R. China. Methods Twenty-fiveelements in Phytolacca including essential elements, toxic elements and rare earth elements intraditional Chinese medicinal herbs from different areas were analyzed by ICP-MS. Results The 25elements in Phytolacca were determined by ICP-MS under optimized conditions. The detection limitswere 0.003 -0.71 ng·mL^(-1). The recoveries were 88% - 118% . The relative standard deviations ofthe measurements were 1.7% - 13.3%. Conclusion The determined concentrations of elements inPhytolacca acinosa Roxb vary from one area to another; however, the distribution tendency ofelements in all the samples is similar. The distribution tendency of rare earth elements inPhytolacca acinosa Roxb is consistent with that in nature.展开更多

Secondly, a dynamic usermodeling is introduced to model the user's sketching habits based on recording and analyzinghistorical information incrementally. The stress and strain in different deformation zones were analyzed. The groove size reduces gradually and the groove shape drives to triangle during the extrusion procedure. The maximum values of equivalent effective stress and radial stress appear in groove zones, and the maximum equivalent effective strain firstly is obtained also in groove zones. The grain size in groove zones is less than that in other zones. The experimental results are consistent with simulation results, which prove that the copper tubes with sketch section are favorable to the metallurgy bond of boundary interface between aluminum and copper.展开更多

Patch spraying to control weeds has advantages of chemical saving, reduced cost and environmental pollution. Advent of electro-optical sensing capabilities has paved the way of using machine vision technologies for patch spraying. Machine vision system has to acquire and process digital images to make control decisions. Proper identification and classification of objects present in image holds the key to make control decisions and use of any spraying operation performed. Recognition of objects in digital image may be affected by background, intensity, image resolution, orientation of the object and geometrical characteristics. A set of 16, including 11 shape and 5 texture-based parameters coupled with predictive discriminating analysis has been used to identify the weed leaves. Geometrical features were indexed successfully to eliminate the effect of object orientation. Linear discriminating analysis was found to be more effective in correct classification of weed leaves. The classification accuracy of 69% to 80% was observed. These features can be utilized for development of image based variable rate sprayer.展开更多

The results of the experiment show that it can get notable effect in segmentation according to the color feature, and the possibility of successful segmentation is 87%-93%. This method can also be widely used in other fields which are complicated in the background of the image and facilely influenced in illumination, such as weed identification, tree species discrimination, fruit picking and so on.展开更多
The status of the social and human sciences as genuine sciences on a par with the natural sciences has widely been held in doubt, and the subject-oriented approach (SOA) to knowledge also shows the traditional scien...The status of the social and human sciences as genuine sciences on a par with the natural sciences has widely been held in doubt, and the subject-oriented approach (SOA) to knowledge also shows the traditional scientific view to be misleaded. Its shows that it is mandatory to dismiss the idea that personal knowledge is a representation of a common world created by some God, and also the mistake to take the seductive noun/verb structure as for given. We need a new methodological paradigm of science--an approach that avoids the pitfalls of dualism and realism--and take the effort to couch its thinking in a re-interpretation of natural language. This line of reasoning paves the way for the SOA--a new epistemology that takes the individual knower and its feelings as the coherent point of departure. The traits of a new foundation are sketched and to that end a bootstrap model is proposed that departs from the early man's first experience. In doing so, we, in a subject-oriented manner, can bring man's living experience and his priverse (or private universe), under the collective umbrella of a consensual science. This approach brings the promise to provide a sound theory of everything-or rather a theory of every thin/kin/g-which in one step removes the cleft between the natural and social sciences.展开更多

The experience gained in mapping of alpine vegetation shows that the schematic small-scale maps reflect only the very common features. In boreal forest mountains of Siberia there are four systems of alpine vegetation including alpine-meadow, goltsy-tundra, island near the Pacific Ocean and tundra-steppe ones. Every system is represented by several geographical variants, characterized by regional phytogeographical peculiar features of this vegetation. The geographical variant includes a number of altitudinal belts (zones), each of them has its floristic peculiarities and complexes of plant formations (for instance, Altai-Sayan variant composes of subalpine-alpine-subnival-nival belts). Such geographical variants of alpine vegetation may be indicated as chorological units on a new Circum-Boreal Vegetation Map. Under discussion is the possible use of ecological-geographical approach to reflect the alpine vegetation as chorological (structural) units of the vegetation cover being exemplified by high mountains of the Siberia. A suitable ehro- matographic system was established using a linear gradient elution with acetonitrile and 0.1% phosphoric acid (H3PO4) as the mobile phase and a detection wavelength of 210 nm. HPLC analysis identified 12 common peaks and 8 common fingerprint peaks among the different parts of the herb. We found that the constituents varied at different harvesting times and in different parts of the plant. The HPLC fingerprint method could be used to evaluate the quality ofM. suaveolens Ledeb and Melilotus genus herbs. The same strategy can also be applied for the development of fingerprints for the quality control of other herbal medicines.展开更多

Here we report a high-quality draft genome sequence of M. oleifera. This assembly represents 91.78% of the estimated genome size and contains 19,465 protein-coding genes. Comparative genomic analysis between M. oleifera and related woody plant genomes helps clarify the general evolution of this species, while the identification of several species-specific gene families and positively selected genes in M. oleifera may help identify genes related to M. oleifera's high protein content, fast-growth, heat and stress tolerance. This reference genome greatly extends the basic research on M. oleifera, and may further promote applying genomics to enhanced breeding and improvement of M. oleifera.展开更多

The long hist...Three Australian tephritid fruit flies (Bactrocera tryoni - Q-fly, Bactrocera neohumeralis - NEO, and Bactrocera jarvisi - JAR) are promising models for genetic studies of pest status and invasiveness. The long history of ecological and physi- ological studies of the three species has been augmented by the development of a range of genetic and genomic tools, including the capacity for forced multigeneration crosses between the three species followed by selection experiments, a draft genome for Q-fly, and tissue- and stage-specific transcriptomes. The Q-fly and NEO species pair is of particular interest. The distribution of NEO is contained entirely within the wider distribution of Q-fly and the two species are ecologically extremely similar, with no known differences in pheromones, temperature tolerance, or host-fruit utilisation. However there are three clear differences be- tween them: humeral callus colour, complete pre-mating isolation based on mating time-of-day, and invasiveness. NEO is much less invasive, whereas in historical times Q-fly has invaded southeastern Australia and areas of Western Australia and the North- em Territory. In southeastern fruit-growing regions, microsatellites suggest that some of these outbreaks might derive from ge- netically differentiated populations overwintering in or near the invaded area. Q-fly and NEO show very limited genome diffe- rentiation, so comparative genomic analyses and QTL mapping should be able to identify the regions of the genome controlling mating time and invasiveness, to assess the genetic bases for the invasive strains of Q-fly, and to facilitate a variety of improve- ments to current sterile insect control strategies for that species展开更多

In combination with local BLAST searches and gap filling techniques, a comprehensive antiSMASH-based method was adopted to assemble the secondary metabolite biosynthetic gene clusters in the draft genome of S. ansochromogenes. A total of at least 35 putative gene clusters were identified and assembled. Transcriptional analysis showed that 20 of the 35 gene clusters were expressed in either or all of the three different media tested, whereas the other 15 gene clusters were silent in all three different media. This study provides a comprehensive method to identify and assemble secondary metabolite biosynthetic gene clusters in draft genomes of Streptomyces, and will significantly promote functional studies of these secondary metabolite biosynthetic gene clusters.展开更多
Objective: The combination effect of Piperbetle (PB) and 5-fluorouracil (5-FU) in enhancing the cytotoxic potential of 5-FU in inhibiting the growth of colon cancer cells was investigated. Methods: HT29 and HCT1...Objective: The combination effect of Piperbetle (PB) and 5-fluorouracil (5-FU) in enhancing the cytotoxic potential of 5-FU in inhibiting the growth of colon cancer cells was investigated. Methods: HT29 and HCT116 cells were subjected to 5-FU or PB treatment. 5-FU and PB were then combined and their effects on both cell lines were observed after 24 h of treatment. PB-5-FU interaction was elucidated by isobologram analysis. Apoptosis features of the treated cells were revealed by annexin V/PI stain. High-performance liquid chromatography (HPLC) was performed to exclude any possible chemical interaction between the compounds. Results: In the presence of PB extract, the cytotoxicity of 5-FU was observed at a lower dose (IC^0 12.5 pmol/L) and a shorter time (24 h) in both cell lines. Both cell lines treated with 5-FU or PB alone induced a greater apoptosis effect compared with the combination treatment. Isobologram analysis indicated that PB and 5-FU interacted synergistically and antagonistically in inhibiting the growth of HT29 and HCT116 cells, respectively. Conclusions: In the presence of PB, a lower dosage of 5-FU is required to achieve the maximum drug effect in inhibiting the growth of HT29 cells. However, PB did not significantly reduce 5-FU dosage in HCT116 cells. Our result showed that this interaction may not solely contribute to the apoptosis pathway.展开更多

In the present study, we obtained the draft genome of an Aerophobetes bac- te...Aerophobetes (or CD12) is a recently defined bacterial phylum, of which the metabolic processes and ecological importance remain unclear. In the present study, we obtained the draft genome of an Aerophobetes bac- terium TCSI from saline sediment near the Thuwal cold seep in the Red Sea using a genome binning method. Analysis of 16S rRNA genes of TCS1 and close relatives revealed wide distribution of Aerophobetes in deep-sea sediments. Phylogenetic relationships showed affinity between Aerophobetes TCS 1 and some thermophilic bac- terial phyla. The genome of TCS1 (at least 1.27 Mbp) contains a full set of genes encoding core metabolic path- ways, including glycolysis and pyruvate fermentation to produce acetyl-CoA and acetate. The identification of cross-membrane sugar transporter genes further indicates its potential ability to consume carbohydrates preserved inthe sediment under the microbial mat. Aerophobetes bac- terium TCS1 therefore probably carried out saccharolytic and fermentative metabolism. The genes responsible for autotrophic synthesis of acetyl-CoA via the Wood-Ljung- dahl pathway were also found in the genome. Phylogenetic study of the essential genes for the Wood-Ljungdahl pathway implied relative independence of Aerophobetes bacterium from the known acetogens and methanogens. Compared with genomes of acetogenic bacteria, Aero- phobetes bacterium TCS 1 genome lacks the genes involved in nitrogen metabolism, sulfur metabolism, signal trans- duction and cell motility. The metabolic activities of TCS 1 might depend on geochemical conditions such as supplies of CO2, hydrogen and sugars, and therefore the TCSI might be a facultative bacterium in anaerobic saline sedi- ments near cold seeps.展开更多
Rainfall,runoff (surface runoff,interflow and groundwater runoff) and soil loss from 5 m × 15 m plots were recorded for 5 years (2001-2005) in an experiment with three treatments (cover,mulch and bare ground) on ...Rainfall,runoff (surface runoff,interflow and groundwater runoff) and soil loss from 5 m × 15 m plots were recorded for 5 years (2001-2005) in an experiment with three treatments (cover,mulch and bare ground) on sloping red soil in southern China.Surface runoff and erosion from the Bahia grass (Paspalum notatum Flugge) cover plot (A) and mulch plot (B) during the 5 years were low,despite the occurrence of potentially erosive rains.In contrast,the bare plot (C) had both the highest surface runoff coefficient and the highest sediment yield.There were significant differences in interflow and surface runoff and no significant difference in groundwater runoff among plots.The runoff coefficients and duration of interflow and groundwater runoff were in the order plot B > plot A > plot C.Effects of Bahia grass cover were excellent,indicating that the use of Bahia grass cover can be a simple and feasible practice for soil and water conservation on sloping red soil in the region.展开更多

The essential oils were analyzed by gas chromatography-mass spectrometry(GC-MS). Data were collected in full scan mode(m/z 60-600). Volatile components were identified based on their retention indices and by comparing their mass spectra with those in the National Institute of Standards and Technology 2005 database assisted by tandem mass spectrometry information. The relative content of each constituent wasdetermined by area normalization.RESULTS: We identified 111 components, of which12 were common to all 5 parts, 30 were found only in roots, 14 were found only in flowers, 6 were found only in leaves, 4 were found only in stems,and 17 were found only in fruits.CONCLUSION: Our results show that the stems,flowers, leaves, and fruits of Chaihu(Radix Bupleuri Chinensis)contain a high concentration of essential oils, and that the exact composition of the essential oils differs among the plant parts. To develop new medicines and make full use of the Chaihu(Radix Bupleuri Chinensis)resource, it is important to characterize the essential oils from different parts of the plant. In future research, it will be important to determine the pharmacological effects of the various components and the essential oil mixtures.展开更多

Comparative genomic analysis between M. oleifera and related woody plant genomes helps clarify the general evolution of this species, while the identification of several species-specific gene families and positively selected genes in M. oleifera may help identify genes related to M. oleifera's high protein content, fast-growth, heat and stress tolerance. This reference genome greatly extends the basic research on M. oleifera, and may further promote applying genomics to enhanced breeding and improvement of M. oleifera.
文摘Three Australian tephritid fruit flies (Bactrocera tryoni - Q-fly, Bactrocera neohumeralis - NEO, and Bactrocera jarvisi - JAR) are promising models for genetic studies of pest status and invasiveness. The long history of ecological and physi- ological studies of the three species has been augmented by the development of a range of genetic and genomic tools, including the capacity for forced multigeneration crosses between the three species followed by selection experiments, a draft genome for Q-fly, and tissue- and stage-specific transcriptomes. The Q-fly and NEO species pair is of particular interest. The distribution of NEO is contained entirely within the wider distribution of Q-fly and the two species are ecologically extremely similar, with no known differences in pheromones, temperature tolerance, or host-fruit utilisation. However there are three clear differences be- tween them: humeral callus colour, complete pre-mating isolation based on mating time-of-day, and invasiveness. NEO is much less invasive, whereas in historical times Q-fly has invaded southeastern Australia and areas of Western Australia and the North- em Territory. In southeastern fruit-growing regions, microsatellites suggest that some of these outbreaks might derive from ge- netically differentiated populations overwintering in or near the invaded area. Q-fly and NEO show very limited genome diffe- rentiation, so comparative genomic analyses and QTL mapping should be able to identify the regions of the genome controlling mating time and invasiveness, to assess the genetic bases for the invasive strains of Q-fly, and to facilitate a variety of improve- ments to current sterile insect control strategies for that species

文摘A draft genome sequence of Streptomyces ansochromogenes 7100 was generated using 454 sequencing technology. In combination with local BLAST searches and gap filling techniques, a comprehensive antiSMASH-based method was adopted to assemble the secondary metabolite biosynthetic gene clusters in the draft genome of S. ansochromogenes. A total of at least 35 putative gene clusters were identified and assembled. Transcriptional analysis showed that 20 of the 35 gene clusters were expressed in either or all of the three different media tested, whereas the other 15 gene clusters were silent in all three different media. This study provides a comprehensive method to identify and assemble secondary metabolite biosynthetic gene clusters in draft genomes of Streptomyces, and will significantly promote functional studies of these secondary metabolite biosynthetic gene clusters.

文摘Objective: The combination effect of Piperbetle (PB) and 5-fluorouracil (5-FU) in enhancing the cytotoxic potential of 5-FU in inhibiting the growth of colon cancer cells was investigated. Methods: HT29 and HCT116 cells were subjected to 5-FU or PB treatment. 5-FU and PB were then combined and their effects on both cell lines were observed after 24 h of treatment. PB-5-FU interaction was elucidated by isobologram analysis. Apoptosis features of the treated cells were revealed by annexin V/PI stain. High-performance liquid chromatography (HPLC) was performed to exclude any possible chemical interaction between the compounds. Results: In the presence of PB extract, the cytotoxicity of 5-FU was observed at a lower dose (IC^0 12.5 pmol/L) and a shorter time (24 h) in both cell lines. Both cell lines treated with 5-FU or PB alone induced a greater apoptosis effect compared with the combination treatment. Isobologram analysis indicated that PB and 5-FU interacted synergistically and antagonistically in inhibiting the growth of HT29 and HCT116 cells, respectively. Conclusions: In the presence of PB, a lower dosage of 5-FU is required to achieve the maximum drug effect in inhibiting the growth of HT29 cells. However, PB did not significantly reduce 5-FU dosage in HCT116 cells. Our result showed that this interaction may not solely contribute to the apoptosis pathway.

文摘Aerophobetes (or CD12) is a recently defined bacterial phylum, of which the metabolic processes and ecological importance remain unclear. In the present study, we obtained the draft genome of an Aerophobetes bac- terium TCSI from saline sediment near the Thuwal cold seep in the Red Sea using a genome binning method. Analysis of 16S rRNA genes of TCS1 and close relatives revealed wide distribution of Aerophobetes in deep-sea sediments. Phylogenetic relationships showed affinity between Aerophobetes TCS 1 and some thermophilic bac- terial phyla. The genome of TCS1 (at least 1.27 Mbp) contains a full set of genes encoding core metabolic path- ways, including glycolysis and pyruvate fermentation to produce acetyl-CoA and acetate. The identification of cross-membrane sugar transporter genes further indicates its potential ability to consume carbohydrates preserved inthe sediment under the microbial mat. Aerophobetes bac- terium TCS1 therefore probably carried out saccharolytic and fermentative metabolism. The genes responsible for autotrophic synthesis of acetyl-CoA via the Wood-Ljung- dahl pathway were also found in the genome. Phylogenetic study of the essential genes for the Wood-Ljungdahl pathway implied relative independence of Aerophobetes bacterium from the known acetogens and methanogens. Compared with genomes of acetogenic bacteria, Aero- phobetes bacterium TCS 1 genome lacks the genes involved in nitrogen metabolism, sulfur metabolism, signal trans- duction and cell motility. The metabolic activities of TCS 1 might depend on geochemical conditions such as supplies of CO2, hydrogen and sugars, and therefore the TCSI might be a facultative bacterium in anaerobic saline sedi- ments near cold seeps.

文摘Rainfall,runoff (surface runoff,interflow and groundwater runoff) and soil loss from 5 m × 15 m plots were recorded for 5 years (2001-2005) in an experiment with three treatments (cover,mulch and bare ground) on sloping red soil in southern China.Surface runoff and erosion from the Bahia grass (Paspalum notatum Flugge) cover plot (A) and mulch plot (B) during the 5 years were low,despite the occurrence of potentially erosive rains.In contrast,the bare plot (C) had both the highest surface runoff coefficient and the highest sediment yield.There were significant differences in interflow and surface runoff and no significant difference in groundwater runoff among plots.The runoff coefficients and duration of interflow and groundwater runoff were in the order plot B > plot A > plot C.Effects of Bahia grass cover were excellent,indicating that the use of Bahia grass cover can be a simple and feasible practice for soil and water conservation on sloping red soil in the region.

文摘OBJECTIVE: To analyze the essential oils from flowers, leaves, stems, roots, and fruits of Chaihu(Radix Bupleuri Chinensis).METHODS: We extracted essential oils from different parts of Chaihu(Radix Bupleuri Chinensis) using a steam distillation method. The essential oils were analyzed by gas chromatography-mass spectrometry(GC-MS). Data were collected in full scan mode(m/z 60-600). Volatile components were identified based on their retention indices and by comparing their mass spectra with those in the National Institute of Standards and Technology 2005 database assisted by tandem mass spectrometry information. The relative content of each constituent wasdetermined by area normalization.RESULTS: We identified 111 components, of which12 were common to all 5 parts, 30 were found only in roots, 14 were found only in flowers, 6 were found only in leaves, 4 were found only in stems,and 17 were found only in fruits.CONCLUSION: Our results show that the stems,flowers, leaves, and fruits of Chaihu(Radix Bupleuri Chinensis)contain a high concentration of essential oils, and that the exact composition of the essential oils differs among the plant parts. To develop new medicines and make full use of the Chaihu(Radix Bupleuri Chinensis)resource, it is important to characterize the essential oils from different parts of the plant. In future research, it will be important to determine the pharmacological effects of the various components and the essential oil mixtures.
