Then, the arsenic in the leaching...A process was proposed for removing and stabilizing arsenic(As) from anode slime. The anode slime with high arsenic concentration was pretreated by circular alkaline leaching process. Then, the arsenic in the leaching solution can be further precipitated as a form of scorodite crystalline(FeAsO4·2H2O). In the precipitating arsenic reaction, in which ferrous ions were oxidized by air gas, the effects of acidity(p H), reaction temperature, air flow rate, initial concentration of arsenic and initial molar ratio of Fe(II) to As(V) on arsenic precipitation were investigated. The results showed that sufficiently stable crystal scorodite could be achieved under the condition of initial arsenic concentration of 10 g/L, pH 3.0-4.0, Fe/As molar ratio of 1.5, the temperature of 80-95 °C, and the air flow rate higher than 120 L/h. Under the optimal condition, more than 78% of arsenic could be precipitated as a form of scorodite crystalline. The As leaching concentration of the precipitates was less than 2.0 mg/L and the precipitates may be considered to be safe for disposal.展开更多

Using DGD embedment_free electron microscopy, ultrastructural observation on the intra_ and intercellular microtrabecular network (MN) of the pollen mother cells (PMC) of the whole meiotic prophase Ⅰ in onion ( Alli...Using DGD embedment_free electron microscopy, ultrastructural observation on the intra_ and intercellular microtrabecular network (MN) of the pollen mother cells (PMC) of the whole meiotic prophase Ⅰ in onion ( Allium cepa L.) was performed. Complex nuclear MN was observed in the nucleus of PMCs, spreading throughout the nuclear region. The nucleolus and chromosomes were connected with the MN filament network. The uniformity of nuclear MN changed with the development of the PMCs. A lamina_like structure surrounded the nucleus and joined the MN in nucleus and in cytoplasm, but it disassembled at the end of prophase Ⅰ. There was also a complex cytoplasmic MN in PMCs, without obvious variation during the prophase Ⅰ. Furthermore, MN in cytoplasmic connections (plasmodesmata and cytoplasmic channels) was noticed to link the frameworks in two neighboring PMCs into one entity. Cytomixis was observed at synizesis of prophase Ⅰ in this experiment, and MN in cytoplasm and in nucleus was noticed to distribute in these granules which migrated from one PMC into its neighboring cell. At this time the nucleus moved aside from center of the PMC, but the rest of the cell was still fulfilled with MN filaments. The relationships of nuclear MN with nucleolus and chromosomes, lamina with nucleus, as well as intra_ and intercellular MN with cytomixis are discussed in this paper.展开更多

[Result] Different concentrations of onion generated different cure rates. When the combination was adding 1.0% - 2.0% medicine into feed and spraying 2.0 g,/m3 - 5.0 g,/m3, the curative result was the best with cure rate was 70% -90% [ Conclusion] The onion was effective on curing bacterial rotted gill disease in grass carp and could be taken as curative medicine.展开更多

[Objective] This study aimed to compare the similarities and dissimilarities in karyotypes of purple and white onions,so as to lay foundation for further studying its genetic improvement and breeding more onion variet...[Objective] This study aimed to compare the similarities and dissimilarities in karyotypes of purple and white onions,so as to lay foundation for further studying its genetic improvement and breeding more onion varieties.[Method] Sections of root-tip cells were prepared by normal wafer method.Photomicrography was adopted to produce the high-quality chromosome specimen.Finally,karyotype analysis was conducted using Adobe Photoshop.[Result] Chromosome karyotype analysis on the root-tip showed that the chromosome number of both purple and white onions was 2n=2x=16.Among the 16 chromosomes of both the onions,no satellite chromosome was found.By measuring and calculating,it was found that the karyotype of the studied white union belonged to the lowly evolutional 2A type,and its karyotype formula was 2n=2x=16=14 m +2St,with the karyotype asymmetry coefficient of 58.67%.No obvious centromere was observed in the sections of purple onion.[Conclusion] The karyotypes of white and purple unions are similar and we can infer from this fact that the reason of different characters between both the onions is not due to the chromosome aberration.This karyotype formula and asymmetrical karyotype coefficient have some reference values on the identification of onion variety resources.展开更多

METHODS: In vivo cytotoxicity was first ...AIM: To investigate the anti-tumor activity of ursolic acid (UA) and its derivatives isolated from Aralia decaisneana on hepatocellular carcinoma both in vitro and in vivo. METHODS: In vivo cytotoxicity was first screened by 3-[4,5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) assay. Morphological observation, DNA ladder, flow cytometry analysis, Western blot and real time PCR were employed to elucidate the cytotoxic mechanism of UA. Implanted mouse hepatoma H22 was used to evaluate the growth inhibitory effect of UA in vivo . RESULTS: UA could significantly inhibit the proliferation of HepG2 and its drug-resistance strain, R-HepG2 cells, but had no inhibitory effect on primarily cultured normal mouse hepatocytes whereas all the six derivatives of UA could not inhibit the growth of all tested cell lines. Further study on mechanism demonstrated that apoptosis and G0/G1 arrest were involved in the cytotoxicity and cleavage of poly-(ADP-ribose)- polymerase (PARP). Downregulation of cyclooxygenase-2 (COX-2) protein and upregulation of heat shock protein (HSP) 105 mRNA correlated to the apoptosis of HepG2 cells treated with UA. In addition, UA also could inhibit the growth of H22 hepatoma in vivo. CONCLUSION: UA is a promising anti-tumor agent, but further work needs to be done to improve its solubility.展开更多

Analysis of the homology, characteristics and functional domains of ADH sequence and the phy- Iogenetic relationship to other dipteran ADH were conducted. [Result] The full length of ADH cDNA is 1 088 bp containing a 771 bp of ORF, encoding 256 amino acids, with a calculated relative molecular weight of 30.80 kDa and a theoretical isoelectric point of 8.22. The deduced amino acid sequence shares the highest homology with Glossina morsitans morsitans based on homological analysis and phylogenetic analysis. [Conclusion] This study provides basis for further research of ADH gene.展开更多

So far there is no repor...Turmeric has long been used as a spice and food colouring agent in Asia. In the present investigation, the antimutagenic potential of curcumin was evaluated in Allium cepa root meristem cells. So far there is no report on the biological properties of curcumin in plant test systems. The root tip cells were treated with sodium azide at 200 and 300 μg/ml for 3 h and curcumin was given at 5, 10 and 20 μg/ml for 16 h, prior to sodium azide treatment. The tips were squashed after colchicine treatment and the cells were analyzed for chromosome aberration and mitotic index. Curcumin induces chromosomal aberration in Allium cepa root tip cells in an insignificant manner, when compared with untreated control. Sodium azide alone induces chromosomal aberrations significantly with increasing concentrations. The total number of aberrations was significantly reduced in root tip cells pretreated with curcumin. The study reveals that curcumin has antimutagenic potential against sodium azide induced chromosomal aberrations in Allium cepa root meristem cells. In addition, it showed mild cytotoxicity by reducing the percentage of mitotic index in all curcumin treated groups, but the mechanism of action remains unknown. The antimutagenic potential of curcumin is effective at 5 μg/ml in Allium cepa root meristem cells.展开更多
To improve stability of scorodite,a method of simultaneous synthesis and in-situ coating of scorodite was proposed.Scorodite particles with polyhedral and raspberry-like morphologies were synthesized in an Fe(Ⅱ).As(...To improve stability of scorodite,a method of simultaneous synthesis and in-situ coating of scorodite was proposed.Scorodite particles with polyhedral and raspberry-like morphologies were synthesized in an Fe(Ⅱ).As(Ⅴ).H2O system at 90℃and pH 1.5 by blowing oxygen gas into the system.When the initial Fe/As molar ratio exceeded 1:1,a coating of sulfate-containing iron(hydr)oxides formed on the surfaces of scorodite particles during synthesis.To evaluate the leaching stability of synthesized scorodite samples,toxicity characteristic leaching procedure(TCLP)tests were conducted at pH 4.93 for 60 h,and long-term leaching tests were conducted for 30.40 d within a pH range of 5.40.10.88.The leaching results indicated that the release of arsenic from scorodite was noticeably postponed by the coating,and the average arsenic concentrations in the leaching solutions were as low as 0.12 mg/L in the TCLP tests and lower than 0.5 mg/L in the long-term leaching tests.展开更多

We detected actin in meristematic cells of Allium cepa with indirect im...The presence of actin in eukaryotic nuclei and chromosomes, and especially in higher plant nuclei and chromosomes, has not been well established. We detected actin in meristematic cells of Allium cepa with indirect immunofluorescence technique and observed bright fluorescence in the intact nuclei and chromosomes, indicating that actin is present in the nuclei and chromosomes of the higher plant. We labeled sections of the meristematic cells of A. cepa with immunogold technique, gold particles were found over the whole nuclei and a number of gold particles were concentrated in condensed chromatin and nucleoli, confirming the results of the immunofluoresence observations. We treated the nuclei and chromosomes of A.cepa with DNase I and 2M NaCl and obtained DNA- and histone-depleted nuclei and chromosomes. Indirect immunofluorescence tests showed that the DNA- and histonedepleted nuclei and chromosomes reacted positively with the anti-actin antibodies. These results demonstrate that actin exists not only in intact nuclei and chromosomes,but also in DNA- and histone-depleted nuclei and chrmosomes of the plant. In addition, our immuno-fluorescence tests indicate that tropomyosin is present in the nuclei and chromosomes of A. cepa.展开更多

Aiming to analyze genotoxic potential ofd. princeps leaves extract, Allium cepa assay and micronucleus test were employed. No genotoxic activity presented J. princeps leaves extract; however, extracts cytotoxic activity over A. cepa meristematic cells and mice marrow cells was observed. J. princeps leaves extract presented antiproliferative activity at concentrations and systems employed indicating their therapeutic potential for cellular cycle inhibition. Moreover, results showed that in regard to its mutagenic effect, usage of J. princeps leaves tea on popular medicine has been a safe procedure, as well.展开更多

[Result] Through orthogonal optimization experiments, the optimum condition was as follows: the ratio of Arabic gum and maltodextrin ration was 1∶1, the core content was 25%, feed composition was 18%, and the microencapsulation efficiency and yield can reach 80.7% and 95.3% respectively. [Conclusion] The microcapsulaiton of onion oil with complex coacervation spray drying method can increase membrane thickness, improve core stability and prolong shelf life of products.展开更多

The present work showed in onion cells the nuclear bodies appeared as a spherical structure about 0.3 to 0.8 microm in diameter. They possibly were formed in nucleolus and subsequently released, and entered into nucleoplasm. Observation through cytochemical staining method at the ultrastructural level confirmed that nuclear bodies consisted of ribonucleoproteins (RNPs) and silver-stainable proteins. Immunocytochemical results revealed that nuclear bodies contained no DNA and ribosomal gene transcription factor (UBF). Based on these data, we suggested that nuclear bodies are not related to the ribosome or other gene transcription activities, instead they may act as subnuclear structures for RNPs transport from nucleolus to cytoplasm, and may also be involved in splicing of pre-mRNAs.展开更多

The occupation of new environments by these plants is a problem to be solved and it is essential to investigate all the aspects that allow this successful to find solutions to this question, such as its mutagenic effects. Thus, this study aimed to evaluate the mutagenic effect of leaves extracts of Acacia mangium Willd, Artocarpus heterophyllus Lam and Eriobothrya japonica (Thunb.) Lindl through Allium cepa bioassay. For this, A. cepa seeds were submitted to continuous and discontinuous (acute and chronic) treatments in medium with water (negative control) or four concentration of each extract (1, 5, 10 and 50 mg/mL). The mitotic index was affected at all concentrations of three extracts tested in all treatments, continuous and discontinuous. Aneugenic effects were not related to any treatment tested. E. japonica extract induced clastogenic effects at 1, 5 and 10 mg/mL in continuous treatment, 5 and 10 mg/mL in acute discontinuous treatment and at 10 mg/mL in chronic discontinuous treatment. Clastogenic effect was also observed at 10 mg/mL ofA. heterophyllus extract in continuous and acute discontinuous treatments.展开更多

Morphological observation, DNA ladder, flow cytometry analysis, Western blot and real time PCR were employed to elucidate the cytotoxic mechanism of UA. Implanted mouse hepatoma H22 was used to evaluate the growth inhibitory effect of UA in vivo . RESULTS: UA could significantly inhibit the proliferation of HepG2 and its drug-resistance strain, R-HepG2 cells, but had no inhibitory effect on primarily cultured normal mouse hepatocytes whereas all the six derivatives of UA could not inhibit the growth of all tested cell lines. Further study on mechanism demonstrated that apoptosis and G0/G1 arrest were involved in the cytotoxicity and cleavage of poly-(ADP-ribose)- polymerase (PARP). Downregulation of cyclooxygenase-2 (COX-2) protein and upregulation of heat shock protein (HSP) 105 mRNA correlated to the apoptosis of HepG2 cells treated with UA. In addition, UA also could inhibit the growth of H22 hepatoma in vivo. CONCLUSION: UA is a promising anti-tumor agent, but further work needs to be done to improve its solubility.

文摘[Objective] This study aims to conduct cloning and sequence analysis of ADH gene in D. Antiqua. [Method] Full-length cDNA of ADH gene in D. antiqua was cloned by using RACE technology (GenBank access number: JQ666006). Analysis of the homology, characteristics and functional domains of ADH sequence and the phy- Iogenetic relationship to other dipteran ADH were conducted. [Result] The full length of ADH cDNA is 1 088 bp containing a 771 bp of ORF, encoding 256 amino acids, with a calculated relative molecular weight of 30.80 kDa and a theoretical isoelectric point of 8.22. The deduced amino acid sequence shares the highest homology with Glossina morsitans morsitans based on homological analysis and phylogenetic analysis. [Conclusion] This study provides basis for further research of ADH gene.

文摘Turmeric has long been used as a spice and food colouring agent in Asia. In the present investigation, the antimutagenic potential of curcumin was evaluated in Allium cepa root meristem cells. So far there is no report on the biological properties of curcumin in plant test systems. The root tip cells were treated with sodium azide at 200 and 300 μg/ml for 3 h and curcumin was given at 5, 10 and 20 μg/ml for 16 h, prior to sodium azide treatment. The tips were squashed after colchicine treatment and the cells were analyzed for chromosome aberration and mitotic index. Curcumin induces chromosomal aberration in Allium cepa root tip cells in an insignificant manner, when compared with untreated control. Sodium azide alone induces chromosomal aberrations significantly with increasing concentrations. The total number of aberrations was significantly reduced in root tip cells pretreated with curcumin. The study reveals that curcumin has antimutagenic potential against sodium azide induced chromosomal aberrations in Allium cepa root meristem cells. In addition, it showed mild cytotoxicity by reducing the percentage of mitotic index in all curcumin treated groups, but the mechanism of action remains unknown. The antimutagenic potential of curcumin is effective at 5 μg/ml in Allium cepa root meristem cells.

文摘To improve stability of scorodite,a method of simultaneous synthesis and in-situ coating of scorodite was proposed.Scorodite particles with polyhedral and raspberry-like morphologies were synthesized in an Fe(Ⅱ).As(Ⅴ).H2O system at 90℃and pH 1.5 by blowing oxygen gas into the system.When the initial Fe/As molar ratio exceeded 1:1,a coating of sulfate-containing iron(hydr)oxides formed on the surfaces of scorodite particles during synthesis.To evaluate the leaching stability of synthesized scorodite samples,toxicity characteristic leaching procedure(TCLP)tests were conducted at pH 4.93 for 60 h,and long-term leaching tests were conducted for 30.40 d within a pH range of 5.40.10.88.The leaching results indicated that the release of arsenic from scorodite was noticeably postponed by the coating,and the average arsenic concentrations in the leaching solutions were as low as 0.12 mg/L in the TCLP tests and lower than 0.5 mg/L in the long-term leaching tests.

文摘Friction stir welding provides better retention of ductility and plasticity in comparison to fusion welding and it increases the stability against cracking.This study aims to enhance both the ductility and strength of the welded joints.Tool profile was found to play an important role in retaining ductility,understanding the metal flow behavior and determining the fracture initiating point.The microstructure of different regions in the onion ring was inferred to correlate the strength with the metal flow pattern.The onion ring pattern strengthened the weld nugget.Shoulder and taper threaded profile tool resulted in superior mechanical properties.Microhardness results confirmed that the fracture runs through low hardness areas such as heat affected zone and thermo-mechanically affected zone.The scanning electron microscopic images revealed elongated grains and dimples,justifying ductile mode of fracture.
文摘The presence of actin in eukaryotic nuclei and chromosomes, and especially in higher plant nuclei and chromosomes, has not been well established. We detected actin in meristematic cells of Allium cepa with indirect immunofluorescence technique and observed bright fluorescence in the intact nuclei and chromosomes, indicating that actin is present in the nuclei and chromosomes of the higher plant. We labeled sections of the meristematic cells of A. cepa with immunogold technique, gold particles were found over the whole nuclei and a number of gold particles were concentrated in condensed chromatin and nucleoli, confirming the results of the immunofluoresence observations. We treated the nuclei and chromosomes of A.cepa with DNase I and 2M NaCl and obtained DNA- and histone-depleted nuclei and chromosomes. Indirect immunofluorescence tests showed that the DNA- and histonedepleted nuclei and chromosomes reacted positively with the anti-actin antibodies. These results demonstrate that actin exists not only in intact nuclei and chromosomes,but also in DNA- and histone-depleted nuclei and chrmosomes of the plant. In addition, our immuno-fluorescence tests indicate that tropomyosin is present in the nuclei and chromosomes of A. cepa.
文摘Evaluation of the genotoxic and cytotoxic potentials is a part of the analysis process where plant products with therapeutic properties must be submitted for being safely employed, mainly through long time points, doannesia princeps, also known as "cotieira" has been used on popular medicine as laxative and several diseases treatment. Aiming to analyze genotoxic potential ofd. princeps leaves extract, Allium cepa assay and micronucleus test were employed. No genotoxic activity presented J. princeps leaves extract; however, extracts cytotoxic activity over A. cepa meristematic cells and mice marrow cells was observed. J. princeps leaves extract presented antiproliferative activity at concentrations and systems employed indicating their therapeutic potential for cellular cycle inhibition. Moreover, results showed that in regard to its mutagenic effect, usage of J. princeps leaves tea on popular medicine has been a safe procedure, as well.

文摘[Objective] The aim was to study the microcapsulation of onion oil with double place separation-coacevation and spray drying. [Method] The effect of processing parameters which included the different wall material combination,the ratio between the wall material,the core material content and the feed concentration on the efficiency and yield of microencapsulated was evaluated by single factor experiments. [Result] Through orthogonal optimization experiments, the optimum condition was as follows: the ratio of Arabic gum and maltodextrin ration was 1∶1, the core content was 25%, feed composition was 18%, and the microencapsulation efficiency and yield can reach 80.7% and 95.3% respectively. [Conclusion] The microcapsulaiton of onion oil with complex coacervation spray drying method can increase membrane thickness, improve core stability and prolong shelf life of products.
文摘Nuclear bodies have long been noted in interphase nuclei of plant cells, but their structural component, origin and function are still unclear by now. The present work showed in onion cells the nuclear bodies appeared as a spherical structure about 0.3 to 0.8 microm in diameter. They possibly were formed in nucleolus and subsequently released, and entered into nucleoplasm. Observation through cytochemical staining method at the ultrastructural level confirmed that nuclear bodies consisted of ribonucleoproteins (RNPs) and silver-stainable proteins. Immunocytochemical results revealed that nuclear bodies contained no DNA and ribosomal gene transcription factor (UBF). Based on these data, we suggested that nuclear bodies are not related to the ribosome or other gene transcription activities, instead they may act as subnuclear structures for RNPs transport from nucleolus to cytoplasm, and may also be involved in splicing of pre-mRNAs.
文摘Invasive alien species are a global threat to biodiversity that affects protected areas around the world. The occupation of new environments by these plants is a problem to be solved and it is essential to investigate all the aspects that allow this successful to find solutions to this question, such as its mutagenic effects. Thus, this study aimed to evaluate the mutagenic effect of leaves extracts of Acacia mangium Willd, Artocarpus heterophyllus Lam and Eriobothrya japonica (Thunb.) Lindl through Allium cepa bioassay. For this, A. cepa seeds were submitted to continuous and discontinuous (acute and chronic) treatments in medium with water (negative control) or four concentration of each extract (1, 5, 10 and 50 mg/mL). The mitotic index was affected at all concentrations of three extracts tested in all treatments, continuous and discontinuous. Aneugenic effects were not related to any treatment tested. E. japonica extract induced clastogenic effects at 1, 5 and 10 mg/mL in continuous treatment, 5 and 10 mg/mL in acute discontinuous treatment and at 10 mg/mL in chronic discontinuous treatment. Clastogenic effect was also observed at 10 mg/mL ofA. heterophyllus extract in continuous and acute discontinuous treatments.
