α7烟碱型乙酰胆碱受体与阿尔茨海默病的关系

背景:α7烟碱型乙酰胆碱受体在大脑皮质和海马高度表达,并在阿尔茨海默病的病理发展过程中起重要调控作用,是阿尔茨海默病治疗的潜在靶点。目的:总结α7烟碱型乙酰胆碱受体和阿尔茨海默病的密切关系和相互作用机制。方法:检索中国知网、PubMed数据库中相关文献,中文检索词为“α7烟碱型乙酰胆碱受体,阿尔茨海默病,β-淀粉样蛋白,激动剂,正变构调节剂,拮抗剂”;英文检索词为“alpha 7 nicotinic acetylcholine receptor,Alzheimer’s disease,beta amyloid protein,agonist,positive allosteric modulator,antagonist”,文献检索时限为各数据库建库至2024年7月,依据入选标准对检索结果进行录用或排除,最终纳入符合标准的83篇文献进行综述。结果与结论:α7烟碱型乙酰胆碱受体通过与β-淀粉样蛋白的相互作用减轻β-淀粉样蛋白的神经毒性,如促进阿尔茨海默病的突触可塑性和胆碱能突触的快速传递、减轻β-淀粉样蛋白诱导的神经中枢炎症反应、抵抗神经细胞凋亡,从而对阿尔茨海默病患者的脑具有保护作用等。α7烟碱型乙酰胆碱受体作为阿尔茨海默病治疗靶标具有很大的潜能,但是又存在一系列问题有待解决,比如α7烟碱型乙酰胆碱受体的脱敏性、适度活性稳定性及基因多态性等问题。筛选高特异、安全性和以α7烟碱型乙酰胆碱受体为核心的多靶点结合作用的药物,将成为未来阿尔茨海默病治疗研究的一个方向。

基于DIgSILENT数据接口DGS的含风电电力系统随机潮流分析

同一风区的不同风电场由于地理位置相近,各风电场的风速和出力具有强相关性。针对该特点,提出了一种考虑风电场之间相关性的含风电电力系统随机潮流算法。首先建立了多个风电场出力的离散化联合概率分布模型,在此基础上采用线性化潮流方法分析随机潮流的各项指标。结合随机潮流算法和DIg SILENT软件的优势,研究了一种基于DIg SILENT数据接口DGS的数据转换方法,有效地提高了计算效率。最后利用IEEE-24节点系统以及我国新疆地区2015年规划电网进行方法验证。算例结果表明,建立的考虑风电场相关性的含风电电力系统随机潮流算法具有可行性和优越性,DGS数据接口技术可以使该潮流算法在实际大型电力系统中推广使用,工程实用性较强。

静默疗法对经皮冠状动脉介入术患者心理应激反应及睡眠障碍的影响

目的评价静默疗法在经皮冠状动脉介入(PCI)术患者中的应用效果。方法选择2023年1月至2024年3月本院收住入院接受PCI术的100例患者为研究对象。对照组接受PCI术后常规护理,观察组在对照组的基础上采用静默疗法。两组患者干预周期均为4周,干预前后采用焦虑自评量表(SAS)、抑郁自评量表(SDS)、匹兹堡睡眠质量指数量表(PSQI)调查患者心理应激反应程度及睡眠质量。结果干预前两组患者SAS、SDS、PSQI得分比较差异无统计学意义(P>0.05);干预后观察组SAS、SDS、PSQI得分低于对照组,差异有统计学意义(P<0.05)。结论将静默疗法应用于PCI术患者中,能降低患者心理应激反应程度及改善睡眠质量,建议护理人员积极推广应用。

E文本在线电网模型自动导入DIgSILENT的研究和开发

为了将DIgSILENT/Power Factory集成为调度运行和规划分析高层应用平台中的核心计算工具,研究在线电网模型自动导入和计算结果自动导出DIgSILENT的技术。采用IEEE9节点电网,研究电网设备模型、拓扑模型和图形模型在DGS接口文件中的数据内容和格式,测试验证利用DGS接口导入全网模型的可行性。对E文本在线电网模型和DGS接口电网模型进行对比分析,研究一种基于电网拓扑数据自动生成DGS格式的电网图形数据的方法,设计E文本在线电网模型转为DGS接口电网模型的实现流程。最后开发相应的软件,并测试软件功能,同时分析总结潮流计算中设置节点类型的调试经验。研究内容和成果对于将DIgSILENT推广应用于在线运行分析和离线快速建模具有指导作用。

PROPER CONDITIONS OF MESHING FOR Hy-Vo SILENT CHAIN AND SPROCKET

Proper meshing of Hy-Vo silent chain and sprocket is important for realizing the transmission of the silent chain with more efficiency and less noise. Based on the study of the meshing theory of the Hy-Vo silent chain with the sprocket and the roll cutting machining principle of the sprocket with the hob, the proper conditions of the meshing for the Hy-Vo silent chain and the sprocket are put forward with the variable pitch characteristic of the Hy-Vo silent chain taken into consideration, and the proper meshing design method on the condition that the value of the link tooth pressure angle is unequal to the value of the sprocket tooth pressure angle is studied. Experiments show that this new design method is feasible. In addition, the design of the pitch, the sprocket tooth pressure angle and the fillet radius of the sprocket addendum circle are studied. It is crucial for guiding the design of the hob which cuts the Hy-Vo silent chain sprocket.

Degradation of Benzene by Using a Silent-Packed Bed Hybrid Discharge Plasma Reactor

In this work, a novel gas phase silent-packed bed hybrid discharge plasma reactor has been proposed, and its ability to control a simulative gas stream containing 240 ppm benzene is experimentally investigated. In order to optimize the geometry of the reactor, the benzene conversion rate and energy yield （EY） were compared for various inner electrode diameters and quartz tube shapes and sizes. In addition, benzene removal efficiency in different discharge regions was qualitatively analyzed and the gas parameter （space velocity） was systematically studied. It has been found that silent-packed bed hybrid discharge plasma reactor can effectively decompose benzene. Benzene removal proved to achieve an optimum value of 60% with a characteristic energy density of 255 J/L in this paper with a 6 mm bolt high-voltage electrode and a 13 mm quartz tube. The optimal space velocity was 188.1 h^-1, which resulted in moderate energy yield and removal efficiency. Reaction by-products such as hydroquinone, heptanoic acid, 4-nitrocatechol, phenol and 4-phenoxy-phenol were identified by mean of GC-MS. In addition, based on these organic by-products, a benzene destruction pathway was proposed.

MESHING THEORY AND DESIGN METHOD OF NEW SILENT CHAIN AND SPROCKET

Based on the study of the meshing theory of a new silent chain and sprockets, and the rolling cutting theory of sprocket and hob, the harmonious relations of dominating dimensions among the new silent chain, sprocket and hob is build, the meshing conditions are expatiated, and the resolved expression, which can instruct design and calculation, is educed. The tests show that the meshing design method is feasible.

miR-211 promotes lens epithelial cells apoptosis by targeting silent mating-type information regulation 2 homolog 1 in age-related cataracts

AIM： To detect the expression of miR-211 in age-related cataract tissue, explore the effects of miR-211 on lens epithelial cell proliferation and apoptosis, and identify its target gene.METHODS： This study used real-time quantitative polymerase chain reaction（RT-q PCR） to measure the expression of miR-211 and its predicted target gene [silent matingtype information regulation 2 homolog 1（SIRT1）] in 46 anterior lens capsules collected from age-related cataract patients. Human lens epithelial cell line（SRA01/04） cells were transfected with either miR-211 mimics, mimic controls, miR-211 inhibitors or inhibitor controls, 72 h after transfection, miR NA and protein expression of SIRT1 were measured using RT-qP CR and Western blotting; then cells were exposed to 200 μmol/L H2O2 for 1h, whereupon cell viability was measured by MTS assay, caspase-3 assay was performed. Dual luciferase reporter assay was performed to verify the relationship between miR-211 of SIRT1.RESULTS： Compared to the control group, expression of miR-211 was significantly increased（P〈0.001）, the miR NA and protein expression of SIRT1 were significantly decreased（P〈0.001） in the anterior lens capsules of patients with age-related cataracts. Relative to the control group, SIRT1 miR NA and protein levels in the miR-211 mimic group were significantly reduced, cell proliferation activity significantly decreased, and caspase-3 activity was significantly increased（P〈0.001）. In the miR-211 inhibitor group, SIRT1 miRNA and protein expression were significantly increased, cell proliferation activity significantly increased, and caspase-3 activity was significantly decreased（P〈0.001）. A dual luciferase reporter assay confirmed that SIRT1 is a direct target of miR-211.CONCLUSION： miR-211 is highly expressed in the anterior lens capsules of patients with age-related cataracts. By negatively regulating the expression of SIRT1, miR-211 promotes lens epithelial cell apoptosis and inhibits lens epithelial cell proliferation.

高校英语专业综合英语课程思政的教学设计研究——以“Silent Spring”一课的教学为例

Protective Effect of Silent Mating Type Information Regulation 2 Homolog 1 on TGF-β1 Pathway via mTOR in Diabetic Nephropathy

Objective: To demonstrate whether the expression of silent mating type information regulation 2 homolog 1 (SIRT1) affects the level of TGF-β1 and Smad3 in HEK293 cells through regulating mTOR. Methods: First, recombinant plasmids DNA (rSIRT1) and siRNA targeting SIRT1 were constructed which were transfected into Human Embryonic Kidney 293 cell (HEK293) cells, respectively. Then, the generation of intracellular ROS in cells was examined by flow cytometry using the oxidation-sensitive probe. Last, the expressions of TGF-β1, smad3, P53, mTOR, p-mTOR, LC3-I and LC3-II in cells were detected to observe the effect of SIRT1 on TGF-β1 Pathway by western blot analysis. Results: We demonstrated that overexpressing of SIRT1 may decrease TGF-β1 and Smad3 expression in HEK293 cells through regulating mTOR. In addition, the result is the opposite when SIRT1 was silent in HEK293 cells. Conclusions: SIRT1 is closely related to TGF-β1/Smad3 pathway that correlates with the regulation of mTOR and ROS generation and causes diabetic nephropathy. The available evidence implies that SIRT1 has great potential as a clinical target for the prevention and treatment of renal fibrosis in the development of DN.

Silent brain infarctions and leuko-araiosis in Chinese patients with first-ever acute lacunar strokes

We report on silent brain infarction (SBI) and leuko- araiosis (LA) of 23 patients with clinically diagnosed “first-ever” acute ischemic lacunar stroke. The lacunar syndromes were pure motor hemiparesis (10), pure sensory syndrome (2), ataxic hemiparesis (3), dysarthria clumsy hand syndrome (3), and sensory- motor deficit (5). Nineteen out of the 23 patients presented with completed strokes on arrival to the hospital, and 4 (17%) developed evolving-stroke within 24 hours of stroke onset. A lacune corresponded to the acute stroke could be found in all patients on brain magnetic resonance imaging (MRI), and in 18 (78%) on brain computed tomography (CT). MRI showed additional subclinical or asymptomatic “silent brain infarctions or lacunes” (SBI) in 19 (83%) of 23 patients, and leuko-araiosis (LA) of moderate to severe degree (> grade 2) was present in 61% of patients although dementia was absent. Hypertension is the risk factor in 78% of cases followed by diabetes mellitus, smoking, and elevated plasma cholesterol level. Independence of the types of lacunar syndromes, patients with hypertension and diabetes mellitus are associated with high grade LA. None with normal blood pressure and plasma glucose had grade 3 or grade 4 LA (p < 0.05). In conclusion, evolving-stroke occurs in one- fifth of patients with “first-ever” lacunar infarct within the first 24 hours of stroke onset. SBI was found in 83% of cases. Hypertension and diabetes mellitus are associated with additional SBI and high grade LA. The severity of leuko-araiosis per se dictates the cerebrovascular risks.

An Extraordinary Case of Silent Extensive Anterior Wall Myocardial Infarction Complicated with Giant Left Ventricular Aneurysm and Dressler Syndrome

Early post-acute myocardial infarction (AMI) pericarditis, pericardial effusion with or without cardiac tamponade, and late post-MI pericarditis (Dressler syndrome), are the major pericardial complications after AMI. It is quite rare and estimated to be only about 0.1% in AMI patients according to a recent report, so it is easily neglected or misdiagnosed and may have tragic result to patient. Clinical features of this post-AMI complication include fever, chest pain, pericarditis and pleurisy occurring 2 to 3 weeks after AMI. Dressler syndrome is rarely associated with left ventricular aneurysm. Contrast enhanced magnetic resonance and echocardiography play important roles in diagnosis of left ventricle aneurysm. We report a 54-year-old male heavy labor worker who had asymptomatic, severe coronary artery disease, complicated with silent myocardial infarction, which resulted in large left ventricular aneurysm, and also systolic heart failure was noted. Patient was diagnosed to have Dressler syndrome after his second cardiology clinic follow-up. He received coronary angiography which revealed triple vessel disease with total occlusion of left anterior descending artery, and a giant left ventricular aneurysm was found. He received surgical intervention with Batista method and followed-up uneventfully at the cardiology clinic.

Application of Silent Way in Foreign Language Teaching

The Silent Way (SW) is language teaching method which teacher keeps silent much of the time making students to produce the new language. This paper aims to apply SW to English teaching and figure out the idea why Silent Way couldn't be a widely-adopted teaching method. In terms of the characteristics of this innovational teaching method, it has some advantages as well as inevitable defects.

白藜芦醇通过调节SIRT1/AMPK信号通路介导自噬反应对膝关节骨性关节炎大鼠细胞凋亡的影响

目的:从沉默信息调节因子1(SIRT1)/腺苷酸活化蛋白激酶(AMPK)信号通路介导自噬角度,探讨白藜芦醇对大鼠膝关节骨性关节炎(KOA)软骨细胞凋亡的影响。方法:50只健康Wistar大鼠随机分为对照组、模型组、白藜芦醇组、白藜芦醇+SIRT1抑制剂组、自噬激活剂组,每组10只。除对照组外其余大鼠均通过注射弗氏完全佐剂造模法复制KOA大鼠模型,白藜芦醇组、白藜芦醇+AMPK抑制剂组、自噬激活剂组分别使用10μmol/kg白藜芦醇、10μmol/kg白藜芦醇+10 mg/kg EX527、2 mg/kg雷帕霉素进行干预,4周后,观察大鼠Lequesne MG膝关节级别;测定大鼠膝关节液中IL-6、肿瘤坏死因子β(TNF-β)水平;HE染色与TUNEL染色观测大鼠膝关节软骨组织形态及凋亡情况;透射电子显微镜观察大鼠软骨细胞自噬情况;Western blot法检测SIRT1、p-AMPK、AMPK、LC3、Beclin-1蛋白表达。结果:与对照组相比,模型组局部反应、步态反应、关节活动、关节肿胀程度加重(P<0.05);与模型组相比,白藜芦醇组、自噬激活剂组局部反应、步态反应、关节活动、关节肿胀程度减轻(P<0.05)。与对照组相比,模型组大鼠软骨组织细胞排列紊乱,粗糙,存在纤维化变性、边缘丘状隆起,细胞器减少,可见空泡变性,自噬小体数目增多,膝关节液IL-6、TNF-β水平、软骨细胞凋亡率、Beclin-1和LC3B/A升高(P<0.05),软骨组织SIRT1、p-AMPK/AMPK降低(P<0.05);与模型组相比,白藜芦醇组、自噬激活剂组大鼠软骨组织细胞排列紊乱、边缘丘状隆起等现象有改善,自噬小体数目增多,膝关节液IL-6、TNF-β水平、软骨细胞凋亡率降低(P<0.05),软骨组织SIRT1、p-AMPK/AMPK、Beclin-1和LC3B/A水平升高(P<0.05);SIRT1抑制剂可逆转白藜芦醇组对大鼠软骨细胞的保护作用。结论:白藜芦醇可能是通过激活SIRT1/AMPK通路介导自噬KOA大鼠软骨细胞凋亡,SIRT1抑制剂可逆转此过程。

THEORETICAL ANALYSIS ON THE TRANSIENT PROCESS OF AC SILENT DISCHARGE

A mathematical model for analysing transient process of AC silent discharge isestablished from its physical model.Then,the breakdown characteristics and the dependence ofdischarge current density on time are calculated theoretically in this kind of AC silent discharge.They are compared with the experimental results and the deviation between the theoretical andexperimental results are discussed briefly.

软骨细胞中SIRT1基因敲减后作用状态不明确信号通路及对相关疾病或功能的影响

背景:沉默信息调节因子1以去乙酰化的方式调控软骨细胞中相关蛋白的功能,参与软骨细胞增殖分化,促进软骨缺损修复。目的:利用高通量技术筛选软骨细胞中沉默信息调节因子1基因敲减后作用状态不明确的信号通路以及产生变化的疾病或功能。方法:取处于对数生长期的小鼠ATDC5软骨细胞,分2组转染:对照组转染沉默信息调节因子1基因敲减阴性对照慢病毒,实验组转染沉默信息调节因子1基因敲减慢病毒。转染72 h后,利用小鼠基因表达谱芯片GeneChip®Mouse Genome 4302.0 Array检测mRNA的基因层面变化情况,应用生物信息学技术筛选激活或抑制不明确的信号通路以及这些信号通路相关因子,并分析疾病或功能模块的富集情况。结果与结论:①沉默信息调节因子1基因敲减后,小鼠ATDC5软骨细胞中激活或抑制状态不明确的信号通路有245条;②根据-log(P-value)排序选择排前20的激活或抑制状态不明确信号通路中的因子情况进行报道,包括IGFBP4、TGFBR1、CTGF、COL4A5、LHX2、IL1RL1、KLF6等;③根据-log(P-value)进行排序,细胞生长和增殖、生物体存活和细胞死亡与存活等14个疾病和功能密切相关模块明显变化;根据差异基因数量排序,生物体损伤和异常、癌症和细胞死亡与存活3个疾病和功能密切相关模块明显变化;根据-log(P-value)与差异基因数量综合排序,固有免疫应答这一疾病和功能模块被显著激活。

Identification of novel silent HIV propagation routes in Pakistan

Human immunodeficiency virus(HIV) is rapidly increasing in both high risk groups and the general population.In this study, silent routes of propagation in teenaged Pakistanis are discussed. In order to promote sexual activity in youths, regular clients write contact details of sex workers on the doors of public washrooms. HIV prevalence is much higher among Hijra sex workers.Hijra sex workers have apparently stepped into the profession of begging at public places, where they earn money by both begging and distributing visiting cards offering unsafe sex. In many educational institutes, sex education is lacking or absent; if delivered via teachers,government agencies and nongovernmental organizations this could prevent a future epidemic of sexually transmitted infections in Pakistan.
