ROLE OF B LYMPHOCYTE AND ITS SUBPOPULATIONS IN PATHOGENESIS OF IMMUNORELATED PANCYTOPENIA 被引量:10

Objective To measure the quantities and apoptosis-related protein levels of B lymphocyte in the patients with immunorelated pancytopenia (IRP) and explore the action of B lymphocyte in the pathogenic mechanism of IRP. Methods Quantifies of whole B lymphocytes and CD5^+ B lymphocytes as well as the expressions of Fas and Bcl-2 in B lymphocytes in 35 patients with untreated IRP, 15 IRP patients in complete remission (CR), and 10 normal controls were assayed by flow cytometry. The percentages of B lymphocyte and CD5^+ B lymphocyte were significantly higher in untreated IRP patients than in CR IRP patients and normal controls ( P 〈 0. 05 ), and there was no significant difference between the latter two groups ( P 〉 0. 05 ). There was no significant difference of Fas expression in B lymphocyte among three groups ( P 〉 0. 05). The expression of Bcl-2 in B lymphocyte was significantly higher in untreated patients than in CR patients or normal controls ( P 〈 0. 01 ), and significantly higher in CR patients than in normal controls ( P 〈 0. 01 ). The apoptosis. related index was significantly lower in untreated patients than in CR patients or normal controls ( P 〈 0. 05 ), and signif. icantly lower in CR patients than in normal controls ( P 〈 0. 05 ). The percentage of B lymphocyte was positively correlated with post-treated response time ( r = 0. 53, P 〈 0. 01 ). Conclusion The production of auto-antibodies in IRP patients probably has some relationship with the abnormal quantifies of B lymphocyte and its subpopulations as well as with the inhibition of B lymphocyte apoptosis. 展开更多

MSH5, a member of the MutS homolog DNA mismatch repair protein family, has been shown to be required for proper homologous chromosome recombination in diverse organisms such as mouse, budding yeast and Caenorhabditis elegans. In this paper, we show that a mutant Arabidopsis plant carrying the putative disrupted AtMSH5 gene exhibits defects during meiotic division, producing a proportion of nonviable pollen grains and abnormal embryo sacs, and thereby leading to a decrease in fertility. AtMSH5 expression is confined to meiotic floral buds, which is consistent with a possible role during meiosis. Cytological analysis of male meiosis revealed the presence of numerous univalents from diplotene to metaphase I, which were associated with a great reduction in chiasma frequencies. The average number of residual chiasmata in the mutant is reduced to 2.54 per meiocyte, which accounts for ~25% of the amount in the wild type. Here, quantitative cytogenetical analysis reveals that the residual chiasmata in Afresh5 mutants are randomly distributed among meiocytes, suggesting that AtMSH5 has an essential role during interferencesensitive chiasma formation. Taken together, the evidence indicates that AtMSH5 promotes homologous recombination through facilitating chiasma formation during prophase Ⅰ in Arabidopsis. 展开更多

Calf spleen extract alleviates cyclophosphamide-induced leucopenia in mice and inhibits the glycolysis of HL60 cells

Calf spleen extract(CSE) has been clinically used as an adjuvant agent in malignant tumor therapy.It can improve the physical status of patients.However,its mechanism of action remains relatively unclear.In this study,we investigated the effect of CSE on leucopenia in mice and on promyelocytic leukemia cells(HL60).CSE in 10 mL/kg(2.5 mg bioactive polypeptides and 190μg ribose/mL) significantly increased leukocyte numbers in leucopenia mice.The effect on neutrophil numbers,among all leukocytes,was the most evident.CSE stimulated the proliferation of bone marrow cells in vitro and decreased the GM-CSF level in serum.In addition,CSE significantly reduced the viability of HL60 cells,decreased the production of lactate and adenosine triphosphate(ATP) of these cells.CSE induced the apoptosis and S-phase arrest in HL60 cells.In conclusion,CSE can enhance leukocyte numbers,which may be attributed to its direct stimulatory effect on bone marrow cells.CSE is an inhibitor of promyelocytic leukemia cell viability,which may be attributed to the induction of the apoptosis,the arrest of cell cycle and the inhibition of the glycolysis of cells. 展开更多

Regulatory Effect of Acupuncture and Moxibustion on the Hematopoiesis of Leukopenia Model in Rats

To investigate the effect of acupuncture or moxibustion at acupoint Geshu (BL 17) on raising leukocyte and promoting marrow proliferation. Methods.. Wister rats were selected to establish the leukopenia and immuno-suppression model by intraperitoneal injection of cytoxan (CTX). After they were grouped and treated, leukocyte and marrow karyocyte were counted in different phases to evaluate the effect of acupuncture or moxibustion at acupoint Geshu (BL 17) on raising leukocyte and promoting marrow hematopoiesis. Results. Acupuncture or moxibustion at acupoint Geshu (BL 17) and routine western medicine could all significantly raise leukocyte and increase marrow karyocyte. The curative effect was better in moxibustion group than in acupuncture group and in routine western medicine group (P〈 0.05). Conclusion. Moxibustion at acupoint Geshu (BL 17) can markedly raise peripheral blood leukocyte and marrow karyocyte of the leucopenia model in rats. 展开更多
