This product was identified as p-hydroxy benzyl alcohol on the basis of its 1H, 13C NMR and EI-MS spectral data. It could be inferred that the enzyme responsible for the biotransforma-tion reaction was a kind of extracellular and constitutive enzyme since the transformation reaction of the substrate could be carried out in cell free extracts of the fermentation broth of the Mucor spinosus. 展开更多

Ultrastructural changes in the sieve element and dense cells of nectariferous tissue during the development of floral nectary in Arabidopsis thaliana L. were investigated with transmission electron microscopy. Samples were prepared with high pressure freezing and freeze substitution techniques. The ultrastructure of dense cell was similar to that of sieve element at its early developmental stage. With the concurrent agglutination of chromatin in the nucleus, the abnormal location of organelles and the high density of cytoplasm, the ultrastructural characteristics in die dense cells of the nectariferous tissue and in the sieve element are matched with those of the programmed cell death in animal and plant reported in recent years. The disorganization of nucleus and most organelles in the differentiation of sieve elements and dense cells is closely associated with the transportation and modification of pre-nectar and the transference of nectar. This suggests that the cytological changes in sieve element and nectariferous tissue are closely associated with the nectary functional activities. 展开更多

[Objective] The aim of this study was to investigate the efficient technique of artificial insemination for silkworm. [Method] Sperms were extracted from bursa copulatrix of female moths mated for 30 min through extruding and centrifugal method, and then the semen was injected into other virgin moths with trypsinase. [ Result] A high-effective collection technology of spermatids from silkworm was established successfully, 50 μl semen could be collected by only one person in each hour. The survival rate of spermatids was over 80% in vito after collected from bursa copulatrix, while the obtained semen was quite pure and the average fertilization rate of silkworm was 76,5%. [ Conclusion] The establishment of high-effective semen extraction technique of silkworm provides the technical basis for studies on other related techniques for silkworm sperm. 展开更多

Anthers of male sterile line and its maintainer line of Zidao type rice, Yingxiang A and Yingxiang B were studied by cytological methods, such as F-KI staining, silver nitrate staining and paraffin sections, It is found that in Yingxiang A, there are some abnormalities in meiosis, represented by unequal division at the meiotic phase; the abortion of CMS line arises at the meiotic phase and is almost completed at the phase of monomicrospore, but only a tiny amount of aborted mi- crospores are observed; the abortion is complete and thorough, and none normal functional pollens are found; the morphology of aborted pollens is similar to that of typically aborted pollens; the tapetal cells still exist in anthers from the microsporocyte stage to the tetrad stage, but they are abnormally hyperplastic. However, in Yingxiang B, there are no abnormalities in meiosis at the meiotic phase; the tapetal cells exist in anthers before the tetrad stage, but they disappear completely when the pollen are mature. It is indicated that the abortion of CMS Yingxiang line mainly occurs at the meiotic phase, and is basically completed at the phase of monomicrospore. The abortion is probably caused by the abnormally hyperplastic tapetal cells. The abnormalities in tapetal cells may affect the normal development of pollens, resulting in the emergence of abortion. In addition, the abortion in Yingxiang A is complete and thorough, which is of great significance in hybrid breeding. 展开更多

Objective: To block the apoptosis of T lymphocytes induced by Fas/FasL in order to establish a method of the large-scale preparation of large amounts of tumor-specific cytoxic T-lymphocytes (CTL). Methods: Liver cancer cells and tumor infiltrating lymphocytes (TIL) were isolated from FasL positive fresh specimens, and co-cultured. Specific CTL were activated and prepared in the presence of the co-stimulation of monoclonal antibody CD28. Then the blocking and activation of apoptosis of T lymphocytes was activated by soluble Fas receptor, which was detected by cytometry and DNA ladder test simultaneously. The apoptosis-blocking effect was compared with the control group. Furthermore, the changes of T cell proliferation and killing activity were detected by the method of ^3H thymidine incorporation and ^51Cr release test. Results: There was a significant increase in apoptosis rate in unblocking group compared with blocking group and quiescent group, with the unblocking group of 47.82%±0.13%, quiescent group of 3.76%±0.25%, and the blocking group of 8.22%±0.26% respectively (P〈0.01). T cell-ladder appeared in unblocking group by DNA ladder test. Both the killing ability and proliferation rate of T cells were significantly increased after blocking. There was significant difference among blocking group, unblocking group and quiescent group (P〈0.01). Conclusion: With this method we obtained large amounts of tumor-specific T lymphocytes, which was able to kill liver cancer cells effectively. 展开更多

At that time, Chinese scientists rarely published their work in non-Chinese journals, and China did not have a major English journal of its own in life sciences. In a way, Cell Research was born at the right time, in the right place, and with the right kind of people guiding its development. The rise of Cell Research as China's leading journal publishing peer-reviewed articles in basic life sciences has paralleled the rapid development of the Chinese economy in general and the science and technology sector in particular over the same period. The current economic trend should allow even more rapid advances in the life sciences which will ensure a healthy growth of Cell Research in years to come. In recent years, the importance of Coix has been realized with the exploration of its effects in treating and preventing cancer. The systematic study of Coix has met a superb development opportunity with the rapid development of biotechnology especially the deep-sequencing technique. Therefore, this paper presented here the current status of origin and evolution, classification, cytology and molecular biology progress of Coix, analyzed the ploidy of Coix and the genetic relationships with Zea, Sorghum and so on from the point of cytology, and then based on the molecular biology, analyzed the important significance of DNA molecular marker, genetic mapping and gene library in the research on identification of genetic relationship between species and genetic breeding, which could provide useful information for further Coix research. 展开更多
