The result...The fresh pollen vitality, the effect of different storage conditions on the pollen vitality, and the difference of vitality among the species of seven species of Syringa were determined in Shenyang, China. The results indicated that the pollen vi-tality (81.5%) of Syringa villosa was the highest among the seven tested species, followed by S. microphylla and S. meyeri, and that of S. oblata var. affinis was the lowest. The low temperature was the best condition for storage of pollen of Syringa, and the most proper temperature for the storage was 0-2 癈. The storability of S. microphylla was the best of all, and it could be stored over 60 days at the temperature of 0-2 癈, next was S. villosa and S. meyeri.展开更多

Franco and Thuja occidentalis L. seem to be of the same type, as based on the morphological study of the change in pollination drop, during the process of pollination. Entering the pollination drop, the pollen may alter the surface of the drop or reduce the secretion of the ovule with concomitant increase of evaporation. It has been further noticed that the dynamiechange of the drop withdrawal rate was closely associated with the different kinds of pollen received. It seemsthat the withdrawal of the pollination drop may be induced by pollen from plants consanguineous to P. orientalis, almost as effective as from pollen of P. orientalis itself and comparatively more effective than the foreignpollen received from distant plant species. The drop withdrawal could he mainly induced by the pollen that causes the reduction of ovule secretion.展开更多

Glucanases were found in the cell wall of Lilium longiflorum Thunb. pollen tubes grown in vitro . The activity of β_glucanases was, in a certain extent, decreased by nojirimycin, an inhibitor of glucosidase. P...Glucanases were found in the cell wall of Lilium longiflorum Thunb. pollen tubes grown in vitro . The activity of β_glucanases was, in a certain extent, decreased by nojirimycin, an inhibitor of glucosidase. Pollen germination percentage reduced dramatically when nojirimycin was applied in the culture medium. In case that nojirimycin was added at 0 or 1 h after the onset of incubation, the inhibition rate was 99.6% and 91.4%, respectively. When 3 mmol/L of nojirimycin was applied in the liquid medium at 0, 1, 1.5 and 2 h after the onset of incubation, the growth of pollen tubes was interrupted, which resulted in the morphological change of the pollen tubes such as the newly grown portion of pollen tubes being bent, curved and swollen. Tracing the growth pattern of the individual pollen tube grown in semi_solid medium by video microscopy, the authors demonstrated that pollen tube growth rate was strongly inhibited by nojirimycin at concentrations ranged from 0.003 to 3 mmol/L. Moreover, the cytoplasmic arrangement and the morphology of the pollen tubes were also affected by nojirimycin. The growth inhibition brought about by nojirimycin was reversible. These results indicated that β_glucanases, which degrade 1,3_β_glucan and/or 1,4_β_glucan or 1,3:1,4_β_glucan constructed in the cell wall, are involved in pollen germination and pollen tube growth. It provides new insight into an understanding of the contribution of β_glucanases to the cell wall extensibility and the crucial role of cell wall in regards to the regulation of pollen tube growth.展开更多

It was observed in Dianthus caryophyllus L. cv. White Sim that protein synthesis in flowers declined and the responsiveness to ethylene increased as the flowers became matured. However, when the flowers were pre-treated with aminooxyacetic acid (AOA), an inhibitor of ethylene synthesis, the observed responsiveness to ethylene in flowers declined with age. These results suggested that responsiveness to ethylene in flowers may be due to a declination in the capacity of protein synthesis in flowers.展开更多

[Method] Adsorption experiment was carried out in a hermetic container, and the effect of calyx amount, contact time, moisture content of basic tea and temperature on the scenting process was studied. [Result] The optimal moisture and temperature for scenting process was 4% and 10 ℃, respectively. [Conclusion] The scenting process accorded pseudo-first-order kinetic model, and the adsorption dynamic data of total process could better fit pseudo-second-order kinetic model.展开更多

[Method] A novel cDNA was cloned by PT-PCR using the total RNA as template, and amplified to the desirable s...[Objective] The aim was to study the cDNA cloning of SMAT gene from Chimonanthus praecox and its expression. [Method] A novel cDNA was cloned by PT-PCR using the total RNA as template, and amplified to the desirable size. The RT-PCR products were reclaimed and transformed into E. coli DH 5o together with the PMD18-T vector after ligating by T-A cloning. Identified by colony PCR and EcoRI and Notl digestion, the recombinant plasmid with target gene was screened out and conducted the sequence analysis. [Result] Results of the sequence analysis showed that the ORF fragment of the SAMT cDNA was successfully cloned from Chimonanthus praecox gene, with the length of 1 196 bp and encoding 380 amino acids fragment which shared 99.2% homology to that of previously reported SAMT cDNA from Chimonanthus praecox (ABU88887). The SAMT cDNA fragment was sub-cloned into the prokaryotic expression vector PGEX1-4T-1, and the obtained re- combinant plasmid was named PGSAMT. After inducting by 0.01mol/L IPTG, the re- sult of the SDS-PAGE analysis showed that the molecular weight of the fusion ex- pression SAMT protein was about 66 kDa, which was close to the predicted fusion protein derived from the 26 kDa GST band and 42.3 kDa SAMT gene of Chimo- nanthus praecox encoded protein. [Conclusion] This study successfully cloned and expressed the SAMT gene of Chimonanthus praecox.展开更多

By hydroxymethylation and etherification reaction, the fluorene can be converted to desired products with high yield and purity. The optimum conditions of the synthetic procedures were determined. For hydroxymethylation, the reaction temperature was 13 ℃and the mole ratio of sodium methoxide to paraformaldehyde was 0.26; and for etherification, the reaction temperature was 40 ℃and the reaction time was 15 h. Essential oil from clove buds Syzygium aromaticum L. Merr. & Perry (Myrtaceae) was extracted using petroleum ether in Soxhlet apparatus. The resultant oil contained eugenol (48.92%), caryophyllene (18.55%), α-...Essential oil from clove buds Syzygium aromaticum L. Merr. & Perry (Myrtaceae) was extracted using petroleum ether in Soxhlet apparatus. The resultant oil contained eugenol (48.92%), caryophyllene (18.55%), α-caryophyllene (3.25%), eugenol acetate (23%), cis-13-docosenamide (3.2 1%), presenting more than 96% of the oil. This oil was examined as a fumigant and repellent agent against adults of the red flour beetle, Tribolium castaneum. Data showed that various concentrations of the oil have revealed dramatic repellent activity against the tested insect, where it gave 100% repellency by 1.0, 0.8, and 0.2% clove oil at 4, 8, 10 min, respectively. The RTs0 and RT95 were 1.1 and 8.0 min at 0.2% and 0.4, 2.6 min at 0.8%, respectively. In addition, fumigation assay has also exhibited strong fumigant activity toward the adults of T. castaneum. At 100 μL oil/L air, mortality was 75, 80 and 100% after 6, 7, and 8 days exposure period with LCs0 and LC95 17 and 70 μL/L air, respectively. The utilization of clove oil for its potential effects against stored product insect is discussed.展开更多
Sixteen priority polycyclic aromatic hydrocarbons(PAHs) in surface water samples were analyzed by gas chromatograph-mass spectrometer(GC-MS) to study their distribution and characterizing sources.The water samples wer...Sixteen priority polycyclic aromatic hydrocarbons(PAHs) in surface water samples were analyzed by gas chromatograph-mass spectrometer(GC-MS) to study their distribution and characterizing sources.The water samples were collected from five sites(J1-J5) in the Jialing River of Chongqing downtown area from September 2009 to August 2010.The results demonstrate that the concentration of total PAHs in three samples upstream are relatively higher than those in other two sites downstream,with average concentration of total PAHs for each site ranging from 811.5 ng/L to 1585.8 ng/L.The 2,3 and 4-ring PAHs for sampling stations account for 13.0%,56.6% and 28.6%,respectively,in total PAHs.There are obvious tendencies of seasonal change for PAHs concentration in surface water.The PAHs concentration in April of wet season is 1 301.6 ng/L,which is 1.3 times the lowest amount of total PAHs in August of flood season.Ratios of specific PAH compounds were used to characterize the possible pollution sources.Experimental results indicate that the PAHs in surface water samples are primarily from pyrolytic PAHs because of factories along these sites,while the direct leakage of petroleum products may be significant for two sites,Jiahua Bridge(J4) and Huanghuayuan Bridge(J5),because of the wharf boat nearby.展开更多
