[Method] An orthogonal test was designed based on 1-year-old clones of F. pennsylvanica Marsh Lula 5. Under dif- ferent growth hormones, hormone concentrations and processing time, F. velutina Lula 2 and F. pennsy/vanica Marsh Yuanla 2 were treated by vacuum pump for 10 min, [Result] The decreasing order of cutting survival rates of Lula 5 under different etnvironments was small plastic flowerpot (94.4%), nutrition cup (92.8%), sand tray (56.1%), and cuffings treated with 100 mg/L ABT, for 2 h, 200 mg/L ABT, for 4 h and 100 mg/L IBA for 6 h under large nutrition cup environment grew the best, showing a survival rate of 100%. Lula 2 and Yuanla 2 were subjected to vacuum pump treatment, the survival rate of cuttings under the sand tray environment was averagely 29%; and under the large plastic flowerpot environment, the survival rate of cuttings was averagaly 64.4%, and cuttings of Lula 2 treated with vacuum pump for 10 min in 200 mg/L ABTI showed a survival rate up to 95%, which was the highest among all treatments. It could be seen that whether cuttings were treated with vacuum pump, the survival rate of cuttings in sand tray was lower than those under other environments, while there was no big difference in survival rate of cut- tings between the nutrition cup and plastic flowerpot environments. [Conclusion] This study is of great significance to improvement of cutting propagation coefficient.展开更多

AIM: TO assess whether polymorphisms in NOD2 and ATG16L1 affect cytokine responses and mycobacterium avium subspecies paratuberculosis (MAP) survival in monocytes from Crohn's disease (CD) patients METHODS: Mon...AIM: TO assess whether polymorphisms in NOD2 and ATG16L1 affect cytokine responses and mycobacterium avium subspecies paratuberculosis (MAP) survival in monocytes from Crohn's disease (CD) patients METHODS: Monocytes were isolated from peripheral blood of CD patients of known genotype for common single nucleotide polymorphisms of NOD2 and ATG16L1, Monocytes were challenged with MAP and bacterial per- sistence assessed at subsequent time-points. Cytokine responses were assayed using a Milliplex multi-analyte profiling assay for 13 cytokines. RESULTS: Monooltes heterozygous for a NOD2 polymorphism (R702W, P268S, or 1007fs) were more permissive for growth of MAP (P = 0.045) than those without. There was no effect of NOD2 genotype on subsequent cytokine expression. The T300A polymorphism of ATG16L1 did not affect growth of MAP in our model (P = 0.175), but did increase expression of cytokines interleukin (IL)-10 (P = 0.047) and IL-6 (P = 0.019). CONCLUSION: CD-associated polymorphisms affected the eliminaUon of MAP from ex v/vo monooltes (NOD2), or expression of certain oltokines (ATG16LI), implying independent but contributory roles in the pathogenesis of CD.展开更多

In this study, efforts were made to standardize vegetative propagation techniques for Lycium shawiL Nitraria retusa and Farsetia aegyptia. Effects of varying concentrations of rooting hormones on the success of rooting of hardwood cuttings of these species were studied in this research. Rooting in hardwood cuttings of Lycium shawii was enhanced (100%) by treating them with 1,000 ppm IBA, Ormone radicante in polvere (5,000 ppm NAA) and hormex (1,000 ppm IBA). Treatment with 1,000 ppm NAA was found to be effective for Nitraria retusa, while marginal success was recorded in rooting of hardwood cuttings of Farsetia aegyptia when treated with 400 ppm NAA. Methods: Mononucl...Objective: To investigate the effects of soluble factors secreted by acute myeloid leukemia (AML) cells on the phenotypical and functional properties of DCs derived from normal mononuclear cells. Methods: Mononuclear cells were cultured with interleukin-4 (IL-4) and granulocyte-macrophage colony-stimulating factor (GM-CSF), in the presence or absence of 24 h culture supematants from fresh pdmary AML cells, to generate immature DCs. The maturation of DCs was induced by cytokines IL-lbeta, IL-6, tumor necrosis factor-alpha (TNF-alpha), and prostaglandin-2 (PGE-2). The phenotypic alterations of DCs and DCs-primed CD4+ T cells were evaluated using flow cytometry. Precursor frequency (PF) was calculated to monitor the allostimulatory effects of DCs on CD4^+ and CD8^+ T cells. Results:AML cell supernatant-treated DCs showed significantly lower expression of co-stimulatory molecules CDS0 and CD86, and reduced response to cytokines IL-1beta, IL-6, TNF-alpha, and PGE-2. The allostimulatory effects of AML cell supematant-treated DCs on CD4^+ and CD8^+ T cells were significantly lower than those of normal mature DCs [PF: (1.8 ±0.5)% vs. (5.2 ± 1.6)% for CD4^+ T cells, (2.1 ±0.6)% vs. (6.5 ± 2.0)% for CD8^+ T cells, P 〈 0.01]. These AML supernatantoinduced DCs could also induce allogeneic CD4^+ T cells to differentiate into CD4^+CD25high T cells, which had immunophenotyping characteristics of regulatory T cells, i.e. they expressed Foxp3 but not active maker CD69. Conclusion: This study demonstrates that soluble factors secreted by AML cells can inhibit development and functions of DCs. In addition, AML supernatant-induced DCs can induce the generation of CD4^+CD25^high T cells from CD4^+ T cells, which may be a mechanism of increased prevalence of CD4^+CD25^high regulatory T cells and immune dysfunction in AML patients.展开更多

While the biochemical function of Rv2302 is still unknown, rece...The Mycobacterium tuberculosis protein Rv2302 has been characterized using nuclear magnetic resonance (NMR) and circular dichroism (CD) spectroscopy. While the biochemical function of Rv2302 is still unknown, recent microarray analyses show that Rv2302 is upregulated in response to starvation and overexpression of heat shock proteins and, consequently, may play a role in the biochemical processes associated with these events. Rv2302 is a monomer in solution as shown by size exclusion chromatography and NMR spectroscopy. CD spectroscopy suggests that Rv2302 partially unfolds upon heating and that this unfolding is reversible. Using NMR-based methods, the solution structure of Rv2302 was determined. The protein contains a five-strand, antiparalle β-sheet core with one C-terminal α-helix (A61 to A75) nestled against its side. Hydrophobic interactions between residues in the α-helix and β-strands 3 and 4 hold the α-helix near the β-sheet core. The electrostatic potential on the solvent-accessible surface is primarily negative with the exception of a positive arginine pocket composed of residues R18, R70, and R74. Steady-state {^1H}-^15N heteronuclear nuclear Overhauser effects indicate that the protein's core is rigid on the picosecond timescale. The absence of amide cross-peaks for residues G 13 to H 19 in the ^1H-^15N heteronuclear single quantum correlation spectrum suggests that this region, a loop between β-strands 1 and 2, undergoes motion on the millisecond to microsecond timescale. Dali searches using the structure closest to the average structure do not identify any high similarities to any other known protein structure, suggesting that the structure of Rv2302 may represent a novel protein fold.展开更多

METHODS Human GC cell lines KATOIII, MKN-28 and SGC-7901 were infected with adenovirus gp96 at a multiplicity of infection of 100. gp96-GC antigen peptide complexes were purified. MTT (3-(4,5-dimethylthiazol-2-yl)2,5- diphenyltetrazolium bromide) assay, lactate dehydrogenase (LDH) release assay and enzyme-linked immunosorbent assay were used to determine allo-reactive T cell stimulation, natural killer (NK) cell activity and expression of cytokines (such as interleukin (IL)-10, IL-12, interferon (IFN)-gamma and tumor necrosis factor (TNF)-alpha), respectively. Effect of cytotoxic T lymphocyte (CTL) on DCs incubated with HSP-gp96 was also evaluated by LDH release. All assays were performed in triplicate and the average values were reported. Comparison between groups was conducted using Student's t test. RESULTS T cells incubated with HSP-gp96 exhibited a marked increase in proliferation in a dose-dependent manner (P < 0.05). NK cell activity after gp96-GC peptide complex treatment was significantly higher than that after antigen peptide treatment (P < 0.05). The activity of CTLs incubated with DCs from three GC cells lines was obviously higher than that stimulated by GC antigen at ratios of 50: 1, 25: 1, 10: 1, and 5: 1 (P < 0.05). Furthermore, the secretion of TNF-alpha, IL-10, IL-12 (P70) and IFN-alpha markedly increased after incubation with HSP-gp96 (P < 0.05). CONCLUSION HSP-gp96 promotes T cell response, enhances DC antigen presentation and induces cytokine secretion, as well. HSP-gp96 has potential as immunotherapy for elimination of residual GC cells.展开更多
