METHODS: NQOt activity was determined in cytosol from normal, APAP and PBC liver specimens. Western blot and immunohistochemical staining were used to determine patterns of NQO1 expression using a specific antibody against NQO1. RESULTS: NQO1 protein was very low in normal human livers. In both APAP and PBC livers, there was strong induction of NQO1 protein levels on Western blot. Correspondingly, significant up-regulation of enzyme activity (16- and 22-fold, P〈0.05) was also observed in APAP and PBC livers, respectively. Immunohistochemical analysis highlighted injury-specific patterns of NQO1 staining in both APAP and PBC livers. CONCLUSION: These data demonstrate that NQO1 protein and activity are markedly induced in human livers during both APAP overdose and PBC. Up-regulation of this cytoprotective enzyme may represent an adaptive stress response to limit further disease progression by detoxifying reactive species.展开更多

METHODS: The series included 37 patients with chronic active HCV-related hepatitis and 11 with HCV-related compensated cirrhosis. Eight-hydroxydeoxyguanosine in liver biopsies was quantified using an electrochemical detector. The mRNA expression of TNF-α, IL-1β, TGF-α and c-myc in liver specimens was detected by semiquantitative comparaUve RT-PCR. RESULTS: TNF-α levels were significantly higher in hepatitis patients than in cirrhosis patients (P=0.05). IL-1β was higher in cirrhosis patients (P=0.05). A sig- nificant correlation was found between TNF-α and staging (P= 0.05) and between IL-1β levels and grading (P=0.04). c-myc showed a significantly higher expression in cirrhosis patients (P=0.001). Eight-hydroxydeoxyguanosine levels were significantly higher in cirrhosis patients (P=0.05) and in HCV genotype 1 (P=0.03). Considering all patients, 8-hydroxydeoxyguanosine levels were found to be correlated with genotype (P=0.04) and grading (P=0.007). Also multiple logistic regression analysis demonstrated a significant correlation among the number of DNA adducts, TNF-α expression and HCV genotype (P= 0.02). CONCLUSION: In chronic HCV-related liver damage, oxidative DNA damage correlates with HCV genotype, grading and TNF-α levels. As HCV-related liver damage progresses, TNF-α levels drop while IL-1β and c-myc levels increase, which may be relevant to liver carcinogenesis.展开更多

It is more reasonable to describe the reversible and the irreversible deformations of sample separately during creep process. A nonlinear elastic-visco-plastic rheological model is presented to characterize the time-based deformational behavior of hard rock. Specifically, a spring element is used to describe reversible instantaneous elastic deformation. A reversible nonlinear visco-elastic (RNVE) model is developed to characterize recoverable visco-elastic response. A combined model, which contains a fractional derivative dashpot in series with another Hook’s body, and a St. Venant body in parallel with them, is proposed to describe irreversible visco-plastic deformation. Furthermore, a three-stage damage equation based on strain energy is developed in the visco-plastic portion and then nonlinear elastic-visco-plastic rheological damage model is established to explain the trimodal creep response of hard rock. Finally, the proposed model is validated by a laboratory triaxial rheological experiment. Comparing with theoretical and experimental results, this rheological damage model characterizes well the reversible and irreversible deformations of the sample, especially the tertiary creep behavior.展开更多

Little is known about the possible pathomechanisms and the clinical course.METHODS: Secondary sclerosing cholangitis was diagnosed in five patients with persistent jaundice after severe trauma (one burn injury, three accidents, one power current injury). Medical charts were retrospectively reviewed with regard to possible trigger mechanisms for cholestasis, and the clinical course was recorded.RESULTS: Diagnosis of secondary sclerosing cholangitis was based in all patients on the primary sclerosing cholangitis (PSC)-Iike destruction of the intrahepatic bile ducts at cholangiography after exclusion of PSC. In four patients, arterial hypotension with subsequent ischemia may have caused the bile duct damage, whereas in the case of power current injury direct thermal damage was assumed to be the trigger mechanism. The course of secondary liver fibrosis was rapidly progressive and proceeded to liver cirrhosis in all four patients with a follow-up 〉2 years. Therapeutic possibilities were limited.CONCLUSION: Posttraumatic sderosing cholangitis is a rare but rapidly progressive disease, probably caused by ischemia of the intrahepatic bile ducts via the peribiliary capillary plexus due to arterial hypotension.Gastroenterologists should be aware of this disease in patients with persistent cholestasis after severe trauma.展开更多

In this experiment using the Ames test, there was antimutagenic effect by the water-soluble chitosan that dissolved in hard water (CHW). The Ames test was performed using Salmonella typhimurium TA98 and TA100. For the antimutagenic test, and the desmutagenic and bioantimutagenic tests, Trp-P-2, 2AA and AF-2 were used as mutagens. Tests for spontaneous mutation rate without mutagens were performed using S. typhimurium TA98 and TA100. When several concentrations of CHW were directly added to Minimal Glucose Plates, there were no changes in the number of His+ revertants of TA98 and TA100. In desmutagenic and bioantimutagenic tests, the number of His^+ revertants of TA98 and TA100 decreased. In the test for spontaneous mutation rate, the number of His+ revertants of TA98 and TA 100 decreased slightly. When a concentration of CHW was added to examine antimutagenic effects, there was no change in the viable cell count of TA98 and TA100. From these results, AF-2 that is a direct mutagen may combine with CHW, and that may also be carrying out the inactivation. CHW possibly acts directly on the DNA by decreasing the rate at which mutation is caused to the strain's promutagens, Trp-P-2 and 2AA. This is evident from the decreased spontaneous mutation rate without a decreased viable cell count.展开更多

We herein introduce a withdrawing method of migrating stiffening tube incidentally inserted into the colorectum.A 65-year-old Japanese woman underwent colonoscopy because of abdominal discomfort. We used stiffening tube to insert the scope to the proximal colon because of her redundant sigmoid colon. When withdrawing the scope,we realized that the tube was fully inside the colorectum. We could not remove the tube instantly, and it reached the splenic flexure, finally. We reinserted the scope through the migrating tube, straightened the scope, and withdrew it holding a slight angle of the scope over the proximal end of the tube. Then, we could safely remove the tube along with the scope through the anus.展开更多

People use wheelchairs for a variety of reasons, the most common one is paralysis from spinal cord injuries. Current estimates indicate there are around 504,000 people alive in Spain with spinal cord injuries. Among other reasons for using wheelchairs are: fatigue from multiple sclerosis, muscle weakness from muscular dystrophy, lower limb spasticity from cerebral palsy, and missing limbs due to amputation. People who use wheelchairs may encounter a variety of obstacles at their workstations depending on their limitations. Some possible accommodations could be adjustable height desk or table for a person who cannot work comfortably at an existing desk, accessible filing system or office supplies, and frequently used materials on most accessible shelves or drawers for a person who cannot reach upper and lower shelves and drawers. This paper presents the graphical design of a workstation for wheelchair users, in this case, technical, anthropometric, ergonomic, aesthetics and scientists aspects have been analyzed. The design of composite structure conduct to explicit safety constraints applied to office furniture requirements kept continuously current and updated as part of innovation. Finally, results are showed in a three-dimensional model.展开更多

Methods: We retrospectively analyzed the clinical data of 32 patients with TSH developing into CSDH and reviewed related literature. Results: 16.7% of TSH developed into CSDH in this study. The time of evolution was from 22 to 100 days after head injury. All the patients were cured with hematoma drainage. Conclusions: TSH is one of the origins of CSDH. The clinical characteristics of TSH developing into CSDH follow that the ages of the patients are polarized, that the evolution often happens in the patients with small chronic hydromas and being treated conservatively, that the patients are usually injured deceleratedly and that the accompanying cerebral damage is often very mild.展开更多
