Schott] were observed, laying foundation for highly-yielding cultivation technology system of Xiangsha taro. The results indicate that germination of the second taro happened from 46 d after sprouting (7-8 leaves); the first taro began to expand, from 125 d after sprouting (12 leaves) when the the second taro was been formed; the third taro began to form from 141 to 150 d after sprouting (15-16 leaves). The fresh weight of the second and third taros quickly increased from the time of 15 leaves to harvest. Early September is a transformation term of Xiangsha taro from vegetative body dominant to corm growth dominant.展开更多

[Method] PDA medium was used to isolate the fungus from C. abscondi...[Objective] This study aimed to uncover the biological characteristics of a parasitical fungus in Chrysosp/enium absconditicapsu/um J. T. Pan leaves. [Method] PDA medium was used to isolate the fungus from C. absconditicapsulum leaves; PDA medium, modified KB medium and Czapek medium were adopted to cultivate the isolated strain. [Result] Colonies of the strain were stretched, white, fedora- shaped with smooth and wavy edges, and showed diameter growth on PDA medi- um, modified KB medium and Czapek medium. At the late stage of culture, the colonies turned into cyanish brown on the above media. Spores were black and born on the surface of colonies on PDA medium with blackened medium. While on modified KB medium and Czapek medium, they were born at the edge of colonies with blackened medium. The spores varied in a wide range of shapes, mostly ob- clavate, sometimes spherical or ellipsoidal. The conidia were muriformly septate with transverse or longitudinal or oblique septations. The pseudo-beaks were short and cylindrical. [Conclusion] According to relevant literatures, the isolated strain is a fungus in Alternaria, Dematiaceae, Hylohomycetales, Hyphomycetes, Deuteromycotina.展开更多

[Method] Based on continuous field observation and destructive sampling over a growing season, detailed organ morphological data were obtained including leaf length, node number, plant height, tiller number, leaf angle, leaf area and specific leaf weight, to compare organ morphological differences among 4 rice cultivars of Baidao (indica), Jinnanfeng (japonica), 9325 (japonica) and 9915 (japonica) with 3 cultivation methods of field planting in Weigang, pot planting in Weigang, and field planting in Jiangpu. [Result] Maximum leaf length of each node gradually increased at the early growth stage and decreased at the later growth stage, the relationship between maximum leaf length and node position can be described by the equations y=a,,-~ and y=ax+b; node number, growth duration, leaf length and plant height of pot planting rice in Weigang were smaller than that of the other two field planting meth- ods; the relationship between plant height and sunshine duration, plant height and GDD (growing degree days) can be described by the equation y=ax+b, 19.23 ℃.d of GDD (≥10 ≥-d) and 8.12 h of sunshine duration were required to increase 1 cm of plant height; plant height, tiller number, and leaf area of Baidao were higher than that of the other 3 laponica rice cultivars, but the specific leaf weight and leaf angle were smaller. [Conclusion] Comparison of morphological characteristic differ- ences among rice cultivars is an important way to select water-saving and drought- tolerant rice varieties. In this study, the experimental results can be integrated into a rice functional-architectural model to simulate rice organ growth dynamics in a three- dimensional space, thereby providing reference for selecting water-saving and drought-tolerant rice cultivars.展开更多

This technology a...Seeds from Tabebuia roseo-alba lose viability very fast. Moreover, the seed germination rate is very low, reaching approximately 40%. This study aimed at the in vitro induction of embryogenic callus. This technology allows subsequent plant regeneration as an alternative for the production of T. roseo-alba seedlings. Seeds were germinated in vitro and after 20 days, cotyledonary leaves, hypocotyls and root segments excised from these seedlings were used as explants. They were inoculated on MS medium supplemented with sucrose (30 g/L), agar (5.0 g/L) and different auxins. The effect of 2,4-D, picloram and NAA at concentrations 0.0, 0.5, 1.0, 2.0 and 4.0 mg/L was evaluated. For the analysis of callus with embryogenic characteristics, ultra-structural study by scanning electron microscopy and cytochemical test with carmine were performed. The results showed that the culture medium supplemented with 4 mg/L NAA presented induction of callus with embryogenic characteristics in all explants used, with cotyledonary leaves showing the highest percentage (70% of explants with embryogenic characteristics). The use of 2, 4-D and picloram was efficient for callus formation in different explants, but no embryogenic characteristics were observed. From the ultra-structural analysis of callus with embryogenic characteristics, it was found that cells from different explant sources had isodiametric format. This format is similar to somatic embryos in globular stage. The cytochemical analysis confirmed the presence of pro-embryogenic cells in callus mass. Five different sites with a soluble salt gradient of 3.0-17.7 g kg^-1 dry soil from the coast to the inland were selected, and the microbial population size, activity and diversity in the rhizospheres of five common p...Five different sites with a soluble salt gradient of 3.0-17.7 g kg^-1 dry soil from the coast to the inland were selected, and the microbial population size, activity and diversity in the rhizospheres of five common plant species and the adjacent bulk soils (non-rhizosphere) were compared in a degraded wetland of the Yellow River Delta, Shandong Province, China to study the effects of soil environment (salinity, seasonality, depth, and rhizosphere) on microbial communities and the wetland's ecological function, thus providing basic data for the bioremediation of degraded wetlands. There was a significant negative linear relationship between the salinity and the total number of microorganisms, overall microbial activity, or culturable microbial diversity. Salinity adversely affected the microbial community, and higher salinity levels resulted in smaller and less active microbial communities. Seasonal changes were observed in microbial activity but did not occur in the size and diversity. The microbial size, activity and diversity decreased with increasing soil depth. The size, activity and diversity of culturable microorganisms increased in the rhizospheres. All rhizospheres had positive effects on the microbial communities, and common seepweed had the highest rhizosphere effect. Three halophilic bacteria (Pseudomonas mendocina, Burkholderia glumae, and Acinetobacter johnsonii) were separated through BIOLOG identification, and common seepweed could be recommended for bioremediation of degraded wetlands in the Yellow River Delta.展开更多

No robust system has been obtained by empiric approaches, and HCV cell culture remained hypothetical until 2005. The construction of functional molecular clones has served as a starting point to reconstitute a consensus infectious cDNA that was able to transcribe infectious HCV RNAs as shown by intrahepatic inoculation in a chimpanzee. Other consen- sus clones have been selected and established in a hu- man hepatoma cell line as replicons, i.e. self-replicating subgenomic or genomic viral RNAs. However, these repli- cons did not support production of infectious virus. Inter- estingly, some full-length replicons could be established without adaptive mutations and one of them was able to replicate at very high levels and to release virus particles that are infectious in cell culture and in vivo. This new cell culture system represents a major breakthrough in the HCV field and should enable a broad range of basic and applied studies to be achieved.展开更多

However, with these metho...In a commercialized, fully artificial plant factory, artificial luminaire is arranged in a unified way using a general illumination theory, an actual measurement, or an empirical methodology. However, with these methods, lightings are implemented without considering specific optical characteristics of lighting or material characteristics of each component that constructs a cultivation system, resulting in an amount of light that becomes irregular. The amount of lighting is closely related with the growth and quality of crops, and the deviation between points where cultivated crops are located causes quality difference in the produced crops, thus impairing the economic feasibility of a plant factory. In this regard, a simulation to figure out an optimum lighting layout was performed. Arrangements based on the spectrum distribution of light source and reflector materials were implemented to ascertain the distance between lighting and height of lighting and gather information in the pre-treatment process to improve the uniformity of light in the plant cultivation system. Improvement of around 15% in light uniformity is achieved compared with the existing system after the simulation is carried out. This result would reduce the deviation in crop growth to make uniform quality crop production possible.展开更多
