Although pericentrin was shown to localize in the cytoplasm and reported to be sensitive to leptomycin B (LMB), a specific inhibitor of Crml, the regions within pericentrin that serve as signals for transporting in and out of the nucleus have not yet been identified. In this study, we identified five novel nuclear export signals (NESs) in pericentrin with diverse export activities. All of the five NESs could bind to Crml in a LMB-sensitive way when mediating the nuclear export of pericentrin. We also demonstrated that the region of amino acids 8-42 in pericentrin contains a tripartite nuclear localization signal (NLS) consisting of three clusters of basic amino acids. The NLS of pericentrin binds to importin β directly or via the adaptor importin α to form the import complex, which could be disrupted by RanQ69L, a dominant-negative Ran GTPase possessing high affinity for importin β. Furthermore, we found that mutation of the NESs in full-length pericentrin results in both nuclear and cytoplasmic localization, and mutation of the NLS abolishes the nuclear import of pericentrin. On the basis of these results, we suggest that the NESs and NLS of pericentrin are essential for its subcellular localization and nucleocytoplasmic trafficking during the cell cycle.展开更多

However, cytogenetic and genomic data is limited in the organism partly because the chromosomes are difficult to isolate and analyze. In this study, fluorescence in-situ hybridization (FISH) was used to identify the chromosomes of F. chinensis. The 5S ribosomal RNA gene (rDNA) of F. chinensis was isolated, cloned and then used as a hybridization probe. The results show that the 5S rDNA was located on one pair of homologous chromosomes in F chinensis. In addition, triploid shrimp were used to evaluate the feasibility of chromosome identification using FISH and to validate the method. It was confirmed that 5S rDNA can be used as a chromosome-specific probe for chromosome identification in E chinensis. The successful application ofFISH in E chinensis shows that chromosome-specific probes can be developed and this finding will facilitate further research on the chromosomes ofpenaeid shrimps.展开更多

The singleshell states and the corresponding matrix elements needed for describing Sn isotopes are reconstructed to calculate the coefficient of fractional parantage by reducing the calculation requirements. This reconstruction allows us to do the shell model calculations of the neutron deficient Sn isotopes in very reasonable time. The results are compared to the recent high-resolution experimental data and found to be in good agreement with experiments.展开更多

It is shown that, even in the presence of the three-body force, the empirical parabolic law of the energy per nucleon vs. isospin asymmetry is fulfilled in the whole asymmetry range and also up to high density. The three-body force provides a strong enhancement of symmetry energy at high density in agreement with relativistic approaches. It also shows that the three-body force leads to a much more rapid increasing of symmetry energy with density in relatively high density region and to a much lower threshold density for the direct URCA process to occur in a neutron star as compared to the predictions adopting only pure two-body force.展开更多
