[Objective] The aim was to explore the relevance between decreased 1 000-grain weight and grain-filling characteristics of rice giant embryo mutant.[Method] Richards equation was used to describe grain filling processes of giant embryo mutant MH-ge1 and its corresponding wild type so as to analyze the parameters of grain-filling characteristics of materials.[Result] The initial filling power of mutant MH-ge1 was higher than the wild type MH86,however,the maximum filling rate,the mean filling rate and the final increment of mutant MH-ge1 was lower than those of MH86.[Conclusion] The grain-filling characteristics of mutant MH-ge1 were poorer than the wild type MH86,thus resulting in the decrease of the grain weight of the giant embryo rice. 展开更多

To evaluate the role of tissue factor (TF) and protease activated receptor (PAR)-2 in liver fibrosis. METHODSUsing CCl<sub>4</sub> administration for eight weeks, we induced hepatic fibrosis in wild-type C57BL/6 mice and in mice with deletion of the cytoplasmic signalling domain of TF (TF<sup>§CT/§CT</sup>), deletion of PAR-2 (PAR-2<sup>-/-</sup>) and combined deletion of TF signalling domain and PAR-2 (TF<sup>§CT/§CT</sup>/PAR-2<sup>-/-</sup>). Hepatic fibrosis area was assessed by quantitative imaging of picrosirius red staining. Hepatic collagen content was assessed by hydroxyproline levels. Hepatic stellate cells (αSMA positive) and hepatic macrophages (CD68 positive) were identified by immunohistochemistry. Hepatic gene expression was determined by PCR and liver TGFβ1 content by ELISA. RESULTSCCl<sub>4</sub> treated mice with deletion of the PAR-2 gene (PAR-2<sup>-/-</sup>) and the cytoplasmic domain of TF (TF<sup>§CT/§CT</sup>) developed significantly less hepatic fibrosis, characterised by reduced liver fibrosis area and hydroxyproline content, compared to control wildtype mice treated with CCl<sub>4</sub>. The observed reduction in histological fibrosis was accompanied by a significant decrease in the hepatic content of TGFβ, the prototypic fibrogenic cytokine, as well as fewer activated hepatic stellate cells and hepatic macrophages. Deletion of the TF cytoplasmic signalling domain reduced hepatic fibrosis to levels similar to those observed in mice lacking PAR-2 signalling but combined deletion provided no added protection against fibrosis indicating a lack of mutual modulating effects that have been observed in other contexts such as angiogenic responses. CONCLUSIONTissue factor cytoplasmic domain is involved in TF-PAR-2 signalling initiating hepatic fibrosis and is a potential therapeutic target, as its deletion would not impact coagulation. 展开更多

A new experimental apparatus was set up to investigate the actual fi-iction characteristics on the basis of speed control of the serve system.A modified friction model was proposed due to real time varying deformation resistance.The approach to identify the parameters of comprehensive friction behaviors based on the modified model was proposed and applied to the forging press.The impacts on parameters which the external load had were also investigated.The results show that friction force decreases with velocity in the low velocity regime whereas the friction force increases with the velocity in the high velocity regime under no external load.It is also shown that the Coulomb friction force,the maximum static friction force and the vicious friction coefficient change linearly with the external load taking the velocity at which the magnitude of the steady state friction force becomes minimum as the critical velocity. 展开更多

Objective To explore the relationship between human cytomegalovirus (HCMV) UL144 sequence variability and clinical disease. Methods HCMV UL144 open reading frame (ORF) was amplified by PCR assay in 72 lowpassage isolates [65 con-genitally infective children and 7 healthy children who were HCMV-DNA positive by quantitative PCR (qPCR)]. All positive PCR products were analyzed by heteroduplex mobility assay and single-stranded conformation polymorphism (HMA-SSCP) and 32 of them were sequenced. Results Fifty-five patient isolates and five healthy children isolates were HCMV-UL144 positive by PCR. Sequencing and HMA-SSCP analysis showed that significant strain-specific variability was present in the UL144 ORF. Phylogenetic analysis indicated that the nucleotide sequences could be separated into 3 major genotypes. Comparing between UL144 se-quences and the corresponding symptoms showed that genotype 2 did not exist in megacolon isolates. And genotype 1 and 3 were the major types among microcephaly and jaundice isolates respectively. Conclusions HCMV-UL144 existed in most of low passage isolates and sequences were hypervariable. The UL144 ORF and its predicted product with the high level of sequence variability in different kinds of isolates suggest that UL144 ORF might play a role in HCMV infectivity and subsequent diseases. 展开更多

Over the past few decades,an increasing number of marine activities have been conducted in the East China Sea,including the construction of various marine structures and the passage of large ships.Marine safety issues are paramount and are becoming more important with respect to the likely increase in size of ocean waves in relation to global climate change and associated typhoons.In addition,swells also can be very dangerous because they induce the resonance of floating structures,including ships.This study focuses on an investigation of swells in the East China Sea and uses hindcast data for waves over the past 5 years in a numerical model,WAVEWATCH III(WW3),together with historical climate data.The numerical calculation domain covers the entire North West Pacific.Next,swells are separated and analyzed using simulated wave fields,and both the characteristics and generation mechanisms of swells are investigated. 展开更多

The effects of selected DNA repair inhibitors on the frequency of human cytomegalovirus (HCMV)-induced chromosome aberrations in human peripheral blood lymphocytes (PBLs) were evaluated. Trealment of HCMV-infected PBLs with camptothecin (0.05 to 0.3μg/ml), an inhibitor of topoisomerase I, for 30 hr resulted in a significant (P<0.01) synergistic enhancement of the frequency of HCMV-induced chromosome damage, on the other hand, a significant increase in the frequency of chromosome damage was not noted for infected PBLs treated with either 3-aminobenzamide (3-AB) (3 to 30μg/ml), an inhibitor of poly (ADP-ribose) poiymerase, ur novobiocin 3 to 30 |ig/ml) an inhibitor of topoiso-merase I or excision repair processes for 30 hr. chromatid-type breaks including chromosome exchanges were the predominant type of chromosome aberrations observed in the HCMV-infected cells treated with camptothecin suggesting that HCMV infection is associated with the induction of single-strand DNA breaks. Furthermore, these findings suggest that HCMV infection does rol inflict dircc: DNA damage which is repaired through 3-AB- or novobiocinsensitive pathways.Human cytomegalovirus (HCMV) is a common pathogen which irferfs about 80% of the world's population causing, for the most part, persistent subcliniclil infeclions (Wellcr, 1971). A relatively small percentage of otherwise healthy immunologically competent people experience clinical HCMV disease (Cohen et al., 1985). Generalized HCMV infection, however, is the bane of individuals whose immune system is compromised (Schooley, 1990) Rubin, 1990). Molecular epidemiological studies strongly suggest that HCMV is one of the most frequent cause of congenital infections and that these infections result in a high incidence of birth defects and developmental abnormalities (Alford et al., 1990). Several studies (Nachtigal et al., 1978; Luleci et al., 1980; AbuBakar et al., 1988) have shown that HCMV infectior can result in an increase in the frequency of chromosome aberrations. Since the ability to cause chromosome damage may be significant in the induction of birth defects and possibly in HCMV-induced malignancy, we undertook the present investigation to evaluate the mechanisms by which HCMV may cause chromosome damage. In ths study, the effects of selected DNA repair inhibitors on the frequency of HCMV-induced chromosome aberrations were evaluated. The results indicae that the presence of camptothecin, but nut 3-aminobenzamidc (3-AB) ro novobiocin, results in a concentration-dependent increase in the frequent) of chromosome aberrations in HCMV-infected peripheral blood lymphocytes (PBLs). To evaluate the therapeutic effects of bone marrow-derived CD11b<sup>+</sup>CD14<sup>+</sup> monocytes in a murine model of chronic liver damage.METHODSChronic liver damage was induced in C57BL/6 mice by administration of carbon tetrachloride and ethanol for 6 mo. Bone marrow-derived monocytes isolated by immunomagnetic separation were used for therapy. The cell transplantation effects were evaluated by morphometry, biochemical assessment, immunohistochemistry and enzyme-linked immunosorbent assay.RESULTSCD11b<sup>+</sup>CD14<sup>+</sup> monocyte therapy significantly reduced liver fibrosis and increased hepatic glutathione levels. Levels of pro-inflammatory cytokines, including tumor necrosis factor-α, interleukin (IL)-6 and IL-1β, in addition to pro-fibrotic factors, such as IL-13, transforming growth factor-β1 and tissue inhibitor of metalloproteinase-1 also decreased, while IL-10 and matrix metalloproteinase-9 increased in the monocyte-treated group. CD11b<sup>+</sup>CD14<sup>+</sup> monocyte transplantation caused significant changes in the hepatic expression of α-smooth muscle actin and osteopontin.CONCLUSIONMonocyte therapy is capable of bringing about improvement of liver fibrosis by reducing oxidative stress and inflammation, as well as increasing anti-fibrogenic factors. 展开更多

Correlation of megafauna extinctions and mega-biosphere disturbances with past supernova explosions has been accomplished by considering a time correction for supernova debris traveling at 88.2325 percent of light speed. Supernova W44 is responsible for the Piora Oscillation which appears to be the biblical event of Noah's Flood. The closest supernova explosion, Vela Jr at 652 light-years, gives the beginning of the greatest historical human disaster, The Black Death. When supernova debris energy input occurs in the northern or southern hemisphere, it causes heating (global warming) in the northern or southern hemisphere, respectively. Long term cooling, the Little Ice Age, occurs in the northern hemisphere when the incoming debris of exploding stars impacts only the southern hemisphere for hundreds of years. Termination of the last ice age results due to melting of numerous supernova impacts that correlate time of impact by changing sea level and geothermal energy released for 2,800 years from the exit crater of Dr. J. Kennet's nano-diamond meteor theory and part of the process involves Dr. O'Keefe's tektite theory. Correlation of Dr Frezzotti's ice melt Antarctica data with supernova impact times over the past 800 years establishes the Global Warming model in conjunction with the November 2016 Antarctic sea ice melt. 展开更多

The Yuncheng salt lake has formed under the setting of stepped subsidence of fault-blocks from the north to the south in Yuncheng Basin. In the phase of red clay accumulation during 7.1–3.6 Ma, the size of palaeo-lake was larger than the present salt lake, and palaeo-monsoon had formed. At 3.6 Ma, the northern basement in the basin raised abruptly due to the radiative effect of Qinghai-Tibet Plateau uplifting, and palaeo-lake was contracting southwards. At ca. 2.6 Ma ancient river flowed into the northern part of the basin. During ca. 2.0–1.9 Ma aerolian effect strengthened and loess started to accumulate on the most part of the basin. Since ca. 1.8–1.0 Ma the subsidence of the lake fault-block has been speeding up abruptly. As under the natural hydrogradient the salt lake received enough groundwater supply, and the rate of loess accumulation in the lake area was lower than that of subsidence of the lake fault-block, the lake could be preserved and becomes the only modern lake on Chinese Loess Plateau. Four large strengthening change records of the monsoon were found in the lake sequence of 5.8–1.9 Ma B.P. 展开更多

Perovskite manganites show exotic functionalities due to the coupling between spin, charge, orbital and lattice, such as metal-insulator transition, colossal magnetoresistance ( CMR ) , charge-orbital order and phase separation. Recently, an extraordinary anisotropic magnetoresistance ( AMR ) has been observed in perovskite manganite single crystals. The AMR value is about 2 orders larger than that of the conventional 3E transition metals and alloys, which is attributed to tunable metal-insulator transition temperature modulated by the magnetic field. This result provides a new route for exploring novel AMR materials and their applications. 展开更多
