Profiling the protein composition of bacteria is essential for understanding their biology,physiology and interaction with environment.Mass spectrometry has become a pivotal tool for protein analysis,facilitating the ...Profiling the protein composition of bacteria is essential for understanding their biology,physiology and interaction with environment.Mass spectrometry has become a pivotal tool for protein analysis,facilitating the examination of expression levels,molecular masses and structural modifications.In this study,we compared the performance of three widely-used mass spectrometry methods,i.e.,matrix-assisted laser desorption/ionization(MALDI)protein fingerprinting,top-down proteomics and bottom-up proteomics,in the profiling of bacterial protein composition.It was revealed that bottom-up proteomics provided the highest protein coverage and exhibited the greatest protein profile overlap between bacterial species.In contrast,MALDI protein fingerprinting demonstrated superior detection reproducibility and effectiveness in distinguishing between bacterial species.Although top-down proteomics identified fewer proteins than bottom-up approach,it complemented MALDI fingerprinting in the discovery of bacterial protein markers,both favoring abundant,stable,and hydrophilic bacterial ribosomal proteins.This study represents the most systematic and comprehensive comparison of mass spectrometry-based protein profiling methodologies to date.It provides valuable guidelines for the selection of appropriate profiling strategies for specific analytical purposes.This will facilitate studies across various fields,including infection diagnosis,antimicrobial resistance detection and pharmaceutical target discovery.展开更多
In this study,circular dichroism(CD)and molecular dynamics(MD)simulation were used to investigate the thermal unfolding pathway of staphylococcal enterotoxin B(SEB)at temperatures of 298–371 and 298–500 K,and the re...In this study,circular dichroism(CD)and molecular dynamics(MD)simulation were used to investigate the thermal unfolding pathway of staphylococcal enterotoxin B(SEB)at temperatures of 298–371 and 298–500 K,and the relationship between the experimental and simulation results were explored.Our computational findings on the secondary structure of SEB showed that at room temperature,the CD spectroscopic results were highly consistent with the MD results.Moreover,under heating conditions,the changing trends of helix,sheet and random coil obtained by CD spectral fitting were highly consistent with those obtained by MD.In order to gain a deeper understanding of the thermal stability mechanism of SEB,the MD trajectories were analyzed in terms of root mean square deviation(RMSD),secondary structure assignment(SSA),radius of gyration(R_(g)),free energy surfaces(FES),solvent-accessible surface area(SASA),hydrogen bonds and salt bridges.The results showed that at low heating temperature,domain Ⅰ without loops(omitting the mobile loop region)mainly relied on hydrophobic interaction to maintain its thermal stability,whereas the thermal stability of domain Ⅱ was mainly controlled by salt bridges and hydrogen bonds.Under high heating temperature conditions,the hydrophobic interactions in domain Ⅰ without loops were destroyed and the secondary structure was almost completely lost,while domain Ⅱ could still rely on salt bridges as molecular staples to barely maintain the stability of the secondary structure.These results help us to understand the thermodynamic and kinetic mechanisms that maintain the thermal stability of SEB at the molecular level,and provide a direction for establishing safer and more effective food sterilization processes.展开更多

[Method] Agar diffusion method was adopted to screen antimicrobial strains and determine the antimicrobial spectrum. Phylogenetic relationship of the strains was analyzed by neighbor-joining method of the Mega 4.0 software. [Result] Twenty antimicrobial strains were screened from seawater of Southern Ocean collected during the 27^th Chinese Antarctic Scientific Expedition. Molecular identification and phyloge- netic analysis indicated that two antimicrobial strains were members of Pseu- domonas, two strains were members of Psychrobacter, and the other 16 trains were members of Pseudoalteromonas. The antimicrobial spectrum of four strains which had higher antimicrobial activity indicated that the strains 312, 83-1 and 195 greatly inhibited the growth of Fusarium oxysporum, Rhizoctonia solani K(Jhn, Phytophthora capsici Leonian, Verticillium dahliae, Alternaria solani, Thanatephoru scucumeris and Phomopsis asparagi (Sacc); strain 312-1 had obvious antimicrobial effect on the six of the plant pathogens except R. solani. [Conclusion] Four strains which had higher antimicrobial effect were obtained and should be further studied for development and application.展开更多

The essential oil wa...To analyze the constituents of essential oil from Chinese eaglewood [resinous wood of Aquilaria sinensis (Lour.) Gilg] and its anti-methicillin-resistant Staphylococcus aureus (MRSA) activity. The essential oil was extracted by water-steam distillation and analyzed by GC/MS method. The relative contents of the compounds were determined by normalization. The compounds were characterized by NIST05 and WILEY275L database matching and comparison of their MS spectra with those of literature data. Antibacterial activity of the oil was assayed by the filter paper disc agar diffusion method. The oil showed significant antibacterial activity against MRSA. Sixty-six chromatographic peaks were detected, among them thirty compounds comprising 59.80% of the total essential oil were characterized. Twenty-six compounds comprising 54.26% of the oil were identified as sesquiterpenes. β-Agarofuran (8.96%), kusunol (7.82%), (-)-jinkoh-eremol (5.04%), agarospirol (4.53%), baimuxifuranic acid (4.09%) were the major sesquiterpenes. Four nor-sesquiterpenes and some other sesquiterpenes, such as 10-epi-γ-eudesmol, α-agarofuran, epi-ligulyl oxide, etc. were detected in Chinese eaglewood oil for the first time. This is the first report about anti-MRSA activity of Chinese eaglewood oil from A. sinensis.展开更多

The investigation result of seedlings before transplanting seedling showed that leaf age,the number of green leaves,the number of tiller of individual plant and the weight of per 100 fresh plants in microbial inoculum treatment group increased 0.11,0.12 ,0. 03 and 2.6 g respectively compared with those of control group. 20 days after transplanting, increased tiller number in microbial inoculum treatment group was 55 500 hm^2 more than that in control group. 30 days after transplanting, tiller number in microbial inoculum treatment group was 42 000 hm^2 more than that in control group. In harvest season, the yield ,weight of I 0130 grain and seed-setting rate increased 300 kg/hm2, 0. 7 g and 0. 23% respectively compared with these in control group. The result of preliminary study demonstrated that microbial inoculum confected by endophytes phomopsis B3 could increase the number of strong seedling and yield.展开更多

The influence of EtOAc extracts on the thermogenic curve of growth of Escherchia coli was obtained by microcalorimetry. The chemical differences of EtOAc extracts of Radix isatidis from various sources in the HPLC fingerprints were probed with hierarchical clustering analysis and similarity analysis. The correlation between the HPLC fingerprints and in vitro antibacterial activities was analyzed with multivafiant correlation analysis. Results Close correlation existed between the HPLC fingerprints and in vitro antibacterial activities of EtOAc extracts of Radix isatidis. Methods Reambinant hirudin 3 was isolated andpurified from the fermentation broth by three column chromatography steps with macroporous resin,DEAE cellulose DES2 and preparative RP-HPLC, respectively, and the optimal conditions were obtained.Purity of the product was determined by SDS-PAGE and analytical RP-HPLC. The molecular weight wasdetermined by mass spec-trometry. The structure of the product was analyzed by peptide map.ResultsThe product with purity of 95.4786% was obtained after three purification steps in the optimumconditions with a total yield of 39%. The molecular weight of the product was 6 913.32 ± 6.55 Da,coincident to the theoretical molecular weight of r-hirudin 3. The structure of the product wascoincident to r-hirudin 3 either. Conclusion The optimized purification steps can be successfullyemployed for purification of r-hirudin 3 from E. coli using batch-type approaches. The productobtained with high purity was confirmed to be r-hirudin 3.展开更多

In recent years, the mapping of protein-protein interactions in different species has been reported, but few reports have focused on the large-scale mapping of protein-protein interactions in human. On the basis of their chemical properties and spectral data, they were identified as eclalbasaponins II(1), I(2), III(3), 3-O-[b-D-glucopyranosyl(12)-b-D-glucopyranosyl]-16a-ethoxy-olean-12-ene-28-oic acid-28-O-b-D-glu-copyranoside(4) and 3-O-[(2-O-sulfuryl-b-D-glucopyranosyl)(12)-b-D-glucopyranosyl]-echinocystic acid-28-O-b-D-glucopyranoside(5). Compounds 4 and 5 are new compounds and named eclalbasaponins XI and XII, respectively. Compounds 1 and 5 induced morphological deformation of Pyricularia oryzae mycelia.
文摘[Objective] The aim was to isolate a prodigiosin producing strain and study its pigment fractions.[Method] Red pigment-producing bacteria was identified by physiological and biochemical characteristics after isolation in plate.By using column chromatography and thin-layer chromatography,pigment fractions were separated and purified from the extractives of the strain after fermentation in flask,and then pigment fractions were analyzed via UV-Vis and LC/MS.[Result] A red pigment-producing Serratia marcescens strain NS-17 sampled from soil of Nanchang was isolated and identified.2 pigment fractions showing similar UV-Vis and LC/MS characters were separated and purified,the characters of fraction 1 were identical to those of prodigiosin,while fraction 2 showed a special UV-Vis absorption spectrum that had not been reported.[Conclusion] A prodigiosin-producing Serratia marcescens strain NS-17 and its 2 pigment fractions were isolated.
文摘Aim To optimize purification conditions of recombinant hirudin 3 in thefermentation broth and characterize the product. Methods Reambinant hirudin 3 was isolated andpurified from the fermentation broth by three column chromatography steps with macroporous resin,DEAE cellulose DES2 and preparative RP-HPLC, respectively, and the optimal conditions were obtained.Purity of the product was determined by SDS-PAGE and analytical RP-HPLC. The molecular weight wasdetermined by mass spec-trometry. The structure of the product was analyzed by peptide map.ResultsThe product with purity of 95.4786% was obtained after three purification steps in the optimumconditions with a total yield of 39%. The molecular weight of the product was 6 913.32 ± 6.55 Da,coincident to the theoretical molecular weight of r-hirudin 3. The structure of the product wascoincident to r-hirudin 3 either. Conclusion The optimized purification steps can be successfullyemployed for purification of r-hirudin 3 from E. coli using batch-type approaches. The productobtained with high purity was confirmed to be r-hirudin 3.
文摘Interactions are the essence of all biomolecules because they cannot fulfill their roles without interacting with other molecules. Hence, mapping the interactions of biomolecules can be useful for understanding their roles and functions. Furthermore, the development of molecular based systems biology requires an understanding of the biomolecular interactions. In recent years, the mapping of protein-protein interactions in different species has been reported, but few reports have focused on the large-scale mapping of protein-protein interactions in human. Here, we review the developments in protein interaction mapping and we discuss issues and strategies for the mapping of the human protein interactome.
