomized selection. Eighteen patients completed the treatment schedule, and hemato-biochemical and virological effects were evaluated. RESULTS: There was no significant difference in biochemical and virological responses between the two groups. However, anemia was significantly reduced in the NYT group compared with the control group. The maximal decrease of Hb in the NYT group (2.59±1.10 g/dL) was significantly (P= 0.026) smaller than that in the control group (3.71±0.97 g/dL). There was no significant difference in serum glutathione peroxidase activity, serum RBV concentration, and Thl/Th2 balance between the two groups. There was no specific adverse effect in NYT administration. CONCLUSION: These results suggest that NYT could be used as a supportive remedy to reduce the RBV-induced anemia in the treatment of chronic hepatitis C.展开更多

To obtain expressed OCP in vitro, a...Sequences analysis revealed Grass carp reovirus (GCRV) s10 was 909 nucleotides coding a 34 kDa protein denoted as VP7, which was determined to be a viral outer capsid protein (OCP). To obtain expressed OCP in vitro, a full length VP7 gene was produced by RT-PCR amplification, and the amplified fragment was cloned into T7 promoted prokaryotic expression vector pRSET. The recombinant plasmid,which was named as pR/GCRV-VP7,was then transformed into E.coli BL21 host cells. The data indicated that the expressed recombinant was in frame with the N-terminal fusion peptide. The over-expressed fusion protein was produced by inducing with IPTG, and its molecular weight was about 37kDa, which was consistent with its predicted size. In addition, the fusion protein was produced in the form of the inclusion body with their yield remaining steady at more than 60% of total bacterial protein. Moreover,the expressed protein was able to bind immunologically to anti-his-tag monoclonal antibody (mouse) and anti-GCRV serum (rabbit). This work provides a research basis for further structure and function studies of GCRV during entry into cells.展开更多

The toxic mechanism of herbicide butachlor to induce extremely high lethality in marine flatfish flounder, Paralichthys Olivaceus, was analyzed by histopathological examination, antioxidant enzymes activities and ATP ...The toxic mechanism of herbicide butachlor to induce extremely high lethality in marine flatfish flounder, Paralichthys Olivaceus, was analyzed by histopathological examination, antioxidant enzymes activities and ATP content assay. Histopathological examination of gill, liver and kidney of exposed fishes showed that gill was a target organ of butachlor. The butachlor seriously impaired the respiration of gills by a series of lesions such as edema, lifting and detachment of lamellar epithelium, breakdown of pillar cells, and blood congestion. The dysfunction of gill respiration caused suffocation to the exposed flounder with extremely high acute lethality. Antioxidant enzyme activity assay of the in vitro cultured flounder gill (FG) ceils exposed to butachlor indicated that butachlor markedly inhibited the antioxidant enzyme activities of Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX). Furthermore, along with the decline of antioxidant enzyme activities, ATP content in the exposed FG cells decreased, too. This infers that the oxidative stress induced by butachlor can inhibit the production of cellular ATP. Similar decrease of ATP content was also observed in the exposed flounder gill tissues. Taken together, as in FG cells, butachlor possibly induced a short supply of ATP in pillar cells by inhibiting the antioxidant enzyme activities and then affecting the contractibility of the pillar cells, which in turn resulted in the blood congestion and suffocation of exposed flounder.展开更多

It was designated as CPV/WH02/06. This isolate was identified as serotype CPV-2a by the hemagglutination test, CPV Ag detection strip, electron microscopy, and PCR. The vp2 gene was cloned and sequenced and assigned GenBank accession number EU377537. A 1242 bp segment of the 5' region of the vp2 gene was cloned and inserted into the binary vector pBI121 and used for Agrobacterium-mediated tobacco transformation. Transgenic tobacco plants were selected on MS medium supplemented with 100 μg/mL kanamycin and 100 μg/mL timentin. Integration of the vp2 gene into the tobacco genome was confirmed by PCR using T1 progeny plants, and the expression of the VP2 protein was confirmed by Western blotting.展开更多

文摘[Objective] This study aimed to identify strawberry anthracnose pathogens and screen appropriate germicides. [Method] Strawberry anthracnose pathogens in Jurong City of Jiangsu Province were isolated and identified. The toxicities of sever-al germicides to Col etotrichum fragariae were determined with indoor mycelial growth inhibition assay. Field control test was preformed to further verify the control efficacy of germicides. [Result] The isolated strawberry anthracnose pathogenic strains were identified as C. gloeosporioide from genus Col etotrichum of subphylum Deuteromycotina. Prochloraz, pyrazole Kresoxim and hexaconazole al exhibited good indoor control effects; 25% pyrazole Kresoxim EC2000-fold solution, 24% azoxys-trobin·hexaconazole (azoxystrobin∶hexaconazole=1∶2) SC1500-fold solution and 25%prochloraz EC1000-fold solution al exhibited significant field control efficacy. [Con-clusion] The study provides reference for the alternate use of germicides in actual production.

文摘[Objective] This study aimed to evaluate the effects of exogenous salicylic acid derivatives on tobacco resistance to TMV and activity of defense enzymes. [Method] The tobboco leaves were treated by exogenous salicylic acid derivatives. Then, the disease occurrence was observed, and the activity of phenylalanin ammo- nia lyase (PAL) and peroxidase (POX) were measured. [Result] Exogenous salicylic acid derivative increased the activities of PAL and POX, while did not influence the resistance to TMV. [Conclusion] The result provides a theoretical basis for the study of plant disease resistance mechanisms.
文摘In this experiment, the effects of beta-cypermethrin on acute toxicity and serum GPT and GOT of crucian were studied. It was indicated that the 96hLC50 of beta-cypermethrin was 11.4 μg L-1. The crucian serum was treated at the levels, 0 μg L-1, 0.114 μg L-1, 0.57 μg L-1, 1.14 μg L-1 for 0 d 5 d 10 d 15 d and 20 d, respectively. The results showed that beta-cypermethrin could make great damage to crucian biochemical characteristics, and suggested that the effects of toxicant on serum GPT and GOT could be used as an index of toxicological assessment.

文摘[Objective] The aim of this study was to detect the acute toxicity of clethodim, cypermethrin and 2,4-D-butylate. [Method] Chironomus sinicus larvae were exposed to different concentrations of clethodim, cypermethrin and 2,4-D-butylate. Survival rate, lethal concentration 50 (LC50) and superoxide dismutase (SOD) activity in the homogenate of the larvae were monitored to detect the toxicity of the pesti- cides. [Result] The survival rates of C. sinicus larvae exposed to cypermethrin and 2,4-D-butylate for 12 h were almost unchanged at different concentrations, but de- creased with concentration increasing 48 h later. For each pesticide, the differences in survival rates of C. sinicus larvae at 48 h were significant. The 48 h LC50 of clethodim, cypermethrin and 2,4-D-butylate to C. sinicus larvae were 1.842, 0.150 and 1.999 mg/L, respectively. The acute toxicity of cypermethrin was the highest a- mong the three pesticides and that of 2,4-D-butylate was the lowest. Clethodim sig- nificantly reduced the SOD content in a dose-dependent manner. 2,4-D-butylate also reduced SOD content in C. sinicus larvae, but the reduction was not significantly re- lated to its dosage. However, cypermethrin showed no significant effect on SOD ac- tivity in C. sinicus larvae.

文摘Karenia mikimotoi (Miyake & Kominami ex Oda) Hansen & Moestrup is associated with harmful algal blooms in temperate and subtropical zones of the world. The hemolytic substances produced by K. mikimotoi are thought to cause mortality in fishes and invertebrates. We evaluated the composition of the hemolytic toxin produced by K. mikimotoi cultured in the laboratory using thin-layer chromatography. In addition, we evaluated the effect of co-occuring algae (Prorocentrum donghaiense and Alexandrium tamarense) and the cladoceran grazer Moina mongoliea on hemolytic toxin production in K. mikimotoi. The hemolytic toxins from K. mikimotoi were a mixture of 2 liposaccharides and 1 lipid. Waterborne clues from P. donghaiense and A. tamarense inhibited the growth of K. mikimotoi but increased the production of hemolytic toxins. Conversely, K. mikimotoi strongly inhibited the growth of caged P. donghaiense and A. tamarense. In addition, the ingestion of K. mikimotoi by M. mongolica induced the production of hemolytic toxins in K. mikimotoi. Taken together, our results suggest that the presence of other microalgae and grazers may be as important as environmental factors for controlling the production of hemolytic substances. K. mikimotoi secreted allelochemicals other than unstable fatty acids with hemolytic activity. The production of hemolytic toxins in dinofiagellates was not only dependent on resource availability, but also on the risk of predation. Hemolytic toxins likely play an important role as chemical deterrents secreted by K. mikimotoi.
文摘Shoot-tip culture of strawberry is the most important way to get virus-free plants.Moreover,detoxification and rapid propagation are the key techniques for producing high-quality seeds and seedlings massively and rapidly and achieving the high-quality,high-yield and high-efficiency development of strawberry industry.We have recently produced virus-free strawberry seedlings by combing tissue culture technology and stolon-reproduction method and this technology has been widely used.In addition,a relatively perfect three breeding system of virus-free strawberry seedlings was established after production practices,guaranteeing the supply of high-quality seeds and seedlings and technical support for strawberry industry in corps.

文摘AIM:Ribavirin (RBV) shows a strong antiviral effect on hepatitis C virus when used in combination with interferon. However, RBV-induced anemia is a major problem in this therapy. It would be of great clinical importance to ameliorate the anemia without reducing the RBV dose. We report here that, Ninjinyoeito (NYT), a herbal medicine can reduce the RBV-induced anemia. METHODS: Twenty-three patients with chronic hepatitis C were treated with interferon alpha 2b plus RBV with (NYT group) or without (control group) NYT by a randomized selection. Eighteen patients completed the treatment schedule, and hemato-biochemical and virological effects were evaluated. RESULTS: There was no significant difference in biochemical and virological responses between the two groups. However, anemia was significantly reduced in the NYT group compared with the control group. The maximal decrease of Hb in the NYT group (2.59±1.10 g/dL) was significantly (P= 0.026) smaller than that in the control group (3.71±0.97 g/dL). There was no significant difference in serum glutathione peroxidase activity, serum RBV concentration, and Thl/Th2 balance between the two groups. There was no specific adverse effect in NYT administration. CONCLUSION: These results suggest that NYT could be used as a supportive remedy to reduce the RBV-induced anemia in the treatment of chronic hepatitis C.

文摘Sequences analysis revealed Grass carp reovirus (GCRV) s10 was 909 nucleotides coding a 34 kDa protein denoted as VP7, which was determined to be a viral outer capsid protein (OCP). To obtain expressed OCP in vitro, a full length VP7 gene was produced by RT-PCR amplification, and the amplified fragment was cloned into T7 promoted prokaryotic expression vector pRSET. The recombinant plasmid,which was named as pR/GCRV-VP7,was then transformed into E.coli BL21 host cells. The data indicated that the expressed recombinant was in frame with the N-terminal fusion peptide. The over-expressed fusion protein was produced by inducing with IPTG, and its molecular weight was about 37kDa, which was consistent with its predicted size. In addition, the fusion protein was produced in the form of the inclusion body with their yield remaining steady at more than 60% of total bacterial protein. Moreover,the expressed protein was able to bind immunologically to anti-his-tag monoclonal antibody (mouse) and anti-GCRV serum (rabbit). This work provides a research basis for further structure and function studies of GCRV during entry into cells.

文摘[Objective] The study aimed to extract the statistical features of complete tobacco necrosis virus genome and conduct clustering analysis.[Method] On the base sequence of complete tobacco necrosis virus genome,by using three base groups which was composed of each base and its subsequent two bases,a new sequence S was arranged;the probability of all 64 kinds of three base groups occurred on S was calculated,a 64-dimensional vector L was obtained;L-vector of each genome was compared,8 three base groups were obtained,their probabilities have significant differences.[Result] The emergence probability of 8 three base groups (CGT;CAA;TAC;TAA;AGG;ATG;ATA;AAG) has significant association with genetic variation of tobacco necrosis virus genome;according to genetic variation result,4 different sources tobacco necrosis virus complete genome formed 2 major categories.[Conclusion] The study will provide theoretical basis for the control of tobacco necrosis virus.

文摘The toxic mechanism of herbicide butachlor to induce extremely high lethality in marine flatfish flounder, Paralichthys Olivaceus, was analyzed by histopathological examination, antioxidant enzymes activities and ATP content assay. Histopathological examination of gill, liver and kidney of exposed fishes showed that gill was a target organ of butachlor. The butachlor seriously impaired the respiration of gills by a series of lesions such as edema, lifting and detachment of lamellar epithelium, breakdown of pillar cells, and blood congestion. The dysfunction of gill respiration caused suffocation to the exposed flounder with extremely high acute lethality. Antioxidant enzyme activity assay of the in vitro cultured flounder gill (FG) ceils exposed to butachlor indicated that butachlor markedly inhibited the antioxidant enzyme activities of Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX). Furthermore, along with the decline of antioxidant enzyme activities, ATP content in the exposed FG cells decreased, too. This infers that the oxidative stress induced by butachlor can inhibit the production of cellular ATP. Similar decrease of ATP content was also observed in the exposed flounder gill tissues. Taken together, as in FG cells, butachlor possibly induced a short supply of ATP in pillar cells by inhibiting the antioxidant enzyme activities and then affecting the contractibility of the pillar cells, which in turn resulted in the blood congestion and suffocation of exposed flounder.
文摘A strain of canine parvovirus(CPV)was isolated from feces of an ill puppy in an animal hospital in Wuhan, China. It was designated as CPV/WH02/06. This isolate was identified as serotype CPV-2a by the hemagglutination test, CPV Ag detection strip, electron microscopy, and PCR. The vp2 gene was cloned and sequenced and assigned GenBank accession number EU377537. A 1242 bp segment of the 5' region of the vp2 gene was cloned and inserted into the binary vector pBI121 and used for Agrobacterium-mediated tobacco transformation. Transgenic tobacco plants were selected on MS medium supplemented with 100 μg/mL kanamycin and 100 μg/mL timentin. Integration of the vp2 gene into the tobacco genome was confirmed by PCR using T1 progeny plants, and the expression of the VP2 protein was confirmed by Western blotting.
