Methods Lymphocyte phenotypes were detected using indirect immunofluorescencetechnique. The proliferation responses were determined by MTT method. Flow cytometry and automaticimage analysis were performed to evaluate the apoptosis of lymphocytes. Results The proliferationresponses of lymphocytes in the elderly were lower than that in young adults. Decreased CD_(45) RA^+cells and increased CD_(45) RO^+ cells were found in lymphocyte population of aged people, comparedwith that of young adults. The CD_(45) RO^+ cells were prone to apoptosis. There is an inhibitoryeffect of PE on apoptosis of lymphocytes in the elderly. Conclusion It is implied that thesusceptibility of lymphocyte in the elderly to apoptosis depends on activation, so called,activation-induced cell death. Present results suggest that apoptosis of lymphocytes in aged peopleplay an important role in the pathogenesis of immunosenescence. METHODS:We conducted a study on 100 cases of gastric cancer (GC),100 cases of chronic gastritis (CG),and 150 controls (C).Deletion of the GSTT1 and GSTM1 genes was assessed by multiplex PCR.CYP2E1/Pst1 genotyping was performed using a PCR-RFLP assay. RESULTS:No relationship between GSTT1/GSTM1 deletion and the c1/c2 genotype of CYP2E1 was observed among the three groups.However,a significant difference between CG and C was observed,due to a greater number of GSTT1/GSTM1 positive genotypes in the CG group.The GSTT1 null genotype occurred more frequently in Negroid subjects,and the GSTM1 null genotype in Caucasians,while the GSTM1 positive genotype was observed mainly in individuals with chronic gastritis infected with H pylori. CONCLUSION:Our findings indicate that there is no obvious relationship between the GSTT1,GSTM1 and CYP2E1 polymorphisms and gastric cancer.展开更多

It is strongly associat...Plasmablastic lymphoma (PBL) is a rare form of diffuse large B-cell lymphoma characterized by weak/absent expression of conventional B-cell markers and strong expression of plasma cell markers. It is strongly associated with human immunodeficiency virus (HIV) and Epstein Barr virus infection, and shows an unusual tropism to the oral cavity. Herein we describe a patient with AIDS who presented with weight loss and dysphagia owing to a large gastroesophageal mass. His radiographic and endoscopic findings and long history of cigarette consumption suggested carcinoma. Biopsy demonstrated a poorly differentiated tumor stained negatively to routine lymphoid markers including CD20. However, gene rearrangement studies confirmed a B-cell process and a more detailed immunohistochemical analysis revealed the cells stained positively for CD138 (plasma cell antigen). These findings were diagnostic of PBL. Our report reviews the wide differential diagnosis of PBL and underscores the importance of a broad array of viral and molecular studies needed to establish this diagnosis.展开更多
AIM: TO investigate toll-like receptor 2 (TLR2) -196 to -274 del, and TLR4 (+896A/G rs4986790 and +1196C/ T rs4986791) polymorphisms at risk of chronic gastritis and gastric cancer in a Brazilian population and...AIM: TO investigate toll-like receptor 2 (TLR2) -196 to -274 del, and TLR4 (+896A/G rs4986790 and +1196C/ T rs4986791) polymorphisms at risk of chronic gastritis and gastric cancer in a Brazilian population and associ-ation of gastric lesions with risk factors such as smoking, alcohol intake and Helicobacter pylori infection.METHODS: In this casecontrol study, polymorphism at TLR2 -96 to -174 del was investigated by using the allele-specific polymerase chain reaction (PCR) method, while the PCR-restriction fragment length polymorphism technique was carried out to identify the TLR4 (rs4986790 and rs4986791) genotypes in 607 Brazilian individuals (208 with chronic gastritis-CG, 174 with gastric cancer-GC and 225 controls -C).RESULTS: The single nucleotide polymorphisms TLR4+1196ClT was not associated with risk of chronic gastritis or gastric cancer and the homozygous genotypes TLR4+896GG and TLR4+1196TF were absent in the studied population. However, the frequency of TLR2 -196 to -174 ins/del + del/del and TLR4+896AGgenotypes was significantly higher (P 〈 0.01 and P = 0.01, respectively) in the cancer group (33.4% and 11.5%, respectively) than in the control group (16.9% and 4.5%, respectively). It was also observed that the G-C haplotype of the TLR4+896A/G+1196C/T (P = 0.02) and the combination of variant alleles of the TLR21TLR4+896G (P = 0.02) are associated with susceptibility to gastric cancer. In addition, the multiple logistic regression showed that male gender [odds ratio (OR) = 2.70; 95% CI: 1.66-4.41; P 〈 0.01], alcohol intake (OR = 2.93; 95% CI: 1.76-4.87, P 〈 0.01), TLR2 -196 to -174 del (OR = 2.64; 95% CI: 1.56-4.44; P 〈 0.01) and TLR4+896G (OR = 3.19; 95% CI: 1.34- 7.61; P 〈 0.01) polymorphisms were associated with a higher susceptibility to developing this neoplasm.CONCLUSION: Our data indicate that T/R2 -196 to -174 del and TLR4+896G may increase the risk of gastric cancer in a Brazilian population.展开更多

Based on the long historical association and the provocative findings of blood pressure effects at low level of lead exposure a study was carried out to determine if an association existed between low blood lead concentration and hypertension. In this study the effects of low-level exposure to lead on blood pressure were examined among 244 adults using atomic absorption spectrometer. For quality assurance purpose certified reference materials i.e., Animal blood A-13, Bovine liver 1577 and cotton cellulose V-9 from IAEA (International Atomic Energy Agency) and NIST (National Institute of Standard Technology) were analyzed under identical experimental conditions. The mean age of hypertensive adults was 52 years (range 43-66). The mean values of systolic and diastolic blood pressure were (209±11.7) (range 170-250) and (117±3.9) (range 105-140) mmHg respectively. Blood lead concentration ranged from 78-201 μg/L with a mean of 139 μg/L and 165-497 μg/L with a mean of 255 μg/k in normal and hypertensive adults respectively. Increase in systolic blood pressure was significantly predictive with increase in blood lead levels. Body mass index (BMI) and lipid profile including total cholesterol, low density lipoprotein cholesterol, high density lipoprotein cholesterol and triglyceride correlated with blood pressure.展开更多

Highe...There is an increased awareness that celiac disease may occur in the elderly although presentations with either diarrhea, weight loss or both may be less common causing delays in diagnosis for prolonged periods. Higher detection rates also seem evident owing to active case screening, largely through serodiagnostic measures. In some elderly patients who are genetically predisposed, it has been hypothesized that celiac disease might be precipitated late in life by an antigen, possibly from an infectious agent. As a result, peptide mimicry or other poorly-defined mechanisms may precipitate an autoimmune gluten-dependent clinical state. Although diarrhea and weight loss occur, only isolated iron deficiency anemia may be present at the time of initial diagnosis. In addition, the risk of other autoimmune disorders, particularly autoimmune thyroiditis, and bone disease, are increased. Osteopenia may also be associated with an increased risk of fractures. Finally, elderly celiacs have an increased risk of malignant intestinal disease, especially lymphoma.展开更多

Production ofmonoclonal antibodies by the traditional hybridoma technology is costly, and the resulting murine antibodies often have the disadvantage of triggering human anti-mouse antibody (HAMA) response. Therefore recombinant Fab antibodies generated by the phage display technology can be a suitable alternative in managing B cell lymphoma. In this study, we extracted total RNA from spleen cells of BALB/c mice immunized with human B lymphoma cells, and used RT-PCR to amplify cDNAs coding for the κ light chains and Fd fragments of heavy chains. After appropriate restriction digests, these cDNA fragments were successively inserted into the phagemid vector pComb3H-SS to construct an immunized Fab phage display library. The diversity of the constructed library was approximately 1.94× 10^7. Following five rounds of biopanning, soluble Fab antibodies were produced from positive clones identified by ELISA. From eight positive clones, FabC06, FabC21, FabC43 and FabC59 were selected for sequence analysis. At the level of amino acid sequences, the variable heavy domains (VH) and variable light domains (VL) were found to share 88-92% and 89-94% homology with sequences coded by the corresponding murine germline genes respectively. Furthermore, reactivity with membrane proteins of the B cell lymphoma was demonstrated by immunohistochemistry and western blotting. These immunized Fab antibodies may provide a valuable tool for further study of B cell lymphoma and could also contribute to the improvement of disease therapy.展开更多

An investigation into the current status of the game was conducted to determine the extent to which it is still played. Although results indicate that the game is still being played in the rural areas, it also shows that it is steeply decreasing in popularity. About 6.6% of respondents interviewed have never heard of the game, while about 77% have played, or still play the game. Overall 17% of all respondents, or 22% of those who have ever played, still play the game. About 26% of respondents are optimistic about the game surviving their generation. The decline in popularity is attributable to decreasing proportion of youths in rural areas due to rural-urban migration, emergence of computer games, and scarcity of the seeds. Cultivation of D. reflexa plant for the game, food and medicinal purposes is recommended.展开更多

In the present study, we investigated the effect of colostrum and mature human milk on E. histolytica parasites in vitro.METHODS:Samples of human milk were collected from 5 healthy lactating mothers.The medium with human milk at concentrations of 2%, 5% and 10% was obtained.RESULTS:The lethal effect of E. histolytica on the medium supplemented with different concentrations of both colostrum and mature human milk was significant during the first 30min. We also detected that the results of colostrum and mature human milk were similar. No statistically significant differences were found between same concentrations of colostrum and mature human milk at the same times.CONCLUSION:Colostrum and mature human milk have significant lethal effect on E. histolytica and protect against its infection in breast fed children.展开更多
This research aims to analyse the contents of Fundo Clamor, located at the Documentation and Scientific Information Centre--CEDIC, from the Pontifical Catholic University of S~_o Paulo--SP, between the years of 1970 a...This research aims to analyse the contents of Fundo Clamor, located at the Documentation and Scientific Information Centre--CEDIC, from the Pontifical Catholic University of S~_o Paulo--SP, between the years of 1970 and 1992, as well as its contribution to the process of rescuing historical memory of child and teenager abduction, imprisonment, and torture in the course of military dictatorships in Brazil, Argentina, Uruguay, and Paraguay. The methodology used in this descriptive and qualitative research intends to take two directions: a bibliographical and a descriptive analytic documental research. Through the consultation and analysis of documents from Fundo Clamor (correspondence, denunciations, bulletins, and leaflets from Abuelas de la Plaza de Mayo, reports, writs of habeas corpus, pieces of reportage, and files from clandestine detention centres and on enforced disappearances), a compilation will be made with information regarding the disappearance of children and the imprisonment and/or abduction of pregnant militants in an attempt to describe the situation of apprehension and incarceration, the historical context, and the repression forces involved in the operation of arrest, kidnapping and/or torture of militants, children, and teenagers. Besides this heritage, a documental research will be made at Terror Archives, the Centre for Documentation and Archives for the Defence of Human Rights (CDyA) of Paraguay Supreme Justice Court, which contains a register of Alfredo Stroessner's thirty-five-year-long military dictatorship in Paraguay. The main argument in this research states that the archives from the Committee for the Defence of Refugees Human Rights from the Southern Cone, available at Fundo Clamor, contribute significantly to the rescue of historical memory from the dictatorial period and to the achievement of complete citizenship in these countries, considering that Brazil was the protagonist of the National Security Doctrine implantation process in South America.展开更多

In the present study, we investigated the effect of colostrum and mature human milk on E. histolytica parasites in vitro.METHODS:Samples of human milk were collected from 5 healthy lactating mothers.The medium with human milk at concentrations of 2%, 5% and 10% was obtained.RESULTS:The lethal effect of E. histolytica on the medium supplemented with different concentrations of both colostrum and mature human milk was significant during the first 30min. We also detected that the results of colostrum and mature human milk were similar. No statistically significant differences were found between same concentrations of colostrum and mature human milk at the same times.CONCLUSION:Colostrum and mature human milk have significant lethal effect on E. histolytica and protect against its infection in breast fed children.
文摘This research aims to analyse the contents of Fundo Clamor, located at the Documentation and Scientific Information Centre--CEDIC, from the Pontifical Catholic University of S~_o Paulo--SP, between the years of 1970 and 1992, as well as its contribution to the process of rescuing historical memory of child and teenager abduction, imprisonment, and torture in the course of military dictatorships in Brazil, Argentina, Uruguay, and Paraguay. The methodology used in this descriptive and qualitative research intends to take two directions: a bibliographical and a descriptive analytic documental research. Through the consultation and analysis of documents from Fundo Clamor (correspondence, denunciations, bulletins, and leaflets from Abuelas de la Plaza de Mayo, reports, writs of habeas corpus, pieces of reportage, and files from clandestine detention centres and on enforced disappearances), a compilation will be made with information regarding the disappearance of children and the imprisonment and/or abduction of pregnant militants in an attempt to describe the situation of apprehension and incarceration, the historical context, and the repression forces involved in the operation of arrest, kidnapping and/or torture of militants, children, and teenagers. Besides this heritage, a documental research will be made at Terror Archives, the Centre for Documentation and Archives for the Defence of Human Rights (CDyA) of Paraguay Supreme Justice Court, which contains a register of Alfredo Stroessner's thirty-five-year-long military dictatorship in Paraguay. The main argument in this research states that the archives from the Committee for the Defence of Refugees Human Rights from the Southern Cone, available at Fundo Clamor, contribute significantly to the rescue of historical memory from the dictatorial period and to the achievement of complete citizenship in these countries, considering that Brazil was the protagonist of the National Security Doctrine implantation process in South America.
