METHODS Fat and lean culture media were developed...AIM To develop a human in vitro model of non-alcoholic fatty liver disease(NAFLD), utilising primary hepatocytes cultured in a three-dimensional(3D) perfused platform. METHODS Fat and lean culture media were developed to directly investigate the effects of fat loading on primary hepatocytes cultured in a 3D perfused culture system. Oil Red O staining was used to measure fat loading in the hepatocytes and the consumption of free fatty acids(FFA) from culture medium was monitored. Hepatic functions, gene expression profiles and adipokine release were compared for cells cultured in fat and lean conditions. To determine if fat loading in the system could be modulated hepatocytes were treated with known anti-steatotic compounds. RESULTS Hepatocytes cultured in fat medium were found to accumulate three times more fat than lean cells and fat uptake was continuous over a 14-d culture. Fat loading of hepatocytes did not cause any hepatotoxicity and significantly increased albumin production. Numerous adipokines were expressed by fatty cells and genes associated with NAFLD and liver disease were upregulated including: Insulin-like growth factorbinding protein 1, fatty acid-binding protein 3 and CYP7A1. The metabolic activity of hepatocytes cultured in fatty conditions was found to be impaired and the activities of CYP3A4 and CYP2C9 were significantlyreduced, similar to observations made in NAFLD patients. The utility of the model for drug screening was demonstrated by measuring the effects of known antisteatotic compounds. Hepatocytes, cultured under fatty conditions and treated with metformin, had a reduced cellular fat content compared to untreated controls and consumed less FFA from cell culture medium.CONCLUSION The 3D in vitro NAFLD model recapitulates many features of clinical NAFLD and is an ideal tool for analysing the efficacy of anti-steatotic compounds.展开更多

The objectives of this study were to measure water-retention curves, θ(ψ), and unsaturated hydraulic conductivity functions, K(ψ), of the soils sampled at different slope positions in three directions, namely, in vertical direction, along the slope and along the contour, and to determine the effects of sampling direction and slope position of two soil catenas. At the upper slope positions, the surface soils (0-10 cm) sampled in the vertical direction had a lower soil water content, 0, at a certain soil water potential (-1 500 kPa 〈 ψ 〈 -10 kPa) and had the greatest unsaturated hydraulic conductivity, K, at ψ 〉 -10 kPa. At the lower slope positions, K at ψ〉 -10 kPa was smaller in the vertical direction than in the direction along the slope. The deep soils (100 110 cm) had similar soil hydraulic properties in all the three directions. The anisotropic variations of the hydraulic properties of the surface soils were ascribed to the effects of natural wetting and drying cycles on the structural heterogeneity. These results suggested that the anisotropy of soil hydraulic properties might be significant in influencing soil water movement along the slope and need to be considered in modeling.展开更多

Methods hAMSCs were isolated from human amniotic membrane by trypsin-collagenase digestion, and cultured in Dulbecco's modified Eagle's medinm/F12 medium supplemented with 10% fetal bovine serum. Phenotypic characteristics of these cells were analyzed by means of immunocytochemistry and flow cytometry. Results The cells successfully isolated from human amniotic membrane expressed representative mesenchymal cell surface markers CD44, CD90, and vimentin, but not CD45. Conclusions This study establishes a potential method for isolation of hAMSCs from human amnion, in vitro culture, and identification. The isolated cells show phenotypic characteristics of mesenchymal stem cells.展开更多

Cultural landscapes in China are faced with the crisis of fragmentation and i...The relationship between cross-regional cultural landscapes does not currently receive enough attention in cultural landscape conservation. Cultural landscapes in China are faced with the crisis of fragmentation and islanding, which makes it necessary to strengthen the idea of integrated conservation. The Great Wall cultural belt in Beijing is a cross-regional linear cultural landscape in the northwest mountainous area of Beijing. With the Great Wall as a link connecting the surrounding natural, social, and cultural resources to form a banded structure, it has the typical and practical significance of integrated conservation. Based on the theory of the cultural landscape security pattern and its ‘node – setting – connection' model, this research studies the spatial structure and shaping mechanism of the Great Wall cultural belt in Beijing from the perspective of tourist flow, using Flickr geo-tagged photos and Wikiloc tracks. The results identify the core nodes and intermediate nodes of the Great Wall cultural belt in Beijing and show that it has a multicore, multi-group hierarchical nesting structure differing from the geographic space, which has groups following the evolution orbit of ‘linear string – circumnuclear star – complete network.' To explain the differences between the cultural association network(CAN) and the geospatial network(GSN), we find that the cultural associations between nodes rely on geophysical channels, including heritage corridors and traffic passages, as physical carriers, but the nodes' attraction and their setting elements, containing natural geography environments, settlements, administrative divisions, etc., change the channel resistance, so CAN and GSN present different characteristics. From the perspective of systematic and spatial network thoughts, this study has made an attempt to adopt new analysis and research methods to achieve the integrated conservation and inheritance of linear cultural landscapes in mountainous areas.展开更多

For many, the living room is the locus of product use, social activities, cultural values, and the person...Consumption is an active process that engages the social world for individuals to make use of certain products. For many, the living room is the locus of product use, social activities, cultural values, and the personal narrative of the people who live in it. Consumers engage with their furniture in order for it to function effectively for them: It is thus used to furnish both the home and the self as well. Creating a certain living space with a particular use of furniture and interior decoration is the precise way in which the resident forms its arrangements, and builds a relationship with the surrounding objects in that living space. This activity, I argue, positions the residents in the process of presenting their personality characteristics and cultural identity. The Ethnographic approach is most appropriate methodology for research into such a personal area: the living room within the domestic environment. The topic of this research falls between different fields of studies including consumerism and media. This research reviews the influence of advertising in magazines and furniture shops on the meaning of living space design and how that encourages higher levels of consumption.展开更多

Emotional two-character Chinese words are used a...This study addresses the problem of classifying emotional words based on recorded electroencephalogram (EEG) signals by the single-trial EEG classification technique. Emotional two-character Chinese words are used as experimental materials. Positive words versus neutral words and negative words versus neutral words are classified, respectively, using the induced EEG signals. The method of temporally regularized common spatial patterns (TRCSP) is chosen to extract features from the EEG trials, and then single-trial EEG classification is achieved by linear discriminant analysis. Classification accuracies are between 55% and 65%. The statistical significance of the classification accuracies is confirmed by permutation tests, which shows the successful identification of emotional words and neutral ones, and also the ability to identify emotional words. In addition, 10 out of 15 subjects obtain significant classification accuracy for negative words versus neutral words while only 4 are significant for positive words versus neutral words, which demonstrate that negative emotions are more easily identified.展开更多
Spatial variation of dissolved organic carbon(DOC) in soils of riparian wetlands and responses to hydro-geomorphologic changes in the Sanjiang Plain were analyzed through in situ collecting soil samples in the Naoli R...Spatial variation of dissolved organic carbon(DOC) in soils of riparian wetlands and responses to hydro-geomorphologic changes in the Sanjiang Plain were analyzed through in situ collecting soil samples in the Naoli River and the Bielahong River. The results showed that the average contents of DOC for soil layer of 0–100 cm were 730.6 mg/kg, 250.9 mg/kg, 423.0 mg/kg and 333.1 mg/kg respectively from riverbed to river terrace along the transverse directions of the Naoli watershed. The content of the soil DOC was the highest in the riverbed, lower in the high floodplain and much lower in the river terrace, and it was the lowest in the low floodplain. The difference in the content and vertical distribution of DOC between the riverbed and the three riparian wetlands was significant, while it was not significant among the low floodplain, the high floodplain and the river terrace. The variability of soil DOC was related to the hydrological connectivity between different landscape position of the riparian wetlands and the adjacent stream. Extremely significant correlations were observed between DOC and total organic carbon(TOC), total iron(TFe), ferrous iron(Fe(II)) whose correlation coefficients were 0.819, –0.544 and –0.709 in riparian wetlands of the Naoli River. With the increase of wetland destruction, soil p H increased and soil DOC content changed. The correlation coefficients between soil DOC and TOC, TFe, Fe(II) also changed into 0.759, –0.686 and –0.575 respectively in the Bielahong River. Under the impact of drainage ditches, the correlations between soil DOC and TFe, Fe(II) were not obvious, while the soil p H was weakly alkaline and was negatively correlated with soil DOC in the previous high floodplain. It indicates that riparian hydro-geomorphology is the main factor that could well explain this spatial variability of soil DOC, and the agricultural environmental hydraulic works like ditching also must be considered.展开更多

Presently, colchicine has been produced by using the seeds of C. autumnale L. and the tubers of G. superba L. through different chemical extraction methods. The most important reason for this is seen in the fact that texts since the advent of print culture have been disseminated and passed on in written form and that in the medium of writing the processual character of language is only insufficiently taken care of by a cultural convention of arranging and approaching the presented signs in an particular sequence. Whereas in an oral culture the dynamic processuality of a speech or recitation was directly experienced by the listeners, the spatial arrangement of signs in writing enables and even entices readers and critics to read this or that part of a written text in a sequence of their own making. What remains out of focus is that in doing so they miss the particular semantic profile and aesthetic character of the work as created by the author--a procedure particularly hurtful in the case of literary works of art. However, the space features of creative industries and its association with local socio-cultural contexts have not been fully understood, especially at a micro-city level. This study attempts to understand the agglomeration of creative industries in Shanghai from the sociology perspective. For this study, this paper utilizes primarily a questionnaire survey to explain the space features of creative industries in Shanghai. The results indicate an extensive socio-cultural embeddedness of the agglomeration of creative industries in Shanghai. First, strong emphasis on face-to-face contacts by creative professionals makes geographical agglomeration necessary for creative industries. Second, the reason why inner city of Shanghai is popular among creative professionals and enterprises lies in the diversity of cultures and special environment of the former colonial zones of Shanghai. Additionally, highly concentrated dining and entertainment facilities in the central city of Shanghai offer creative workers social networking places and nightlife venues. Third, as the educational attainment of local citizens and the protection of intellectual property are highly stressed by creative professionals, research and design specialized creative industries are more likely located near universities and research institutes.展开更多

Although he raised questions about cultural mores, morality, and religiosity, none are judgmental. On the contrary, it is the viewer who might or might not articulate a moral judgment according to personal experiences. Creating a certain living space with a particular use of furniture and interior decoration is the precise way in which the resident forms its arrangements, and builds a relationship with the surrounding objects in that living space. This activity, I argue, positions the residents in the process of presenting their personality characteristics and cultural identity. The Ethnographic approach is most appropriate methodology for research into such a personal area: the living room within the domestic environment. The topic of this research falls between different fields of studies including consumerism and media. This research reviews the influence of advertising in magazines and furniture shops on the meaning of living space design and how that encourages higher levels of consumption.
文摘The difference between the Chinese time view and the western time view originates from agricultural civilization's inertia and industrial civilization's feeling way. This time coordinate controls people's behavior, awareness of concepts, industrial relationship, interpersonal relationship and value orientation. Different cultures bred different time conceptions and time behaviors. This paper mainly talks about the aspects of their differences. When different time views are reflected in cross-cultural communication, conflicts and obstacles of interpersonal communication may arise. With the development of social economy and cultural communication, the time views of Chinese culture and western culture have impacts on each other and are gradually integrated with each other.
文摘In the thirteenth century the Mongols created a vast, transcontinental empire that intensified cultural, art, craftwork, garment and commercial contact throughout Eurasia. Hats were so indispensable parts of formal Mongol dress that were seen as a symbol of Mongol traditional garment. Roughly Mongolian men's headgear can be divided into two groups, which are Li and Mao. In addition, Mongolian women's Gu-gu-gwan, a kind of bonnet, was the head-dress for the empress, concubines of the emperor, and wife of high ranking ministers. This study, based on the analysis of image resources and existing evidence, discusses the hats of Yuan period in the context of its group, origin, decoration and cultural influences.

文摘This study addresses the problem of classifying emotional words based on recorded electroencephalogram (EEG) signals by the single-trial EEG classification technique. Emotional two-character Chinese words are used as experimental materials. Positive words versus neutral words and negative words versus neutral words are classified, respectively, using the induced EEG signals. The method of temporally regularized common spatial patterns (TRCSP) is chosen to extract features from the EEG trials, and then single-trial EEG classification is achieved by linear discriminant analysis. Classification accuracies are between 55% and 65%. The statistical significance of the classification accuracies is confirmed by permutation tests, which shows the successful identification of emotional words and neutral ones, and also the ability to identify emotional words. In addition, 10 out of 15 subjects obtain significant classification accuracy for negative words versus neutral words while only 4 are significant for positive words versus neutral words, which demonstrate that negative emotions are more easily identified.

文摘Spatial variation of dissolved organic carbon(DOC) in soils of riparian wetlands and responses to hydro-geomorphologic changes in the Sanjiang Plain were analyzed through in situ collecting soil samples in the Naoli River and the Bielahong River. The results showed that the average contents of DOC for soil layer of 0–100 cm were 730.6 mg/kg, 250.9 mg/kg, 423.0 mg/kg and 333.1 mg/kg respectively from riverbed to river terrace along the transverse directions of the Naoli watershed. The content of the soil DOC was the highest in the riverbed, lower in the high floodplain and much lower in the river terrace, and it was the lowest in the low floodplain. The difference in the content and vertical distribution of DOC between the riverbed and the three riparian wetlands was significant, while it was not significant among the low floodplain, the high floodplain and the river terrace. The variability of soil DOC was related to the hydrological connectivity between different landscape position of the riparian wetlands and the adjacent stream. Extremely significant correlations were observed between DOC and total organic carbon(TOC), total iron(TFe), ferrous iron(Fe(II)) whose correlation coefficients were 0.819, –0.544 and –0.709 in riparian wetlands of the Naoli River. With the increase of wetland destruction, soil p H increased and soil DOC content changed. The correlation coefficients between soil DOC and TOC, TFe, Fe(II) also changed into 0.759, –0.686 and –0.575 respectively in the Bielahong River. Under the impact of drainage ditches, the correlations between soil DOC and TFe, Fe(II) were not obvious, while the soil p H was weakly alkaline and was negatively correlated with soil DOC in the previous high floodplain. It indicates that riparian hydro-geomorphology is the main factor that could well explain this spatial variability of soil DOC, and the agricultural environmental hydraulic works like ditching also must be considered.

In the book The Silence Language, Edward hall first uses the term proxemics, and he has given a detailed analysis of proxemics. Proxemics is the study of spatial distances between individuals in different cultures and situations. Cultural factors for proxemics has a great influence, and the factors such as gender and religion also has a profound significance on proximics. This thesis will mainly analyze this phenomenon in the way of the definition and theory ofproximics, especially, the ways how people deal with the international culture situation. In order to improve the readers cultural accomplishment and ability to communicate, prevent the errors in the protocol, this paper will further explain how to solve these cross-cultural problems in cross-cultural context, especially for Chinese learners. With some fresh insights into international culture communication research, hopefully the readers may benefit from this paper and communicate with foreign people in a better way.
文摘The secondary metabolites synthesized by plants are economically important chemical compounds in the agricultural and industrial areas such as food, perfumery and pharmaceutical sectors. In recent years, attempts for their production by in vitro plant cell and tissue cultures have been accelerated considerably. Colchicine, the principle secondary metabolite of Colchicum autumnale L. and Gloriosa superba L., is an important alkaloid that has poison effect used for treatment of various diseases and plant breeding studies. Presently, colchicine has been produced by using the seeds of C. autumnale L. and the tubers of G. superba L. through different chemical extraction methods. Applying in vitro plant cell and tissue cultures together with metabolic and genetic engineering techniques, large-scale production of colchicine can be achieved from the above two plant species.
文摘The objective of this paper is to demonstrate that the processual aspect of literary works of art deserves much more attention than it normally receives by readers, critics, and theorists. The most important reason for this is seen in the fact that texts since the advent of print culture have been disseminated and passed on in written form and that in the medium of writing the processual character of language is only insufficiently taken care of by a cultural convention of arranging and approaching the presented signs in an particular sequence. Whereas in an oral culture the dynamic processuality of a speech or recitation was directly experienced by the listeners, the spatial arrangement of signs in writing enables and even entices readers and critics to read this or that part of a written text in a sequence of their own making. What remains out of focus is that in doing so they miss the particular semantic profile and aesthetic character of the work as created by the author--a procedure particularly hurtful in the case of literary works of art. There is hope, however, that this will somewhat change by our moving towards a performance culture
文摘Since time and beyond, when humans first developed the understanding of staying in communities, a sense of affection and adoration grew towards fellow beings. The law of nature of the cycle of life gave rise to the belief in life after death and a sense of intimidation for the same, gave way to supernatural beliefs. Such beliefs gave rise to multifarious customary activities. A study to understand them on the basis of symbolism has paved the way for simplification of the complex rituals, and hence, perceive the prevalent socio-cultural aspects of those times.
文摘Rapidly emerged creative industries receive increasing attention from a variety of disciplines. However, the space features of creative industries and its association with local socio-cultural contexts have not been fully understood, especially at a micro-city level. This study attempts to understand the agglomeration of creative industries in Shanghai from the sociology perspective. For this study, this paper utilizes primarily a questionnaire survey to explain the space features of creative industries in Shanghai. The results indicate an extensive socio-cultural embeddedness of the agglomeration of creative industries in Shanghai. First, strong emphasis on face-to-face contacts by creative professionals makes geographical agglomeration necessary for creative industries. Second, the reason why inner city of Shanghai is popular among creative professionals and enterprises lies in the diversity of cultures and special environment of the former colonial zones of Shanghai. Additionally, highly concentrated dining and entertainment facilities in the central city of Shanghai offer creative workers social networking places and nightlife venues. Third, as the educational attainment of local citizens and the protection of intellectual property are highly stressed by creative professionals, research and design specialized creative industries are more likely located near universities and research institutes.
文摘Jan Haicksz Steen (1626-1679), a Dutch actor, poet and painter, engaged the viewer with various innuendos and double entendres in his paintings about Woman at Her Toilet. Decoding the conceits introduces the viewer to Dutch artistic astuteness and popular culture. Steen drew the observer into his picture plane through a series of emblematic vignettes. Although he raised questions about cultural mores, morality, and religiosity, none are judgmental. On the contrary, it is the viewer who might or might not articulate a moral judgment according to personal experiences. For Steen, the imagery, filled with complex conceits, is an artistic visual representation to delight the eyes, tantalize the senses, and maybe even spark the intellect.
