AIM:To confirm the value of blocking treatment by zenshengping(ZSP),a Chinese herb composite and Riboflavin for esophageal epithelia dysplasia cases screened out in high risk area in northern china by exfoliative ball...AIM:To confirm the value of blocking treatment by zenshengping(ZSP),a Chinese herb composite and Riboflavin for esophageal epithelia dysplasia cases screened out in high risk area in northern china by exfoliative balloon cytology(EBC),so to reduce the incidence rate of esophageal cancer(EC).METHODS:Esophageal epithelium dysplasia cases including mind esophageal epithelium dysplasia(MEED),stage one severe esophageal epithelium dysplasia(SEED Ⅰ),and stage two severe esophageal epithelium dysplasia(SEEDⅡ) were screened out from people aged 40 years and older in the high risk area of Chixian.These cases were randomly divided into a treatment and control group.Subjects in the treatment and control groups took ZSP,riboflavin,and placebo daily for three years.EC cases registered by cancer registry and identified by EBC re-screening in the treatment and control groups were used to calculate incidence and blocking rates to demonstrate effects of blcking medication.RESULTS:It was found that 31.92%and 24.15% of people aged 40 years and older in Cixian could been diagnosed as MEED and SEED cases.The severity of dysplasia increased with age ,ZSP had blocked EC occurrence by 47.79% after 3 year medication among the SEED cases.CONCLUSION:ZSP can block the development from SEED I and SEED Ⅱto EC by 47.79%.Efforts should by made to screen and treat dysplasia cases in people aged 40 years and older in high risk areas to reduce the mortality figres.展开更多

METHODS: Forty-one specimens were obtained from esophageal cancer (EC) patients. Histopathological assessment identified 23 squamous cell carcinomas (SCC) and 18 adenocarcinomas (ADC), including only 8 ADC with Barrett esophageal columnar epithelium (metaplasia) and dysplasia adjacent to ADC. Paraffin-embedded normal squamous epithelium, Barrett esophageal columnar epithelium (metaplasia), dysplasia and esophageal tumor tissues were dissected from the surrounding tissues under microscopic guidance. DNA was extracted using proteinase K digestion buffer, and DNA was diluted at 1:100, 1:1000, 1:5000, 1:10 000 and 1:50 000, respectively. Seven microsatellite markers (D2S123, D3S1616, D3S1300, D5S346, D17S787, D18S58 and BATRII loci) were used in this study. Un-dilution and dilution polymerase chainreactions (PCR) were performed, and microsatellite analysis was carried out. RESULTS: No statistically significant difference was found in microsatellite instability (MSI) and loss of heterozygosity (LOH) of un-diluted DNA between SCC and ADC. The levels of MSI and LOH were high in the metaplasia-dysplasia-adenocarcinoma sequence of diluted DNA. The more the diluted DNA was, the higher the rates of MSI and LOH were at the above 7 loci, especially at D3S1616, D5S346, D2S123, D3S1300 and D18S58 loci. CONCLUSION: The sequence of metaplasia-dysplasia-adenocarcinoma is associated with microsatellite alterations, including MSI and LOH. The MSI and LOH may be the early genetic events during esophageal carcinogenesis, and genetic alterations at the D3S1616, D5S346 and D3S123 loci may play a role in the progress of microsatellite alterations.展开更多

Methods We studied the expression of Notch1,2,3 isforms ...Objective To investigate the temporal expression of Notch receptors in developing lungs of rats and to explore the regulating role of Notch in lung development. Methods We studied the expression of Notch1,2,3 isforms in embryonic days 18,20,21 and postnatal days 1,4,7,14, 21 rat lungs. Six rats of each group were used to assess lung histologic changes by HE staining and expression of Notch in lungs by immunohistochemistry. Total RNA was extracted by Trizol reagent from the frozen lung tissues. mRNA levels of Notch were measured by reverse transcription polymerase chain reaction (RT-PCR). Results It is showed that Notch_ 1-3 mainly localized in the airway surface epithelium、alveolar epithelium during the psdueoglandular stage, and reached the peaks at canalicular period. The expression patterns of Notch_ 1-3 were changed with the fetal age. Conclusion These results support multiple roles for Notch1,2,and 3 receptor activation during lung development, probably not only modulating the process of branching morphogenesis but also involved in determining the cell differentiation fate in fetal alveolar epithelium.展开更多
