Objective To examine the relationship between Tpeak- Tend interval (Tpe) and Tpe/QT ratio with occurrence of ventricular premature beats (VPBs) and left ventricular remodeling in hypertension. Methods A total of 52 patients with mild to moderate essential hypertension were included, undergoing echocardiography and 24-hours Holter monitoring. Ventricular remodeling was assessed by left ventricular mass index (LVMI) using the Devereux formula and diastolic fimction by transmitral E and A wave velocities and E/A ratio. Tpe was measured in the precordial leads. The end of the T wave was set by the method of the tangent to the steepest descending slope of the T wave. Results Tpe and Tpe/QT in leads V2 (r = 0.33, P = 0.01; r = 0.27, P = 0.04 respectively) and V3 (r = 0.40, P = 0.002; r = 0.40, P = 0.003, respectively) correlated significantly with LVMI. A significant inverse relationship was observed between E/A ratio and QT (r = -0.33, P = 0.01), Tpe in V3 (r = -0.39, P = 0.003) and Tpe/QT in V3 (r = -0.31, P = 0.02). Tpe in V3, V5, mean Tpe and maximum Tpe with cut-offvalues of 60 ms, 59 ms, 62 ms and 71 ms, respectively, associated with the occurrence of ventricular premature beats. Conclusions The repolarization parameters Tpe interval and Tpe/QT ratio correlate with LVMI and indices of left ventricular diastolic function and show better predictive values than traditional parameters such as QT interval and QT dispersion. Lead V3 is the best lead for measuring Tpe and Tpe/QT. These ECG indices can therefore be used in clinical practice to monitor LV remodeling and predict occurrence of VPBs. 展开更多

However, the genetic model of fruit shapes has not been proposed. Therefore, in the present study, analysis of genetic effects for fruit shape traits (fruit length and fruit perimeter) in sponge gourd was conducted by employing a developmental genetic model including fruit direct effects and maternal effects. Analysis approaches of unconditional and conditional variances were applied to evaluate the genetic behavior of fruit shape traits at economical and physiological maturation times. The results of variance analysis indicated that fruit length and fruit perimeter were simultaneously affected by fruit direct genetic effects and maternal effects. Fruit direct genetic effects were relatively more important for fruit shape traits at whole developmental period. The gene expression was most active at the economical maturation stage (1-12 d after flowering) for two shape traits, and the activation of gene was mostly due to direct dominance effects at physiological maturation stage (13-60 datter flowering). The coefficients due to different genetic effects, as well as the phenotypic correlation coefficients, varied significantly between fruit shape traits themselves at various maturation stages. The results showed that it was relatively easy to improve fruit shape traits for industrial purpose by carefully selecting the parents at economical maturation stage instead of that at physiological maturation stage. 展开更多

No p34cdc2, H1 kinase activity was detected in the oocytes after progesterone stimulation or OA microinjection; Western blotting analysis showed that the level of p3cdc2, and p33 in the oocytes are significantly lower than those in the oocytes derived from the hibernating toads (below 10 ℃). 25S-Met incorporation analysis showed that when the oocytes were incubated at 6℃, synthesis of about thirty defferent polypeptides was promoted or induced, including p34cdc2 and some other p13suc1-binding proteins. All these results indicated that a low temperature environment is essential for the oocytes of Bufo Bufo gargarizans to express and store some cell cycle drivers and its regulators, and to gain the maturation competence. These results will also provide a new clue for explaining the molecular mechanisms why gametogenesis of some organisms depends on a relative low temperature and how to maintain the geographical distribution of some animals. 展开更多

We conducted a phenological evaluation of Sargassum thunbergii, a common algal species, along the coast of Lidao Bay, Rongcheng, China. The local S. thunbergii population entered a maturation period from early June to mid-August, when seawater temperature was 14.4 to 25. 1℃. Our results suggest an increase in temperatttre and day length during this period initiated thalli maturation. Inhabitants of the low tidal zone had higher thalli length and biomass compared with those from the mid- and high tidal zones. We observed a switch in the length frequency distribution and contribution of length classes to biomass between the small (〈20 cm) and long (20-79.9 cm) length classes at the time of thallus maturity. This suggests there is a trade-off between sexual reproduction and vegetative regeneration. 展开更多

Human pluripotent stem cells represent a potentially unlimited source of functional pancreatic endocrine lineage cells. Here we report a highly efficient approach to induce human embryonic stem (ES) cells and induced pluripo- tent stem (iPS) cells to differentiate into mature insulin-producing cells in a chemical-defined culture system. The differentiated human ES cells obtained by this approach comprised nearly 25% insulin-positive cells as assayed by flow cytometry analysis, which released insulin/C-peptide in response to glucose stimuli in a manner comparable to that of adult human islets. Most of these insulin-producing cells co-expressed mature β cell-specific markers such as NKX6-1 and PDX1, indicating a similar gene expression pattern to adult islet β cells in vivo. In this study, we also demonstrated that EGF facilitates the expansion of PDXl-positive pancreatic progenitors. Moreover, our protocol also succeeded in efficiently inducing human iPS cells to differentiate into insuIin-producing ceils. Therefore, this work not only provides a new model to study the mechanism of human pancreatic specialization and maturation in vitro, but also enhances the possibility of utilizing patient-specific iPS cells for the treatment of diabetes. 展开更多

Plantains (Musa paradisiaca L.) are a major food staple in West Africa and are cooked in various forms. The objective of this work was to evaluate the frying characteristics of plantains at different stages of ripening. The plantains used in the study were at the unripe stage through four different ripening stages. The samples were peeled and sliced into 2 mm thickness and blanched in hot water at 70 ℃ for 3 min. The slices were then deep flied in canola oil at 180 ℃. The result showed that ripening stage significantly affected moisture loss and fat absorption profiles of the plantain chips. Fully ripened plantain absorbed up to 34% (db) oil during 4 min frying, much higher than unripe plantain. The oil uptake and moisture loss during frying of the plantain chip samples were modelled using 1st order kinetics. The kinetic parameters including rates of moisture loss and oil uptake varied according to the different stages of ripening. Ripening had a significant effect on the colour lightness (L) of the chips. Similarly, the redness (a), the yellowness (b) and textural characteristics were significantly affected by ripening stage. 展开更多

BF's were analyzed for pH, total soluble solids (TSS), water holding capacity (WHC) and oil holding capacity (OHC) at 40℃, 60 ℃ and 80 ℃, color values L*, a* and b*, back extrusion force and viscosity. Physicochemical data were analyzed by Multivariate Analysis of Variance, discriminant analysis and cluster analysis. All statistical analyses showed that physicochemical properties of BF prepared from different variety and stage of ripeness were different from each other. Viscosity, WHC40, WHC60 and TSS were recommended methods for discrimination between banana flour prepared from the two varieties, whilst viscosity, WHC60 and WHC80 were suggested for differentiation of banana flour prepared using green and ripe banana. 展开更多

In wine production, the typical characteristics of variety, defined by its place of their origin, contribute to the development of distinctive and unique wines. In the current study, we analyzed the effect of the environment using vine response and grape composition as indicators. Four cv. Tannat vineyards in three different climatic regions of Uruguay with similar soil conditions were studied in 2008 and 2009. Vines grafted onto SO4 (Vitis berlandieri × Vitis riparia) rootstock and were trained on a trellis system. Weather information was obtained from weather stations (MMO standards). At each vineyard, we recorded: yield per plant, pruning weight, leaf area and pre-dawn leaf water potential. We analyzed sugars, total acidity and pH, polyphenolic potential, organic acids and berry weight. Analysis of variance, Pearson correlations and discriminant analysis were carried out. The climate factors with the highest discriminant weight were water balance, degree days (〉 10 ℃) of maturation and rainfall during the vegetative growth period. Plant response allowed us to discriminate between vineyards regardless of the year and was consistent with climate. Exposed leaf area and length of maturation period were the indexes with the highest values, followed by leaf water potential and grape yield. The total anthocyanin content, sugar contents and their daily accumulation, and acid composition statistically separate regions regardless of the year. We concluded that plant response and grape composition were strongly influenced by water supply and thermal conditions during ripening. 展开更多

Effect of Pork Meat pH on Iron Release from Heme Molecule during Cooking

This study investigated the relationship between the level of iron chemical forms and the initial raw meat pH values during cooking, including the effect of the sodium nitrite presence. The pH value, the heme iron (HI) and non-heme iron (NHI) concentrations were measured on one ground portion of five loin, five masseter, five semimembranosus, five neck muscles and five shoulder muscles. Afterwards, all raw meats were halved and mixed with NaCl (2%), ascorbic acid (0.05%) and 0 mg/kg of sodium nitrite (mix1) or 150 mg/kg of sodium nitrite (mix2). Both mixes were divided into 50 g-portions, vacuum packed in cryovac bags and cooked in thermostatic bath up to F (10, 71 ℃) = 30 minutes at the core. The NHI and HI concentrations were determined both on the cooked meats and on the juice lost during cooking. The cooking process caused the release of NHl and H1 on all cooked sample juices. The HI overall percentage was significantly lower than raw meat one (P 〈 0.005) and the NHI significantly higher (P 〈 0.005) in all cooked mix 1. The raw and cooked HI percentage variances depended on the pork meat pH values (ra = 0.70). The overall HI percentage was unchanged respect to raw meats on cooked mix 2, while the NHI amount was not quantitatively estimated. These results emphasized the role of sodium nitrite on tying up NHI in the cooked meats and safeguarding the oxidative stability of cooked meat products. 展开更多

The spindle behavior and MPF activity changes in the progression of oocyte maturation were investigated and compared with cytological observation and kinase assay between gynogenetic silver crucian carp and amphimictic colored crucian carp. MPF activity was measured by using histone H1 as phosphorylation substrate. There were two similar oscillatory MPF kinase activity changes during oocyte maturation in two kinds of fishes with different reproductive modes, but there existed some subtle difference between them. The subtle difference was that the first peak of MPF kinase activity was kept to a longerlasting time in the gynogenetic silver crucian carp than in the amphimictic colored crucian carp. It was suggested that the difference may be related to the spindle behavior changes, such as tripolar spindle formation and spindle rearrangement in the gynogenetic crucian carp. 展开更多

This study was carried out in 2007 and 2008, to evaluate responses of three chickpea cultivars (Hashem and Arman from kabuli and Pirooz from desi type) under well watering (I1: 70mm evaporation from class A pan), gradual water deficit (12 and 13: 70→90→ 110→130 and 70→100→130mm evaporation from class A pan, respectively) and severe water stress (14: 130mm evaporation from class A pan). Result showed that days to flowering and plant height were decreased, as water limitation increased. This reduction was significant under gradual water stress (I2 and I3) and Severe water deficit (I4), compared with control (I1). There were no significant differences in grain filling period and grain yield among I~, I2 and I3 irrigation treatments. No significant differences in days to physiologic maturity and number of sub branches were recorded among irrigation treatments. Interactions of year×cultivar for days to physiologic maturity, grain filling period and grain yield (P≤0.01) and for days to flowering and plant height (P≤0.05) were significant. The superiority of Arman in producing comparatively greater grain yield could be attributed to higher grain filling period of this cultivar in both years. 展开更多

The following instruments were identified and selected: (a) the methodology for the analysis of metropolitan transport systems; (b) the knowledge management maturity model (Siemens); and (c) Documentation and Information Centre. The simulation, using the tools in the knowledge management to analyze the Company Metropolitan of S^io Paulo--METRO, identified the need for improvements in eight areas and the problems: (1) rethink the knowledge management area; (2) develop new knowledge management policies; (3) define responsibilities in knowledge management area; (4) implement new knowledge management processes; (5) explore new ways and structures of knowledge; (6) consider the internal and external actors in knowledge management actions; (7) expand the dissemination of the collaborative culture; and (8) integrate in the knowledge management area current and future initiatives. The analysis shows that the transposition of the current situation to the desired situation for the company can be obtained with the implementation of an Information and Documentation Centre. The conclusion is that this set of tools allows to characterize, evaluate, and model the knowledge management in this business. 展开更多

This study aimed at the physical, chemical and biochemical changes during ripening of Sweetsop (Annona squamosa L.) and Golden Apple (Spondias citherea Sonner) fruits during ripening as important features to better understand their postharvest handling. It was carried out physical analysis such as firmness and chemical analysis such as total chlorophyll, total carotenoids, soluble solids, pectins and titrable acidity and biochemical analysis such as pectin methyl esterase, polygalacturonase, cellulase, and peroxidase and polyphenoloxidase activities in crude extract. Fruits were harvested at different stages of ripening. Experimental design was completely randomized and was carried out analysis of variance and Tukey tests, Total chlorophyll was decreasing in later stages of ripening, total soluble solid contents increased as the fruits ripen, while the acidity expressed percentage of citric acid decreased during fruits ripening. The loss of firmness and soluble solids content increased as the fruit get ripped stage, while the content of pectin decreased. Activity was observed for pectin methyl esterase and polygalacturonase enzymes during all stages of maturation, presenting the highest activity for both enzymes in the mature state. No cellulase activity detected at any stage during the ripening of these fruits. Activity of the enzyme polyphenoloxidase and peroxidase, associated with pulp browning was higher in the last stages of ripening of these fruits. Physical, chemical and biochemical patterns during ripening were different according to fruit species suggesting differential postharvest handling requirements. 展开更多

OBJECTIVE: To determine whether affected reticulocytes could be a reliable marker for the diagnosis of paroxysmal nocturnal hemoglobinuria (PNH), we analyzed CD59-antigen expression on the membranes of reticulocytes and erythrocytes. METHODS: We studied 10 PNH patients and 5 healthy volunteers by two-color flow cytometry with a membrane permeable fluorescent dye, thiazole orange (TO), and anti-CD59 monoclonal antibodies (MoAb). TO was introduced to gate reticulocytes and anti-CD59 MoAb were used to identify glycosylphosphatidylinositol (GPI)-deficient cells. RESULTS: Cells from healthy individuals were only CD59 positive. However, in all PNH patients, CD59-antigen expression could be divided into 3 types: type I cells (CD59 normally positive), type II cells (CD59 partly positive) and type III cells (CD59 negative). The majority of reticulocytes belonged to type III cells, GPI-deficient cells (61.0%). In addition, the percentage of affected reticulocytes was higher than that of erythrocytes. CONCLUSIONS: Analyzing PNH reticulocytes was important, because most patients had elevated numbers of reticulocytes, which represent more closely the recent erythroid output of BM. However, circulating mature erythrocytes were subject to complement-mediated intravascular lysis. Therefore, the percentage of abnormal erythrocytes may not accurately reflect the proliferation rate of normal and abnormal erythroid progenitor cells. OBJECTIVE:To establish a quantification model of Traditional Chinese Medicine(TCM)syndromes by sampling patients undergoing idiopathic precocious puberty(IPP)and early puberty.METHODS:A questionnaire for classifying and quantifying TCM syndromes was designed and administered.All the results were analyzed;the relationship between 3 types of syndrome and 47symptoms were summated.Meanwhile,the frequency distribution of each symptom or sign was aggregated.Fuzzy mathematics was used to develop a quantification model ofTCM syndromes.RESULTS:We found that precocious puberty had 3types of syndrome,including hyperactivity of fire due to Yin deficiency(Syndrome I),depressed liver Qi transforming into fire(Syndrome II),and end retention of damp heat(Syndrome III).In the IPP group,Syndrome I was the most common principal syndrome(100%).Forty-six patients(43.81%)werediagnosed with Syndrome I accompanied by Syndrome II and 11(10.48%)were diagnosed with Syndrome I accompanied by Syndrome III.In the early puberty group,Syndrome I was also the main syndrome(98.39%).The degrees of most symptoms were mild to moderate.Reddened tongue was the most common tongue manifestation(62.86%prevalence)in the IPP group.The most common pulse manifestations were slippery pulse,thread pulse,and taut pulse.The Asymptotic Normalization Coefficient(ANC)method was used to quantify the TCM syndromes in 167 cases.Diagnostic accuracy rate reached 91%,comparable to expert diagnosis.CONCLUSION:We find that there are 3 types of syndrome in the IPP group and in the early puberty group.Syndrome I(hyperactivity of fire due to Yin deficiency)is the main syndrome in the two groups.ANC may be an appropriate for quantification model ofTCM syndromes. 展开更多

In plants, each pollen mother cell undergoes two rounds of cell divisions to form a mature pollen grain, which contains a vegetative cell(VC) and two sperm cells(SC). As a companion cell, the VC carries the SCs to an ovule by germinating a pollen tube. In-depth sequencing analyses of mature pollen showed that micro RNAs(mi RNAs) and short interfering RNAs(si RNAs) are present in both the VC and SCs. Additionally, epigenetically-regulated transposable elements(TEs) are reactivated in the VC and these TE m RNAs are further processed into 21-nt epigenetically reactivated si RNA(easiR NA) in SCs, which prevent 24-nt si RNA accumulation and sequester mi RNA loading. Small RNAs are thought to move from the VC to SCs, where they regulate gene expression and reinforce TE silencing. Here, we summarize current knowledge of the biogenesis and function of mi RNAs, si RNAs, and easi RNAs in pollen, emphasizing how these different small RNAs coordinately contribute to sperm cell formation and TE silencing. 展开更多

Mature adipocytes are terminally differentiated somatic cells. Here, we report the successful generation of induced pluripotent stem (iPS) cells from mouse mature adipocytes by forced expression of six transcription factors (Oct4, Sox2, c-Myc, Klf4, Rarγ, and Lrh1) with a piggyBac transposon-based strategy. The resulting iPS cells were pluripotent as evidenced by the fact that they stained positive for alkaline phosphatase, expressed high levels of key pluripotency markers including Oct4, Nanog, and SSEA1, and remained pluripotent on a 2i media. In vitro differen- tiation of the iPS cells showed that the cell derivatives of all three germ layers could be readily obtained through forma- tion of embryoid bodies. Most importantly, these adipocyte- derived iPS cells were capable of producing chimera with high frequencies when reintroduced into early-stage em- bryos and transmitted through the germ line. This study demonstrates that the new six-factor reprogramming tech- nology facilitates the reset of the terminally differentiated adipocytes to the ground state of pluripotency, enabling us to fully explore the potential of mature adipocytes as a viable cell source for regenerative medicine. 展开更多
