Among the 16 kinds of FAAs, Ala, Arg, Glu and Asp, four kinds of flavor amino acids, showed the highest contents, and their total content ranged from 36.05 to 53.96 mg/g, accounting for 69.11%-86.47% of the total FAA content. The total contents of flavor amino acids and FAAs all reached the peaks in all the test ma- terials, except the control cultivar, at middle harvest period. The essential amino acid content was trended to be increased in all the test materials with the extended harvest period, and it reached the peak at the late harvest period. Sutong showed the highest flavor amino acid content and FAAs content, followed by Y-H002, Suyan and Sulian, while the control cultivar showed the lowest contents.展开更多

The uptake of K^+ reduced in the presence of both salts whereas uptake of Ca^2+ retarded mainly by Na2SO4. Chlorophyll content was affected mainly by NaCl, while Na2SO4 treatment lowered the rate of photosynthetic activity. Both salt compounds caused accumulation of free amino acids including free proline but reduced the protein content in the leaves. The salt tolerance capacity of the broad beans plant in which the observations of the effect of salinity determined by different concentrations of NaCI and Na2SO4 on growth, chloroplast pigments, photosynthetic, free amino acids, free proline, protein and inorganic elements on plants are combined with observations on growth and yield, in order to arrive at a better understanding of their salt tolerance, has been studied and discussed in this paper. S...The objective of this work was to investigate the effect of six individual strains of fungi on the reduction of gossypol levels and nutritional value during solid substrate fen'aentation of cottonseed meal (CSM). Six groups of disinfected CSM substrate were incubated for 48 h after inoculation with either of the fungi C. capsuligena ZD- 1, C. tropicalis ZD-3, S. cerevisae ZD-5, A. terricola ZD-6, A. oryzae ZD-7, or A. niger ZD-8. One not inoculated group (substrate) was used as a control. Levels of initial and final free gossypol (FG), crude protein (CP), amino acids (AA) and in vitro digestibility were assayed. The experiment was done in triplicate. The experimental results indicated that microbial fermentation could greatly decrease (P〈0.05) FG levels in CSM. The detoxification efficiency differed between the species of microorganisms applied. From the perspective of reducing CSM potential toxicity, C. tropicalis ZD-3 was most successful followed by S. cerevisae ZD-5 and A. niger ZD-8. They could reduce FG levels of CSM to 29.8, 63.07 and 81.50 mg/kg based on DM (dry matter), respectively, and their detoxification rates were 94.57%, 88.51% and 85.16%, respectively. If crude protein, amino acids content and their in vitro digestibility were also taken into account, Aniger ZD-8 may be the best choice. The CP content of CSM substrate fermented by C. tropicalis ZD-3 and A. niger ZD-8 were improved by 10.76% and 22.24%; the TAA (total amino acids) contents were increased by 7.06% and 11.46%, and the EAA (essential amino acids) were raised by 7.77% and 12.64%, respectively. Especially, the levels of methionine, lysine and threonine were improved greatly (P〈0.05). The in vitro CP digestibility of CSM fermented by C. tropicalis ZD-3 and A. niger ZD-8 was improved by 13.42% and 18.22%, the TAA were increased by 17.75% and 22.88%, and the EAA by 16.61% and 21.01%, respectively. In addition, the in vitro digestibility of methionine, lysine and threonine was also improved greatly (P〈0.05).展开更多

[Method] The phys...[Objective] This study aimed to screen out the methods and indices that could quickly and accurately detect the maturity of materials through evaluating the mature status of rapeseed meal composting. [Method] The physicochemical proper- ties variation of rapeseed meal composting was analyzed by testing emission of carbon dioxide, degradation of organic matter, conductivity values, coefficient of seed germination and free amino acids during the fermentation process. [Result] In the process of fermentation, the release amount of carbon dioxide per unit time was drastically reduced at the beginning and then the change became smoothly later. The organic matter was degraded quickly and then slowed down, and the total amount of degradation was 7.34%, total degradation rated of 9.06%. The seed ger- mination coefficient showed a trend of gradual decline, and after the start of fermen- tation, the germination coefficient was below the critical coefficient of 0.8. The con- ductivity value and the free amino acid content changed significantly, which showed strong regularity, and the increase amount was close to 1 times. [Conclusion] The detection of conductivity value and free amino acid content in rapeseed meal com- posting can be used as the methods and indices for the quick and accurate detec- tion of the maturity of organic fertilizer fermentation.展开更多

In various types of cells, saturated free fatty acids (FFAs), such as palmitate, have been shown to induce cellular apoptosis by several mechanisms. Palmitate down-regulates the activity of PKR and thereby decreases the level of Bcl-2 protein, mediated in part by reduced activation of the NF-KB transcription factor. In addition to the level of Bel-2 protein, the phosphorylation of Bcl-2 at different amino acid residues, such as Ser70 and Ser87, is also important in regulating cellular apoptosis. The decrease in the phosphorylatiou of Bcl-2 at Ser70 upon exposure to palmitate is mediated by inhibition of PKR and possibly by c-Jun N-terminal kinase (JNK), whereas the phosphory- lation of Bcl-2 at Ser87 is unaffected by palmitate or PKR. In summary, PKR mediates the regulation of the protein level and the phosphorylation status of Bel-2, providing a novel mechanism of palmitate-induced apoptosis in HepG2 cells.展开更多

Mass spectroscopy was used to detect the products of sonolytic degradation of 2-chlorobiphenyl. The results show that the products of sonolytic degradation, such as biphenyl, ethyl benzene, diethylbiphenyl, dibutylbiphenyl, phenol, propylphenol and di-tert-butyl phenol are produced by thermolysis and hydroxyl free radical reactions, in which biphenyl counts for almost 40%(mole fraction) of the mother compound and others are at trace level. Rapid accumulation of chloride ion shows quick dechlorination, and 78% organic chlorine is converted into chloride ion. Free radical scavengers, bicarbonate and carbonate, decrease the reaction rate of sonolytic degradation of 2-chlorobiphenyl significantly, and the pseudo 1st order rate constant of sonolytic degradation of 2-chlorobiphenyl decreases linearly with the natural logarithm of the concentration of the added free radical scavenger, showing that the pyrolysis and hydroxyl free radical reaction are the two major pathways for the sonolytic degradation of 2-chlorobiphenyl, in which the hydroxyl radical concentration is estimated to be 1×10-10 (mol/L.)展开更多

Juvenile tongue sole were directly transferred from a salinity of 30 to 0, 10, 20, ...A serial of salinity transferring treatments were performed to investigate the osmoregulation of tongue sole(Cynoglossus semilaevis). Juvenile tongue sole were directly transferred from a salinity of 30 to 0, 10, 20, 30, 40 and 50. Blood sampling was performed for each treatment after 0, 1, 6 and 12 h, as well as after 1, 2, 4, 8, 16 and 32 d. The plasma osmolality, cortisol and free amino acids were assessed. Under the experimental conditions, no fish died after acute salinity transfer. The plasma cortisol level increased 1 h after the abrupt transfer from a salinity of 30 to that of 0, 40 and 50, and decreased from 6 h to 8 d after transfer. Similar trends were observed in the changes of plasma osmolality. The plasma free amino acids concentration showed a ‘U-shaped' relationship with salinity after being transferred to different salinities for 4 days. More obvious changes of plasma free amino acid concentration occurred under hyper-osmotic conditions than under hypo-osmotic conditions. The concentrations of valine, isoleucine, lysine, glutamic acid, glycine, proline and taurine increased with rising salinity. The plasma levels of threonine, leucine, arginine, serine, and alanine showed a ‘U-shaped' relationship with salinity. The results of this study suggested that free amino acids might have important effects on osmotic acclimation in tongue sole.展开更多
Surface seawater samples were collected from Jiaozhou Bay,China,during six cruises(MarchMay 2010,September-November 2010) to study the distribution of dissolved organic matter including dissolved organic carbon(DOC),t...Surface seawater samples were collected from Jiaozhou Bay,China,during six cruises(MarchMay 2010,September-November 2010) to study the distribution of dissolved organic matter including dissolved organic carbon(DOC),total dissolved carbohydrates,namely monosaccharides(MCHO) and polysaccharides(PCHO) and total hydrolysable amino acids.These included dissolved free amino acids(DFAA) and combined amino acids(DCAA).The goal was to investigate possible relationships between these dissolved organic compounds and environmental parameters.During spring,the concentrations of MCHO and PCHO were 9.6(2.8-22.6) and 11.0(2.9-42.5) μmol C/L,respectively.In autumn,MCHO and PCHO were 9.1(2.6-27.0) and 10.8(2.4-25.6) μmol C/L,respectively.The spring concentrations of DFAA and DCAA were 1.7(1.1-4.1) and 7.6(1.1-31.0) μmol C/L,respectively,while in autumn,DFAA and DCAA were 2.3(1.1-8.0) and 3.3(0.6-7.2) μmol C/L,respectively.Among these compounds,the concentrations of PCHO were the highest,accounting for nearly a quarter of the DOC,followed by MCHO,DCAA and DFAA.The concentrations of the organic compounds exhibited a decreasing trend from the coastal to the central regions of the bay.A negative correlation between concentrations of DOC and salinity in each cruise suggested that riverine inputs around the bay have an important impact on the distribution of DOC in the surface water.A significant positive correlation was found between DOC and total bacteria count in spring and autumn,suggesting bacteria play an important role in the marine carbon cycle.展开更多

[Method]Chinese kale was grown in hydroponic under three different Light Emitting Diode(LEDs) conditions [red∶blue=8∶1(8R1B), red∶blue=6∶3(6R3B) and red∶green∶blue=6∶2∶1(6R2G1B), 12 h light, 50 μmol/(m2·s)]. Then its growth and quality indices including root and shoot fresh weight and dry weight, concentrations of soluble protein, vitamin C, nitrate, soluble phenols, flavonoids, soluble sugar, free amino acids, and activity of nitrate reductase were measured. [Result] There was no significant difference in plant height, diameter of flower stalk and leaf number among three LED treatments. The fresh weight of shoot, root and plant in 8R1 B and 6R2G1 B was significantly higher than in 6R3 B. The dry weight of shoot and plant in 8R1 B was significantly higher than in 6R3 B. The concentrations of vitamin C, soluble protein and soluble sugar in flower stalk of 6R3 B treatment were significantly higher than those in 8R1 B and 6R2G1 B, while there was no remarkable difference in concentrations of reducing sugar, soluble phenol, flavonoids and free amino acid among the three treatments. The nitrate concentration in flower stalk of 6R3 B was significantly lower than in the other two treatments, and the activity of nitrate reductase in6R3 B was significantly higher than in the other two treatments. [Conclusion] The LED treatment of red:blue=6:3 was more suitable for the growth of Chinese kale.展开更多

Despite of the fact, only a little information is available about the free amino acid contents in foods. To utilize information of free amino acids in food, we have carried out the experiments to quantitate the free amino acids by derivatized with NBD-F (4-fluoro-7-nitrobenzofurazan) and analyzed on reversed-phase UHPLC (ultra-high pressure liquid chromatography) equipped with ultraviolet visible detector. Almost all of food extracts contained free amino acids including GABA (T-amino butyrate). Contents of free amino acids vary considerably depending upon vegetables and fruits. Principal free amino acids found in vegetables and fruits were asparagine, glutamine, arginine and GABA, which are involved in important metabolic pathways in human. About 140 species of vegetables and fruits were subjected for the data base. All of the plants and fruits we examined exhibit significant amount of free amino acids, those are clearly distinct from data bases obtained after acid hydrolysis treated food samples. Since glutamate and GABA act as excitatory and inhibitory neurotransmitters in CNS, respectively; free amino acids in vegetables and fruits that we eat daily, should be an important source for the cellular metabolic activities.展开更多
[Objective] This study to aimed to investigate the effects of Cu stress on root exudates and microbial activities in rhizosphere of grafted and ungrafted cucum-ber seedlings, and therefore to elucidate the microbial m...[Objective] This study to aimed to investigate the effects of Cu stress on root exudates and microbial activities in rhizosphere of grafted and ungrafted cucum-ber seedlings, and therefore to elucidate the microbial mechanism of grafting for in-creasing cucumber plants tolerance to Cu stress [Method] Four treatments: (1) un-grafted seedlings + test soil (U0); (2) ungrafted seedlings + test soil + CuSO4·5H2O (U1); (3) grafted seedlings + test soil (G0); (4) grafted seedlings + test soil + Cu-SO4·5H2O (G1) were set in the pot culture experiment. The contents of free amino acids, organic acids, phenolic acid and sugars, microbial population and enzyme ac-tivity in the four treatment were measured, respectively. [Result] The secretion of amino acids and organic acids were increased under Cu stress. The amino acids secretions of grafted seedlings roots were obviously higher than ungrafted seedlings except for Phe and Val. At the same time, the secretion of oxalic acid, malic acid, acetic acid, citric acid, cinnamic acid, ρ-hydroxybenzoic acid and benzoic acid of grafted seedlings were significantly higher than ungrafted seedlings as wel . There-fore, more Cu2+ were restricted in soil by chelating, complexing and precipitation with root exudates, and its toxicity was decreased. The soil microbial biomass C and N in grafted cucumber rhizosphere were significantly higher than those in ungrafted cu-cumber rhizosphere, whereas basal respiration and metabolic quotient were signifi-cantly lower. Under Cu stress, the numbers of actinomyces and nitrogen fixing bac-teria decreased and the number of fungi increased significantly, whereas there was no significant difference in amounts of bacteria. The numbers of bacteria, actino-myces, and nitrogen fixing bacteria in grafted cucumber rhizosphere were significant-ly higher than those in ungrafted cucumber rhizosphere, but the number of fungi was opposite. The activities of soil urease, phosphatase, sucrase and catalase in grafted cucumber rhizosphere were significantly higher than those in ungrafted cu-cumber rhizosphere. [Conclusion] These indicated that the soil microbial environment and soil enzymes activities were improved by grafting under Cu stress, and as a re-sult, the adaptability of cucumber to Cu stress was improved.展开更多

The paper deals with application proposition of protein concentrates, free AA (amino acids), HMB (hydroxy-methyle-butyrate), creatine and camitine. If the athlete is involved in strength and power sport (e.g. throwing events in track and field, olympic weightlifting etc.), the explosive and maximum strength is of primary importance. For strength athletes, these mentioned substances are those legal preparates, supplements, which can help effectively in performance improvement of sport results. The main reason is that using protein concentrates, AAs, HMB, creatine and camitine, the anabolic (and anticatabolic) effect will enhance the protein biosynthesis of the organism, improve the aerob and anaerob capacity of the athletes, activate and stimulate the own hormonal system. And the final result is creating higher loadability because of the faster recovery, and higher performance level on the competition. Experiments were conducted at Entomology division of Directorate of Rice research to study the qualitative and quantitative changes in biochemical constituents in the rice plant treated with different xenobiotics, viz...Experiments were conducted at Entomology division of Directorate of Rice research to study the qualitative and quantitative changes in biochemical constituents in the rice plant treated with different xenobiotics, viz. insecticides like deltamethrin, imidacloprid, herbicide like 2,4-Dichlorophenoxyacetic acid, fungicide like carbendazim, green manure Calotropis gigantea leaves, botanicals like Neem Seed Powder and fertilizer urea along with control plants of TN1, PTB33 and Swama varieties comprising of total 15 treatments. Application of butachlor, carbendazim, deltamethrin and excessive nitrogen fertilization resulted in increased population build up of brown plant hopper (BPH). Significantly, lower honeydew excretion (13 mme area) in the BPH nymphs was observed on resistant PTB33 variety compared to susceptible TN1 and Swarna varieties (116 and 110 mm2 respectively). In susceptible TN1 variety, the sucrose content was significantly higher (9.35 mg/g tissue) compared to BPH resistant PTB33 (5.32 mg/g tissue). The BPH resistant PTB33 variety exhibited significantly lower free amino acid content (1.23 mg/g tissue) as compared to TN1 (2.51 mg/g). The C/N ratio was comparatively high in the resistant PTB33 alone (4.32), but was low in PTB33 applied with 2,4-D + carbendazim (2.73), 2,4-D + imidacloprid ~ carbendazim (2.3), excess urea fertilizer (2.18). The total phenol contents measured in resistant PTB33 (7.5 mg/g dry wt), susceptible TN1 (5.865 mg/g dry wt) and the popular variety Swarna (5.57 mg/g dry wt) were almost same and no statistical difference was observed. Further, treatment of these varieties with different xenobiotics and botanicals also did not significantly alter their total phenol content. However, the phenol content of the resistant PTB 33 alone and treated with xenobiotics was higher compared to the susceptible TN 1 and Swarna varieties alone and treated with xenobiotics. The plant biochemical constituents such as free amino acids, sucrose were lower in untreated resistant PTB33 compared to susceptible TN1, and PTB33 treated with urea and deltamethrin. The high feeding rate and population build up on the susceptible variety TN1 is positively correlated with high quantities of sucrose, free amino acids and lower quantities of phenols and low C/N ratio.展开更多
Objective To investigate the role of acyl-coenzyme A:cholesterol acyltransferase inhibitor(ACATI) in apoptosis induced by lipids and whether lipids-induced apoptosis is accompanied by increase of free cholesterol in e...Objective To investigate the role of acyl-coenzyme A:cholesterol acyltransferase inhibitor(ACATI) in apoptosis induced by lipids and whether lipids-induced apoptosis is accompanied by increase of free cholesterol in endoplasmic reticulum(ER),in order to further understand the mechanism of lipids-induced apoptosis in advanced atherosclerosis.Methods Human vascular smooth muscle cells(VSMCs) and phorbol 12-myristate 13-acetate(PMA) differentiated THP-1 macrophages were used.Tritiated thymidine incorporation was applied to detect cell proliferation.Cytotoxicity was assessed by lactate dehydrogenase(LDH) release.4',6-diamidino-2-phenylindole(DAPI) staining,caspase-3,-7 assay,and Annexin-V/propidium iodide(PI) staining were used to detect apoptosis.High performance liquid chromatography was used in intracellular free cholesterol and cholesterol ester assay.ER free cholesterol was quantified.Results Different lipids had different effects on proliferation and cytotoxicity of VSMCs.25-hydroxycholesterol(25OHC) had biphasic effects on the proliferation of VSMCs.At low concentration,it stimulated cell proliferation,but turned to proliferation inhibition as concentration reached 15 μg/mL.25OHC and acetylated low density lipoprotein(AcLDL) could respectively induce apoptosis in human VSMCs and PMA differentiated THP-1 macrophages,which was aggravated by ACATI,accompanied by increase of intracellular free cholesterol content.There was also an increase of cholesterol content in ER with AcLDL-induced apoptosis in THP-1 macrophages.Conclusions Lipids could induce apoptosis,accompanied by increase of intracellular free cholesterol content,which could be augmented by ACATI,suggesting that insults resulting in ER free cholesterol rise might be the initiator of apoptosis.展开更多

How- ever, the contents of free glycine (Gly) and proline (Pro) were very sensitive to grazing. Glycine content in grazing population was higher than that in non-grazing population at flowering stage; the content of free proline in non-grazing population was higher than that in grazing population at tillering, heading and especially flower- ing stage; the content of free proline under grazing pressure at maturity stage was higher than that of non-grazing population.
文摘Salinity treatments (NaCl and Na2SO4) reduced plant growth and increased the accumulation of Na^+, P^3+,Fe^3+ , Mn^2+, and CIin root, stem, leaf and legume. The uptake of K^+ reduced in the presence of both salts whereas uptake of Ca^2+ retarded mainly by Na2SO4. Chlorophyll content was affected mainly by NaCl, while Na2SO4 treatment lowered the rate of photosynthetic activity. Both salt compounds caused accumulation of free amino acids including free proline but reduced the protein content in the leaves. The salt tolerance capacity of the broad beans plant in which the observations of the effect of salinity determined by different concentrations of NaCI and Na2SO4 on growth, chloroplast pigments, photosynthetic, free amino acids, free proline, protein and inorganic elements on plants are combined with observations on growth and yield, in order to arrive at a better understanding of their salt tolerance, has been studied and discussed in this paper. The results for the latter only (fruiting stage) are presented.

文摘The objective of this work was to investigate the effect of six individual strains of fungi on the reduction of gossypol levels and nutritional value during solid substrate fen'aentation of cottonseed meal (CSM). Six groups of disinfected CSM substrate were incubated for 48 h after inoculation with either of the fungi C. capsuligena ZD- 1, C. tropicalis ZD-3, S. cerevisae ZD-5, A. terricola ZD-6, A. oryzae ZD-7, or A. niger ZD-8. One not inoculated group (substrate) was used as a control. Levels of initial and final free gossypol (FG), crude protein (CP), amino acids (AA) and in vitro digestibility were assayed. The experiment was done in triplicate. The experimental results indicated that microbial fermentation could greatly decrease (P〈0.05) FG levels in CSM. The detoxification efficiency differed between the species of microorganisms applied. From the perspective of reducing CSM potential toxicity, C. tropicalis ZD-3 was most successful followed by S. cerevisae ZD-5 and A. niger ZD-8. They could reduce FG levels of CSM to 29.8, 63.07 and 81.50 mg/kg based on DM (dry matter), respectively, and their detoxification rates were 94.57%, 88.51% and 85.16%, respectively. If crude protein, amino acids content and their in vitro digestibility were also taken into account, Aniger ZD-8 may be the best choice. The CP content of CSM substrate fermented by C. tropicalis ZD-3 and A. niger ZD-8 were improved by 10.76% and 22.24%; the TAA (total amino acids) contents were increased by 7.06% and 11.46%, and the EAA (essential amino acids) were raised by 7.77% and 12.64%, respectively. Especially, the levels of methionine, lysine and threonine were improved greatly (P〈0.05). The in vitro CP digestibility of CSM fermented by C. tropicalis ZD-3 and A. niger ZD-8 was improved by 13.42% and 18.22%, the TAA were increased by 17.75% and 22.88%, and the EAA by 16.61% and 21.01%, respectively. In addition, the in vitro digestibility of methionine, lysine and threonine was also improved greatly (P〈0.05).

文摘[Objective] This study aimed to screen out the methods and indices that could quickly and accurately detect the maturity of materials through evaluating the mature status of rapeseed meal composting. [Method] The physicochemical proper- ties variation of rapeseed meal composting was analyzed by testing emission of carbon dioxide, degradation of organic matter, conductivity values, coefficient of seed germination and free amino acids during the fermentation process. [Result] In the process of fermentation, the release amount of carbon dioxide per unit time was drastically reduced at the beginning and then the change became smoothly later. The organic matter was degraded quickly and then slowed down, and the total amount of degradation was 7.34%, total degradation rated of 9.06%. The seed ger- mination coefficient showed a trend of gradual decline, and after the start of fermen- tation, the germination coefficient was below the critical coefficient of 0.8. The con- ductivity value and the free amino acid content changed significantly, which showed strong regularity, and the increase amount was close to 1 times. [Conclusion] The detection of conductivity value and free amino acid content in rapeseed meal com- posting can be used as the methods and indices for the quick and accurate detec- tion of the maturity of organic fertilizer fermentation.
文摘The present study shows that double-stranded RNA-dependent protein kinase (PKR) regulates the protein expres- sion level and phosphorylation of Bcl-2 and plays an anti-apoptotic role in human hepatocellular carcinoma ceils (HepG2). In various types of cells, saturated free fatty acids (FFAs), such as palmitate, have been shown to induce cellular apoptosis by several mechanisms. Palmitate down-regulates the activity of PKR and thereby decreases the level of Bcl-2 protein, mediated in part by reduced activation of the NF-KB transcription factor. In addition to the level of Bel-2 protein, the phosphorylation of Bcl-2 at different amino acid residues, such as Ser70 and Ser87, is also important in regulating cellular apoptosis. The decrease in the phosphorylatiou of Bcl-2 at Ser70 upon exposure to palmitate is mediated by inhibition of PKR and possibly by c-Jun N-terminal kinase (JNK), whereas the phosphory- lation of Bcl-2 at Ser87 is unaffected by palmitate or PKR. In summary, PKR mediates the regulation of the protein level and the phosphorylation status of Bel-2, providing a novel mechanism of palmitate-induced apoptosis in HepG2 cells.
文摘The sonolytic degradation of 2-chlorobiphenyl was investigated. Mass spectroscopy was used to detect the products of sonolytic degradation of 2-chlorobiphenyl. The results show that the products of sonolytic degradation, such as biphenyl, ethyl benzene, diethylbiphenyl, dibutylbiphenyl, phenol, propylphenol and di-tert-butyl phenol are produced by thermolysis and hydroxyl free radical reactions, in which biphenyl counts for almost 40%(mole fraction) of the mother compound and others are at trace level. Rapid accumulation of chloride ion shows quick dechlorination, and 78% organic chlorine is converted into chloride ion. Free radical scavengers, bicarbonate and carbonate, decrease the reaction rate of sonolytic degradation of 2-chlorobiphenyl significantly, and the pseudo 1st order rate constant of sonolytic degradation of 2-chlorobiphenyl decreases linearly with the natural logarithm of the concentration of the added free radical scavenger, showing that the pyrolysis and hydroxyl free radical reaction are the two major pathways for the sonolytic degradation of 2-chlorobiphenyl, in which the hydroxyl radical concentration is estimated to be 1×10-10 (mol/L.)
文摘By employing TCLs (thin cell layers) culture, the floral gradient in flowering tobacco of different developmental stages was confirmed. The TCLs from early flowering tobacco regenerated more floral buds than those from the tobacco plants in full blooming or fruiting stages. Analysis of free amino acid levels revealed the acropetal gradient of Pro in flowering tobacco stem. L-Pro. L-Trp. D,L-Met and L-Arg were respectively added into the culture medium for testing their influence on floral bud formation from tobacco pedicel segments. Only L-Trp evidently enhanced the floral bud neoformation.

文摘A serial of salinity transferring treatments were performed to investigate the osmoregulation of tongue sole(Cynoglossus semilaevis). Juvenile tongue sole were directly transferred from a salinity of 30 to 0, 10, 20, 30, 40 and 50. Blood sampling was performed for each treatment after 0, 1, 6 and 12 h, as well as after 1, 2, 4, 8, 16 and 32 d. The plasma osmolality, cortisol and free amino acids were assessed. Under the experimental conditions, no fish died after acute salinity transfer. The plasma cortisol level increased 1 h after the abrupt transfer from a salinity of 30 to that of 0, 40 and 50, and decreased from 6 h to 8 d after transfer. Similar trends were observed in the changes of plasma osmolality. The plasma free amino acids concentration showed a ‘U-shaped' relationship with salinity after being transferred to different salinities for 4 days. More obvious changes of plasma free amino acid concentration occurred under hyper-osmotic conditions than under hypo-osmotic conditions. The concentrations of valine, isoleucine, lysine, glutamic acid, glycine, proline and taurine increased with rising salinity. The plasma levels of threonine, leucine, arginine, serine, and alanine showed a ‘U-shaped' relationship with salinity. The results of this study suggested that free amino acids might have important effects on osmotic acclimation in tongue sole.

文摘Surface seawater samples were collected from Jiaozhou Bay,China,during six cruises(MarchMay 2010,September-November 2010) to study the distribution of dissolved organic matter including dissolved organic carbon(DOC),total dissolved carbohydrates,namely monosaccharides(MCHO) and polysaccharides(PCHO) and total hydrolysable amino acids.These included dissolved free amino acids(DFAA) and combined amino acids(DCAA).The goal was to investigate possible relationships between these dissolved organic compounds and environmental parameters.During spring,the concentrations of MCHO and PCHO were 9.6(2.8-22.6) and 11.0(2.9-42.5) μmol C/L,respectively.In autumn,MCHO and PCHO were 9.1(2.6-27.0) and 10.8(2.4-25.6) μmol C/L,respectively.The spring concentrations of DFAA and DCAA were 1.7(1.1-4.1) and 7.6(1.1-31.0) μmol C/L,respectively,while in autumn,DFAA and DCAA were 2.3(1.1-8.0) and 3.3(0.6-7.2) μmol C/L,respectively.Among these compounds,the concentrations of PCHO were the highest,accounting for nearly a quarter of the DOC,followed by MCHO,DCAA and DFAA.The concentrations of the organic compounds exhibited a decreasing trend from the coastal to the central regions of the bay.A negative correlation between concentrations of DOC and salinity in each cruise suggested that riverine inputs around the bay have an important impact on the distribution of DOC in the surface water.A significant positive correlation was found between DOC and total bacteria count in spring and autumn,suggesting bacteria play an important role in the marine carbon cycle.

文摘[Objective] This study was conducted to investigate the effects of light quality on growth and quality of Chinese kale(Brassica alboglabra Bailey). [Method]Chinese kale was grown in hydroponic under three different Light Emitting Diode(LEDs) conditions [red∶blue=8∶1(8R1B), red∶blue=6∶3(6R3B) and red∶green∶blue=6∶2∶1(6R2G1B), 12 h light, 50 μmol/(m2·s)]. Then its growth and quality indices including root and shoot fresh weight and dry weight, concentrations of soluble protein, vitamin C, nitrate, soluble phenols, flavonoids, soluble sugar, free amino acids, and activity of nitrate reductase were measured. [Result] There was no significant difference in plant height, diameter of flower stalk and leaf number among three LED treatments. The fresh weight of shoot, root and plant in 8R1 B and 6R2G1 B was significantly higher than in 6R3 B. The dry weight of shoot and plant in 8R1 B was significantly higher than in 6R3 B. The concentrations of vitamin C, soluble protein and soluble sugar in flower stalk of 6R3 B treatment were significantly higher than those in 8R1 B and 6R2G1 B, while there was no remarkable difference in concentrations of reducing sugar, soluble phenol, flavonoids and free amino acid among the three treatments. The nitrate concentration in flower stalk of 6R3 B was significantly lower than in the other two treatments, and the activity of nitrate reductase in6R3 B was significantly higher than in the other two treatments. [Conclusion] The LED treatment of red:blue=6:3 was more suitable for the growth of Chinese kale.
文摘Significant amounts of free amino acids exist in commercially sold vegetables and fruits. Despite of the fact, only a little information is available about the free amino acid contents in foods. To utilize information of free amino acids in food, we have carried out the experiments to quantitate the free amino acids by derivatized with NBD-F (4-fluoro-7-nitrobenzofurazan) and analyzed on reversed-phase UHPLC (ultra-high pressure liquid chromatography) equipped with ultraviolet visible detector. Almost all of food extracts contained free amino acids including GABA (T-amino butyrate). Contents of free amino acids vary considerably depending upon vegetables and fruits. Principal free amino acids found in vegetables and fruits were asparagine, glutamine, arginine and GABA, which are involved in important metabolic pathways in human. About 140 species of vegetables and fruits were subjected for the data base. All of the plants and fruits we examined exhibit significant amount of free amino acids, those are clearly distinct from data bases obtained after acid hydrolysis treated food samples. Since glutamate and GABA act as excitatory and inhibitory neurotransmitters in CNS, respectively; free amino acids in vegetables and fruits that we eat daily, should be an important source for the cellular metabolic activities.

文摘[Objective] This study to aimed to investigate the effects of Cu stress on root exudates and microbial activities in rhizosphere of grafted and ungrafted cucum-ber seedlings, and therefore to elucidate the microbial mechanism of grafting for in-creasing cucumber plants tolerance to Cu stress [Method] Four treatments: (1) un-grafted seedlings + test soil (U0); (2) ungrafted seedlings + test soil + CuSO4·5H2O (U1); (3) grafted seedlings + test soil (G0); (4) grafted seedlings + test soil + Cu-SO4·5H2O (G1) were set in the pot culture experiment. The contents of free amino acids, organic acids, phenolic acid and sugars, microbial population and enzyme ac-tivity in the four treatment were measured, respectively. [Result] The secretion of amino acids and organic acids were increased under Cu stress. The amino acids secretions of grafted seedlings roots were obviously higher than ungrafted seedlings except for Phe and Val. At the same time, the secretion of oxalic acid, malic acid, acetic acid, citric acid, cinnamic acid, ρ-hydroxybenzoic acid and benzoic acid of grafted seedlings were significantly higher than ungrafted seedlings as wel . There-fore, more Cu2+ were restricted in soil by chelating, complexing and precipitation with root exudates, and its toxicity was decreased. The soil microbial biomass C and N in grafted cucumber rhizosphere were significantly higher than those in ungrafted cu-cumber rhizosphere, whereas basal respiration and metabolic quotient were signifi-cantly lower. Under Cu stress, the numbers of actinomyces and nitrogen fixing bac-teria decreased and the number of fungi increased significantly, whereas there was no significant difference in amounts of bacteria. The numbers of bacteria, actino-myces, and nitrogen fixing bacteria in grafted cucumber rhizosphere were significant-ly higher than those in ungrafted cucumber rhizosphere, but the number of fungi was opposite. The activities of soil urease, phosphatase, sucrase and catalase in grafted cucumber rhizosphere were significantly higher than those in ungrafted cu-cumber rhizosphere. [Conclusion] These indicated that the soil microbial environment and soil enzymes activities were improved by grafting under Cu stress, and as a re-sult, the adaptability of cucumber to Cu stress was improved.
文摘Peformance enhancing drugs are widely used today in different sport fields, and therefore the successful anti-doping activity should be based not only on regular doping controls, but also it should show the real alternative: how to replace the forbidden doping substances with legal and effective supplements. The paper deals with application proposition of protein concentrates, free AA (amino acids), HMB (hydroxy-methyle-butyrate), creatine and camitine. If the athlete is involved in strength and power sport (e.g. throwing events in track and field, olympic weightlifting etc.), the explosive and maximum strength is of primary importance. For strength athletes, these mentioned substances are those legal preparates, supplements, which can help effectively in performance improvement of sport results. The main reason is that using protein concentrates, AAs, HMB, creatine and camitine, the anabolic (and anticatabolic) effect will enhance the protein biosynthesis of the organism, improve the aerob and anaerob capacity of the athletes, activate and stimulate the own hormonal system. And the final result is creating higher loadability because of the faster recovery, and higher performance level on the competition. Of course, the athletes--as a minimum requirement--need an adequate nutrition (good balanced nourishment), as well, with appropriate application and supplementation of all vitamins and essential minerals.
文摘Experiments were conducted at Entomology division of Directorate of Rice research to study the qualitative and quantitative changes in biochemical constituents in the rice plant treated with different xenobiotics, viz. insecticides like deltamethrin, imidacloprid, herbicide like 2,4-Dichlorophenoxyacetic acid, fungicide like carbendazim, green manure Calotropis gigantea leaves, botanicals like Neem Seed Powder and fertilizer urea along with control plants of TN1, PTB33 and Swama varieties comprising of total 15 treatments. Application of butachlor, carbendazim, deltamethrin and excessive nitrogen fertilization resulted in increased population build up of brown plant hopper (BPH). Significantly, lower honeydew excretion (13 mme area) in the BPH nymphs was observed on resistant PTB33 variety compared to susceptible TN1 and Swarna varieties (116 and 110 mm2 respectively). In susceptible TN1 variety, the sucrose content was significantly higher (9.35 mg/g tissue) compared to BPH resistant PTB33 (5.32 mg/g tissue). The BPH resistant PTB33 variety exhibited significantly lower free amino acid content (1.23 mg/g tissue) as compared to TN1 (2.51 mg/g). The C/N ratio was comparatively high in the resistant PTB33 alone (4.32), but was low in PTB33 applied with 2,4-D + carbendazim (2.73), 2,4-D + imidacloprid ~ carbendazim (2.3), excess urea fertilizer (2.18). The total phenol contents measured in resistant PTB33 (7.5 mg/g dry wt), susceptible TN1 (5.865 mg/g dry wt) and the popular variety Swarna (5.57 mg/g dry wt) were almost same and no statistical difference was observed. Further, treatment of these varieties with different xenobiotics and botanicals also did not significantly alter their total phenol content. However, the phenol content of the resistant PTB 33 alone and treated with xenobiotics was higher compared to the susceptible TN 1 and Swarna varieties alone and treated with xenobiotics. The plant biochemical constituents such as free amino acids, sucrose were lower in untreated resistant PTB33 compared to susceptible TN1, and PTB33 treated with urea and deltamethrin. The high feeding rate and population build up on the susceptible variety TN1 is positively correlated with high quantities of sucrose, free amino acids and lower quantities of phenols and low C/N ratio.

文摘Objective To investigate the role of acyl-coenzyme A:cholesterol acyltransferase inhibitor(ACATI) in apoptosis induced by lipids and whether lipids-induced apoptosis is accompanied by increase of free cholesterol in endoplasmic reticulum(ER),in order to further understand the mechanism of lipids-induced apoptosis in advanced atherosclerosis.Methods Human vascular smooth muscle cells(VSMCs) and phorbol 12-myristate 13-acetate(PMA) differentiated THP-1 macrophages were used.Tritiated thymidine incorporation was applied to detect cell proliferation.Cytotoxicity was assessed by lactate dehydrogenase(LDH) release.4',6-diamidino-2-phenylindole(DAPI) staining,caspase-3,-7 assay,and Annexin-V/propidium iodide(PI) staining were used to detect apoptosis.High performance liquid chromatography was used in intracellular free cholesterol and cholesterol ester assay.ER free cholesterol was quantified.Results Different lipids had different effects on proliferation and cytotoxicity of VSMCs.25-hydroxycholesterol(25OHC) had biphasic effects on the proliferation of VSMCs.At low concentration,it stimulated cell proliferation,but turned to proliferation inhibition as concentration reached 15 μg/mL.25OHC and acetylated low density lipoprotein(AcLDL) could respectively induce apoptosis in human VSMCs and PMA differentiated THP-1 macrophages,which was aggravated by ACATI,accompanied by increase of intracellular free cholesterol content.There was also an increase of cholesterol content in ER with AcLDL-induced apoptosis in THP-1 macrophages.Conclusions Lipids could induce apoptosis,accompanied by increase of intracellular free cholesterol content,which could be augmented by ACATI,suggesting that insults resulting in ER free cholesterol rise might be the initiator of apoptosis.
