Colloid stationary phase was prepared by dissolving the polymer in the mixe...An insoluble β-cyclodextrin polymer cross-linked with epichlorohydrin was prepared, and its structure was identified with infrared spectrum. Colloid stationary phase was prepared by dissolving the polymer in the mixed solvent of diisopropyl ether, methylene dichloride and benzene and treated for 0.5 h by ultrasonication, and then was coated on a fused silica capillary column. The optimun reaction conditions are as follows: the mole ratio of epichlohydrin to β-cyclodextrin is 12.1:1, reacting at 65 °C for 24 h. The Chromatographic performance such as column efficiency, thermal stabilities and polarity were studied, two kinds of disubstituted benzene isomers and eight pairs of enantiomers were separated on the capillary column. The results show that the β-cyclodextrin polymer is suitable for use as a capillary gas chromatographic stationary phase, and possess excellent chromatographic properties in separating enantiomers and position isomers.展开更多
The conventional silk fibroin(SF)films obtained by casting from SF aqueous solution are amorphous,and easily dissolved in water.This defect limits the practical use of SF films in biomedical field.A new water-insolubl...The conventional silk fibroin(SF)films obtained by casting from SF aqueous solution are amorphous,and easily dissolved in water.This defect limits the practical use of SF films in biomedical field.A new water-insoluble silk fibroin film(an SF-FA film)could be simply prepared by casting from SF formic acid solution.The physical properties and moisture permeability of SF-FA films were examined.The results show that the SF-FA film swollen in water exhibits outstanding pliability.The weight loss of SF-FA films in water is also very little.The unique physical properties for the SF-FA film in the wet relate to the intrinsic structure of the fibroin film.The cell viability of rat embryo dermal fibroblasts seeded on the control films and the SF-FA films was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Results show that the optical density(OD)value of the SF-FA film is almost the same as that of the conventional SF film treated with alcohol,therefore,the SF-FA film has good cell compatibility.In addition,the SF-FA film is also a biodegradable material.Because of their good transparency,it is helpful in study of clinical experiment with the SF-FA films.展开更多

The dissolution of the expressed IFN 2b in inclusion body was carried out and it was found that the optimal condi...Alpha-interferon 2b (IFN 2b) was produced both in soluble and insoluble forms from recombinant E. coli. The dissolution of the expressed IFN 2b in inclusion body was carried out and it was found that the optimal condition to dissolve the expressed protein was 7 mol稬1guanidinium salt solution at pH 3.0. The resultant solution was diluted 20 times using pH 6.0 buffer to refold the protein correctly. The cation exchange column was employed to purify both refolded and soluble IFN 2b. For soluble IFN sample, high IFN 2b recovery yield (92.1%) with 91.7% purity was obtained in the eluate. However, for refolded IFN sample, only 72.7% of IFN 2b was recovered with relatively low purity (56.8%) by cation exchange chromatography. Although the expression level of insoluble IFN was higher than that of co-expressed soluble IFN in this recombinant E. coli cells, the productivity of bioactive IFN 2b was higher with soluble expressed IFN after primary purification process. Soluble expression of foreign proteins in recombinant bacteria might be an alternative strategy for efficient production of heterogeneous pro-teins due to high bioactivity and simple downstream protein purification process.展开更多
