Their adsorption performance...Five tryptophan analogues with a hydrophobic indole ring and an amino group on each molecule were used as functional ligands of mixed-mode resins for human serum albumin(HSA) purification. Their adsorption performance was evaluated and the effects of p H and salt addition on HSA adsorption were studied. The resins prepared showed typical p H-dependent adsorption and the highest adsorption capacity and affinity were found at pH 5.0for all the resins tested. The saturated adsorption capacity was 138.02 mg·g^(-1)with the tryptaminefunctionalized resin, which significantly decreased at p H below 4.0 due to electrostatic repulsion between ligands and HSA. Moreover, the addition of Na Cl or(NH_4)_2SO_4in media reduced HSA adsorption capacity, although the two salts showed different affecting profiles. The tryptamine-functionalized resin showed the best salt-tolerant performance, and its high adsorption capacity was maintained under high salt concentrations. In addition, the five resins prepared showed good adsorption selectivity for recombinant HSA from Pichia pastoris broth. Molecular docking results between tryptamine and HSA indicated that tryptamine was favorable to bind on Site II(indole-binding site) of HSA.展开更多

Methods The C57BL/6J mice bone ma...Objective To explore the influence of oxidized high-density lipoprotein (oxHDL) on the maturation and migration of bone marrow-derived dendritic cells (BMDCs) from C57BL/6J mice. Methods The C57BL/6J mice bone marrow cell suspension was prepared and purified. Recombinant granulocyte-macrophage colony-stimulating factor (rmGM-CSF) and recombinant interleukin-4 (rmlL-4) were used to promote monocytes to differentiate and suppress lymphocytes. Then 50μg/mL oxHDL was added to stimulate BMDCs, using 50μg/mL high-density lipoprotein (HDL) as homologous protein control, PBS as negative control, and 1 μg/mL lipopolysaccharide (LPS) as positive control. The CD86 and MHCII expression rates were detected with fluorescence-activated cell sorting (FACS). Liquid scintillation counting (LSC) was used in mixed lymphocyte reactions (MLRs) to reflect the ability of BMDCs in stimulating the proliferation of homologous T cells, Levels of cytokines IL-12 and IL-10 were detected by ELISA. The cell migration was evaluated with the transwell system. Results Compared with PBS group, the expressions of CD86 and MHCII, counts per minute of MLRs, secretion of IL-12 and IL-10, and number of migrated cells in oxHDL group and LPS group significantly increased (all P 〈 0.05), while the increment was less in oxHDL group than LPS group. The number of migrated cells in oxHDL group was about twice of that in HDL group. Conclusion OxHDL may promote the maturation and migration of BMDCs in vitro.展开更多

The longer one co...A rabbit anti serum to tree shrew apolipoprotein C I (apo C I) was used to screen an expression cDNA library constructed by us from tree shrew (TS)liver tissue. Two apo C I cDNA clones were obtained. The longer one consistsof 380 nucleotides, including 21 bp and 95 bp at the 5’ and 3’ end of the non translated regions respectively, and a 264 bp fragment in an open reading frame encoding 88 amino acids prepropeptide which contains 26 amino acids of signal peptide and a mature protein (62 amino acids). Comparing the amino acid sequence deduced from this cDNA with those of the published mammalian apo C Is reveals that it shared some structural similarity with rat, mouse and dog apo C I, but it had 5 more amino acids than that of human and baboon. The expression of apo C I mRNA in 8 different tissues were also assayed with Northern blot. The results demonstrated that liver had the highest expression, intestine had much less expression and no expression in other tissues, which is much different from human and other species. This study has laid down a good foundation for further studying on the function and the stucture of tree shrew apo C I gene.展开更多

Sheet molding compounds for automotive applications are ty...We present comparative life-cycle assessments of three fiber-reinforced sheet molding compounds (SMCs) using kenaf fiber, glass fiber and soy protein resin. Sheet molding compounds for automotive applications are typically made of unsaturated polyester and glass fibers. Replacing these with kenaf fiber or soy protein offers potential environmental benefits. A soy-based resin, maleated acrylated epoxidized soy oil (MAESO), was synthesized from refined soybean oil. Kenaf fiber and polyester resins were used to make SMC 1 composites, while SMC2 composites were made from kenaf fiber and a resin blend of 20% MASEO and 80% unsaturated polyester. Both exhibited good physical and mechanical properties, though neither was as strong as glass fiber reinforced polyester SMC. The functional unit was defined as mass to achieve equal stiffness and stability for the manufacture of interior parts for automobiles. The life-cycle assessments were done on SMCI, SMC2 and glass fiber reinforced SMC. The material and energy balances from producing one functional unit of three composites were collected from lab experiments and the literature. Key environmental measures were computed using SimaPro software. Kenaf fiber-reinforced SMC composites (SMC1 and SMC2) performed better than glass fiber-reinforced SMC in every environmental category. The global warming potentials of kenaf fiber-reinforced SMC (SMCI) and kenaf soy resin-based SMC (SMC2) were 45% and 58%, respectively, of glass fiber-reinforced SMC. Thus, we have demonstrated significant ecological benefit from replacing glass fiber reinforced SMC with soy-based resin and natural fiber.展开更多
