Effects of Nourishing “Yin”-Removing “Fire” Chinese Herb Mixture on the Differential Expression of GABA<sub>A</sub>Receptor <i>α</i>Subunits in Hypothalamus of Precocious Puberty Female Rats

GABAergic input to Gonadotropin-releasing hormone (GnRH) neurons is necessary to initiate the onset of puberty and its action mainly depends on GABAA receptor of which the subunit composition, properties and consequently function varies during this period. Nourishing “Yin”-Removing “Fire” Chinese herb mixture, a Chinese herb-based formulation, has been proved that it may retard the initiation of pubertal development in female precocious puberty rats. Our objective is to investigate the effects of Nourishing “Yin”-Removing “Fire” Chinese herb mixture on the expression of GABAA receptor α subunits in hypothalamus. Female Sprague-Dawley rats were divided into normal (N), precocious puberty model (M) induced by danazol, model exposed to saline (MS) and model exposed to Chinese herb mixture (CHM) groups. All rats were administered by the Chinese herb mixture from P15 on. Coefficients of reproductive organs and serum gonadotropins and estradiol levels in M were significantly enhanced while they were significantly decreased in CHM. The hypothalamic GnRH mRNA was also significantly increased in M and in CHM, as well as ERα mRNA. At the mean time, the hypothalamic GABAA receptor α1 and α3 subunits mRNA were more significantly decreased in M than those of N, while they were more significantly enhanced in CHM than those in M (p 0.01), the protein expression of which in hypothalamus had the same trend as the mRNA expression. The evidence suggests that Nourishing “Yin”-Removing “Fire” Chinese herb mixture could significantly retard the sexual development of the precocious rats, and up-regulate the expressions of hypothalamic GABAA receptor α1 and α3 subunits. Our result indicated that GABAA receptor α1 and α3 subunits might involve in the effective treatment of herb mixture on idiopathic precocious puberty. 展开更多

背景:六味地黄丸以“三补三泻”为配伍特点,补泻兼施以滋阴补肾;左归丸以“阳中求阴”为配伍特点,以阳促阴补肾。二者同属于滋阴补肾法,其在证候和细胞分子水平等都发挥较好的疗效。目的:观察六味地黄丸和左归丸在骨代谢中的作用,探讨... 背景:六味地黄丸以“三补三泻”为配伍特点,补泻兼施以滋阴补肾;左归丸以“阳中求阴”为配伍特点,以阳促阴补肾。二者同属于滋阴补肾法,其在证候和细胞分子水平等都发挥较好的疗效。目的:观察六味地黄丸和左归丸在骨代谢中的作用,探讨其在破骨通路骨保护素(osteoprotegerin,OPG)/核因子κB受体活化因子配体(receptoractivatorofnuclearfactor-κBligand,RANKL)的作用机制。方法:将32只SD随机大鼠分为模型组、六味地黄丸组、左归丸组、假手术组,每组8只。除假手术组大鼠外,其余大鼠切除双侧卵巢制备骨质疏松模型。术后30 d开始六味地黄丸组大鼠灌胃六味地黄丸1.125 g/(kg·d);左归丸组大鼠灌胃左归丸2.25 g/(kg·d);假手术组和模型组灌胃生理盐水10 mL/(kg·d)。灌胃12周后,取大鼠胫骨检测骨密度,采用ELISA检测血清中雌激素E2、骨碱性磷酸酶、cAMP/cGMP水平,Westernblot法检测股骨中OPG/RANKL的表达情况。结果与结论:①与假手术组相比,模型组骨密度及雌激素E2、骨碱性磷酸酶水平降低(P<0.05),cAMP/cGMP水平升高(P<0.05)。②与模型组相比,六味地黄丸组和左归丸组骨密度显著提高(P<0.05);骨碱性磷酸酶均显著提高(P<0.05);左归丸组cAMP/cGMP水平明显降低(P<0.05)和上调OPG蛋白表达(P<0.05),六味地黄丸组上调OPG蛋白表达和下调RANKL蛋白表达均有作用(P<0.05);两组均不能显著提高雌激素E2水平(P>0.05)。③结果显示,“阳中求阴”的左归丸能够有效提高OPG表达,不能下调RANKL蛋白,而“三补三泻”的六味地黄丸能双向调节OPG和RANKL蛋白的表达,提示了六味地黄丸对抑制破骨功能方面可能比左归丸显著,此结论仍有待进一步的研究加以验证。 展开更多

We defined generalized relations and generalized reasoning, introduced the concept of steady multilateral systems with two non-compatibility relations, and discussed its energy properties. Later based on the intervention principle in image mathematics of TCMath and treated the research object of the image mathematics as a steady multilateral system, it has been proved that the mathematical intervening principle is true. Objective:To explore the nature of deficiency in spleen-yin syndrome,which could provide scientific theoretical support and practical guidance for clinical Traditional Chinese Medicine(TCM)syndrome differentiation based on biology,and had a strong clinical significance.Methods:Serum Cu and Zn were detected by atomic absorption spectrophotometer,serum vitamin E by high performance liquid chromatography,serum vitamin C by 2,4-Dinitrophenylhydrazine Colorimetry,total superoxide dismutase(SOD)and Cu and Zn- SOD by the xanthine oxidase method,and malondialdehyde(MDA)by the 2-thiobarbituric acid method(TBA).Total antioxidant capacity was detected by a colorimetry kit.Amylase Activity was detected by an automatic biochemical analyzer.Lysozyme was detected by lysozyme detection plate,the diameter of bacteriolysis circle was measured and the corresponding content of lysozyme was obtained from a table of standard curve values.Results: No significant difference in total SOD and Cu,Zn-SOD was found between deficiency in spleen-yin group and normal group.However,such factors in deficiency in kidney-yin group were significantly lower than the other groups(P〈0.05).The MDA content in both deficiency in spleen-yin group and deficiency in kidney-yin group were significantly higher than that of normal group(P〈0.05),while the total antioxidant capacity was significantly lower than normal group(P〈0.05).The vitamin E content in deficiency in kidney-yin group was significantly lower than that in the other two groups(P〈0.05).No significant difference in the contents of vitamin C,Cu and Zn were observed in these groups.The Zn/Cu level in deficiency in kidney-yin group and the vitamin E level in deficiency in spleen-yin group decreased,but with no significant difference.Amylase activity in unit time in cases with deficiency in spleen-yin was lower than and had significant differences with that in normal cases,and higher than that in cases with deficiency in kidney-yin.The sectional velocity of saliva and the ratio of lysozyme in normal case group were significantly higher than other two groups,while deficiency in the spleen-yin group was significantly higher than the deficiency in kidney-yin group.Conclusion:All the results indicated that the objective pathological mechanism between the deficiency in spleen-yin and deficiency in kidney-yin was different. 展开更多

A 3D dynamic core of the non-hydrostatic model GRAPES(Global/Regional Assimilation and Prediction System) is developed on the Yin-Yang grid to address the polar problem and to enhance the computational efficiency. Three-dimensional Coriolis forcing is introduced to the new core, and full representation of the Coriolis forcing makes it straightforward to share code between the Yin and Yang subdomains. Similar to that in the original GRAPES model, a semi-implicit semi-Lagrangian scheme is adopted for temporal integration and advection with additional arrangement for cross-boundary transport. Under a non-centered second-order temporal and spatial discretization, the dry nonhydrostatic frame is summarized as the solution of an elliptical problem. The resulting Helmholtz equation is solved with the Generalized Conjugate Residual solver in cooperation with the classic Schwarz method. Even though the coefficients of the equation are quite different from those in the original model, the computational procedure of the new core is just the same. The bi-cubic Lagrangian interpolation serves to provide Dirichlet-type boundary conditions with data transfer between the subdomains. The dry core is evaluated with several benchmark test cases, and all the tests display reasonable numerical stability and computing performance. Persistency of the balanced flow and development of both the mountain-induced Rossby wave and Rossby–Haurwitz wave confirms the appropriate installation of the 3D Coriolis terms in the semi-implicit semi-Lagrangian dynamic core on the Yin-Yang grid. 展开更多

AIM To evaluate the therapeutic mechanism of Yinchenhao Chengqi (YCHCQ) decoction (containing mainly Herba Artemisia capillaris) combined with endoscopic sphincterotomy (EST) for endotoxemia (ETM) in acute cholangitis.METHODS Twenty-one cases of acute cholangitis with ETM were divided randomly into two groups: group A, 10 patients treated with YCHCQ decoction combined with EST, group B, 11 patients treated with EST. The incidence rate of ETM, plasmic ET, serum superoxide dismutase (SOD) activity, malonyldialdehyde (MDA), complement C3 and C-reactive protein (CRP) were studied respectively.RESULTS The ET level of group A (35.92ng/L±8.30ng/L) was significantly reduced after 7 days of treatment (P<0.05) in contrast to that of group B (47.8ng/L±11.62ng/L), so did the level of MDA and CRP. But the SOD activity and C3 level in group A increased significantly (P<0.05).CONCLUSION YCHCQ decoction combined with EST had a beneficial effect for ETM in acute cholangitis.INTRODUCTIONEndotoxemia (ETM) is one of the most important physiopathologic causes of acute cholangitis and it is the trigger of cytokines and inflammatory factors. In recent studies it has been found that Yinchenhao Chengqi (YCHCQ) decoction has a beneficial effect on ETM in acute cholangitis. With the development of endoscopic surgery, endoscopic sphincterotomy (EST) has become an effective replacement for some operations in the treatment of acute cholangitis[1]. The effect of YCHCQ decoction combined with EST on ET, oxygen free radical and complement C3 was observed in order to find out its therapeutic mechanism. 展开更多

Apoptosis of PANC-1 cells before and after treatment with YCHD was determined by TUNEL staining. Expression of the apoptosisassociated genes, Bcl-2 and Bax, was detected by immunohistochemical staining and reverse transcription-PCR.RESULTS: YCHD inhibited the growth of PANC-1 cells. Following treatment with YCHD for 24-96 h, the apoptotic rate of PANC-1 cells increased with time. In addition, the positive rate of Bcl-2 protein expression decreased in a time-dependent manner, whereas the positive rate of Bax protein expression increased in a time-dependent manner. Following treatment of with YCHD for 24-96 h, expression of BAX m RNA increased gradually and BCL-2 m RNA reduced gradually with time.CONCLUSION: YCHD induces apoptosis of PANC-1 cells mediated in part via up-regulation of BAX and down-regulation of BCL-2. 展开更多

AIM To investigated the mechanism of the association between the TBX21 T-1993 C promoter polymorphism and autoimmune hepatitis type 1(AIH-1) development.METHODS In vivo,In vivo,and reporter analyses were performed to determine the function of transcription factors binding to the T-1993 C element of the TBX21 promoter in human CD4+ T and B cell lines. Flow cytometry and quantitative real-time PCR were used to analyze T-box transcription factor(T-bet) and interferon-γ(IFN-γ) expressions in CD4+ T cells,B cells and monocytes from the peripheral blood of AIH-1 patients including 5-1993 TC and 15-1993 TT genotype carriers,and healthy controls including 10-1993 TC and 25-1993 TT genotype carriers. Furthermore,a range of biochemical indices was measured simultaneously in the blood of AIH-1 patients.RESULTS TBX21-1993 C allele created a strong Yin-Yang 1(YY1)-binding site and decreased transcriptional activity of TBX21 promoter in human CD4+ T and B cells.Higher levels of T-bet and IFN-γ were detected in the circulating CD4+ T cells and B cells of AIH-1 patients carrying the TBX21-1993 TT genotype compared with the patients carrying the-1993 TC genotype and controls with the-1993 TC genotype. T-bet expression levels of circulating T cells and B cells were positively correlated with AIH-1 disease activity. Knockdown of YY1 with si RNA caused increased expression of T-bet and IFN-γ in peripheral blood mononuclear cells in AIH-1 patients. CONCLUSION The repression of TBX21 expression by high-affinity binding of YY1 to the-1993 C allele may contribute to a decreased development of AIH-1 via suppression of type 1 immunity. 展开更多

Yin-mountain Hilly Area is one of the ideal regions for potato (Solanum tuberosum) production in China. However, potato yield is severely limited as a result of rain-fed crop mode due to water deficiency, as well as an inadequate farming practices. In this study, yield gaps were determined by using attainable yield (Ya) as a benchmark under optimized management practices, i.e., micro-ridge and side planting with plastic-mulching (MS), and flat planting with plastic-mulching (PM). The yields under MS and PM modes are defined as Yal and Ya2, respectively. Under the same field with MS and PM modes but different densities and fertilizer usages and so on, it was defined as simulated farmers' practices. The yield of simulated farmers' practices (Yfl) reached 57.3 and 69.6% of Yal and Ya2, respectively, while the average yield of 298 randomly surveyed farmers (Yf2) reached only 37.0 and 47.8% of Yal and Ya2 for rain-fed potato, respectively. The gaps of water use efficiency exhibited similar pattern. Further analysis shows that improper measures in rainwater conservation and accumulation, and other management practices contributed to 18.5, 18.2, and 42.6% of yield gap between Yal and Yf2. Improper nutrition management, including overuse of nitrogen and the deficiency of phosphorus andpotassium supplication, was one of the important reasons of yield gap. The results indicate the possibilities of increasing rain-fed potato yields by optimized water and fertilizermanagements in the Yin-mountain Hilly Area. 展开更多

A global transport model is proposed in which a multimoment constrained finite volume (MCV) scheme is applied to a Yin-Yang overset grid. The MCV scheme defines 16 degrees of freedom (DOFs) within each element to build a 2D cubic reconstruction polynomial. The time evolution equations for DOFs are derived from constraint conditions on moments of line-integrated averages (LIA), point values (PV), and values of first-order derivatives (DV). The Yin-Yang grid eliminates polar singularities and results in a quasi-uniform mesh. A limiting projection is designed to remove nonphysical oscillations around discontinuities. Our model was tested against widely used benchmarks; the competitive results reveal that the model is accurate and promising for developing general circulation models. 展开更多

AIM To investigate the secreting functions of saliva in patients with TCM Piyinxu (Spleen yin deficiency). METHODS According to the traditional Chinese medical diagnositic standard, 25 cases of Piyinxu (male 15, female 10, aged from 26~70, average 45 years) were observed and compared with 20 cases of Shenyinxu (Kidney yin deficiency) patients (male 11, female 9, aged from 35~75, average 50) and 30 normal persons (male 17, female 13, aged from 35~65, average 49). After acid stimulation, the secretion flow was measured, the saliva amylase and protein were surveyed using the automatic biochemical analyzer. Statistical analysis was carried out with Kruskal Wallis test and one way factorial ANOVA test. RESULTS In Piyinxu patients, the saliva′s secreting flow rate (0 27ml/min±0 016ml/min) and amylase per min (2134 13IU/min±343 51IU/min) was lower than that in normal subjects (0 46ml/min±0 027ml/min and 3501 63IU/min±1099 63IU/min, P <0 01) and higher than that in Shenyinxu group (0 13ml/min±0 051ml/min and 951 62IU/min±383 17IU/min, P <0 01). But there was no significant difference between each group in total salivary protein (Piyinxu group 3 07g/L±0 60g/L, Shenyinxu 3 01g/L±0 90g/L and control 2 94g/L±1 13g/L, P =0 869), amylase per volume and the ratio of the amylase to the protein ( P =0 173 and 0 436). CONCLUSION The secreting functions of saliva in Piyinxu patients are low. 展开更多

Methods: The mouse model of bronchial asthma with Yin deficiency syndrome was established by the treatment with injecting ovalbumin(OVA) two times to sensitize, inhaling OVA 14 times to stimulate, and using thyroxin through lavage during late stimulation. This model was evaluated through body weight, asthmatic behaviors, respiratory function, autonomous activity, lung pathology, and pulmonary fluid clearance. Results: OVA combined with thyroxin was an appropriate method to induce the mouse model with increased food and water intake, autonomous activity, asthmatic behaviors score, and respiratory rate, decreased body weight, tidal volume, and wet/dry ratio of lung, and changed with pathology of lung tissue. The changes of the above mentioned parameters indicated that the model was the bronchial asthma with Yin deficiency syndrome. Conclusion: The OVA combined with thyroxin is a good pattern to establish a mouse model of bronchial asthma with Yin deficiency syndrome successfully, which can highly simulate the clinical symptoms of this disease. 展开更多

In the past decade, research has mostly focused on the Yang aspect of erectile function. However, in recent years, the Yin side is attracting increasingly greater attention. This is due to the realization that penile flaccidity is no less important than penile erection and is actively maintained by mechanisms that play critical roles in certain types of erectile dysfunction (ED); for example, in diabetic patients. In addition, there is evidence that the Yin and Yang signaling pathways interact with each other during the transition from flaccidity to erection, and vice versa. As such, it is important that we view erectile function from not only the Yang but also the Yin side. The purpose of this article is to review recent advances in the understanding of the molecular mechanisms that regulate the Yin and Yang of the penis. Emphasis is given to the Rho kinase signaling pathway that regulates the Yin, and to the cyclic nucleotide signaling pathway that regulates the Yang. Discussion is organized in such a way so as to follow the signaling cascade, that is, beginning with the extracellular signaling molecules (e.g., norepinephrin and nitric oxide) and their receptors, converging onto the intracellular effectors (e.g., Rho kinase and protein kinase G), branching into secondary effectors, and finishing with contractile molecules and phosphodiesterases. Interactions between the Yin and Yang signaling pathways are discussed as well. 展开更多
