Studying the skin care efficacy of recombinant humanized collagen based on in vitro level.The stability of the recombinant humanized collagen was first analyzed by treating at different temperatures,then its skincare ...Studying the skin care efficacy of recombinant humanized collagen based on in vitro level.The stability of the recombinant humanized collagen was first analyzed by treating at different temperatures,then its skincare efficacy based on in vitro level was evaluated by detecting the inhibition rate of elastase,the inhibition rate of collagenase,the protein content of type I collagen in human fibroblasts,the inhibition of reactive oxygen species(ROS)with human keratinocytes,and the effects of the recombinant humanized collagen on the expression of hyaluronic acid(HA),filaggrin(FLG)and transglutaminase 1(TGM1)in keratinocytes.The results showed that recombinant humanized collagen was able to maintain stability at temperatures below 70℃.With regard to its skincare efficacy,recombinant humanized collagen could inhibit elastase and collagenase activities and promote the increase of type I collagen content in human fibroblasts.It also showed good inhibition of ROS in keratinocytes in vitro and could increase the expression of HA,FLG,and TGM1 in keratinocytes.In short,the recombinant humanized collagen exhibited a favourable skin care effect in vitro level.This study proved that it has potential firming,anti-wrinkle,moisturizing,and repairing efficacy,and is a valuable cosmetic raw material.展开更多

[Result] Except Higuchi model and Logistic model, other models could fit BSA release profile better. Influence of high light stress on the photosynthesis of flag leaves of indica subspecies (cv. “Shanyou 63', sensitive to photoinhibition) and japonica subspecies (cv. “Wuyujing', resistant to photoin...Influence of high light stress on the photosynthesis of flag leaves of indica subspecies (cv. “Shanyou 63', sensitive to photoinhibition) and japonica subspecies (cv. “Wuyujing', resistant to photoinhibition) of rice ( Oryza sativa L.) was comparatively investigated. In both cultivars of rice, the excitation energy distribution between two photosystems was altered and the excitation energy transfer from light harvesting chlorophyll protein complexes to PSⅡ was inhibited by high light stress. These decreases were more pronounced in indica rice cultivar as compared to japonica one. The analysis of mild SDS_PAGE showed that in indica rice, high light stress almost disaggregated the trimer of light harvesting chlorophyll protein complexes of PSⅡ (LHC Ⅱ 1). The stress reduced the contents of internal antennae chlorophyll protein complexes of PSⅡ (CPa), light harvesting chlorophyll protein of PSⅠ (CPⅠa) and Chl a protein complex of PSⅠ reaction center (CPⅠ) as well as dimer of LHCⅡ (LHCⅡ 2) in indica rice. In japonica subspecies, however, high light stress depressed the contents of LHCⅡ 1, CPa and CPⅠa, but slightly impacted on CPⅠ content. Moreover, the increase in the contents of monomer of LHCⅡ by high light stress was found in both subspecies. In consistent with above results, analysis of polypeptide indicated that the amounts of 27 kD and 25 kD polypeptide of LHCⅡ in particular, as well as that of 21 kD polypeptide of CPⅠa were reduced by high light stress in both subspecies. It was found that, comparing with japonica rice, the stress pronouncedly diminished 43 kD and 47 kD proteins of CPa and 23 kD extrisic protein in indica rice.展开更多

The molecular mechanisms of trefoil factors (TFFs), which involved in mucosal healing and tumorigenesis, have remained elusive. βγ-CAT is a novel multifunctional protein complex of non-lens βγ-crystallin and trefoil factor from frog skin secretions. Here we report that βγ-CAT could induce sustained contraction of isolated rabbit aortic rings in dosage (2-35nmol/L) and endothelium dependent manners (P〈0.01 ). In addition, in situ immunofluorescence indicated that positive TNF-α signals were mainly detected at the endothelial cell layer of βγ-CAT (25nmol/L) treated rings. Furthermore, βγ-CAT induced primary cultured rabbit thoracic aortic endothelial cells (RAECs) rapidly to release TNF-α. After βγ-CAT (25nmol/L) treated for 10 and 30min, the levels of the endothelial cells released TNF-ct were 34.17±5.10 pg/mL and 98.01±4.67 pg/mL (P〈0.01), respectively. In conclusion, βγ-CAT could induce sustained contraction of isolated aortic rings, and the contractile effect might be partially explained by the release of TNF-α. These findings will give new insight into understanding the functions and physiological roles of non-lens βγ-crystallins and trefoil factors.展开更多

SDS PAGE analysis of c...The changes of chlorophyll_protein complexes and photosynthetic activities of chloroplast isolated from lotus ( Nelumbo nucifera Gaertn.) seeds germinating under illumination were studied. SDS PAGE analysis of chlorophyll_protein complexes showed that there was only the light harvesting chlorophyll a/b protein complex from PSⅡ (LHCⅡ) precursor in chloroplast from lotus seeds germinated for 2 to 6 days, while LHC Ⅱ 1, and the chlorophyll_protein complex of PSⅠ (CPⅠ) appeared on the 8th day of germination and PSⅡ reaction center complex appeared later. Studies on the polypeptides composition of the chloroplast revealed the following results: 1) Small amount of the 27 kD polypeptide was synthesized in invisible light; 2) The 30 kD polypeptide existed previously in the plumules of the dry seeds; 3) The amount of the 30 kD polypeptide was more than any other polypeptides before germination and decreased gradually throughout germination, while the 27 kD polypeptide changed in the opposite way; 4) In the process of germination, measurement of the electron transport rate and the fluorescence induction kinetics at room temperature showed that PSⅡ activities and efficiency of primary light energy transformation were only experimentally measurable after 7 days of germination and gradually increased afterwards. At the same time, the chl a/b ratio rose from the lower value to normal; 5) The changes of chloroplast membrane components and its functions are concomitant in concert with that of the ultrastructure of chloroplast membranes during germination, as shown in our earlier work . The results have proved again that a different developmental pathway of chloroplast is likely to exist in the lotus plumules, which might provide an important clue for N. nucifera in having an unique position in the phylogeny of the angiosperm.展开更多

The leaf color beca...A green-revertible albino mutant-Qiufeng M was found from the japonica rice (Oryza sativa L. ssp. japonica) Qiufeng in the field. The first three leaves of the mutant were albino with some green. The leaf color became pale green since the fourth leaf and the glume had the same phenomenon as the first three leaves. The measuring data of the pigment content confirmed the visually observed results. It truly had a remarkable changing process in the leaf color in Qiufeng M. Comparison of the main agronomic characters between Qiufeng and Qiufeng M indicated that the neck length and grain weight showed significant difference at the 1% level, and other characters were not different. Genetic analysis showed that the green-revertible albino trait was controlled by a single recessive nucleic gene. Using 209 recessive mutant individuals in the F2 population derived from the cross Pei'ai 64S × Qiufeng M, a gene, tentatively named gra(t), was located between the SSR markers of RM475 and RM2-22 on the long arm of chromosome 2. The genetic distance were 17.3 cM and 2.9 cM respectively.展开更多
To understand metabolic adaptations,the basal metabolic rate(BMR) of Mrs Hume's Pheasant(Syrmaticus humiae) and Elliot's Pheasant(Syrmaticus ellioti) were investigated.Metabolic rate(MR),body temperature(Tb...To understand metabolic adaptations,the basal metabolic rate(BMR) of Mrs Hume's Pheasant(Syrmaticus humiae) and Elliot's Pheasant(Syrmaticus ellioti) were investigated.Metabolic rate(MR),body temperature(Tb) and thermal conductance(C) were determined in both species at a temperatrue range of 5-35 ℃,respectively.Oxygen consumption was measured with a closed circuit respirometer.The thermal neutral zones(TNZ) were 24.5-31.6 ℃,and 23.0-29.2 ℃,respectively.With a temperature range of 5-35 ℃,Mrs Hume's Pheasant and Elliot's Pheasant could maintained stable Tb at a mean of(40.47±0.64) and(40.36±0.10) ℃,respectively.Mean BMRs within TNZs were(1.36±0.84) mLO2/(g.h) for Mrs Hume's Pheasant and(2.03±0.12) mLO2/(g.h) for Elliot's Pheasant,which were 77% and 86% of the expected value based on their body mass,respectively.Thermal conductance of Mrs Hume's Pheasant and Elliot's Pheasant were(0.12±0.01) and(0.17±0.01) mLO2/(g.h.℃),below the lower critical temperature,respectively,which were 119% and 124% of the expected value based on their body mass,respectively.The ecophysiological characteristics of these species were low metabolic rate,high body temperature,and high thermal conductance,which allow both species to better adapt to the warmer climate environment in south China.展开更多

The complete cDNAs of mitochondrial and endoplasmic reticulum small heat shock protein ( shsp) were selected out from the cDNA library. Furthermore, the temperature responses of these shsp genes were determined. Northern hybridization showed that the heat response temperatures of both genes in tomato flower were lower than that in leaf and that mitochondria shsp in leaf was cold-inducible. In this paper, the molecular features of the cloned genes, the causes of the uncommon heat response temperatures of sHSP in newer and the cold inducible character of mitochondria shsp gene in leaf were discussed.展开更多

FATB is a member of FAT enzy...Acyl-ACP thioesterases (FATs) terminates the fatty acid synthesis and allow the transport of fatty acids out of the plastids, which are the important determinants of cellular metabolism. FATB is a member of FAT enzymes that has been described previously in most of the plants. In silico cloning is a new method that utilizes the bioinformatics on the complete genome and available EST database. In this study, a full-length cDNA clone of PtFATB gene was isolated from Populus tomentosa using this approach. It is 1,450 bp in length and the open reading frame encodes a peptide of 421 amino acids. The predicted amino acid sequence shows significant homology with those from other plant species, which contain typical domains owned by FATB proteins. The transcripts of PtFATB were abundant in leaves, and less in roots detected by using semiquantitative RT-PCR. When the shoots were subjected to the stress treatments (cold, dry, NaC1) and ABA (Abscisic acid), the expression of PtFATB was only slightly reduced under the treatment of low temperature. This suggests that the expression of PtFATB is in a constitutive fashion. This study provides the basis not only for the identification and characterization of this gene but also for the improvement of cold tolerance by controlling the expression of the PtFATB gene in trees in near future.展开更多

Ten positive porcine foot-and-mouth disease serums and more than two hundreds negative serum were tested, and the results were the same as the background of samples. The sensitivity test and replicate test indicated that this method was stable and sensitive, which was suitable for monitoring Asia 1 type porcine foot-and-mouth disease virus antibody.展开更多
To clarify the taxonomic status of Phlebotomus chinensb, the morphological characters of Ph. chinensis from Shaanxi, Henan, Sichuan and Gansu Provinces, and Ph. wui and Ph. longiductus from Xinjiang were examined and ...To clarify the taxonomic status of Phlebotomus chinensb, the morphological characters of Ph. chinensis from Shaanxi, Henan, Sichuan and Gansu Provinces, and Ph. wui and Ph. longiductus from Xinjiang were examined and the rDNA-ITS2 region and mtDNA Cyt b gene sequences were analyzed simultaneously. It was found that morphological variation was very low in Ph. chinensis among the different localities; the wings of adult samples distributed in high-altitude areas were merely longer. The rDNA-ITS2 and mtDNA Cyt b sequences had an intraspecific variability. The relationships among species in the genus Phlebotomus can be inferred by molecular markers. These results suggest that there is an obvious genetic divergence at the molecular level among the Ph. chinensis populations in China.展开更多

文摘Using the purified VP1 protein of Asia 1 type foot-and-mouth disease virus as the antigen, the purified monoclonal antibody was labeled by the sodium periodate method and the monoclonal antibody competitive ELISA was established in this study. Ten positive porcine foot-and-mouth disease serums and more than two hundreds negative serum were tested, and the results were the same as the background of samples. The sensitivity test and replicate test indicated that this method was stable and sensitive, which was suitable for monitoring Asia 1 type porcine foot-and-mouth disease virus antibody.

文摘To clarify the taxonomic status of Phlebotomus chinensb, the morphological characters of Ph. chinensis from Shaanxi, Henan, Sichuan and Gansu Provinces, and Ph. wui and Ph. longiductus from Xinjiang were examined and the rDNA-ITS2 region and mtDNA Cyt b gene sequences were analyzed simultaneously. It was found that morphological variation was very low in Ph. chinensis among the different localities; the wings of adult samples distributed in high-altitude areas were merely longer. The rDNA-ITS2 and mtDNA Cyt b sequences had an intraspecific variability. The relationships among species in the genus Phlebotomus can be inferred by molecular markers. These results suggest that there is an obvious genetic divergence at the molecular level among the Ph. chinensis populations in China.
