为了探讨肺大细胞癌(PLCC)的分化表型,通过电镜检查并采用5种与分化表型有关的抗体CK-HMW,CK-LMW,SC,CgA,Syn进行免疫组织化学观察,报告了6例具有三向(腺、鳞、神经内分泌)分化特征的PLCC。免疫组化CK-HMW阳性者4例,CK-... 为了探讨肺大细胞癌(PLCC)的分化表型,通过电镜检查并采用5种与分化表型有关的抗体CK-HMW,CK-LMW,SC,CgA,Syn进行免疫组织化学观察,报告了6例具有三向(腺、鳞、神经内分泌)分化特征的PLCC。免疫组化CK-HMW阳性者4例,CK-LMW、SC、Syn阳性者各3例,CgA阳性者2例。在免疫组化水平上,具有腺鳞神经内分泌、腺神经内分泌及鳞分化表型特征者各为2例。在电镜水平上,5例中3例为腺鳞神经内分泌分化特征,余2例为腺鳞分化持征。6例具有三向分化特征的PLCC,5例分别通过免疫组化或电镜证实,1例为免疫组化和电镜结果综合证实。本结果表明:①PLCC的异质性可显示在其同一肿瘤的不同区域甚至同一细胞内;②结果支持肿瘤的多能干细胞起源学说;③因各有其优劣性,免疫组化和电镜均为鉴别PLCC分化表型不可缺少的重要方法。 展开更多

An organic layer prepared from the seed of Aceriphyllum rossii was studied to identify the active compounds for protein tyrosine phosphatase 1B(PTP1B) inhibition.Bioassay guided fractionation resulted in the isolation of PTP1B inhibitory activity of triterpenes(1-4).These four compounds were identified as aceriphyllic acid C(1),aceriphyllic acid D(2),aceriphyllic acid E(3) and aceriphyllic acid F(4).The isolated 1-4 compounds inhibited PTP1B with IC50 values ranged from(2.1±1.5) μmol/L to(11.2±2.5) μmol/L.Kinetic analysis of PTP1B inhibition by aceriphyllic acid C(1) and aceriphyllic acid D(2) suggested that oleanane-type triterpenes inhibited PTP1B activity in a mixed-type manner. 展开更多

The influences of subsurface cracks,distributing along the axial direction,on the rolling contact fatigue(RCF)faliure in a bearing ring are investigated.A realistic three-dimensional model of the bearing ring containing three subsurface cracks is used to evaluate the fatigue crack propagation based on stress intensity factor(SIF)calculations.Moreover,the distributions of the subsurface cracks along the axial direction are varied to study their effects on RCF.The results provide valuable guidelines for enhanced understanding of RCF in bearings. 展开更多
