Some biological properties were compared between nit mutants and their parental isolates. The results showed that there were no significant differences in growth rate, cultural characters or pathogenicity between JS399-19-resistant nit mutants and their parental isolates. But the conidial production and the sexual reproduction ability changed to some extent. There was no cross resistance toward chlorate and JS399-19 in F. graminearum and the resistance could be stable through 20-time subcultures. Therefore, the nit could be used as a genetic marker for studying the genetics of JS399-19 resistance in E graminearum, which was used to study JS399-19 resistance transferability in hyphal fusion. Resistance in JS399-19 could not be transferred by hyphal fusion or could be transferred with low chance between two compatible isolates, which would delay the development of JS399-19 resistance in the field.展开更多
Embryogenic calli of Kentucky bluegrass, named Md, were induced from mature seeds and embryos, and proliferated on medium K3 containing 2,4-dichlorophenoxyacetic acid (2,4-D, 10.0 mumol/L), 6-benzylaminopurine (BAR, 0...Embryogenic calli of Kentucky bluegrass, named Md, were induced from mature seeds and embryos, and proliferated on medium K3 containing 2,4-dichlorophenoxyacetic acid (2,4-D, 10.0 mumol/L), 6-benzylaminopurine (BAR, 0.5 mumol/L) and K5 which was the K3 medium supplemented with cupric sulfa (0.5 mumol/L) under dim-light condition (20-30 mumol.m(-2).s-1, 16 h light) at 24 degreesC. Embryogenic calli were transformed with plasmids pDM805 Carring bar and gus genes, Which was mediated by an Agrobacterium strain AGL1, four transgenic lines were obtained. The important factors that affect the transformation efficiency and obtain desirable number of transgenic plants included: (1) the quality of embryogenic calli; (2) light condition and time of co-cultivation; (3) concentration of antibiotics used for suppressing the overgrowth of Agrobacterium in the course of transformed plant regeneration; (4) selection pressure, etc. The micro nutrient of cupric had significant influence on the quality of embryogenic calli. This presentation is the first successful protocol of Kentucky bluegrass transformation mediated by Agrobacterium.展开更多

Such characteristics of Octoploid tnticalwere between those of common wheat and Rye. [Objective] This study aimed to explore the effects of spores and crude toxins of Helminthosporium gramineum Rabenh f. sp. echinochloae(HGE) on the ac- tivity of defensive enzymes of barnyardgrass [Echinochloa crus-...[Objective] This study aimed to explore the effects of spores and crude toxins of Helminthosporium gramineum Rabenh f. sp. echinochloae(HGE) on the ac- tivity of defensive enzymes of barnyardgrass [Echinochloa crus-galli (L.) Beauv.]. [Method] The effects of spores and crude toxins of HGE, as well as the mixture of spores and crude toxins on the activity of defensive enzymes in barnyardgrass were determined under laboratory conditions. [Result] Spores and crude toxins of HGE had varying degrees of effects on PAL and POD activity, and no obvious effect on SOD activity in barnyardgrass. In addition, spores and toxins had some similar im- pacts on the defensive enzymes in barnyardgrass. [Conclusion] Since toxins have similar effects on the hosts as spores of fungal pathogen do, they can be a substi- tute for the fungal pathogen in studying the partial pathogenic mechanism of this pathogen due to its complexity in pathogenic process.展开更多
