In previous studies, isoenzyme, RAPD and SSR techniques had been used to conduct the genetic identification of watermelon ( Citrullus lanatus (Thunb.) Mansf.), but their effectiveness was limited due to the extremely narrow genetic background among watermelon genotypes. In this research, amplified fragment length polymorphism (AFLP), which was reported as a reliable technique with high efficiency in detecting polymorphism, was used to conduct genetic analysis and variety identification of thirty genotypes of watermelon core collection that represent a wide range of breeding and commercially available germplasm. As a result, a DNA fingerprint based on 15 bands amplified with four primer combinations was developed. In this fingerprint, each genotype has its unique fingerprint pattern and can be distinguished from each other. Furthermore, in or der to facilitate the utilization of AFLP marker in practice, one specific AFLP band of genotype 'PI296341' coming from fragment amplified by primer combination E-AT/M-CAT was successfully converted into a sequence characterized amplified region (SCAR) marker.展开更多

The polymorphism information content (PIC) values varied from 0.19 to 0.74, with an average of 0.50. The clustering analysis by UPG-MA indicated that the genetic distance among the tested lines was 0.22-0.63, with an average of 0.43. The utilization and changes of forest resources were studied in the Lishuihe Forest Bureau. Based on remote sensing images in 1985 and 1999, changes of major forest resources were analyzed by statistical and overlap m...The utilization and changes of forest resources were studied in the Lishuihe Forest Bureau. Based on remote sensing images in 1985 and 1999, changes of major forest resources were analyzed by statistical and overlap method and classified quantitatively. The results showed that in recent 15 years, logging spots and man-made young forest changed violently, which was due to human activities. Different forest management manners and harvesting intensity played an important role in forest resources change. Dongsheng and Xilinhe tree farms were typical cases of different forest status and management for the Bu-reau, where forest succession was intervened by either human or natural disturbance. Dongsheng Tree Farm underwent a light harvest intensity and maintained a unit stock volume of 536.27 m3hm-2, as much as that of broadleaf/Korean pine forest of Changbai Mountain Natural Reserve; Xilinhe Tree Farm underwent an intense harvest and was composed of secondary forests, where mature forests just had a small percentage and the unit stock volume was low. The study was useful to guide future forest management. What抯 more, problems found in the research were also analyzed and reasonable advice was given to the local forest management.展开更多

Under such a high Cd concentration, the subcellular accumulation of Cd in root, stem and leaf of this plant was found to be the highest in the cell wall, less in the soluble fraction and lowest in the cell organs. The mode of subcellular distribution of Cd in the mining ecotype S. alfredii was similar to other hyper accumulators of heavy metals, in which Cd was distributed more in the aerial part of plant. The results suggest that the mining ecotype S. alfredii is a new species of Cd hyperaccumulator.展开更多

[Objective] This study was to reveal the genetic diversity and genetic relationship of the kenaf(Hibiscus cannabinus L.) resources from different origins, thus providing basis for genetic improvement and molecular mar...[Objective] This study was to reveal the genetic diversity and genetic relationship of the kenaf(Hibiscus cannabinus L.) resources from different origins, thus providing basis for genetic improvement and molecular marker-assisted breeding of kenaf. [Method] Ninety one ISSR molecular markers were used for amplification on 44 shares of kenaf germplasm resources, of which 21 showing good diversity and clear bands were chosen for PCR amplification. Based on amplification results, the genetic similarity coefficients among kenaf germplasm resources were calculated by using analytic software NTSYSpc-2.10e, and phylogenetic tree was then established via UPGMA. [Result] Totally 169 bands were amplified using the 21 screened primers, averagely 8.05 bands were amplified from each primer. Of them, 141 bands were polymorphic, accounting for 83.4%. When genetic similarity coefficient 0.887 was used as criterion L1, these 44 shares of kenaf germplasm could be classified to be 32 shares of cultivars and 12 shares of wild type or half-wild type varieties. When genetic similarity coefficient 0.897 was used as criterion L2, these 32 shares of cultivars could be further grouped into four sub-clusters. The genetic diversities between cultivars and wild type or half-wild type varieties were between 0.46-0.91, showing huge hereditary difference; while that among 32 cultivars were between 0.85-0.97, suggesting that genetic relationships among cultivars are relatively close and their genetic similarities are rather narrow. [Conclusion] ISSR could well determine the genetic similarities among kenaf germplasm resources and provide valuable molecular information for selecting parents of hybrid cross, which can lay a good foundation for DNA mapping of kenaf germplasm resources.展开更多
[Objective] This study aimed to investigate the genetic diversity of agronomic traits and genetic relationships among core collections of bitter gourd.[Method] Total 141 germplasms of bitter gourd were selected,and th...[Objective] This study aimed to investigate the genetic diversity of agronomic traits and genetic relationships among core collections of bitter gourd.[Method] Total 141 germplasms of bitter gourd were selected,and the genetic diversity of 13 agronomic traits was analyzed.In addition,total 46 core collections of bitter gourd were employed,and their genetic relationships were analyzed based on the phenotypic values and genotypic values of 5 agronomic traits,respectively.[Result] The genetic diversity analysis of agronomic traits showed that the genetic diversity indexes of the 4 qualitative traits of bitter gourd germplasms ranged from 0.46 to 1.34;the distribution of the 9 quantitative traits data was more dispersed with average coefficient of variation of 20.02%.The genetic relationship analysis showed that based on the phenotypic values and genotypic values of the 5 quantitative traits,the genetic distances among the 46 core collections of bitter gourd were different.Based on the genotypic values,the genetic distances among the 46 bitter gourd core collections ranged from 0.84 to 10.71.The 46 germplasms were divided into 17 groups with the rescaled distance of 8.5,which further classified the relationships among different germplasms.[Conclusion] This study will lay a solid foundation for the effective utilization of core collections and new variety breeding in bitter gourd.展开更多

The results showed that there were significant differences in seed oil content between different cucumber varieties with the amplitude of variation rangeing from 31.03% to 40.59%, among which the seed oil content of south China cucumber was the highest of 40.59%, while the seed oil content of north China cucumber was the lowest of 31.03%. [Method] Comprehensive analysis combining cl...[Objective] This study aimed to screen out hot pepper germplasms highly resistant to Meloidogyne incognita, thereby providing resistant resources for hot pep- per breeding. [Method] Comprehensive analysis combining cluster analysis and sub- ordinate function was conducted through determining related resistance indexes of 67 hot pepper germplasms 50 days after inoculated with M. incognita. [Result] The effects of M. incognita on related resistance indexes were significantly different am- ong the hot pepper germplasms. Egg index and gall index had abundant genetic variation with variation coefficients of 143.16% and 118.95%, respectively. Based on the gall indexes, cluster analysis of hot pepper germplasms was performed. The 67 hot pepper germplasms were divided into 4 groups (resistant, moderately resistant, susceptible and high susceptible). The resistance intensity of the hot pepper germplasms were ranked according to the sum of subordinate function values of various resistance indexes. The total function values of Rela 2 and L506M were the largest (2.00), indicating that these two germplasms were immune to M. incognita. The total function values of L287-2, L522-1M, L504M, L515-2, 13SM100-1, L512M, L292-1, L319, L316, L317, 13SM87-1 and Rela 5 were larger than 1.95, indicating that these germplasms were highly resistant to M. incognita. [Conclusion] This study could provide certain resistant resources for resistance breeding of hot pepper to M. incognita.展开更多

We found significant but various depressing levels of germination rate under salt stress among different germplasms, and only 5% of 128 germplasms were highly resistant to salinity. We also found that the relative germination index was a useful evaluation index for salt tolerance besides salt damage rate and rela- tive germination rate. After all, we comprehensively screened out 7 materials (JS011, JS024, JS125, JS491, JS523, JS524 and JS525) as salt tolerance germplasms for further research. Key展开更多

文摘The cashew tree is a typical tropical nut tree species in China, and the collection, identification and innovated utilization of excellent cashew germplasm resources are of great significance to increase the income of farmers and agricultural benefits in the tropical area of China. This study summarized the major advances on cashew germplasm resources in China, analyzed the major research progress and existing problems from the aspects of the collection and evaluation, resistances, genetic diversity of cashew germplasm resources, and put forward the work ideas to analyze the construction of core germplasm bank and the genetic stability of the major biological characteristics of cashew germplasm resources, which were of important significance for the improvement of the cultivar classification and resource evaluation system of cashew germplasm resources in China.

文摘[Objective] The paper was to establish tea germplasm database of Yunnan Province,and promote sharing of the tea germplasm resources.[Method] Eight hundred and thirty copies of tea germplasm resources of Yunnan Province were first systematically documented by using Access database software,the generic description of 631 tea resources and characteristic description of 300 tea resources were submitted for e-platform,then linked with the national e-platform for natural scientific and technological resources,and the tea germplasm database of Yunnan Province was established.[Result] Based on the conservation and utilization status of tea germplam resources,the sharing and utilization framework of tea germplam resources was presented.Many problems and suggestion about tea germplasm resources in the process of conservation,documentation concordance and sharing were pointed out.For example,conservation areas were separated and system was not completed;the main traits assessment and identification researching work had not completely accomplished and sharing was inefficient.[Conclusion] The paper laid foundation for standardized,digitized and information-based management of tea germplasm resources.

文摘[Objective] The aim was to screen out excellent Aceraceae germplasm resources which can be applied in gardens and urban landscape. [Method] The set of 17 biological traits of 24 Aceraceae species were observed and recorded. Taking into consideration aesthetic evaluation of different groups of people, the studied species were graded in quantitative scale. The species with higher overall score is considered better germplasm resource. [Result] The Aceraceae species with top ten highest overall scores were: Acer japonicum 'Aconitifolium' (45.5), Acer palmatum var. thunbergii (43.8), Acer palmatum (41.8), Acer elegantulum (39.2), Acer elegantulum var. macrurum (39.2), Acer rubrum (39.1), Acer Cordatum(38.8), Acer truncatum (38.3), Acer olivaceum (37.7) and Acer acutum (37.3). [Conclusion] The above ten species can be used preferentially as excellent Aceraceae germplasm resources for landscaping.

文摘[Objective] This study was to reveal the genetic diversity and genetic relationship of the kenaf(Hibiscus cannabinus L.) resources from different origins, thus providing basis for genetic improvement and molecular marker-assisted breeding of kenaf. [Method] Ninety one ISSR molecular markers were used for amplification on 44 shares of kenaf germplasm resources, of which 21 showing good diversity and clear bands were chosen for PCR amplification. Based on amplification results, the genetic similarity coefficients among kenaf germplasm resources were calculated by using analytic software NTSYSpc-2.10e, and phylogenetic tree was then established via UPGMA. [Result] Totally 169 bands were amplified using the 21 screened primers, averagely 8.05 bands were amplified from each primer. Of them, 141 bands were polymorphic, accounting for 83.4%. When genetic similarity coefficient 0.887 was used as criterion L1, these 44 shares of kenaf germplasm could be classified to be 32 shares of cultivars and 12 shares of wild type or half-wild type varieties. When genetic similarity coefficient 0.897 was used as criterion L2, these 32 shares of cultivars could be further grouped into four sub-clusters. The genetic diversities between cultivars and wild type or half-wild type varieties were between 0.46-0.91, showing huge hereditary difference; while that among 32 cultivars were between 0.85-0.97, suggesting that genetic relationships among cultivars are relatively close and their genetic similarities are rather narrow. [Conclusion] ISSR could well determine the genetic similarities among kenaf germplasm resources and provide valuable molecular information for selecting parents of hybrid cross, which can lay a good foundation for DNA mapping of kenaf germplasm resources.
文摘[Objective] This study aimed to investigate the genetic diversity of agronomic traits and genetic relationships among core collections of bitter gourd.[Method] Total 141 germplasms of bitter gourd were selected,and the genetic diversity of 13 agronomic traits was analyzed.In addition,total 46 core collections of bitter gourd were employed,and their genetic relationships were analyzed based on the phenotypic values and genotypic values of 5 agronomic traits,respectively.[Result] The genetic diversity analysis of agronomic traits showed that the genetic diversity indexes of the 4 qualitative traits of bitter gourd germplasms ranged from 0.46 to 1.34;the distribution of the 9 quantitative traits data was more dispersed with average coefficient of variation of 20.02%.The genetic relationship analysis showed that based on the phenotypic values and genotypic values of the 5 quantitative traits,the genetic distances among the 46 core collections of bitter gourd were different.Based on the genotypic values,the genetic distances among the 46 bitter gourd core collections ranged from 0.84 to 10.71.The 46 germplasms were divided into 17 groups with the rescaled distance of 8.5,which further classified the relationships among different germplasms.[Conclusion] This study will lay a solid foundation for the effective utilization of core collections and new variety breeding in bitter gourd.

文摘With 35 different ecological types of cucumber germplasm resources as test materials, the soxhlet extraction was used to extract the crude fat from the cu- cumber seeds to analyze the seed oil content, with the aim to lay foundations for the application and new variety breeding related with cucumber seed oil. The results showed that there were significant differences in seed oil content between different cucumber varieties with the amplitude of variation rangeing from 31.03% to 40.59%, among which the seed oil content of south China cucumber was the highest of 40.59%, while the seed oil content of north China cucumber was the lowest of 31.03%. Four va.rieties with oil content of greater than 38.50% and four varieties with oil content of less than 32.01% werescreened out.

文摘[Objective] This study aimed to screen out hot pepper germplasms highly resistant to Meloidogyne incognita, thereby providing resistant resources for hot pep- per breeding. [Method] Comprehensive analysis combining cluster analysis and sub- ordinate function was conducted through determining related resistance indexes of 67 hot pepper germplasms 50 days after inoculated with M. incognita. [Result] The effects of M. incognita on related resistance indexes were significantly different am- ong the hot pepper germplasms. Egg index and gall index had abundant genetic variation with variation coefficients of 143.16% and 118.95%, respectively. Based on the gall indexes, cluster analysis of hot pepper germplasms was performed. The 67 hot pepper germplasms were divided into 4 groups (resistant, moderately resistant, susceptible and high susceptible). The resistance intensity of the hot pepper germplasms were ranked according to the sum of subordinate function values of various resistance indexes. The total function values of Rela 2 and L506M were the largest (2.00), indicating that these two germplasms were immune to M. incognita. The total function values of L287-2, L522-1M, L504M, L515-2, 13SM100-1, L512M, L292-1, L319, L316, L317, 13SM87-1 and Rela 5 were larger than 1.95, indicating that these germplasms were highly resistant to M. incognita. [Conclusion] This study could provide certain resistant resources for resistance breeding of hot pepper to M. incognita.
文摘Rhododendron simsii(Ericaeae:Rhododendron) has high ornamental value and ecological value.In this study,7 pairs of novel EST-SSR markers were developed from the genomic sequence of R.simsii,and they were used to investigate the genetic diversity of 32 natural R.simsii samples from Guifeng Mountain,Hubei Province.Results showed that a total of 31 polymorphic bands were amplified with allele number per locus of 4.43.Mean values of heterozygosity(Ho) and expected heterozygosity(He) were 0.679 58 and 0.723 14,respectively.This research will not only enrich the existing SSR database,but also lay a foundation for subsequent studies about molecular marker-assisted breeding,genetic diversity analysis,genetic structure analysis and phylogenetic analysis.

文摘For screening out salt tolerance germplasm and providing materials for ge- netic research of peanut, based on the indexes including relative germination poten- tial, relative germination rate, relative germination index and salt-injury rate, 128 peanut germplasms were selected for salt-tolerant identification and estimation under 2.5% NaCI. We found significant but various depressing levels of germination rate under salt stress among different germplasms, and only 5% of 128 germplasms were highly resistant to salinity. We also found that the relative germination index was a useful evaluation index for salt tolerance besides salt damage rate and rela- tive germination rate. After all, we comprehensively screened out 7 materials (JS011, JS024, JS125, JS491, JS523, JS524 and JS525) as salt tolerance germplasms for further research. Key

文摘[Objective] To utilize the germplasm resources,new systematic evidence for the classification of sect.Paracamellia,genus Camellia is found in this article.[Method] Fruit anatomy of 10 species representing sect.Paracamellia was investigated.And the main component of sect.Paracamellia in the genus Camellia was studied by MVSP software.[Result] The exocarp was thin;mesocarp was broad,having a large number of stone cells;endocarp was lignified,and separated easily from seed.The principle components analysis suggested that the distance between species was far,showing far relation;the current study did not support the formation of Camellia grijsii(C.grijsii)and Camellia yuhsienensis(C.yuhsienensis),Camellia shensiensis(C.shensiensis)should not be recognized as a variety of C.yuhsienensis.In addition,it also proved Ming's combining of Camellia confusa(C.confusa)and Camellia kissi(C.kissi)was unnatural.[Conclusion] The study provided theoretical basis for the development and application of sect.Paracamellia in the genus Camellia.
