There were wrong ideas about the location of the three frontier fortresses. Through textual research the conclusion has been made that the west section of Upper Zhang was the present place form Huang Zhou to Zhang Xiang Li in the south west of Pingrang City Proper and extended to Suian, the Yellow Sea North Course and from Wenhu Li to Fanchuanli was called Lower Zhang, which was also called Manfanhan. 展开更多

AIM: To determine whether Lactobacillus plantarum can modify the deleterious effects of tumor necrosis factor-α (TNF-α) on intestinal epithelial cells. METHODS: Caco-2 cells were incubated with TNF-α alone or in the presence of L. plantarum. Transepithelial electrical resistance was used to measure epithelial barrier function. Interleukin 8 (IL-8) secretion by intestinal epithelial cells was measured using an ELISA. Cellular lysate proteins were immunoblotted using the anti-extracellular regulated kinase (ERK), anti-phospho- ERK and anti-IκB-α. RESULTS: A TNF-α-induced decrease in transepithelial electrical resistance was inhibited by L. plantarum. TNF- α-induced IL-8 secretion was reduced by L. plantarum. L. plantarum inhibited the activation of ERK and the degradation of IκB-α in TNF-a-treated Caco-2 cells. CONCLUSION: Induction of epithelial barrier dysfunction and IL-8 secretion by TNF-α is inhibited byL. plantarum. Probiotics may preserve epithelial barrier function and inhibit the inflammatory response by altering the signal transduction pathway. 展开更多

AIM To investigate potential effects of poly I:C on mucosal injury and epithelial barrier disruption in dextran sulfate sodium (DSS)-induced acute colitis. METHODS Thirty C57BL/6 mice were given either regular drinking water (control group) or 2% (w/v) DSS drinking water (model and poly I:C groups) ad libitum for 7 d. Poly I:C was administrated subcutaneously (20 mu g/mouse) 2 h prior to DSS induction in mice of the poly I:C group. Severity of colitis was evaluated by disease activity index, body weight, colon length, histology and myeloperoxidase (MPO) activity, as well as the production of proinflammatory cytokines, including tumor necrosis factor-a (TNF-alpha), interleukin 17 (IL-17) and interferon-. (IFN-gamma). Intestinal permeability was analyzed by the fluorescein isothiocyanate labeled-dextran (FITC-D) method. Ultrastructural features of the colon tissue were observed under electron microscopy. Expressions of tight junction (TJ) proteins, including zo-1, occludin and claudin-1, were measured by immunohistochemistry/immunofluorescence, Western blot and real-time quantitative polymerase chain reaction (RT-qPCR). RESULTS DSS caused significant damage to the colon tissue in the model group. Administration of poly I:C dramatically protected against DSS-induced colitis, as demonstrated by less body weight loss, lower disease activity index score, longer colon length, colonic MPO activity, and improved macroscopic and histological scores. It also ameliorated DSS-induced ultrastructural changes of the colon epithelium, as observed under scanning electron microscopy, as well as FITC-D permeability. The mRNA and protein expressions of TJ protein, zo-1, occludin and claudin-1 were also found to be significantly enhanced in the poly I:C group, as determined by immunohistochemistry/immunofluorescence, Western blot and RT-qPCR. By contrast, poly I:C pretreatment markedly reversed the DSS-induced up-regulated expressions of the inflammatory cytokines TNF-alpha, IL-17 and IFN-gamma. CONCLUSION Our study suggested that poly I:C may protect against DSS-induced colitis through maintaining integrity of the epithelial barrier and regulating innate immune responses, which may shed light on the therapeutic potential of poly I:C in human colitis. 展开更多

The apoptosis of lens epithehal cells has been proposed as the common basis of cataract formation, with oxidative stress as the major cause. This study was performed to investigate the protective effect of the herbal constituent parthenolide against oxidative stress-induced apoptosis of human lens epithelial (HLE) cells and the possible molecular mechanisms involved. HLE cells (SRA01-04) were incubated with 50 μM H2O2 in the absence or presence of different doses of parthenolide (10, 20 and 50 μM). To study apoptosis, the cells were assessed by morphologic examination and Annexin V-propidium iodide double staining flow cytometry; to investigate the underlying molecular mechanisms, the expression of caspase-3 and caspase-9 were assayed by Western blot and quantitative RT-PCR, and the activities of caspase-3 and caspase-9 were measured by a Chemicon caspase colorimetric activity assay kit. Stimulated with H202 for 18 h, a high fraction of riLE cells underwent apoptosis, while in the presence ofparthenolide of different concentrations, dose-dependent blocking of HLE cell apoptosis was observed. The expression of caspase-3 and caspase-9 induced by H202 in HLE cells was significantly reduced by parthenolide both at the protein and mRNA levels, and the activation ofcaspase-3 and caspase-9 was also suppressed by parthenolide in a dose-dependent manner. In conclusion, parthenolide prevents HLE cells from oxidative stress-induced apoptosis through inhibition of the activation ofcaspase-3 and caspase-9, suggesting a potential protective effect against cataract formation. 展开更多

Although the prognosis is generally favorable, acute fatal complications have been reported in a small subset of patients. Some patients died with acute cardiac complications, on the other hand, other patients died with underlying critical illness which triggered stress-induced cardiomyopathy. 展开更多

In other words, they compensate their divergent intrinsic emotions with imitated convergences (eg., the monotonous "convergence obsessed" logic of hammering, wringing hands etc.) Today's science cannot declare this to be convergent, especially for the fact that psychotic autist patients prove to be weak at convergence, but the male/female proportion reflects on notable facts (with Kanner syndrome it is 3:1 or 4:1, while Rett syndrome only affects females). Can we declare Kanner and Rett syndromes to be basically female brain disorders? Asperger-autism has been scientifically considered as a type of "male-brain disorder" since 199l (Baron-Cohen theory). The proportion of male-female is approximately 6:1 with this disease. The author would like to demonstrate a very special case, the Asperger-autism as a "cognitive autism." It is common to address autistic disorder as "pervasive" or "comprehensive" ontogenetic disorders because they affect all areas of adolescent psychological development negatively. But as the expression itself suggests, we are not aware of the specific disorders directly. As it will turn out we cannot deal with autism as on complex disorder, we should rather use the term in plural, i.e., autisms and autistic disorders. 展开更多

China's economic strategy, in particular Chinese maritime policy, shows a high tendency of urging to develop China's strategic shipping, which aims to safeguard China's national interest. This paper explores the approach of balancing China's maritime interests safeguarding and the compliance of international maritime regulations by examining three dimension of strategic shipping capacity which includes shipping, port and lane. In addition, this paper creatively proposes the idea of merchant ships and ports for military application and gives some advice about the implementation measures and legal system of mechanism safeguarding. 展开更多

Expression and proteolytic activity of calpain in lens epithelial cells of oxidative cataract

Methods: Rat lenses were cultured in vitro and cataract was induced by H2O2. The level of intracellular free Ca2+ was measured by fluorescence determination with fura-2/AM. The expression of m-calpain protein in LECs was detected with immunohistochemical method. The proteolytic activity in LECs was measured using a fluorogenic synthetic substrate. Results: There were significant differences of the level of intracellular free Ca2+ (P=0.001, 0.000, 0.000), the expression of m-calpain (P=0.001, 0.000, 0.000) and the proteolytic activity of calpain (P=0.001, 0.000, 0.000) between H2O2-induced and control group at 6, 12 and 24 h, respectively. Conclusions: H2O2 can increase intracellular free Ca2+, then enhance the expression and proteolytic activity of calpain which may play a role in the mechanism of oxidative cataract of rat. 展开更多

AIM:To evaluate the impacts of Schistosoma japoni-cum(S.japonicum) ova on the tight junction barriers in a trinitrobenzenesulfonic acid(TNBS)-induced colitis model.METHODS:Balb/c mice were randomly divided into three groups:control group;TNBS + ovagroup and TNBS + ova + group.TNBS was used intracolonic to in-duce colitis and mice of the TNBS + ova + group were pre-exposed to S.japonicum ova as a prophylactic intervention.Colon inflammation was quantified using following variables:mouse mortality,weight loss,co-lon extent and microscopic inflammation score.Serum expression of tumor necrosis factor-αand interferon-γ were assessed to evaluate the systemic inflamma-tory response.NOD2 and its mRNA were also tested.Bacterial translocations were tested by culturing blood and several tissues.ZO-1 and occludin were chosen as the representations of tight junction proteins.Both the proteins and mRNA were assessed.RESULTS:Ova pre-treatment contributed to the reliefof colitis and decreased the mortality of the models.NOD2 expression was significantly downregulated when pretreated with the ova.The TNBS injection caused a significant downregulation of ZO-1 and oc-cludin mRNA together with their proteins in the colon;ova pre-exposure reversed these alterations.Treat-ment with S.japonicum ova in the colitis model caused lower intestinal bacterial translocation frequency.CONCLUSION:S.japonicum ova can maintain epithelial barrier function through increasing tight junction pro-teins,thus causing less exposure of NOD2 to the lumi-nal antigens which may activate a series of inflamma-tory factors and induce colitis. 展开更多

METHODS: Enucleated eyes were subjected to immunostaining for bFGF protein. Human cataract LECs were cultured in vitro, and treated with bFGF for 48 hr. Proliferation was estimated by the positive area ratio of proliferating cell nuclear antigen (PCNA) in immunohistochemistry. In this paper, a real-time collision-free path planning of the rust removal robot in a ship environment is proposed, which is based on an improved biologically inspired neural network algorithm. This improved algorithm is based on the biologically inspired neural network and modified with obstacle detection sensors and kinematic state templates, and is implemented in a ship rust removal robot planning system for dynamic trajectory generation. The real-time optimal trajectory is generated by the biologically inspired neural network, and the moving obstacle detection process of a ship robot working on the wall is simulated with the obstacle detection sensors models. The local real-time trajectory can be re-planned by the updated local map information, where the obstacle detection sensors are used to inspect partial environment information and update the robot nearby information in real time in the original neural network algorithm. At the same time, the method of the kinematic state templates matching and searching is used to solve the pipes’ influence of the rust removal robot climbing on the wall, which can not only provide a smooth path, but also can judge the motion direction and turning angle of the robot. Comparison of the proposed approach with the simulation shows that the improved algorithm is capable of planning a real-time collision-free path with achieving the local environmental information and judging the rust removal robot’s motion direction and turning angle.
