B501 promoted the production of artemisinin in Artemisia annua L. hairy root culture. When hairy roots of 22-day-old cultures (later growth phase) were exposed to the elicitor (20 mg/L) for 4 d, the maximum content of artemisinin reached 1.15 mg/g, a 64.29% increment over the control. The electron X-ray microanalysis disclosed the rapid accumulation of Ca 2+ in the elicited cortical cells of hairy root. The electronic microscope observation revealed the high electron density area in vacuole of elicited cells. During the first day of elicitation the peroxidase activity of hairy roots was improved sharply. Some cellular morphological changes including cell shrinkage, condensation of cytoplasm and nuclear fragmentation, coincident with the appearance of DNA ladders, were observed after the third day of elicitation. It was suggested that the oligosaccharide elicitor triggered the programmed cell death, which may provide the substance or chemical signal for artemisinin biosynthesis.展开更多

Based on the geochemical data obtained from the national project about the prevention and control of soil contamination, this paper explored the properties of soil chemical elements in Huanghuaihai Plain, Shandong Pro...Based on the geochemical data obtained from the national project about the prevention and control of soil contamination, this paper explored the properties of soil chemical elements in Huanghuaihai Plain, Shandong Province. The results showed that among the grade-one nutritive elements in soil, organic matter, nitrogen and phosphorus were relatively deficient while potassium was rich. Meanwhile, as the grade-two nutritive elements, calcium oxide and magnesium oxide were relatively short and sulfur’s content was abundant. About the other beneficial and trace nutri-tive elements, iron oxide, manganese, molybdenum and boron were deficient, but the content of chlorine was high, hardly lack. The main barriers to improving land productivity were soil salinization and soil heavy metal contamination. The values of soil integrated fertility index that most of the soil in the study area is middle-lower fertilized. Specifical y, the low fertility area and lower fertility area are 6 1604 and 1 244 km2 respectively, occupying about 97.43% and 1.97% of the total area. The moderate fertility soil has an area of 172 km2, occupying about 0.27% of the total area. The higher fertility soil covers an area of 128 km2, while the high fertility area of only 76 km2. This article proposed scientific fertilization, elimination of soil obsta-cle, remediation of heavy-metal-contaminated soil and other effective measures to improve land productivity according to the basic investigation results, which provides a good technological support for the planning and development of good-quality and high-benefit agriculture.展开更多

The effects of leaching temperature, mechanical agitation rate, concentration of ethanol and feed particle size on leaching kinetics were examined. It is found that the smaller the feed particle size or the higher the leaching temperature, the higher the leaching rate. The leaching process can be described by the shrinking-core model. The apparent activation energy is 10.8kJ·mol^-1, suggesting that the leaching process is controlled by the inner diffusion. An empirical equation relating the flavonoids leaching rate constant to the feed particle size and leaching temperature was expected.展开更多

The apparent molecular mass of the vitellin is 546 ku based on the data of native-PAGE. Under denatured condition (SDS-PAGE), it was found that vitellin was composed of four polypeptides each at 120, 100, 65, and 55 ku. One disulfide bond was detected in the binding of polypeptide subunits. The purified vitellin, contained 4.47% phosphor and 10.6% polysaccharides, and was identified as glyco-lipo-carotenoprotein, according to the PAGE staining data. The purified vitellin can be used as antigen to raise polyclonal antisera in further application.展开更多

The contents of motilin and somatostatin in small intestine of mice were determinated by radioimmunoassay. The electrical potential difference (PD) related to Na+ and glucose transport was measured across the wall of reverted intestinal sacs. Na+-K+-ATPase activity of small intestinal mucosa was measured by spectroscopic analysis.RESULTS: Different dosages of emodin can improve small intestinal peristalsis of mice. Emodin increased the content of motilin, while reduced the content of somatostatin in small intestine of mice significantly. Emodin 0.2, 0.4, 0.8, and 1.6 g/L decreased PD when there was glucose. However, emodin had little effect when glucose was free. The Na+-K+-ATPase activity of small intestinal mucosa of mice in emodin groups was inhibited obviously. CONCLUSION: Emodin can enhance the function of small intestinal peristalsis of mice by mechanisms of promoting secretion of motilin, lowering the content of somatostatin and inhibiting Na+-K+-ATPase activity of small intestinal mucosa.展开更多

Variables tested were volume rati...A uniform experimental design procedure was used to investigate the effects of some operating parameters on the extraction of emodin from Polygonum cuspidatum Sieb. et Zucc. products. Variables tested were volume ratio of material to solvent, size of material, extraction time and temperature and composition of extraction solvent (mixtures of acetone-water). Each variable was tested at seven levels; 7 experiments were performed in random order. Analyses of the extracts were performed by high-performance liquid chromatography with diode array detection(HPLC-DAD). Analytical responses were processed by using a forward regression analysis, in order to find polynomial function describing the relationship between variables and responses. For all the analytes the experimental conditions for providing the highest extraction yield inside the experimental domain considered were found. Finally, a simple, rapid and accurate analytical method was developed for the determination of emodin by high performance liquid chromatography. The separation is achieved within 25 rain on an ODS column using methanol and water as gradient mobiles. Emodin can be quantified by using external standard method detecting at 436 nm. Good linearity is obtained with correlation coefficient exceeding 0.9986 and the detection limit and the quantification limit are 1.53 and 3.23 mg/L respectively. This method shows good reproducibility for the quantification of the emodin with intra-day and inter-day relative standard deviation less than 2.3% and 5.6% respectively. Under optimized extraction conditions, the recovery of the standard is 96.5%. The validated method is successfully applied to quantify the emodin in seven Polygonum cuspidatum sieb. Et zucc. products, which provided an idea for the pre-treatment of determination of active compounds in traditional Chinese medicines.展开更多

The dust had no significant effect on the two insects, none of them died even at 3 d after treatment. The methanol extracts, however, had rapid lethal effects on both C. maculatus and S. zeamais. The mortality of C. maculatus by the lowest concentration of methanol extracts ranged from 95%-100% whereas in S. zeamais, the mortality ranged from 87.5%-100% and 70%-100% in concentrations of 1 g extract+3 ml methanol and 1 g extract+5 ml methanol, respectively, from 24 to 48 h. The least concentration of 1 g extract+l 5 ml methanol had no significant lethal effect on Sitophilus zeamais.展开更多

In order to investigate the enrichment of ferric iron bound by extracellular polymeric substance (EPS) on the mineral surface during bioleaching of chalcopyrite, several methods including sonication, heating and vor...In order to investigate the enrichment of ferric iron bound by extracellular polymeric substance (EPS) on the mineral surface during bioleaching of chalcopyrite, several methods including sonication, heating and vortexing were used and sonication at 48℃ was shown as a good way to extract ferric iron. Scanning electron microscope (SEM) and energy dispersive X-ray spectrometer (EDX) analysis showed that lots of cracks and pits can be found on the chalcopyrite surface after bioleaching and that iron oxide was filled in these cracks and pits. The variations of contents of ferric iron and EPS on the chalcopyrite surface were investigated. The results indicated that the content of EPS increased rapidly in the first 10 d and then maintained at a stable level, while ferric iron content increased all the time, especially in the later stage of bioleaching.展开更多

In fact, an alternative copper extraction route from mixed sulphide ores, chalcopyrite and chalcocite using mesophilic biomass consortium at 33.3 °C and ferric leaching process were attempted. Bioleaching experiments were settled with a fraction size of-75+53 μm. Bacteria were used as the catalyst. A copper yield of 65.50% was obtained. On the other hand, in ferric leaching process, with a fraction size of-53+38 μm, when the temperature was increased to 70 °C, the copper leaching rate increased to 78.52%. Thus, comparatively, the mesophilic bioleaching process showed a more obvious advantage in copper extraction than leaching process with a high temperature. However, it has been resolved from the characterization performed using SEM-EDS, FTIR and XRD observations coupled with different thermodynamic approaches that, the indirect mechanism is the main leaching mechanism, with three transitory mechanisms(polysulphide, thiosulphate and elemental sulphur mechanisms) for the mixed chalcopyrite-chalcocite ore. Meanwhile, the speciation turns into Cu2 S-Cu S-Cu5 Fe S4-Cu2 S before turning into Cu SO4. While ferrous oxidation and the formation of ferric sulphate occur, and there is a formation of strong acid as bacteria digest sulphide minerals into copper sulphate at low temperature, which is why this copper production scenario requires a redox potential more than 550 m V at room temperature for high copper leaching rate.展开更多
Shoulder lines are the most important landform demarcations for geographical analysis,soil erosion modeling and land use planning in the Loess Plateau area of China.This paper proposes an automatic,effective and accur...Shoulder lines are the most important landform demarcations for geographical analysis,soil erosion modeling and land use planning in the Loess Plateau area of China.This paper proposes an automatic,effective and accurate method of determining loess shoulder line from DEMs by integrating a hydrological D8 algorithm and a snake model.The watershed boundary line is adopted as the initial contour which evolves to identify the exact position of loess shoulder-line by the guidance of an external force of snake model from DEMs.Experiments show that the method overcomes the difficulties in both threshold selection for edge detection and the disconnecting issues in former extraction approaches.The accuracy evaluation of shoulder-line maps from the two test sites of the loess plateau area show obvious improvements in the extraction.The average contour matching distance of the new method is 12.0 m on 5 m resolution DEM,and shows improvement in the accuracy and continuity.The comparisons of accuracy evaluations of the two test sites show that the snake model method performs better in the loess plain area than in the area with high gully density.展开更多

In this project, an in vitro hypoxic model using human brain epithelial cells (HBEC) was studied with treatment of the tea extract before inducing hypoxia. We have tested the hypothesis that flavonoids extracted from Ceylon green tea act as potential therapeutic ingredient (s) to reduce oxidative stress in hypoxic cells through its antioxidant properties and its ability to reduce cerebral cellular death. The biochemical antioxidant tests show that the Ceylon green tea has 68% ± 2.8% inhibition property of scavenging of ABTS. The inhibition of pyrogallol red bleaching by HOCI from Ceylon tea was 79% ± 4.5%. After exposing to hypoxia, the cell viability was 29% ± 2.3% in the hypoxia control group but 41% ± 4.7% for flavonoids extract treated group. In LDH assay, flavonoids extract treated group had 75% ± 3.7% reducing of LDH release. The flavonoids extract treated groups significantly increased in antioxidant enzyme activity assays: the activity level of SOD [(1.5 ± 0.6) μmol/min/mg protein], CAT [(0.61 ± 0.06) μmol/min/mg protein], GPx [(2.6 ± 0.41) μmol/min/mg protein] and GST [(6.0 ± 2.4) μmol/min/mg protein] are significantly increased as compared with hypoxic control [(0.5 ± 0.52, 0.51 ± 0.04, 1.2 ± 0.35 and 3.1 ± 1.6) μmol/min/mg protein, respectively]. The study demonstrated a great clinical potential and opened a new avenue to prevent stroke by drinking Ceylon tea.展开更多

In this research, a process has been developed to integratively extract ergosterol, (1→3)-α-D-glucan, and chitosan from Penicillium chrysongenum mycelium. First the mycelia are pretreated with 0.1mol·L^-1 of NaOH. After recovery by centrifugation the solid portion is made to undergo saponification and deacetylation reactaons by addition of 2mol·L^-1 NaOH and et anol.After reaction, extraction is carried out by addition of petroleum ether, which separates the reaction mixture into two phases. The upper layer of petroleum ether contains extracted ergosterol, and the .bottom layer of NaOH solution contains (1→3)-α-DEglucan; the chitosan is on the mycelia residuum. After isolation, the recovery yield of ergosterol is 0.52% of dry mycelium. That of (1→3)-α-D-glucan is about 8.2%; and chitosan is 5.7% with 86% deacetylation. The compositions have been characterized by 1R, HPLC analyses.展开更多

It is found that under the premise of no influence on root growth of cucumber,extracts from Picrorhiza scrophulariiflora and Punica granatum showed strong prevention and nematicidal activity,and had the similar efficacy of Avermectin; while the extracts from Cibotium barometz,Aucklandia lappa Decne and Fructus cnidii showed low nematicidal activity and various degrees inhibition effect on plant growth.

文摘The kinetics of leaching flavonoids from Pueraria Lobata with ethanol was investigated. The effects of leaching temperature, mechanical agitation rate, concentration of ethanol and feed particle size on leaching kinetics were examined. It is found that the smaller the feed particle size or the higher the leaching temperature, the higher the leaching rate. The leaching process can be described by the shrinking-core model. The apparent activation energy is 10.8kJ·mol^-1, suggesting that the leaching process is controlled by the inner diffusion. An empirical equation relating the flavonoids leaching rate constant to the feed particle size and leaching temperature was expected.

文摘Vitellin of mature female marine crab Charybdis japonica was purified by high-performance liquid chromatography (HPLC, DEAE-cellulose-16 anion exchange column). The apparent molecular mass of the vitellin is 546 ku based on the data of native-PAGE. Under denatured condition (SDS-PAGE), it was found that vitellin was composed of four polypeptides each at 120, 100, 65, and 55 ku. One disulfide bond was detected in the binding of polypeptide subunits. The purified vitellin, contained 4.47% phosphor and 10.6% polysaccharides, and was identified as glyco-lipo-carotenoprotein, according to the PAGE staining data. The purified vitellin can be used as antigen to raise polyclonal antisera in further application.
文摘AIM: To investigate the effect of emodin on small intestinal peristalsis of mice and to explore its relevant mechanisms.METHODS: The effect of emodin on small intestinal peristalsis of mice was observed by charcoal powder propelling test of small intestine. The contents of motilin and somatostatin in small intestine of mice were determinated by radioimmunoassay. The electrical potential difference (PD) related to Na+ and glucose transport was measured across the wall of reverted intestinal sacs. Na+-K+-ATPase activity of small intestinal mucosa was measured by spectroscopic analysis.RESULTS: Different dosages of emodin can improve small intestinal peristalsis of mice. Emodin increased the content of motilin, while reduced the content of somatostatin in small intestine of mice significantly. Emodin 0.2, 0.4, 0.8, and 1.6 g/L decreased PD when there was glucose. However, emodin had little effect when glucose was free. The Na+-K+-ATPase activity of small intestinal mucosa of mice in emodin groups was inhibited obviously. CONCLUSION: Emodin can enhance the function of small intestinal peristalsis of mice by mechanisms of promoting secretion of motilin, lowering the content of somatostatin and inhibiting Na+-K+-ATPase activity of small intestinal mucosa.

文摘A uniform experimental design procedure was used to investigate the effects of some operating parameters on the extraction of emodin from Polygonum cuspidatum Sieb. et Zucc. products. Variables tested were volume ratio of material to solvent, size of material, extraction time and temperature and composition of extraction solvent (mixtures of acetone-water). Each variable was tested at seven levels; 7 experiments were performed in random order. Analyses of the extracts were performed by high-performance liquid chromatography with diode array detection(HPLC-DAD). Analytical responses were processed by using a forward regression analysis, in order to find polynomial function describing the relationship between variables and responses. For all the analytes the experimental conditions for providing the highest extraction yield inside the experimental domain considered were found. Finally, a simple, rapid and accurate analytical method was developed for the determination of emodin by high performance liquid chromatography. The separation is achieved within 25 rain on an ODS column using methanol and water as gradient mobiles. Emodin can be quantified by using external standard method detecting at 436 nm. Good linearity is obtained with correlation coefficient exceeding 0.9986 and the detection limit and the quantification limit are 1.53 and 3.23 mg/L respectively. This method shows good reproducibility for the quantification of the emodin with intra-day and inter-day relative standard deviation less than 2.3% and 5.6% respectively. Under optimized extraction conditions, the recovery of the standard is 96.5%. The validated method is successfully applied to quantify the emodin in seven Polygonum cuspidatum sieb. Et zucc. products, which provided an idea for the pre-treatment of determination of active compounds in traditional Chinese medicines.
文摘Insecticidal effects of different doses of the dust and methanol extracts of Garcinia kolae on Callosobruchus maculatus and Sitophilus zeamais were tested. The dust had no significant effect on the two insects, none of them died even at 3 d after treatment. The methanol extracts, however, had rapid lethal effects on both C. maculatus and S. zeamais. The mortality of C. maculatus by the lowest concentration of methanol extracts ranged from 95%-100% whereas in S. zeamais, the mortality ranged from 87.5%-100% and 70%-100% in concentrations of 1 g extract+3 ml methanol and 1 g extract+5 ml methanol, respectively, from 24 to 48 h. The least concentration of 1 g extract+l 5 ml methanol had no significant lethal effect on Sitophilus zeamais.

文摘[Objective]The aim of this study was to explore allelopathic effects of the water-extracted crude of cucumber plant parts on seed germination and seedling growth of lettuce(Lactuca sativa L.),cabbage(Brassica pekinensis L.),radish(Raphanus sativus L.) and scallion(Allium fistulosum L.).[Method]The allelopathic effects of different cucumber tissues aqueous extracts were investigated on lettuce,cabbage,radish and scallion using the water-soluble allelopathic principle by seedling bioassay tests.[Result]The trends of inhibitory effects of different cucumber parts increased with increasing extract concentrations.The inhibition effects and the inhibition rate of three aqueous extracts were more significant on root growth than shoot length.Leaf aqueous extracts represented the most exhibited and root aqueous extracts representing the least inhibitory on seed germination,shoot length and root length of lettuce,cabbage,radish and scallion.[Conclusion]This study had provided basis for allelopathic character of different cucumber plant parts.

文摘In order to investigate the enrichment of ferric iron bound by extracellular polymeric substance (EPS) on the mineral surface during bioleaching of chalcopyrite, several methods including sonication, heating and vortexing were used and sonication at 48℃ was shown as a good way to extract ferric iron. Scanning electron microscope (SEM) and energy dispersive X-ray spectrometer (EDX) analysis showed that lots of cracks and pits can be found on the chalcopyrite surface after bioleaching and that iron oxide was filled in these cracks and pits. The variations of contents of ferric iron and EPS on the chalcopyrite surface were investigated. The results indicated that the content of EPS increased rapidly in the first 10 d and then maintained at a stable level, while ferric iron content increased all the time, especially in the later stage of bioleaching.
文摘This paper elaborated on the sustainability of the copper extraction process. In fact, an alternative copper extraction route from mixed sulphide ores, chalcopyrite and chalcocite using mesophilic biomass consortium at 33.3 °C and ferric leaching process were attempted. Bioleaching experiments were settled with a fraction size of-75+53 μm. Bacteria were used as the catalyst. A copper yield of 65.50% was obtained. On the other hand, in ferric leaching process, with a fraction size of-53+38 μm, when the temperature was increased to 70 °C, the copper leaching rate increased to 78.52%. Thus, comparatively, the mesophilic bioleaching process showed a more obvious advantage in copper extraction than leaching process with a high temperature. However, it has been resolved from the characterization performed using SEM-EDS, FTIR and XRD observations coupled with different thermodynamic approaches that, the indirect mechanism is the main leaching mechanism, with three transitory mechanisms(polysulphide, thiosulphate and elemental sulphur mechanisms) for the mixed chalcopyrite-chalcocite ore. Meanwhile, the speciation turns into Cu2 S-Cu S-Cu5 Fe S4-Cu2 S before turning into Cu SO4. While ferrous oxidation and the formation of ferric sulphate occur, and there is a formation of strong acid as bacteria digest sulphide minerals into copper sulphate at low temperature, which is why this copper production scenario requires a redox potential more than 550 m V at room temperature for high copper leaching rate.

文摘Shoulder lines are the most important landform demarcations for geographical analysis,soil erosion modeling and land use planning in the Loess Plateau area of China.This paper proposes an automatic,effective and accurate method of determining loess shoulder line from DEMs by integrating a hydrological D8 algorithm and a snake model.The watershed boundary line is adopted as the initial contour which evolves to identify the exact position of loess shoulder-line by the guidance of an external force of snake model from DEMs.Experiments show that the method overcomes the difficulties in both threshold selection for edge detection and the disconnecting issues in former extraction approaches.The accuracy evaluation of shoulder-line maps from the two test sites of the loess plateau area show obvious improvements in the extraction.The average contour matching distance of the new method is 12.0 m on 5 m resolution DEM,and shows improvement in the accuracy and continuity.The comparisons of accuracy evaluations of the two test sites show that the snake model method performs better in the loess plain area than in the area with high gully density.

文摘In this paper a simple preparative method for isolation and purification of ginkgolides A and B was developed,As starting material,a commercially available standardized ginkgo extract (EGb761,containing 24% flavonoid and 6% terpene trilactones) was used,After a pretreatment step,optimized by the uniform design method ,the concentrated intermediate extract with high content of GA and gb(+90%) was separated into the individual terpenes by preparative liquid chromatography eluted with petroleum ether-ethylacetate,Analysis of products was carried out by means of HPLC-ELSD(evaporative light -scattering detector),The results show that ginkgolides A and B are obtained in higher yield and better purity.
文摘We have extracted and purified flavonoids as active ingredients from Ceylon green tea (Dilmah). In this project, an in vitro hypoxic model using human brain epithelial cells (HBEC) was studied with treatment of the tea extract before inducing hypoxia. We have tested the hypothesis that flavonoids extracted from Ceylon green tea act as potential therapeutic ingredient (s) to reduce oxidative stress in hypoxic cells through its antioxidant properties and its ability to reduce cerebral cellular death. The biochemical antioxidant tests show that the Ceylon green tea has 68% ± 2.8% inhibition property of scavenging of ABTS. The inhibition of pyrogallol red bleaching by HOCI from Ceylon tea was 79% ± 4.5%. After exposing to hypoxia, the cell viability was 29% ± 2.3% in the hypoxia control group but 41% ± 4.7% for flavonoids extract treated group. In LDH assay, flavonoids extract treated group had 75% ± 3.7% reducing of LDH release. The flavonoids extract treated groups significantly increased in antioxidant enzyme activity assays: the activity level of SOD [(1.5 ± 0.6) μmol/min/mg protein], CAT [(0.61 ± 0.06) μmol/min/mg protein], GPx [(2.6 ± 0.41) μmol/min/mg protein] and GST [(6.0 ± 2.4) μmol/min/mg protein] are significantly increased as compared with hypoxic control [(0.5 ± 0.52, 0.51 ± 0.04, 1.2 ± 0.35 and 3.1 ± 1.6) μmol/min/mg protein, respectively]. The study demonstrated a great clinical potential and opened a new avenue to prevent stroke by drinking Ceylon tea.

文摘Abstract Ergosterol,(1→3)-α-D-glucan and chitosan are important biomaterials. In this research, a process has been developed to integratively extract ergosterol, (1→3)-α-D-glucan, and chitosan from Penicillium chrysongenum mycelium. First the mycelia are pretreated with 0.1mol·L^-1 of NaOH. After recovery by centrifugation the solid portion is made to undergo saponification and deacetylation reactaons by addition of 2mol·L^-1 NaOH and et anol.After reaction, extraction is carried out by addition of petroleum ether, which separates the reaction mixture into two phases. The upper layer of petroleum ether contains extracted ergosterol, and the .bottom layer of NaOH solution contains (1→3)-α-DEglucan; the chitosan is on the mycelia residuum. After isolation, the recovery yield of ergosterol is 0.52% of dry mycelium. That of (1→3)-α-D-glucan is about 8.2%; and chitosan is 5.7% with 86% deacetylation. The compositions have been characterized by 1R, HPLC analyses.
