To study the genetic effects of inhibin a on apoptosis and proliferation of goose granulosa cells cultured in vitro, two RNA interference (RNAi) expression vectors, psiRNA-INHal and psiRNA-INHα2, were constructed to knock down inhibin α gene expression. After 48 h of transfection, the efficiency of these two RNAi expression vectors was examined by fluorescence microscopy. Meanwhile, inhibin protein expression levels, apoptosis indexes (AI) and proliferation indexes (PI) of granulosa cells were analyzed by flow cytometry. In addition, the supernatants were collected to assay the concentrations of estrogen (E2) and progesterone (P) by radioimmunoassay. The results showed that the expression level of inhibin a in the RNAi group were decreased 30%--40% than those in the control groups (P 〈0.05) and the apoptosis indexes and proliferation indexes in the RNAi groups were significantly higher than those in the control groups (P 〈0.05). However, the E2 concentrations in the RNAi groups were lower than those in the control groups (P 〈0.05). These results indicate that inhibin a has antagonistic effect on granulosa cell apoptosis.展开更多

Liu community may be affected by wind-sand disturbance. On the basis of field survey and experiments that took eight years from 1994, it was found that the plant community was formed in wet and low-lying land, developed in sand burying land, and died out in the land with strong wind erosion and severe sand burying. In the initial stage of the community formation, the low-lying land that was formed by wind erosion provided a suitable habitat for seed germination and plant rooting, but salt stress from ground water reduced seedling survival rate. In the developing stage of the community, sand burying stimulated vigorous growth of the shrubs. In the degenerative stage of the community, the wind erosion and sand burying resulted in dying out of the shrubs. It is therefore concluded that wind-sand disturbance has a positive role in the formation and development of the community, and is in a close association with the process of the community degeneration. Taking the habitat of T taklamakanensis into account, it may be appropriate to note that, under the conditions of frequent disturbance and severe stress, desert pioneer species are adaptable and able to survive. This might be a supplement to the CSR strategy.展开更多

Nowadays, more and more EDCs were released into the environment. EDCs go into water body via atmosphere sedi-mentation, surface runoff, soil eluviation, etc., so water body becomes the main place for existing. In order to attract scientific and public attention worldwide and to prevent EDCs pol ution, in this study we reviewed the classification of EDCs and their concentrations in natural water bodies, drinking water sources and water plants, and the reproductive toxicity of EDCs to fish were reviewed. EDCs could disturb the endocrine system and make reproductive organs and reproduction abnor-mal, resulting in fertility descending, reproduction function damage, community quan-tity decrease and even species extinction. In addition, EDCs could disrupt the homeostasis maintained by hormones, which would result in defects of neural de-velopment and abnormalities of the endocrine and reproductive systems. The exact molecular mechanisms have not been completely reported, but researches have suggested that multiple mechanisms were involved in the action of EDCs. Although there have been researches on the biohazard of EDCs, there stil exist problems of weakness in fundamental researches, difficulties in recognizing and identifying EDCs and high cost, which restraint the knowledge on them.展开更多

[Method]HEp-2/VSV system was used to test the antiviral activity of three batches of rPoIFN-α. Using recombinant human IFN-α as reference, the titer of interferon was measured. The semi-finished product of rPoIFN-α with the known titer were treated with 0.25% trypsin,HCl and mouse anti-porcine IFN-α monoclonal antibody. And the anti-viral activity of each batch of rPoIFN-α was detected. The physicochemical characters of rPoIFN-α were evaluated. The inhibition of induced cytopathic effect of rPoIFN-α on PPV (Porcine parvovirus) and PRV (Porcine pseudorabies) on swine kidney cell (PK-15) was determined. And the antiviral activity of rPoIFN-α in vitro was observed. [Result]The titers of semi-finished products of rPoIFN-α titrated by HEp-2/VSV system could reach 1.5×105 IU/ml,with the specific activity of 1.1×106 IU/mg. The residual rate of the tier of rPoIFN-α treated by 0.25% trypsin at 37 ℃ for 1 h was less than 1%. And that treated with HCl (pH of 2.0) for 72 h was up to 95%. And that treated at 56 ℃ for 30 min and that treated by mouse anti-porcine IFN-α monoclonal antibody at 37 ℃ for 1 h were higher than 47% and about 1% respectively. The antiviral test in vitro showed that 50 and 500 IU/ml rPoIFN-α could inhibit the induced cytopathic effect of PRV and PPV on PK-15 cell lines. [Conclusion]rPoIFN-α had the basic physicochemical characters of IFN-α. And it could inhibit the induced cytopathic effect of PRV and PPV on PK-15 cell lines, but there was dosage difference.展开更多

First, the impedance magnitude is achieved through an exciting probe and a detecting probe, or through calculations based on insertion loss measurement results when inserting a series nigh-value known impedance or a shunt low-value known impedance in the circuit. Then the impedance phase is extracted by the Hilbert transform (HT) of the logarithm of the obtained impedance magnitude. Performance studies show that the estimated phase error can increase greatly at a zero frequency in the Hilbert transform because of the existence of a singular point, and this effect can be eliminated by introducing a zero-point when the noise source does not include a series-connected capacitive component. It is also found that when the frequency is nigher than 150 kHz, the estimated phase error is not sensitive to the inductive source but sensitive to the capacitive source. Finally, under the conditions of the same measurement accuracies for impedance magnitude, the accuracy of complex impedance based on the HT can be improved about 10 times when compared with the accuracy of estimated parameters based on the impedance magnitude fitting method (IMFM).展开更多

Methods A sinusoidal FM fuze was analyzed in time domain and frequency domain and the concept of channel leak was presented. Results It was proved that information channel leak exists in FM fuze because of the nonlinear property of the mixer. The jamming signal was designed based on the channel leak and the jamming mechanism was analyzed in detail. Conclusion This kind of jamming signal can jam the sinusoidal FM fuzes effectively just depending on the jamming signal's feature itself. It's different from the traditional jamming way of simulating echo. Though the sinusoidal FM fuze was just analyzed, the principle is applicable to all FM fuzes. At the same time, it may be used as the reference for FM radar and communication countermeasures.展开更多

We analyze the potential of the proposed method for suppressing the airwave, and compare the proposed method with traditional methods in their effectiveness. A method to select synthetic source length is derived and the effect of the water depth on RAC is examined via numerical simulations. The results suggest that 3DD interferometry method with a synthetic source can effectively suppress the airwave and enhance the potential of marine CSEM to hydrocarbon exploration.展开更多
