《流感》:戏剧化灾难叙事中的人性救赎

菊蓝抗流感片治疗新型冠状病毒感染的药效物质及作用机制整合研究

目的通过网络药理学和现代药理基础分析菊蓝抗流感片(JLKLGT)治疗新型冠状病毒感染(COVID-19)的药效物质及作用机制。方法采用网络药理学和分子对接技术,从TCMSP、BATMAN-TCM数据库检索菊蓝抗流感片各药物的活性成分及其靶点信息,使用Ge... 目的通过网络药理学和现代药理基础分析菊蓝抗流感片(JLKLGT)治疗新型冠状病毒感染(COVID-19)的药效物质及作用机制。方法采用网络药理学和分子对接技术,从TCMSP、BATMAN-TCM数据库检索菊蓝抗流感片各药物的活性成分及其靶点信息,使用GeneCards、OMIM和DrugBank数据库检索COVID-19疾病靶点。采用Cytoscape 3.7.2软件构建“菊蓝抗流感片药物-活性成分-靶点”网络图,通过STRING在线平台构建蛋白互作网络。通过Metascape数据库进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路分析,推测其药效物质及作用机制。结果共筛选菊蓝抗流感片中52种活性成分,对应靶点468个,COVID-19疾病靶点1410个和交集靶点107个。推测槲皮素、阿司匹林、木犀草素、金合欢素、胆固醇、豆甾醇和胆酸为治疗COVID-19主要药效物质,TP53、RELA、AKT1、TNF、MAPK1、NFKB1和MAPK14为关键核心靶点。分子对接结果表明,主要药效物质与关键核心靶点、血管紧张素转化酶2(ACE2)结合构象稳定。结论菊蓝抗流感片通过多成分、多靶点、多通路发挥防治COVID-19的作用。 展开更多

Five monoclonal antibodies(Mabs) to nuclear protein of avain influenza virus(AIV) were developed by syncretizing SP 2/0 and the spleen cells from BALB of mice immuized with H9 subtype AIV. Specificity of these Mabs were identified by immunofluorescent assay(IFA) and enzyme linked immunosorbent assay (ELISA). These five Mabs which were named as AIV-NP-2C3, AIV-NP-6A5, AIV-NP-3 H9, AIV-NP-7B4, AIV-NP-2H4 could react with all viruses of AIV-H9 strains in tests. The result of Western blotting showed that only the 60 ku protein antigen of AIV-H9 could be recognized by the Mabs but never recognized by New castle disease virus, REV and infectious bursa disease virus. The result of preliminary application showed that avian influenza viruses could be deetected bv Mabs in IFA and ELISA. All these Mabs will probably play important roles in preventing and monitoring avian influenza viruses. 展开更多

[Result] The multiplex RT-PCR amplification, agarose gel electrophoresis and sequencing results showed that the self-designed primers are successful for detecting AIV. [Method] 2009 influenza A virus was introduced from the aspects of classification and host, virology, molecular characteristics and vaccine. [Result] A novel influenza A/H1N1 virus emerged in early April 2009 quickly spread worldwide through human-to-human transmission. The virus contained a group of novel gene segments, the nearest known precursor was the virus found in swine. The virus appeared to retain the potential to infect swine again and thus continued reassort with swine viruses. All registered 2009 influenza A vaccines were tested for safety and immunogenicity in clinical trials on human volunteers, and all vaccines were found to be safe, single dose of vaccine could cause protective antibody responses. [Objective] The study aimed to investigate the genetic variation characters of entire sequences between two H9N2 subtype avian influenza virus strains and other reference strains.[Method] The entire sequences of 8 genes were obtained by using RT-PCR,and these sequences were analyzed with that of six H9N2 subtype avian influenza isolates in homology comparison and genetic evolution relation.[Result] The results showed that the nucleotide sequence of entire gene of the strain shared 91.1%-95.4% homology with other seven reference strains,and PG08 shared the highest homology 91.3% with C/BJ/1/94;ZD06 shared the highest homology 92.3% with D/HK/Y280/97.HA cleavage sites of two H9N2 subtype avian influenza virus isolated strains were PARSSR/GLF,typical of mildly pathogenic avian influenza virus.[Conclusion] Phylogenetic tree for entire gene of eight strains showed that the genetic relationship was the closest between ZD06 and C/Pak/2/99 strains,which belonged to the Eurasian lineage;PG08 shared the highest homology 91.3% with ZD06,it may be the product of gene rearrangements of other sub-lines. 展开更多

[Objective] The aim of the present study was to develop a direct immunofluorescence method for the diagnosis of canine distemper (CD) with FITC-conjugated monoclonal antibodies (FITC-McAb).[ Metbod] The McAb against CDV, designated as CE3, was purified with protein G and labeled with FITC through agitation method. After purification and identification, the optimal working concentration of FITC-labeled CE3 was determined. Then 61 clinical samples of suspected canine distemper were detected by direct immunofluorescence assay. [ Result] The absorption test, blocking test and specificity test showed that the labeled antibody had high specificity and sensitivity, but didn't have cross reaction with canine parvovirus (CPV), canine parainfluenza virus (CPIV), canine adenovirus (CAV) and rabies virus (RV). The optimal working concentration was 1:80. The positive rate of clinical suspected samples was 48%. [ Conclusion] The direct immunofluorescence assay developed in this study was rapid, specific and convenient, and had great significance for the early diagnosis of canine distemper. 展开更多

Sequence Comparison and Analysis of HA Gene of Four H9N2 Avian Influenza Virus Isolates

[ Objective] To determine the HA gene sequences of four H9N2 Avian influenza virus (AIV) strains and carry out comparative analysis so as to understand the difference and variation pattern of each strain from the angle of molecular biology and to know the distribution and epidemic law of H9N2 AIV. [Method] One pair of primers was designed referring to HA gene sequences of H9N2 AIV. The HA genes of A/Chicken/Hebei/WD/98 (H9N2; WD98 for short), A/Chicken/Hebei/ZD/04 (H9N2; ZD04 for short)), A/Chicken/Beijing/MY/06 (H9N2; MY06 for short) ), and A/Chicken/Beijing/PG/08 (H9N2; PG08 for short)) were amplified, cloned and sequenced. Then the HA gene sequences of these strains were compared with that of 10 H9N2 AIV stains in GenBank. [Result] The ORF of HA genes of the four strains was 1 683 bp in size, encoding 516 amino acids. The HA gene sequences of the four strains, WD98, MY06, PG08, and ZD04, were 82.6% -95.1%, 83.0% -99.0%, 82.7% -95.5%, and 81.3% -95.7% homologous to that of the 10 H9N2 AIV stains, respectively. And the homology of amino acid was respectively 86.6% -96.3%, 86.6% -97.9%, 87.0% -97.1%, and 86.9% -97.3%. [ Conclusion] The HA gene has greatly high homology among different strains. 展开更多

Objective Cervical spinal cord stimulation (SCS) has been found to augment cerebral blood flow (CBF) in a number of animal models. However, the effective use of SCS is hampered by a lack of understanding of its mechanism(s) of action. In this paper, we focus on the sympathetic and parasympathetic effects of SCS on CBF. Method SpragueDawley rats were selected for the experimental series. The animals were divided into 5 groups to underwent SCS and laser Doppler flowmeter (LDF) recordings. Control group, the animal underwent SCS and LDF recordings without any surgery of the nerve fibers and ganglia. V 1 group, the animal underwent bilateral resection of the nasociliary and post-ganglionic parasympathetic nerve fibbers. SCG group, the animal underwent bilateral resection of supper cervical ganglion. V 1 + SCG group, the animal underwent both surgeries as V1- and SCG-group animals did. Sham group, the animal underwent the carotid manipulation with blunt-tipped forceps as well as the dissection of nasociliary and post-ganglionic parasympathetic nerve fibers around the ethmoidal foramen, but without cutting any nerves. Results During the SCS, the LDF was no statistical difference between the V 1 or SCG group and the control group. Yet, the effects of SCS on CBF are completely abolished in V1+ SCG group. Conclusions Surgical interruption of both the parasympathetic and sympathetic pathways has the contradict effect on SCS-induced CBF augmentation. 展开更多

Cloning and Phylogenetic Analysis of NS1 Genes from Different Isolates of H9N2 Subtype Duck Influenza Virus

[ Objective] The study aimed to lay a foundation for the further studies on function mechanism of NS1 protein in the interspecies transmission of waterfowl influenza virus. [Method] Using the serologic assay and the specific RT-PCR method, some strains of H9 subtype waterfowl influenza virus were isolated from the 12 to 20 day-old muscovy duck flocks without any clinical symptoms in different areas of Guangdong Province. Four of these strains, including A/duck/ZQ/303/2007(H9N2) (A3 for short), A/Duck/FJ/301/2007 (H9N2) (C1 for short), A/Duck/NH/306/2007(H9N2) ( D6 for short), A/duck/SS/402/2007(H9N2) ( E2 for short), and a strain named A/duck/ZC/2007(H9N2) (L1 for short) from a muscovy duck died of avian influenza virus (AIV), were used for NSl gene cloning and sequencing. Subsequently, the obtained NSl gene sequences were compared with other NS1 sequences registered in GenBank, and the phylogenetic analysis was also conducted. [Result] When compared with the H9N2 AIV NS1 sequences in GenBank, the NSl genes of the four AIV strains A3, C1, 136 and E2 displayed homologies ranging from 99% to 100% at nucleotide level, and 95% to 100% at amino acid level; while the NSl gene of L1 strain displayed homology ranging from 94% to 97% at nucleotide level, and 93% to 98% at amino acid level. The phylogenetic tree demonstrated that A3, C1, D6 and E2 were highly resemblant, and L1 was closest to AY66473 (chicken, 2003). By comparison with the NS1 gene sequences of L1, AF523514 (duck), AY664743 (chicken) and EF155262.1 (quail) using DNAstar, A3, C1, D6 and E.2 presented nucleotide variations at site 21 ( R→Q), 70, 71 ( KE→EG), 86 ( A→S), 124 (V→M) and 225 ( S→N), and amino acid variations at site 21,70, 71 and 86 in dsRNA- dependent protein kinase (PKR) binding domain of NSl gene, which induced the evident variations of antigenic determinant and surface proba- bility plot of NS1 protein. [ Conclusion] This study suggested that the amino acid sequence variation in PKR binding domain of NS1 protein had something to do with the virus pathogenicity. 展开更多

A channel assignment algorithm with awareness of link traffic is proposed in multi-radio multi-channel wireless mesh networks. First, the physical interference model based on the signal-to-interference-plus-noise ratio and successful transmission condition is described. The model is more suitable for a wireless communication environment than other existing models. Secondly, a pure integer quadratic programming (PIQP) model is used to solve the channel assignment problem and improve the capacity of wireless mesh networks. Consequently, a traffic- aware static channel assignment algorithm(TASC) is designed. The algorithm adopts some network parameters, including the network connectivity, the limitation of the number of radios and the successful transmission conditions in wireless communications. The TASC algorithm can diminish network interference and increase the efficiency of channel assignment while keeping the connectivity of the network. Finally, the feasibility and effectivity of the channel assignment solution are illustrated by the simulation results. Compared witb similar algorithms, the proposed algorithm can increase the capacity of WMNs. 展开更多

To decrease the metal losses of RF spiral inductor,a novel layout structure with gradually reduced metal line width and space from outside to inside is presented. This gradual changed inductor has less eddy-current effect than the conventional inductor of fixed metal width and space. So the series resistance can be reduced and the quality (Q) factor of the inductor relating to metal losses is increased. The obtained experimental results corroborate the validity of the proposed method. For a 6nH inductor on high-resistivity silicon at 2.46GHz,Q factor of 14.25 is 11.3% higher than the conventional inductor with the same layout size. This inductor can be integrated with radio frequency integrated circuits to gain better performance in RF front end of a wireless communication system. 展开更多

Oil reservoirs with low permeability and porosity that are in the middle and late exploitation periods in China's onshore oil fields are mostly in the high-water-cut production stage.This stage is associated with severely non-uniform local-velocity flow profiles and dispersed-phase concentration(of oil droplets) in oil-water two-phase flow,which makes it difficult to measure water holdup in oil wells.In this study,we use an ultrasonic method based on a transmission-type sensor in oil-water two-phase flow to measure water holdup in lowvelocity and high water-cut conditions.First,we optimize the excitation frequency of the ultrasonic sensor by calculating the sensitivity of the ultrasonic field using the finite element method for multiphysics coupling.Then we calculate the change trend of sound pressure level attenuation ratio with the increase in oil holdup to verify the feasibility of the employed diameter for the ultrasonic sensor.Based on the results,we then investigate the effects of oildroplet diameter and distribution on the ultrasonic field.To further understand the measurement characteristics of the ultrasonic sensor,we perform a flow loop test on vertical upward oilwater two-phase flow and measure the responses of the optimized ultrasonic sensor.The results show that the ultrasonic sensor yields poor resolution for a dispersed oil slug in water flow(D OS/W flow),but the resolution is favorable for dispersed oil in water flow(D O/W flow) and very fine dispersed oil in water flow(VFD O/W flow).This research demonstrates the potential application of a pulsed-transmission ultrasonic method for measuring the fraction of individual components in oil-water two-phase flow with a low mixture velocity and high water cut. 展开更多

To solve the excessive huge scale problem of the traditional multi-bit digital artificial neural network(ANN) hardware implementation methods,a bit-stream ANN hardware implementation method based on sigma delta(ΣΔ) modulation is presented.The bit-stream adder,multiplier,threshold function unit and fully digital ΣΔ modulator are implemented in a field programmable gate array(FPGA),and these bit-stream arithmetical units are employed to build the bit-stream artificial neuron.The function of the bit-stream artificial neuron is verified through the realization of the logic function and a linear classifier.The bit-stream perceptron based on the bit-stream artificial neuron with the pre-processed structure is proved to have the ability of nonlinear classification.The FPGA resource utilization of the bit-stream artificial neuron shows that the bit-stream ANN hardware implementation method can significantly reduce the demand of the ANN hardware resources. 展开更多
