Tracing Changes in the Microbial Community of a Hydrocarbon-Polluted Soil by Culture-Dependent Proteomics 被引量:1

Hydrocarbon contamination may affect the soil microbial community, in terms of both diversity and function. A laboratory experiment was set-up, with a semi-arid control soil and the same soil but artificially contaminated with diesel oil, to follow changes in the dominant species of the microbial community in the hydrocarbon-polluted soil via proteomics. Analysis of the proteins extracted from enriched cultures growing in Luria-Bertani (LB) media showed a change in the microbial community. The majority of the proteins were related to gIycolysis pathways, structural or protein synthesis. The results showed a relative increase in the complexity of the soil microbial community with hydrocarbon contamination, especially after 15 days of incubation. Species such as Ralstonia solanacearum, Synechococcus elongatus and different Clostridium sp. were adapted to contamination, not appearing in the control soil, although Bacillus sp. dominated the growing in LB in any of the treatments. We conclude that the identification of microbial species in soil extracts by culture-dependent proteomics is able to partially explain the changes in the diversity of the soil microbial community in hydrocarbon polluted semi-arid soils, but this information is much more limited than that provided by molecular methods. 展开更多

Chemical compositions of rainwater collected in three stations in Taihu Lake in Jiangsu Province, China between March and May 2003 were analyzed through numerical simulations and field observation data. In terms of average ion deposition rate in spring at the air/water interface, of all anions, that of so4^2- was the largest followed by YO3^-, whereas among all cations, Ca^2+ concentration and the rate was the largest, and then NH4^+ was the next. The correlation of ion concentration indicated that the catchment of the lake has been artificially polluted considerably. Using backward trajectory analysis, the raining water in the stations in Taihu Lake was classified. In spring, marine-originated rain is the main contribution to this area, counting for 92.7% of the total precipitation, in which so4^2-, NO3^- and NH4^+ contributed 89.2%, 88.1%, and 88.3% respectively to the total spring-rain chemicals, whereas land-originated rains contributed in a small amount. However, the ion concentration in the land-originated rain was higher and acidic, causing considerable harm to local ecosystem. The analysis of backward trajectory analysis shows that three types of air masses influenced the chemical composition of the lake water, namely, air mass from NE direction, air mass from SW direction, and local air mass. Although the local air masses often produced small rainfall amount, but the nature of high ion concentration and high acidity impacted the local ecosystem remarkably. It has become a part of modern life that cannot be stopped or rejected. Then, how to correctly face the changes in the Internet era is placed in front of everyone. Students need to “learn how to use the media to get their all-round development, effectively prevent the foreign information pollution, and actively guide students to a truly rational attitude for WeChat, blog and micro-blog”. It is a kind of media literacy being confronted with the network’s ability to consciously resist the harmful information, but also to benefit their own development and firmly grasp the information, so as to make full use of the convenience of network self promotion. “Screen reading” network plays an important role in college students’ life, and the fragmentation, the text features of the writing ability of college students also have a strong impact; this paper focuses on the influence of college students in the network reading and writing patterns on their writing ability and influence. From the perspective of media literacy education for college students, writing ability and reading ability provide a feasible way. 展开更多

Several recent studies have pointed out that northern parts of Jordan are exposed to high levels of the effect of the hot spring, which is widely known to have severe impacts on health, vegetation and structures. North Jordan has a wealth of ancient heritage including six out of the ten Decapolis Cities. Air pollution that comes out from the hot springs is alleged to deteriorate and erode buildings, structures, statues and monuments through acid deposition. Therefore, this research was conducted to assess air quality at Gadara, the capital of the ancient Decapolis and suggest mitigation measures that have to be adopted in order to save the ancient heritage against further deterioration. Measurements revealed that most criteria air pollutants including sulfur dioxide, nitrogen dioxide and carbon monoxide attain corresponding national ambient air quality standards. Atmospheric concentrations of NO are strongly linked to the amount of 03. Simultaneous exposure to SO2 and NO2 demonstrated that a catalytic reaction takes place which increases the rate of sulphate production after 30 h exposure. 展开更多

AIM:To conduct a bacterial culture study for monitoring decontamination of automated endoscope reprocessors(AERs) after high-level disinfection(HLD).METHODS:From February 2006 to January 2011,authors conducted randomized consecutive sampling each month for 7 AERs.Authors collected a total of 420 swab cultures,including 300 cultures from 5 gastroscope AERs,and 120 cultures from 2 colonoscope AERs.Swab cultures were obtained from the residual water from the AERs after a full reprocessing cycle.Samples were cultured to test for aerobic bacteria,anaerobic bacteria,and mycobacterium tuberculosis.RESULTS:The positive culture rate of the AERs was 2.0%(6/300) for gastroscope AERs and 0.8%(1/120) for colonoscope AERs.All the positive cultures,including 6 from gastroscope and 1 from colonoscope AERs,showed monofloral colonization.Of the gastroscopeAER samples,50%(3/6) were colonized by aerobic bacterial and 50%(3/6) by fungal contaminations.CONCLUSION:A full reprocessing cycle of an AER with HLD is adequate for disinfection of the machine.Swab culture is a useful method for monitoring AER decontamination after each reprocessing cycle.Fungal contamination of AERs after reprocessing should also be kept in mind. 展开更多
