Rapid and prolific shoot regeneration and very viable and yellowish callus formation occurred on a MS medium supplemented with 2 mg/L 6-benzylaminopurine(BAP) and 0.5 mg/L β-naphthenacetic(NAA). It was found that BAP was essential to adventitious shoot regeneration from hypocotyl explants. Both low(0.1 mg/L) and high(6 mg/L) concentration of BAP in MS medium were able to stimulate shoot development. Optimal concentration of BAP for shoot development was 2-3 mg/L. 2,4-D alone could stimulate formation of healthy and yellowish callus which, however, did not differentiate to generate shoot on BAP (0.1-6 mg/L) or BAP(2 mg/L) and NAA(0.5 mg/L) media. NAA alone could stimulate root development at the ends of hypocotyl explants. Addition of silver nitrate(AgNO3) to the medium inhibited shoot regeneration,and made all explants brown and die.Histological analysis indicated that adventitious shoot regeneration originated from epidermic cells of hypocotyl.Regenerated shoots rooted readily in MS medium without any growth regulators.展开更多
