[Objective] The aim of this study was to investigate the dry matter accumulation,moisture content in maize kernel and their influences on mechanical harvesting.[Methods] Using Zhengdan 958,Xianyu 335,Hongda 8 and Liyu 16 as experimental materials,we preliminarily measured the dynamic changes of dry matter accumulation and moisture content with the days after pollination lasted,and investigated the effect of moisture content in kernels on mechanical harvesting of Zhengdan 958 and Liyu 16.[Results] During summer growing season,Zhengdan 958 and Liyu 16 became physiologically mature and suitable for harvesting at about 51 days after pollination,Xianyu 335 and Hongda 8 required 58 days to become physiologically mature and suitable for harvesting after pollination.At physiologically mature stage,the moisture contents in kernels of Zhengdan 958,Xianyu 335,Hongda 8 and Liyu 16 were respectively 33.74%,28.86%,32.05% and 35.24%,respectively.Adopting mechanical harvesting at physiologically mature stage,the kernel loss rate of Zhengdan 958 and Liyu 16 was consistent with the range of mechanical operation index(kernel loss rate of 0.84%≤2%),while the percentage of broken kernels did not accord with the range of mechanical operation index(kernel crashing rate of 10.12%≧1%).The results indicate that Zhengdan 958 and Liyu 16 are not suitable for mechanical harvesting using full feeding combine havester.[Conclusion] This study provided theoretical basis for improving the maize harvest index under delayed harvesting. 展开更多

Objective The aim was to explore callus induction and plant regeneration of perennial ryegrass, as well as provide the foundation for transgenic research on perennial ryegrass.[ Methed] Mature seeds of perennial ryegrass were used as explants to study the effects of different hormone compositions on callus induction, proliferation and plant differentiation. Result The result showed that the induction rate achieved its highest on 2,4-D of 8 mg/L combining with 6-BA of 0.025 mg/L, which was up to 56.42%. Callus were differentiated after two to three generations, the highest differentiation rate 34.14% was achieved in the medium contained MS medium with 6-BA of 2 mg/L, and the differentiation rate was obviously affected by the callus condition after proliferation. The root inducing medium, containing 0.5 mg/L NAA and MS medium with half of macroelement, gained 98% root inducing rate. Conclusien A high frequency genetic regeneration system was established. 展开更多

In this study we studied the factors influencing the callus induction from mature embryos of maize inbred lines Qi 319, Zhen 58, Chang 7 -2, Lx 9801 and 81162, such as genotype, combination of plant growth regulators, and low-temperature pretreatment. The results showed that the induction rate of Qi 319 was the highest among the four genotypes tested; combination of 4.0 mg/L 2,4-D + 0.5 mg/L 6-BA was suitable for inducing callus from mature embryos; three days of 4℃ pretreatment can promote the callus induction significantly. The indices optimized in the present study are helpful for establishing genetic transformation system in maize without considering seasonal variation. 展开更多

[ Method] 2,4-D and 6-BA were set at different concentrations in callus induction and differentiation mediums to study their effects on callus induction, seedling formation and regenerated seedlings rooting. [ Result] In the callus induction medium treated with 0.5 mg/L 2,4-D, the callus induction effects on the varieties like Jiayu 948, Yanghui 559, Yangxian 6547, Zhong'erruanzhan, Minghui 86, Guanghui 998 and Zunxian 3 were the best; If 0.2 mg/L 6-BA was added into the callus induction medium containing the optimum level of 2,4-D, there was no obvious effect on induction rate of callus, but the differentiation and seedling of callus were inhibited; If the concentration of 6-BA was reduced appropriately in the differentiation medium, the seedling rate of callus would be not only no decreased but increased, meanwhile the quality of regenerated plants would be improved. To counter the defect of traditional genetic algorithms, an improved adaptivegenetic algorithm with the criterion of premature convergence is provided. The occurrence ofpremature convergence is forecasted using colony entropy and colony variance. When prematureconvergence occurs, new individuals are generated in proper scale randomly based on superiorindividuals in the colony. We use these new individuals to replace some individuals in the oldcolony. The updated individuals account for 30 percent - 40 percent of all individuals and the sizeof scale is related to the distribution of the extreme value of the target function. Simulationtests show that there is much improvement in the speed of convergence and the probability of globalconvergence. 展开更多

[Objective] This study aimed to identify the difference in squalene content among different olive varieties and the law of squalene content change in the same olive variety of different degrees of maturity, with the objective to provide technical support for the harvest and processing of olive fruits. [Method] Taking 13 introduced olive varieties including three varieties of eight different grades of maturity, fat acid w3s first extracted by Soxhlet extraction and then squalene was quantitatively and qualitatively analyzed by gas chromatography-mass spectrometry (GC-MS) method. [Result] The results showed that squalene content was the highest in CG-32, and the lowest in Leccino among the 13 olive varieties. And squalene content increased with the degree of maturity in the three olive varieties including CG-32, Frantoio and Ascolana Tenera. [Conclusion] A more simple and rapid method for the determina- tion of squatene content was established. 展开更多
