Background,aim,and scope Certain physicochemical indexes of topsoil are closely related to climatic factors including temperature,and precipitation.Understanding the relationship between modern topsoil properties and ...Background,aim,and scope Certain physicochemical indexes of topsoil are closely related to climatic factors including temperature,and precipitation.Understanding the relationship between modern topsoil properties and climatic factors is essential for quantitative paleoclimate reconstruction.Motuo located in the eastern Himalayas,exhibits a significant elevation gradient of over 7000 m from Nnamjagbarwa Peak(7782 m a.s.l.)to the Baxika(150 m a.s.l.).This region features a complete vertical zonation of vegetation,from alpine meadow to tropical forest,presenting an ideal place to investigate the relationships among vegetation,soil and climate conditions across altitudinal gradients.This study aims to explore the vertical variations in the physicochemical composition of topsoil and its relationship with temperature and precipitation.Materials and methods Twenty-seven topsoil samples were collected at 100 m intervals from 800 m to 3600 m a.s.l.along the southern slope of the Himalayas.Grain size,magnetic susceptibility and geochemical elements were measured to discuss the vertical variation characteristics of topsoil composition and their correlation with climatic factors.Results(1)The grain size of topsoil at different altitudes in Motuo is mainly composed of sand accounting for an averaged 53.2%,followed by silt and clay.(2)In the mixed forest zone,frequency dependent magnetic susceptibility(χfd%)shows a clear relationship with altitude,and clay is positively correlated with both altitude and climatic factors.(3)The oxides of topsoil in this area mainly consist of SiO_(2),Al_(2)O_(3) and Fe_(2)O_(3),followed by MgO,CaO,Na_(2)O and K_(2)O,with slight variations in the primary components at different altitudes.The sensitivity of elements to climate varies across different altitudes and vegetation zone,likely due to the region’s complex topography and vegetation.(4)Physical and biological weathering dominates in the broad-leaved forest zone of Motuo,while chemical weathering is more prominent in the coniferous forest zone,with the mixed forest zone falling in between.Discussion The formation of topsoil across the three vegetation zones is influenced by various factors,including parent material,vegetation,and climate.In the broad-leaved forest zone,physical weathering(precipitation,root wedging etc.)and pedogenesis dominate,resulting in finer grain size.The χ_(fd)% increases with altitude likely due to the high temperature and abundant precipitation in this zone,which facilitate the transformation of strong magnetic miners into weaker ones,particularly when the soil is oversaturated.Zirconium(Zr),primarily found in zircon,is depleted at lower altitudes by strong current erosion.Barium(Ba)is similarly reduced at low altitudes in this zone.In mixed forest zone,clay content is the lowest,indicating weaker physical weathering conditions than broad-leaved forest zone.The coarser grain size may result from the combined effects of topography and vegetation coverage.Magnetic susceptibility and organic matter show a positive correlation with altitude.Zr concentration is higher than that in the broad-leaved forest zone,likely resulting from decreased precipitation.In the coniferous zone,the clay content shows considerable fluctuations,with grain size generally becoming finer as altitude increases.This trend may be explained by intensified soil disintegration from seasonal freeze-thaw cycles.The χ_(fd)% values lack a clear trend or pattern,possibly due to soil erosion causing the migration of magnetic minerals or insufficient iron precipitation,which reduces the concentration of magnetic minerals in the soil.Additionally,a positive correlation is observed between altitude and organic matter content,with higher altitudes associated with greater organic matter accumulation.This may be attributed to lower microbial activity in colder conditions,which slows the decomposition and transformation of organic matter.Conclusions The variations in grain size,magnetic susceptibility,and geochemical elements differ across altitudes and vegetation zones,closely connected to the complex interplay of terrain,vegetation,and climate in Motuo.In the mixed forest,altitude has a significant impact onχfd%,and the clay component is particularly sensitive to changes in altitude,mean annual temperature,and precipitation.Zr shows a strong correlation with altitude and climate factors,making it a valuable indicator for assessing changes in atmospheric precipitation within specific altitude ranges.Recommendations and perspectives This study enhances our understanding of the relationships between the physicochemical properties of topsoil and climate conditions,offering valuable insights for paleoclimate reconstruction in Motuo.展开更多

Our present works show that the expression of E6/E7 oncogenes of human papillomavirus in HeLa cells is inhibited in the presence of anti-tumor reagent tripchlorolide (TC), which results in the up-regulation of p53 in HeLa cells. Interestingly, under the same TC-treatment, the cells at the early S-phase are more susceptible to apoptosis than those at the middle S-phase although p53 protein is stabilized to the same level in both situations. Significant difference is exhibited between the two specified expression profiles. Further analysis demonstrates that anti-apoptotic gene survivin is up-regulated by p53 in the TC-treated middle-S cells, whereas it is down-regulated by p53 in the TC-treated early-S cells. Taken together, the present study indicates that the differential p53-regulated expres- sion of survivin at different stages of the cell cycle results in different cellular outputs under the same apoptosis-inducer.展开更多
In this study,the reversal of monthly East Asian winter air temperature(EAWT) in 2020/21 and its predictability were investigated.The reversal of monthly EAWT in 2020/21 was characterized by colder temperatures in ear...In this study,the reversal of monthly East Asian winter air temperature(EAWT) in 2020/21 and its predictability were investigated.The reversal of monthly EAWT in 2020/21 was characterized by colder temperatures in early winter(December 2020 to mid-January 2021) and warmer temperatures in late winter(mid-January to February 2021).Results show that the reversal in the intensity of the Siberian high(SH) also occurred between early and late winter in 2020/21.In early winter,as the Barents-Laptev sea ice in the previous September(i.e., in2020) reached a minimum for the period 1981-2020,the SH was strengthaned via a reduction of the meridional gradient between the Arctic and East Asia.In late winter,as a sudden stratospheric warming occurred on 5 January 2021,the stratospheric polar vortex weakened,with the weakest center shifting to North America in January.Subsequently,the negative Arctic Oscillation-like structure shifted towards North America in the middle and lower troposphere,which weakened the SH in late winter.Furthermore,the predictability of the reversal in EAWT in 2020/21 was validated based on monthly and daily predictions from NCEP-CFSv2(National Centers for Environment Prediction-Climate Forecast System,version 2).The results showed that the model was unable to reproduce the monthly reversal of EAWT.However,it was able to forecast the reversal date(18 January 2021)of EAWT at lead times of 1-20 days on the daily scale.展开更多

RT-nested-PCR and Western blot assay were used to testify the HCV gene expression in Hela cells. The Hela cells expressing HCV structural protein were named Hela D cells. Prescriptions of Xiao chaihu Decoction (XCHD),Fufang Huangqi (FFHQ) and Bingganling (BGL) wererespectively added to Hela D cells in various concentrations. Semi-quantitative RT-nested-PCR product analysis was performed according to the fluorescent density between HCV DNA band and GAPDH DNA band in gel electrophoresisafter screened. RESULTS: Recombinant pBK-CMV-HCV could correctly express the HCV structural gene in Hela D cells. After coculture of Hela D cells with three prescriptional different concentrations for 48 h respectively, the transcription of HCVgene decreased with increasing of the concentration of each prescription. The lightness ratio of HCV product bands to GAPDH product bands was 0.24, 0.10 and 0.12 in Hela D cells incubated with 0.1 g/mL of XCHD, FFHQand BGL respectively and the lightness ratio HCV product bands to GAPDH product bands was 0.75, 0.67 and 0.61respectively in the control cells. CONCLUSION: The prescriptions of XCHD, FFHQ and BGL partly inhibit the transcription of HCV structural gene inHela D cells.展开更多

Methods: By polymerase chain reaction (PCR) technique, SjAslp was amplified from the constructed recombinant plasmid pBCSK+/SjAslp, and inserted into cloning vector pUCm-T. Then, SjAslp was subcloned into an eukaryotic expression vector pcDNA3.1(+). After identifying it by PCR, restrictive enzymes digestion and DNA sequencing, the recombinant plasmid was transfected into HeLa cells using electroporation, and the expression of the recombinant protein was analyzed by immunocytochemical assay. Results: The specific gene fragment of 558 bp was successfully amplified. The DNA vaccine of SjAslp was successfully constructed. Immunocytochemical assay showed that SjAslp was expressed in the cytoplasm of HeLa cells. Conclusion: SjAslp gene can be expressed in eukaryotic system, which lays the foundation for development of the SjAslp DNA vaccine against schitosomiasis.展开更多
To investigate the effects of elevated seawater p CO_2 on the early developmental stages of marine benthic calcifying organisms, we exposed the eggs and larvae of Argopecten irradias, an important bivalve species in C...To investigate the effects of elevated seawater p CO_2 on the early developmental stages of marine benthic calcifying organisms, we exposed the eggs and larvae of Argopecten irradias, an important bivalve species in Chinese aquaculture, in seawater equilibrated with CO_2-enriched(1000 ppm) gas mixtures. We demonstrated that elevated seawater pCO_2 significantly interfered with fertilization and larval development and resulted in an increased aberration rate. Fertilization in the treatment(pH 7.6) was 74.3% ± 3.8%, which was 9.7% lower than that in the control(p H 8.3)(84.0% ±3.0%). Hatching success decreased by 23.7%, and aberration rate increased by 30.3% under acidic condition. Larvae in acidified seawater still developed a shell during the post-embryonic phase. However, the shell length and height in the treatment were smaller than those in the control. The development of embryos differed significantly at 12 h after fertilization between the two experimental groups. Embryos developed slower in acidified seawater. Nearly half of the embryos in the control developed into D-shaped larvae at 48 h after fertilization, which were considerably more than those in the treatment(11.7%). Results suggest that future ocean acidification(OA) would cause detrimental effects on the early development of A. irradias.展开更多

The finite element model of a large-scale prestressing bucket founda- tion for offshore wind turbines is set up and the structural characteristics of the arc transition structure of the founda- tion are analyzed for 40-60 channels(20-30 rows) arranged with prestressing steel strand under the same ultimate load and boundary conditions. The mechanical characteristics of the key parts of the foundation structures are illus- trated by the peak of the principal tensile stress, the peak of the principal compressive stress and the distribution areas where the principal tensile stress is larger than 2.00 MPa. It can be concluded that the maximum principal tensile stress of the arc transition decreases with the increasing number of channels, and the amplitude does not change signifi- cantly; the maximum principal compressive stress increases with the increasing number of channels and the amplitude changes significantly; however, for the distribution areas where the principal tensile stress is larger than 2.00 MPa, with different channel numbers, the phenomenon is not obvious. Furthermore, the principal tensile stress at the top of the foundation beams fluctuantly increases with the increasing number of channels and for the top cover of the bucket, the principal tensile stress decreases with the increasing number of channels.展开更多

QY203. CgkP showed relatively high activity at acidities ranging from pH6.0 to pH9.0 and temperatures ranging from 30℃ to 50℃ with the highest activity at 45℃ and pH7.2. Sodium chloride increased its activity markedly, and KC1 increased its activity slightly. The divalent and trivalent metal ions including Cu^2+, Ni^2+, Zn^2+, Mn^2+, Al^3+ and Fe^3+ significantly inhibited its activity, while Mg^2+ did not. CgkP remained 70% of original activity after being incubated at 40℃ for 48h, and remained 80% of the activity after being incubated at 45℃ for 1 h. It exhibited endo-κ-carrageenase activity, mainly depolymerizing the κ-carrageenan into disaccharide and tetrasaccharide. CgkP was more thermostable than most of previously reported κ-carrageenases with a potential of being used in industry.展开更多

Information...This study was performed in five villages of Sandwip Upazila, Chittagong district with a view to identify the tree resources, utilization pattern and economic return of major fruit and timber tree species. Information collected from a total of 60 households ranging from marginal, small, medium and large categories. Number of plant species increased with the increase of homestead area. A total of 33 plant species were recorded from the homegarden, of which 19 were fruit and 14 were timber tree species. Considerable number of vegetables was also planted under the shade of the homestead trees. The investment analysis showed that average benefit-cost ratios were greater than one, net present values were positive and internal rate of returns were more than 10%. Long term investment on horticulture and timber tree species is highly profitable if species like Artocarpus heterophyllus, Cocos nucifera, Tectona grandis and Swietenia macrophylla are planted.展开更多
Using data from Argo and simple ocean data assimilation (SODA), the role of the barrier layer (BL) in the southeastern Arabian Sea (SEAS: 60°E-75°E, 0°-10°N) is investigated during the development ...Using data from Argo and simple ocean data assimilation (SODA), the role of the barrier layer (BL) in the southeastern Arabian Sea (SEAS: 60°E-75°E, 0°-10°N) is investigated during the development of positive Indian Ocean Dipole (IOD) events from 1960 to 2008. It is found that warmer sea surface temperature (SST) in the northern Indian Ocean appears in June in the SEAS. This warm SST accompanying anomalous southeastern wind persists for six months and a thicker BL and a corresponding thinner mixed layer in the SEAS contribute to the SST warming during the IOD formation period. The excessive precipitation during this period helps to form a thicker BL and a thinner mixed layer, resulting in a higher SST in the SEAS. Warm SST in the SEAS and cold SST to the southeast of the SEAS intensify the southeasterly anomaly in the tropical Indian Ocean, which transports more moisture to the SEAS, and then induces more precipitation there. The ocean-atmosphere interaction process among wind, precipitation, BL and SST is very important for the anomalous warming in the SEAS during the development of positive IOD events.展开更多

It occurs in two forms. Red Noctiluca is heterotrophic and fills the role of one of the microzooplankton grazers in the foodweb. In contrast, green Noctiluca contains a photosynthetic symbiont Pedinomonas noctilucae (a prasinophyte), but it also feeds on other plankton when the food supply is abundant. In this review, we document the global distribution of these two forms and include the first maps of their global distribution. Red Noctiluca occurs widely in the temperate to sub-tropical coastal regions of the world. It occurs over a wide temperature range of about 10℃ to 25℃ and at higher salinities (generally not in estuaries). It is particularly abundant in high productivity areas such as upwelling or eutrophic areas where diatoms dominate since they are its preferred food source. Green Noctiluca is much more restricted to a temperature range of 25℃-30℃ and mainly occurs in tropical waters of Southeast Asia, Bay of Bengal (east coast of India), in the eastern, western and northern Arabian Sea, the Red Sea, and recently it has become very abundant in the Gulf of Oman. Red and green Noctiluca do overlap in their distribution in the eastern, northern and western Arabian Sea with a seasonal shift from green Noctiluca in the cooler winter convective mixing, higher productivity season, to red Noctiluca in the more oligotrophic warmer summer season.展开更多
The distribution of hydrography and circulation in the eastern tropical Indian Ocean(ETIO) during April-May 2011 were analyzed using cruise observations,satellite observations,and historic hydrographic data.It was obs...The distribution of hydrography and circulation in the eastern tropical Indian Ocean(ETIO) during April-May 2011 were analyzed using cruise observations,satellite observations,and historic hydrographic data.It was observed that warm water(>28℃) occupies the upper 50-m layer in the ETIO.Low-salinity surface water was observed at the mouth of the Bay of Bengal(BOB),which further extends to the Arabian Sea and off Sumatra via the Sri Lanka coast and the eastern bay mouth.Arabian Sea high-salinity water(ASHSW) is carried eastward along the equator to around 90°E by the equatorial undercurrent(EUC).It also runs south of Sri Lanka(north to 3°N) and in the western bay mouth(west to 87°E) but is much shallower than its counterpart at the equator.It is suggested to be the residual of the ASHSW,which intrudes into the BOB during the preceding southwest monsoon.Our results also show that,in the south of Sri Lanka,just below this subsurface high-salinity water,very-low-salinity water(about 34.8) occurs at depths of 100-200 m.Further analysis suggests that this low-salinity water comes from the BOB.展开更多

Royal HaskoningDHV has developed regional hydrodynamic and wave models covering the Northern Arabian Sea to address these issues. Cyclone modelling was carried out on 11 major cyclones since 1945 and the tsunami modelling on an earthquake along the Makran Fault Line in 1945. Sample results from these modelling studies are presented in this paper. The methodology described in this article for modelling cyclones and tsunamis in the Arabian Sea could be applied to simulate these natural hazards at other sites around the world.展开更多

Kz7 by using degenerate and site-finding PCR. The gene was comprised of an open reading frame of 1224bp, encoding 407 amino acid residues, with a signal peptide of 24 residues. Based on the deduced amino acid sequence, the κ-carrageenase CgkS was classified into the Glycoside Hy- drolase family 16. The cgkS gene was expressed in Escherichia coli, and the recombinant enzyme was purified to homogeneity with a specific activity of 716.8 U mg-1 and a yield of 69%. Recombinant CgkS was most active at 45 ℃ and pH 8.0. It was stable at pH 6.0-9.0 and below 30℃. The enzyme did not require NaCl for activity, although its activity was enhanced by NaCI. CgkS degraded κ-carrageenan in an endo-fashion releasing tetrasaccharides and disaccharides as main hydrolysis products.展开更多

Cell a...OBJECTIVE To study the mechanism of apoptosis induced by cisplatin in vitro in HeLa cells cervical cancer cell line. METHODS The inhibitory effect of cisplatin on HeLa cell growth was analyzed by the MTT assay. Cell apoptosis was measured with flow cytometry and Hoechst 33258 staining following cisplatin treatment. The effect of cisplatin on transcription of HPV E6 was analyzed by RT-PCR and protein expression of E6, P53, p21, Bax and Bcl-2 was studied by Western blots. RESULTS Cisplatin inhibited cellular proliferation in a time and dose- dependant manner. The sub-G1 peak by flow cytometry showed a higher apoptotic rate in the experimental group compared to the controls and Hoechst 33258 staining indicated that apoptosis was induced by cisplatin. Results of RT-PCR demonstrated that cisplatin decreased tran- scription of E6. Western Blots showed that cisplatin decreased protein expression of E6 and increased protein expression of P53, p21 and Bax but had no effect on protein expression of Bcl-2. CONCLUSION Cisplatin induces apoptosis and death of HeLa cells through the suppression of HPV E6 and restoration of p53 function.展开更多
AIM: To investigate the gastrointestinal bleeding (GIB) in people from lowland to high altitude and in workers on Mountain Tanggula and its causes as well as treatment and prophylaxis.METHODS: From 2001 to October 200...AIM: To investigate the gastrointestinal bleeding (GIB) in people from lowland to high altitude and in workers on Mountain Tanggula and its causes as well as treatment and prophylaxis.METHODS: From 2001 to October 2003, we studied GIB in 13 502 workers constructing the railroad on Mountain Tanggula which is 4905 m above the sea level. The incidence of GIB in workers at different altitudes was recorded. Endoscopy was performed when the workers evacuated to Golmud (2808 m) and Xining (2261 m). The available data on altitude GIB were analyzed.RESULTS: The overall incidence of GIB was 0.49% in 13 502 workers. The incidence increased with increasing altitude. The onset of symptoms in most patients was within three weeks after arrival at high altitude. Bleeding manifested as hematemesis, melaena or hematochezia, and might be occult. Endoscopic examination showed that the causes of altitude GIB included hemorrhage gastritis, gastric ulcer, duodenal ulcer, and gastric erosion. Experimental studies suggested that acute gastric mucosal lesion (AGML) could be induced by hypoxic and cold stress, which might be the pathogenesis of altitude GIB. Those who consumed large amount of alcohol, aspirin or dexamethasone were at a higher risk of developing GIB. Persons who previously suffered from peptic ulcer or high-altitude polycythemia were also at risk of developing GIB. Early diagnosis, evacuation, and treatment led to early recovery. CONCLUSION: GIB is a potentially life threatening disease, if it is not treated promptly and effectively. Early diagnosis, treatment and evacuation lead to an early recovery. Death due to altitude GIB can be avoided if early symptoms and signs are recognized.展开更多

The effect of surface tension at the surface below the ice-cover is neglected. There exists only one wave number propagating at just below the ice-cover. A perturbation analysis is employed to solve the boundary value problem governed by Laplace's equation by a method based on Green's integral theorem with the introduction of appropriate Green's function and thereby evaluating the reflection and transmission coefficients approximately up to first order. A patch of sinusoidal ripples is considered as an example and the related coefficients are determined.展开更多
Features of atmospheric circulation and thermal structures are discussed using the NCAR/NCEP data to reveal the reasons for the late onset and anomalous southward persistence of the South China Sea Summer Monsoon(SCSS...Features of atmospheric circulation and thermal structures are discussed using the NCAR/NCEP data to reveal the reasons for the late onset and anomalous southward persistence of the South China Sea Summer Monsoon(SCSSM) in 2005.The results show that three factors are crucial.First,a strong Arabian High overlaps with a high-latitude blocking high and channels strong cold air to southern Asia.Second,the Tibetan Plateau has a bigger snow cover than usual in spring and the melting of snow cools down the surface.Third,the Somali Jet breaks out at a much later date,being not conducive to convection over Indochina.The former two factors restrict atmospheric sensible heating over the Tibetan Plateau and nearby regions while the third one limits latent heating over Indochina.All of the factors slow down atmospheric warming and postpone the onset of SCSSM.Long after the onset of SCSSM,strong cold air over India advances the Southwest Monsoon northward slowly,resulting in weaker convection and latent heating over the Tibetan Plateau and nearby areas.The negative feedback conversely inhibits further northward movement of Southwest Monsoon.展开更多
文摘Background,aim,and scope Certain physicochemical indexes of topsoil are closely related to climatic factors including temperature,and precipitation.Understanding the relationship between modern topsoil properties and climatic factors is essential for quantitative paleoclimate reconstruction.Motuo located in the eastern Himalayas,exhibits a significant elevation gradient of over 7000 m from Nnamjagbarwa Peak(7782 m a.s.l.)to the Baxika(150 m a.s.l.).This region features a complete vertical zonation of vegetation,from alpine meadow to tropical forest,presenting an ideal place to investigate the relationships among vegetation,soil and climate conditions across altitudinal gradients.This study aims to explore the vertical variations in the physicochemical composition of topsoil and its relationship with temperature and precipitation.Materials and methods Twenty-seven topsoil samples were collected at 100 m intervals from 800 m to 3600 m a.s.l.along the southern slope of the Himalayas.Grain size,magnetic susceptibility and geochemical elements were measured to discuss the vertical variation characteristics of topsoil composition and their correlation with climatic factors.Results(1)The grain size of topsoil at different altitudes in Motuo is mainly composed of sand accounting for an averaged 53.2%,followed by silt and clay.(2)In the mixed forest zone,frequency dependent magnetic susceptibility(χfd%)shows a clear relationship with altitude,and clay is positively correlated with both altitude and climatic factors.(3)The oxides of topsoil in this area mainly consist of SiO_(2),Al_(2)O_(3) and Fe_(2)O_(3),followed by MgO,CaO,Na_(2)O and K_(2)O,with slight variations in the primary components at different altitudes.The sensitivity of elements to climate varies across different altitudes and vegetation zone,likely due to the region’s complex topography and vegetation.(4)Physical and biological weathering dominates in the broad-leaved forest zone of Motuo,while chemical weathering is more prominent in the coniferous forest zone,with the mixed forest zone falling in between.Discussion The formation of topsoil across the three vegetation zones is influenced by various factors,including parent material,vegetation,and climate.In the broad-leaved forest zone,physical weathering(precipitation,root wedging etc.)and pedogenesis dominate,resulting in finer grain size.The χ_(fd)% increases with altitude likely due to the high temperature and abundant precipitation in this zone,which facilitate the transformation of strong magnetic miners into weaker ones,particularly when the soil is oversaturated.Zirconium(Zr),primarily found in zircon,is depleted at lower altitudes by strong current erosion.Barium(Ba)is similarly reduced at low altitudes in this zone.In mixed forest zone,clay content is the lowest,indicating weaker physical weathering conditions than broad-leaved forest zone.The coarser grain size may result from the combined effects of topography and vegetation coverage.Magnetic susceptibility and organic matter show a positive correlation with altitude.Zr concentration is higher than that in the broad-leaved forest zone,likely resulting from decreased precipitation.In the coniferous zone,the clay content shows considerable fluctuations,with grain size generally becoming finer as altitude increases.This trend may be explained by intensified soil disintegration from seasonal freeze-thaw cycles.The χ_(fd)% values lack a clear trend or pattern,possibly due to soil erosion causing the migration of magnetic minerals or insufficient iron precipitation,which reduces the concentration of magnetic minerals in the soil.Additionally,a positive correlation is observed between altitude and organic matter content,with higher altitudes associated with greater organic matter accumulation.This may be attributed to lower microbial activity in colder conditions,which slows the decomposition and transformation of organic matter.Conclusions The variations in grain size,magnetic susceptibility,and geochemical elements differ across altitudes and vegetation zones,closely connected to the complex interplay of terrain,vegetation,and climate in Motuo.In the mixed forest,altitude has a significant impact onχfd%,and the clay component is particularly sensitive to changes in altitude,mean annual temperature,and precipitation.Zr shows a strong correlation with altitude and climate factors,making it a valuable indicator for assessing changes in atmospheric precipitation within specific altitude ranges.Recommendations and perspectives This study enhances our understanding of the relationships between the physicochemical properties of topsoil and climate conditions,offering valuable insights for paleoclimate reconstruction in Motuo.

文摘Recent studies indicate that cell-cycle checkpoints are tightly correlated with the regulation of apoptosis, in which p53 plays an important role. Our present works show that the expression of E6/E7 oncogenes of human papillomavirus in HeLa cells is inhibited in the presence of anti-tumor reagent tripchlorolide (TC), which results in the up-regulation of p53 in HeLa cells. Interestingly, under the same TC-treatment, the cells at the early S-phase are more susceptible to apoptosis than those at the middle S-phase although p53 protein is stabilized to the same level in both situations. Significant difference is exhibited between the two specified expression profiles. Further analysis demonstrates that anti-apoptotic gene survivin is up-regulated by p53 in the TC-treated middle-S cells, whereas it is down-regulated by p53 in the TC-treated early-S cells. Taken together, the present study indicates that the differential p53-regulated expres- sion of survivin at different stages of the cell cycle results in different cellular outputs under the same apoptosis-inducer.

文摘In this study,the reversal of monthly East Asian winter air temperature(EAWT) in 2020/21 and its predictability were investigated.The reversal of monthly EAWT in 2020/21 was characterized by colder temperatures in early winter(December 2020 to mid-January 2021) and warmer temperatures in late winter(mid-January to February 2021).Results show that the reversal in the intensity of the Siberian high(SH) also occurred between early and late winter in 2020/21.In early winter,as the Barents-Laptev sea ice in the previous September(i.e., in2020) reached a minimum for the period 1981-2020,the SH was strengthaned via a reduction of the meridional gradient between the Arctic and East Asia.In late winter,as a sudden stratospheric warming occurred on 5 January 2021,the stratospheric polar vortex weakened,with the weakest center shifting to North America in January.Subsequently,the negative Arctic Oscillation-like structure shifted towards North America in the middle and lower troposphere,which weakened the SH in late winter.Furthermore,the predictability of the reversal in EAWT in 2020/21 was validated based on monthly and daily predictions from NCEP-CFSv2(National Centers for Environment Prediction-Climate Forecast System,version 2).The results showed that the model was unable to reproduce the monthly reversal of EAWT.However,it was able to forecast the reversal date(18 January 2021)of EAWT at lead times of 1-20 days on the daily scale.

文摘AIM: To investigate the inhibitory effect of Chinese herbal medicine on the transcription of hepatitis C virus (HCV) structural gene in Hela D cells.METHODS: Hela cell line was transfected with recombinant pBK-CMV-HCV containing HCV structural gene by Lipofectamine. RT-nested-PCR and Western blot assay were used to testify the HCV gene expression in Hela cells. The Hela cells expressing HCV structural protein were named Hela D cells. Prescriptions of Xiao chaihu Decoction (XCHD),Fufang Huangqi (FFHQ) and Bingganling (BGL) wererespectively added to Hela D cells in various concentrations. Semi-quantitative RT-nested-PCR product analysis was performed according to the fluorescent density between HCV DNA band and GAPDH DNA band in gel electrophoresisafter screened. RESULTS: Recombinant pBK-CMV-HCV could correctly express the HCV structural gene in Hela D cells. After coculture of Hela D cells with three prescriptional different concentrations for 48 h respectively, the transcription of HCVgene decreased with increasing of the concentration of each prescription. The lightness ratio of HCV product bands to GAPDH product bands was 0.24, 0.10 and 0.12 in Hela D cells incubated with 0.1 g/mL of XCHD, FFHQand BGL respectively and the lightness ratio HCV product bands to GAPDH product bands was 0.75, 0.67 and 0.61respectively in the control cells. CONCLUSION: The prescriptions of XCHD, FFHQ and BGL partly inhibit the transcription of HCV structural gene inHela D cells.

文摘Objective: To clone and construct the recombinant plasmid containing ATP synthase lipid-binding protein-like protein gene of Schistosomajaponicum,(SjAslp) and transfer it into mammalian cells to express the objective protein. Methods: By polymerase chain reaction (PCR) technique, SjAslp was amplified from the constructed recombinant plasmid pBCSK+/SjAslp, and inserted into cloning vector pUCm-T. Then, SjAslp was subcloned into an eukaryotic expression vector pcDNA3.1(+). After identifying it by PCR, restrictive enzymes digestion and DNA sequencing, the recombinant plasmid was transfected into HeLa cells using electroporation, and the expression of the recombinant protein was analyzed by immunocytochemical assay. Results: The specific gene fragment of 558 bp was successfully amplified. The DNA vaccine of SjAslp was successfully constructed. Immunocytochemical assay showed that SjAslp was expressed in the cytoplasm of HeLa cells. Conclusion: SjAslp gene can be expressed in eukaryotic system, which lays the foundation for development of the SjAslp DNA vaccine against schitosomiasis.

文摘To investigate the effects of elevated seawater p CO_2 on the early developmental stages of marine benthic calcifying organisms, we exposed the eggs and larvae of Argopecten irradias, an important bivalve species in Chinese aquaculture, in seawater equilibrated with CO_2-enriched(1000 ppm) gas mixtures. We demonstrated that elevated seawater pCO_2 significantly interfered with fertilization and larval development and resulted in an increased aberration rate. Fertilization in the treatment(pH 7.6) was 74.3% ± 3.8%, which was 9.7% lower than that in the control(p H 8.3)(84.0% ±3.0%). Hatching success decreased by 23.7%, and aberration rate increased by 30.3% under acidic condition. Larvae in acidified seawater still developed a shell during the post-embryonic phase. However, the shell length and height in the treatment were smaller than those in the control. The development of embryos differed significantly at 12 h after fertilization between the two experimental groups. Embryos developed slower in acidified seawater. Nearly half of the embryos in the control developed into D-shaped larvae at 48 h after fertilization, which were considerably more than those in the treatment(11.7%). Results suggest that future ocean acidification(OA) would cause detrimental effects on the early development of A. irradias.

文摘The key in the force transmission between the tower and the foundation for offshore wind turbines is to transfer the large moment and horizontal loads. The finite element model of a large-scale prestressing bucket founda- tion for offshore wind turbines is set up and the structural characteristics of the arc transition structure of the founda- tion are analyzed for 40-60 channels(20-30 rows) arranged with prestressing steel strand under the same ultimate load and boundary conditions. The mechanical characteristics of the key parts of the foundation structures are illus- trated by the peak of the principal tensile stress, the peak of the principal compressive stress and the distribution areas where the principal tensile stress is larger than 2.00 MPa. It can be concluded that the maximum principal tensile stress of the arc transition decreases with the increasing number of channels, and the amplitude does not change signifi- cantly; the maximum principal compressive stress increases with the increasing number of channels and the amplitude changes significantly; however, for the distribution areas where the principal tensile stress is larger than 2.00 MPa, with different channel numbers, the phenomenon is not obvious. Furthermore, the principal tensile stress at the top of the foundation beams fluctuantly increases with the increasing number of channels and for the top cover of the bucket, the principal tensile stress decreases with the increasing number of channels.

文摘A new extracellular κ-carrageenase, namely CgkP, 34.0 kDa in molecular weight, was purified from Pseudoalteromonas sp. QY203. CgkP showed relatively high activity at acidities ranging from pH6.0 to pH9.0 and temperatures ranging from 30℃ to 50℃ with the highest activity at 45℃ and pH7.2. Sodium chloride increased its activity markedly, and KC1 increased its activity slightly. The divalent and trivalent metal ions including Cu^2+, Ni^2+, Zn^2+, Mn^2+, Al^3+ and Fe^3+ significantly inhibited its activity, while Mg^2+ did not. CgkP remained 70% of original activity after being incubated at 40℃ for 48h, and remained 80% of the activity after being incubated at 45℃ for 1 h. It exhibited endo-κ-carrageenase activity, mainly depolymerizing the κ-carrageenan into disaccharide and tetrasaccharide. CgkP was more thermostable than most of previously reported κ-carrageenases with a potential of being used in industry.
文摘Tidal residual is very important to the transport of water particles, nutrients, plankton, etc. in the coastal sea. Eulerian scheme and Lagrangian scheme are two different ways to get the time averaged residual. Solution of the Bohai Sea’s hydrodynamic system using a semi implicit layer averaged numerical model yielded different direction Eulerian and Lagrangian tidal residuals. The latter were stronger than the former in most sea areas. Their different directions produced different circulation pattern in some areas. Compared with the Eulerian residual, the Lagrangian residual seemed to be more in accord with the observation.
文摘This study was performed in five villages of Sandwip Upazila, Chittagong district with a view to identify the tree resources, utilization pattern and economic return of major fruit and timber tree species. Information collected from a total of 60 households ranging from marginal, small, medium and large categories. Number of plant species increased with the increase of homestead area. A total of 33 plant species were recorded from the homegarden, of which 19 were fruit and 14 were timber tree species. Considerable number of vegetables was also planted under the shade of the homestead trees. The investment analysis showed that average benefit-cost ratios were greater than one, net present values were positive and internal rate of returns were more than 10%. Long term investment on horticulture and timber tree species is highly profitable if species like Artocarpus heterophyllus, Cocos nucifera, Tectona grandis and Swietenia macrophylla are planted.

文摘Using data from Argo and simple ocean data assimilation (SODA), the role of the barrier layer (BL) in the southeastern Arabian Sea (SEAS: 60°E-75°E, 0°-10°N) is investigated during the development of positive Indian Ocean Dipole (IOD) events from 1960 to 2008. It is found that warmer sea surface temperature (SST) in the northern Indian Ocean appears in June in the SEAS. This warm SST accompanying anomalous southeastern wind persists for six months and a thicker BL and a corresponding thinner mixed layer in the SEAS contribute to the SST warming during the IOD formation period. The excessive precipitation during this period helps to form a thicker BL and a thinner mixed layer, resulting in a higher SST in the SEAS. Warm SST in the SEAS and cold SST to the southeast of the SEAS intensify the southeasterly anomaly in the tropical Indian Ocean, which transports more moisture to the SEAS, and then induces more precipitation there. The ocean-atmosphere interaction process among wind, precipitation, BL and SST is very important for the anomalous warming in the SEAS during the development of positive IOD events.

文摘The dinoflagellate Noctiluca scintillans is one of the most important and abundant red tide organisms and it is distributed world-wide. It occurs in two forms. Red Noctiluca is heterotrophic and fills the role of one of the microzooplankton grazers in the foodweb. In contrast, green Noctiluca contains a photosynthetic symbiont Pedinomonas noctilucae (a prasinophyte), but it also feeds on other plankton when the food supply is abundant. In this review, we document the global distribution of these two forms and include the first maps of their global distribution. Red Noctiluca occurs widely in the temperate to sub-tropical coastal regions of the world. It occurs over a wide temperature range of about 10℃ to 25℃ and at higher salinities (generally not in estuaries). It is particularly abundant in high productivity areas such as upwelling or eutrophic areas where diatoms dominate since they are its preferred food source. Green Noctiluca is much more restricted to a temperature range of 25℃-30℃ and mainly occurs in tropical waters of Southeast Asia, Bay of Bengal (east coast of India), in the eastern, western and northern Arabian Sea, the Red Sea, and recently it has become very abundant in the Gulf of Oman. Red and green Noctiluca do overlap in their distribution in the eastern, northern and western Arabian Sea with a seasonal shift from green Noctiluca in the cooler winter convective mixing, higher productivity season, to red Noctiluca in the more oligotrophic warmer summer season.

文摘The distribution of hydrography and circulation in the eastern tropical Indian Ocean(ETIO) during April-May 2011 were analyzed using cruise observations,satellite observations,and historic hydrographic data.It was observed that warm water(>28℃) occupies the upper 50-m layer in the ETIO.Low-salinity surface water was observed at the mouth of the Bay of Bengal(BOB),which further extends to the Arabian Sea and off Sumatra via the Sri Lanka coast and the eastern bay mouth.Arabian Sea high-salinity water(ASHSW) is carried eastward along the equator to around 90°E by the equatorial undercurrent(EUC).It also runs south of Sri Lanka(north to 3°N) and in the western bay mouth(west to 87°E) but is much shallower than its counterpart at the equator.It is suggested to be the residual of the ASHSW,which intrudes into the BOB during the preceding southwest monsoon.Our results also show that,in the south of Sri Lanka,just below this subsurface high-salinity water,very-low-salinity water(about 34.8) occurs at depths of 100-200 m.Further analysis suggests that this low-salinity water comes from the BOB.
文摘Significant loss of life and damage to properties, ecosystems and marine facilities occur due to various natural hazards such as cyclones and tsunamis. Royal HaskoningDHV has developed regional hydrodynamic and wave models covering the Northern Arabian Sea to address these issues. Cyclone modelling was carried out on 11 major cyclones since 1945 and the tsunami modelling on an earthquake along the Makran Fault Line in 1945. Sample results from these modelling studies are presented in this paper. The methodology described in this article for modelling cyclones and tsunamis in the Arabian Sea could be applied to simulate these natural hazards at other sites around the world.

文摘A new κ-carrageenase gene cgkS was cloned from marine bacterium Shewanella sp. Kz7 by using degenerate and site-finding PCR. The gene was comprised of an open reading frame of 1224bp, encoding 407 amino acid residues, with a signal peptide of 24 residues. Based on the deduced amino acid sequence, the κ-carrageenase CgkS was classified into the Glycoside Hy- drolase family 16. The cgkS gene was expressed in Escherichia coli, and the recombinant enzyme was purified to homogeneity with a specific activity of 716.8 U mg-1 and a yield of 69%. Recombinant CgkS was most active at 45 ℃ and pH 8.0. It was stable at pH 6.0-9.0 and below 30℃. The enzyme did not require NaCl for activity, although its activity was enhanced by NaCI. CgkS degraded κ-carrageenan in an endo-fashion releasing tetrasaccharides and disaccharides as main hydrolysis products.
文摘OBJECTIVE To study the mechanism of apoptosis induced by cisplatin in vitro in HeLa cells cervical cancer cell line. METHODS The inhibitory effect of cisplatin on HeLa cell growth was analyzed by the MTT assay. Cell apoptosis was measured with flow cytometry and Hoechst 33258 staining following cisplatin treatment. The effect of cisplatin on transcription of HPV E6 was analyzed by RT-PCR and protein expression of E6, P53, p21, Bax and Bcl-2 was studied by Western blots. RESULTS Cisplatin inhibited cellular proliferation in a time and dose- dependant manner. The sub-G1 peak by flow cytometry showed a higher apoptotic rate in the experimental group compared to the controls and Hoechst 33258 staining indicated that apoptosis was induced by cisplatin. Results of RT-PCR demonstrated that cisplatin decreased tran- scription of E6. Western Blots showed that cisplatin decreased protein expression of E6 and increased protein expression of P53, p21 and Bax but had no effect on protein expression of Bcl-2. CONCLUSION Cisplatin induces apoptosis and death of HeLa cells through the suppression of HPV E6 and restoration of p53 function.

文摘AIM: To investigate the gastrointestinal bleeding (GIB) in people from lowland to high altitude and in workers on Mountain Tanggula and its causes as well as treatment and prophylaxis.METHODS: From 2001 to October 2003, we studied GIB in 13 502 workers constructing the railroad on Mountain Tanggula which is 4905 m above the sea level. The incidence of GIB in workers at different altitudes was recorded. Endoscopy was performed when the workers evacuated to Golmud (2808 m) and Xining (2261 m). The available data on altitude GIB were analyzed.RESULTS: The overall incidence of GIB was 0.49% in 13 502 workers. The incidence increased with increasing altitude. The onset of symptoms in most patients was within three weeks after arrival at high altitude. Bleeding manifested as hematemesis, melaena or hematochezia, and might be occult. Endoscopic examination showed that the causes of altitude GIB included hemorrhage gastritis, gastric ulcer, duodenal ulcer, and gastric erosion. Experimental studies suggested that acute gastric mucosal lesion (AGML) could be induced by hypoxic and cold stress, which might be the pathogenesis of altitude GIB. Those who consumed large amount of alcohol, aspirin or dexamethasone were at a higher risk of developing GIB. Persons who previously suffered from peptic ulcer or high-altitude polycythemia were also at risk of developing GIB. Early diagnosis, evacuation, and treatment led to early recovery. CONCLUSION: GIB is a potentially life threatening disease, if it is not treated promptly and effectively. Early diagnosis, treatment and evacuation lead to an early recovery. Death due to altitude GIB can be avoided if early symptoms and signs are recognized.
文摘Scattering of surface waves by the edge of a small undulation on a porous bed in an ocean of finite depth, where the free surface has an ice-cover being modelled as an elastic plate of very small thickness, is investigated within the framework of linearized water wave theory. The effect of surface tension at the surface below the ice-cover is neglected. There exists only one wave number propagating at just below the ice-cover. A perturbation analysis is employed to solve the boundary value problem governed by Laplace's equation by a method based on Green's integral theorem with the introduction of appropriate Green's function and thereby evaluating the reflection and transmission coefficients approximately up to first order. A patch of sinusoidal ripples is considered as an example and the related coefficients are determined.

文摘Features of atmospheric circulation and thermal structures are discussed using the NCAR/NCEP data to reveal the reasons for the late onset and anomalous southward persistence of the South China Sea Summer Monsoon(SCSSM) in 2005.The results show that three factors are crucial.First,a strong Arabian High overlaps with a high-latitude blocking high and channels strong cold air to southern Asia.Second,the Tibetan Plateau has a bigger snow cover than usual in spring and the melting of snow cools down the surface.Third,the Somali Jet breaks out at a much later date,being not conducive to convection over Indochina.The former two factors restrict atmospheric sensible heating over the Tibetan Plateau and nearby regions while the third one limits latent heating over Indochina.All of the factors slow down atmospheric warming and postpone the onset of SCSSM.Long after the onset of SCSSM,strong cold air over India advances the Southwest Monsoon northward slowly,resulting in weaker convection and latent heating over the Tibetan Plateau and nearby areas.The negative feedback conversely inhibits further northward movement of Southwest Monsoon.
