A relatively large genetic diversity was detected among the samples based on estimation of DNA products amplified from 15 selected ISSR primers, with the similarity coefficient varying from 0.17 to 0.89. The mean expected heterozygosity (He) was 0.171 4 per locus, and Shannon index (1) was 0.271 4. The Principal Coordinate Analysis (PCA) further indicated that genetic diversity of the Jiangwan wild soybean population was not evenly distributed, instead, was presented by a mosaic or clustered distribution pattern. Correlation study between genetic diversity and number of samples demonstrated that genetic diversity increased dramatically with the increase of number of samples within 40 individuals, but the increase became slow and rapidly reached a plateau when more than 40 individuals were included in the analysis. It is concluded that (i) a sample set of approximately 35-45 individuals should be included to represent possibly high genetic diversity when conservation of a wild soybean population ex situ is undertaken; and (ii) collection of wild soybean samples should be spread out as wide as possible within a population, and a certain distance should be kept as intervals among individuals for sampling.展开更多

Variables tested were volume ratio of material to solvent, size of material, extraction time and temperature and composition of extraction solvent (mixtures of acetone-water). Each variable was tested at seven levels; 7 experiments were performed in random order. Analyses of the extracts were performed by high-performance liquid chromatography with diode array detection(HPLC-DAD). Analytical responses were processed by using a forward regression analysis, in order to find polynomial function describing the relationship between variables and responses. For all the analytes the experimental conditions for providing the highest extraction yield inside the experimental domain considered were found. Finally, a simple, rapid and accurate analytical method was developed for the determination of emodin by high performance liquid chromatography. The separation is achieved within 25 rain on an ODS column using methanol and water as gradient mobiles. Emodin can be quantified by using external standard method detecting at 436 nm. Good linearity is obtained with correlation coefficient exceeding 0.9986 and the detection limit and the quantification limit are 1.53 and 3.23 mg/L respectively. This method shows good reproducibility for the quantification of the emodin with intra-day and inter-day relative standard deviation less than 2.3% and 5.6% respectively. Under optimized extraction conditions, the recovery of the standard is 96.5%. The validated method is successfully applied to quantify the emodin in seven Polygonum cuspidatum sieb. Et zucc. products, which provided an idea for the pre-treatment of determination of active compounds in traditional Chinese medicines.展开更多

The contents of motilin and somatostatin in small intestine of mice were determinated by radioimmunoassay. The electrical potential difference (PD) related to Na+ and glucose transport was measured across the wall of reverted intestinal sacs. Na+-K+-ATPase activity of small intestinal mucosa was measured by spectroscopic analysis.RESULTS: Different dosages of emodin can improve small intestinal peristalsis of mice. Emodin increased the content of motilin, while reduced the content of somatostatin in small intestine of mice significantly. Emodin 0.2, 0.4, 0.8, and 1.6 g/L decreased PD when there was glucose. However, emodin had little effect when glucose was free. The Na+-K+-ATPase activity of small intestinal mucosa of mice in emodin groups was inhibited obviously. CONCLUSION: Emodin can enhance the function of small intestinal peristalsis of mice by mechanisms of promoting secretion of motilin, lowering the content of somatostatin and inhibiting Na+-K+-ATPase activity of small intestinal mucosa.展开更多

METHODS: Ten CDs were treated orally with autologous CEP thrice weekly for 16 wk. Subjects were monitored for CDAI and IBDQ. Immune modulatory effect was assessed by T-lymphocyte FACS analysis, CEP-specific IFNγ ELISPOT assay and cytokine levels. RESULTS: Induction of oral immune regulation significantly ameliorated disease activity. All (10/10) subjects had clinical response (CDAI≤70) and 7/10 achieved clinical remission (CDAI≤150). Significant increase in mean IBDQ score was noted (134±9 vs 164±12). No treatment-related adverse events were noted. High levels of CEP-specific IFNγ spot forming colonies were detected in five subjects prior to treatment and in all five, a marked decrease was observed. The CD4+/CD8+ lymphocyte ratio and peripheral NKT cell numbers increased significantly, in 7/10 and in 5/10 subjects, respectively. Significant increase in serum IL-10 and IL-4 levels was observed in 7/10 subjects during treatment period. CONCLUSION: Immune regulation via oral administration of CEP is a safe and possibly effective treatment for subjects with moderate CD and may provide means of antigen-specific immune modulation.展开更多

To carry out these aims, in the first instance the additive was incorporated into meat patties (elaborated with 80% beef and 20% fat plus 2% salt over total mass) at 0, 0.5, 1, 5 and 10 g/kg, respectively. The lipid oxidation was evaluated by measurement of peroxide value during 15 days at 8 ℃ and the results were expressed as reduction percentage of peroxidation (RP). RP obtained were 55, 49, 73 and 59 respectively. As 5 g/kg was the optimum concentration of antioxidant, this was chosen to add to meat products enriched with soybean oil. In this case the RP obtained was 78%, indicating that the extract decreased lipid oxidation effectively.展开更多

Antimicrobial activities for individual crude extract were well established. Crude methanolic extracts of Alpinia galanga, Eurycoma longifolia Jack and Syzygium aromaticum and the combination for all extracts were tested using well diffusion techniques against Saccharomyces cerevisiae (ATCC 9763) and Escherichia coli (ATCC 25922). The mixed extracts were prepared based on the concentration ratio of 50 μg/μL which are 1:1, 1:1:1 and 1:2:2. Single Syzygium aromaticum extract showed higher inhibition zone on Saccharomyces cerevisiae compared to Escherichia coli. There is reduction in diameter of inhibition zone for single extract and mixture extracts either in combination of two or three extracts tested on Saccharomyces cerevisiae and Escherichia coli growth but they are not significant. In conclusion, Syzygium aromaticum showed highest activity against Saccharomyces cerevisiae and Escherichia coli. Reduction in diameter of inhibition zone indicated that Alpinia galanga and Eurycoma longifolia Jack extracts had antagonistic effect with Syzygium aromaticum.展开更多

RAPD results have shown that among 30 selected primers for gametophytes, 297 loci ranging from 200 to 3 000 bp were obtained in the average of 9.9 loci for each primer. This indicated a high polymorphic rate with RAPD detection. UPGMA (unweighted pair-group method arithmetic average) analysis showed that each male and female gametophyte of a generation could be clustered into one pair separately. The genetic distances of the Kelp 901 generation were 0.321 2–0.476 7, and the maximum was between F7 and F8 (0.476 7). Identity analysis showed that F6 generation was more close to the female parent (0.659 3), and F7 generation was more close to the male parent (0.578 8). To the sporophytes study in 24 selected primers for RAPD amplification, 191 loci ranging from 230-2 800 bp were obtained, in the average to each primer of 8.0 loci. The heterozygosity to six populations were male parent (0.223 9), female parent (0.107 2), F6 (0.216 4), F7 (0.228 6), F8 (0.229 6) and F9 (0.317 2). The nearest genetic distance was 0.083 5 (F8, F9). Total heterozygosity (HT) of F6, F7, F8 and F9 generations was 0.318 6, the average heterozygosity (HS) for F6, F7, F8 and F9 generations was 0.248 0, and deduced coefficient of population differentiation (Gst ) was 22.2%. Six sequence characterized amplified regions (SCAR) were preliminary screened through RAPD analysis. It needed to be verified in detail as they are significant for molecular marker assistance in breeding and selecting Laminaria.展开更多

The plant is a raw material for these studies. Zygophyllum gemini is a species widely used in Algeria to treat this disease. Our aim is to investigate the antidiabetic activity of aqueous extract and its fractions in induced diabetic Wistar rats by streptozotoein. The three drugs caused a decrease in blood sugar for 14 days. Butanolic fractions (BF) fraction gives significant results on blood glucose after seven days and significant regulating oral glucose tolerance. This preliminary study shows that Z. geslini is endowed with a remarkable antidiabetic activity and that further studies are needed.展开更多

Bongkok Wistar male rates. Antihyperuricemic investigation on Wistar male rats showed that administration of ethanol extract at doses of 200 mg/kg bw decreased serum uric acid level significantly compared to control group at hour 6 and 7 (P 〈 0.05) after inducing with potassium oxonate intraperitoneally simultaneously with uric acid orally. Whereas, administration of ethanol extract at doses of 100 mg/kg bw did not decrease serum uric acid level significantly different compared to control group at hour 6 and 7 (P 〈 0.05). Determination of uric acid level in urine, administration of ethanol extract at a dose of 200 mg/kg bw, or probenecid as a standard drug, at a dose of 45 mg/kg bw increased excretion of urine uric acid level significantly different compared to control group in day of 7 (P 〈 0.05) after inducing with potassium oxonate intraperitoneally simultaneously with uric acid orally. However, increase of uric acid excretion by ethanol extract was lower compared to that of probenecid at a dose of 45 mg/kg bw. Mechanism of action of the ethanol extract as an antihyperuricemia has been proposed by inhibition of xanthine oxidase and finally decreased the synthesis of uric acid and increased the excretion of urine uric acid level.展开更多

Diabetes was induced intraperitonially by a single injection of STZ (streptozotocin) (60 mg/kg bw (body weight)). Twenty diabetic rats, weighing 263 ± 5 g, were divided into two groups fed a casein diet supplemented or not with Po extract (1 g/kg bw), for four weeks. Control group (n = 6) received 0.23-0.25 mL of citrate buffer and was fed a standard diet during the experiment. The study was carried out at Oran University, Algeria and the entire experiments lasted from September 2011 to July 2012. Blood was obtained from the abdominal aorta of rats after fasting overnight and standard methods were used for the extraction of spices, determination of glycemia, insulinemia, lipid peroxidation and antioxidant enzymes activities. Portulaca oleracea treated compared to untreated rats, glycemia and HbAIc values were respectively 2.8- and 1.7-fold lower. TBARS (thiobarbituric acid reactive substances) concentrations were reduced in RBC (red blood cells) (-54%) and plasma (-65%). Moreover, in liver and kidney, TBARS values were respectively 1.8- and 2-fold lower. SOD (superoxide dismutase) and GSH-Px (glutathione peroxidase) activities were increased respectively by +38% and +85%, in liver. GSSG-Red (glutathione reductase) activity was 1.9-fold higher in kidney, while CAT (catalase) was improved in kidney (+48%). In RBCs, SOD, GSH-Px, GSSH-Red and CAT activities were increased by 31%, 42%, 56% and +50%, respectively. These data have cast a new light on the actions of Portulaca oleracea and its antioxidant potential benefits in preventing diabetes and its complications.展开更多

The assaying and mineralogical studies carried out on the nickel laterite sample, showed the 0.88% Ni, and principally consisted of oxide and silicate crystalline phases i.e. dolomite, quartz, magnetite, and goethite. Among numerous factors affecting such process, four major parameters i.e. temperature, agitator speed(r/min), leaching agents and their concentration were considered in a two-level full factorial experimental design. The agitation leach tests showed that the ore could be leached at atmospheric pressure with sulfuric acid while citric acid was almost unpromising. Analysis of variance(ANOVA) using DX7 software was employed to identify effective parameters. Sulfuric acid concentration and temperature were the most effective parameters on Ni extraction. Furthermore, the factorial models for experiment responses were developed. The results showed 83% Ni extraction after 4 h leaching, under optimized conditions i.e.temperature at 95 ℃, acid concentration at 5 N and agitator speed at 1000 r/min. This study revealed that factorial experimental design can be implemented to identify effective parameters on the agitation leaching process of nickel laterite.展开更多

Fish was the most important prey at each of the colonies sampled, followed by octopods and gastropods. Among colonies, there were marked differences in the size of the fish consumed, which was smaller at Py Point. This was mainly due to the larger number of speci-mens of the smallest fish prey species consumed there. Shags from Py Point performed longer forag-ing trips and spent more time per day foraging. Although at the beginning of the study the number of chicks per nest observed at the three colonies was similar, the number surviving to fledge at Py Point was markedly lower. The relative higher foraging effort and lower breeding success observed at Py Point might be related to the differences in the fish consumed between colonies. My results suggest that the decline in the inshore fish populations observed around the South Shetland Islands could be one of the reasons explaining the steady decrease in the number of breeding Antarctic Shags observed at colonies monitored in this archipelago.展开更多

Reduction in diameter of inhibition zone indicated that Alpinia galanga and Eurycoma longifolia Jack extracts had antagonistic effect with Syzygium aromaticum.

文摘Based on DNA extraction and optimization of random amplified reaction (RAPD) to the gametophytes and sporophytes of Kelp “901” strain, genetic study on variation was conducted to its parents and offsprings of F6, F7, F8, and F9 generation. It needed to be verified in detail as they are significant for molecular marker assistance in breeding and selecting Laminaria.
文摘Diabetes mellitus is a major public health concern. Finding a cure for the disease without its side-effects is the objective of modem medicine. This preliminary study shows that Z. geslini is endowed with a remarkable antidiabetic activity and that further studies are needed.
文摘The aim of the study was to investigate antihyperuricemic effect of snake fruit (Salacca edulis Reinw.) var. Mechanism of action of the ethanol extract as an antihyperuricemia has been proposed by inhibition of xanthine oxidase and finally decreased the synthesis of uric acid and increased the excretion of urine uric acid level.
文摘The aim of this study was to investigate the antihyperglycemic and antioxidant effects of Po (Portulaca oleracea) lyophilised aqueous extract in diabetic male Wistar rats. These data have cast a new light on the actions of Portulaca oleracea and its antioxidant potential benefits in preventing diabetes and its complications.
文摘This research work deals with the extraction of nickel from a low grade nickel laterite ore, taken from a deposit located in southwestern of Iran, through agitation leaching at atmospheric pressure. This study revealed that factorial experimental design can be implemented to identify effective parameters on the agitation leaching process of nickel laterite.
文摘Information on diet composition, foraging effort and breeding success of the Antarctic Shag (Phalacrocorax bransfieldensis) was obtained at three colonies on the Antarctic Peninsula, dur-ing the 1997/98 breeding season. My results suggest that the decline in the inshore fish populations observed around the South Shetland Islands could be one of the reasons explaining the steady decrease in the number of breeding Antarctic Shags observed at colonies monitored in this archipelago.
