目的探讨中性粒细胞胞外诱捕网(NETs)与静脉血栓栓塞(VTE)的关系及糖皮质激素(GC)的调节作用。方法将40只KM小鼠随机分为对照组(n=20)和实验组(n=20)。实验组小鼠通过神经血管钳钳夹联合结扎下腔静脉建立小鼠VTE模型,对照组采用假手术... 目的探讨中性粒细胞胞外诱捕网(NETs)与静脉血栓栓塞(VTE)的关系及糖皮质激素(GC)的调节作用。方法将40只KM小鼠随机分为对照组(n=20)和实验组(n=20)。实验组小鼠通过神经血管钳钳夹联合结扎下腔静脉建立小鼠VTE模型,对照组采用假手术法处理。术后7 d,处死小鼠,取肺组织进行病理切片观察,取眼眶静脉血,石蜡切片HE染色检测肺组织病理变化;采用免疫荧光染色法观察肺组织瓜氨酸化的组蛋白H3(Cit H3)与细胞核共定位情况。检测血浆中游离DNA含量,及小鼠外周血髓过氧化物酶(MPO)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-4(IL-4)、γ-干扰素(IFN-γ)、重组人B细胞活化因子(BAFF)的含量。分离并观察外周血中性粒细胞集落形成能力及对体外NETs形成的影响。检测细胞因子白介素-1(IL-1)、白介素-6(IL-6)及TNF-α的表达及分泌变化,检测细胞活性氧产生情况、脂质过氧化产物产生情况及细胞内NF-κB信号转导激活改变。结果VTE术后,对照组小鼠的肺组织无明显的病理学变化,实验组小鼠的肺组织改变明显。实验组的血浆游离DNA水平较对照组水平明显升高,ELISA检测MPO可见实验组的血浆MPO水平较对照组水平明显升高。免疫荧光标记Cit H3发现对照组小鼠中,Cit H3的阳性信号较少,而在VTE造模实验组的小鼠肺组织中,Cit H3的阳性信号显著增多。实验组的血清TNF-α水平及IL-1β水平较对照组水平明显升高,但是IFN-γ、IL-4和BAFF无显著差异。结论中性粒细胞胞外诱捕网与血栓形成密切相关,其可激活巨噬细胞促进血栓形成。 展开更多

目的:探索术前中性粒细胞与淋巴细胞比值(NLR)对胃肠间质瘤(GIST)预后的影响。方法:检索Pub Med、EMBASE、Cochrane、CNKI、中国生物医学文献数据库、维普、万方等数据库建库至2018年5月公开发表的相关中英文文献,按纳入标准筛选相关文... 目的:探索术前中性粒细胞与淋巴细胞比值(NLR)对胃肠间质瘤(GIST)预后的影响。方法:检索Pub Med、EMBASE、Cochrane、CNKI、中国生物医学文献数据库、维普、万方等数据库建库至2018年5月公开发表的相关中英文文献,按纳入标准筛选相关文献,提取临床效应指标,并进行文献质量评分,提取文献数据用Review Manager5. 3软件进行统计分析,发表性偏倚采用漏斗图进行评价分析。结果:最终纳入10篇符合标准的研究文献,共计2 264例胃肠间质瘤患者,Meta分析结果显示,NLR升高的胃肠间质瘤患者,其总体生存期(OS)(HR=1. 97,95%CI:0. 56-6. 93,P=0. 29)、无复发生存期(RFS)(HR=1. 25,95%CI:1. 06-1. 47,P=0. 008)和无病生存期(DFS)(HR:3. 42,95%CI:1. 58-7. 43,P=0. 002)。结论:术前NLR可能是GIST患者预后的生物标志物,而且NLR升高的GIST患者预后较差,临床上应重视NLR指标,使胃肠间质瘤患者得到有效的治疗。 展开更多

目的探讨抗中性粒细胞胞质抗体(ANCA)对中性粒细胞表面FcγⅡ/Ⅲ(CD32/CD16)受体表达的影响及临床意义。方法提纯活动期ANCA相关性小血管炎(AASV)患者的ANCAIgG,分离健康人新鲜中性粒细胞,以ANCA刺激粒细胞1h后,流式细胞仪检... 目的探讨抗中性粒细胞胞质抗体(ANCA)对中性粒细胞表面FcγⅡ/Ⅲ(CD32/CD16)受体表达的影响及临床意义。方法提纯活动期ANCA相关性小血管炎(AASV)患者的ANCAIgG,分离健康人新鲜中性粒细胞,以ANCA刺激粒细胞1h后,流式细胞仪检测CD32/CD16的表达。流式细胞仪检测18例系统性小血管炎患者CD32/CD16的表达,以35名健康人为对照。同时对18例患者临床表现及血管炎活动情况进行评分,与CD32/CD16表达进行相关分析。结果ANCAIgG与正常人IgG相比显著上调中性粒细胞表面CD16的表达(Mnx67±23vss54±21,P〈0.01),但对CD32的表达无影响(Mnx21±8vs16±8,P〉0.05)。系统性小血管炎患者CD16表达的平均荧光强度显著高于正常人(Mnx62±12vs53±10,P〈0.01),而CD32的表达与正常人差异无统计学意义。CD16表达与伯明翰小血管炎呈显著正相关(P〈0.01)。结论ANCA相关性小血管炎患者高表达Fcγ受体,ANCA可以干预CD32/CD16受体的表达,调控Fcγ受体表达可能是系统性小血管炎的发病机制之一,监测CD16的表达水平对于判断疾病活动具参考价值。 展开更多

复方克感灵治疗急性热病作用机理的实验研究

[目的]以现代医学微观方法探讨复方克感灵作用机理。[方法]使用大肠杆菌内毒素制造外感发热家兔动物模型30只随机分为五组,以插管灌胃形式给药。实验过程中,每只家兔分别于造模后0.5h、1h、1.5h及给药后1h、2h、3h时各测体温1次。每只家兔的血液标本分别于造模前、发热高峰时及用药后3h采集。[结果]实验得出复方克感灵高剂量组的疗效最好。复方克感灵治疗急性外感热病具有能降低外周血白细胞总数、嗜中性粒白细胞比例,血浆中内生致热原(肿瘤坏死因子-α和白细胞介素1)的含量,改善血液流变,提高红细胞的免疫功能,从而起到增强免疫力、退热、抗感染的作用。[结论]以现代医学微观方法探讨复方克感灵作用机理,从而为其治疗急性外感热病提供了客观依据。 展开更多

The aetiology of primary sclerosing cholangitis (PSC) is not known and controversy exists as to whether PSC should be denominated an autoimmune disease. A large number of autoantibodies have been detected in PSC patients,but the specifi city of these antibodies is generally low,and the frequencies vary largely between different studies. The presence of autoantibodies in PSC may be the result of a nonspecifi c dysregulation of the immune system,but the literature in PSC points to the possible presence of specifi c antibody targets in the biliary epithelium and in neutrophil granulocytes. The present review aims to give an overview of the studies of autoantibodies in PSC,with a particular emphasis on the prevalence,clinical relevance and possible pathogenetic importance of each individual marker. 展开更多

Yet, inap-propriate neutrophil activation causes tissue damage andvarious inflammatory diseases. These physiological andpathological functions of neutrophils depend on the en-gagement of certain surface receptors, especially αMβ2,the ma jor β2 integrin receptor present on neutrophil sur-face. Understanding of the molecular mechanisms under-lying ligand binding by αMβ2, as well as the roles of αMβ2-ligand interactions in neutrophil functions will enable usto regulate more precisely neutrophil activities: that is,to promote their host defense functions, and at the sametime to minimize their deleterious effects on normal cells. 展开更多

Helicobacter pylori (H. pylori) virulence factors pro- mote the release of various chemoattractants/inflam- matory mediators, including mainly the neutrophil- attractant chemokine interleukin-8 and neutrophil- activating protein (NAP), involved in H. pylor/-induced gastric pathologies. Co-administration of Chios mastic gum (CMG), which inhibits H. pylor/NAP, with an H. pylori eradication regimen might add clinical benefits against H. pylori-related gastric pathologies, but pos- sibly not CMG as main therapy. Although H. pylori NAP and other H. pylori-related cytotoxins [i.e., vaculating cytotoxin (VacA)] appear to play a major role in gener- ating and maintaining the H. pylori-associated gastric inflammatory response and H. pylor/NAP is a promising vaccine candidate against H. pylori infection (H. pylori-1), concerns regarding its potential drawbacks, particularly neurogenic ones, due to possible cross- mimicry, should be considered. Possible cross-mimicry between H. p, vlor/ NAP and/or bacterial aquaporin (AQP) and neural tissues may be associated with the anti-AQP-4 antibody-related neural damage in multiple sclerosis (MS)/neuromyelitis optica patients. Moreover, the sequence homology found between H. pylori VacA and human Na+/K+-ATPase A subunit suggests that antibodies to VacA involve ion channels in abaxonal Schwann cell plasmalemma resulting in demyelination in some patients. A series of factors have been im- plicated in inducing blood-brain barrier (BBB) disrup- tion, including inflammatory mediators (e.g., cytokines and chemokines induced by H. pylor/-I) and oxidative stress. BBB disruption permits access of AQP4-specific antibodies and T lymphocytes to the central nervous system, thereby playing a major role in multiple sclero- sis pathogenesis. Relative studies show a strong asso- ciation between H. pylori-I and MS. H. pylor/-I induces humoral and cellular immune responses that, owing to the sharing of homologous epitopes (molecular mim- icry), cross-react with components of nerves, thereby contributing and perpetuating neural tissue damage. Finally, H. pylori NAP also plays a possible pathoge- netic role in both gastric and colon oncogenesis. 展开更多

AIM: To detect and evaluate the antibodies against Helicobacter pylori (H pylori) neutrophil-activating protein (HP-NAP) in patients with gastric cancer and other gastroduodenal diseases.METHODS: Recombinant HP-NAP was prepared from a prokaryotic expression system in Escherichia coll. Serum positivity and level of HP-NAP-specific antibodies in sera from 43 patients with gastric cancer, 28 with chronic gastritis, 28 with peptic ulcer, and 89 healthy controls were measured by rHP-NAP-based ELISA. rHP-NAP-stimulated production of interleukin-8 (IL-8) and growth-related oncogene (GROα) cytokines in the culture supernatant of SGC7901 gastric epithelial cells was also detected.RESULTS: The serum positivity and mean absorbancevalue of HP-NAP-specific antibodies in the gastriccancer group (97.7% and 1.01 ± 0.24) were significantly higher than those in the chronic gastritisgroup (85.7% and 0.89 ± 0.14, P 〈 0.005) and healthy control group (27.7% and 0.65 ± 0.18, P 〈 0.001). The sensitivity and specificity of ELISA for the detection of HP-NAP-specific antibodies were 95.5% and 91.5%, respectively. HP-NAP could slightly upregulate IL-8 production in gastric epithelial cell lines but had no effect on GROα production.CONCLUSION: Infection with virulent H py/ori strains secreting HP-NAP is associated with severe gastroduodenal diseases, and HP-NAP may play a role in the development of gastric carcinoma, rHP-NAP- based ELISA can be used as a new method to detect H pylori infection. The direct effect of HP-NAP on gastric epithelial cells may be limited, but HP-NAP may contribute to inflammatory response or carcinogenesis by activating neutrophils. 展开更多

The aim of this study was to evaluate morphometric and hematological parameters of juveniles Piracanjubas treated with two distinct photoperiod. Twenty nine fish with 100 g, in a factorial design were performed in 18 h light:6 h dark (18L:6D) and 6L:lSD two different photoperiod, and morphometric and hematological parameters were evaluated at 45 d and 90 d after photoperiod treatment. Benzocaine 1% was used for euthanasia. Fish were weighed and measured, so as visceral fat, liver and gonads. Hematocrit, total count of white cells, lymphocytes and neutrophils were counted. Results showed that there was no significant relationship between bodyweight and standard length. Significant interaction (P = 0.00018) had been found between the fat index (FI) and gonado-somatic index (GSI) and the photoperiod and time. At 90 d, the fish kept in 18L:6D had a lower rate of fat than the fish kept in 6L:18D (P 〈 0.05). The GSI was higher in the group 6L.'18D compared with the 18L:6D (P 〈 0.005). There was no interaction between photoperiod and period of treatment for total leukocyte count, however with 90 d, the total leukocyte count was decreased (P 〈 0.05). Moreover, decrease of the hematocrit from 58.1% to 46.9% and an increase of lymphocytes from 31.5% to 43.5% had been found in fish kept in 18L:6D after 90 d (P 〈 0.05). Fish kept in 6L:ISD presented an increase in segmented neutrophils and lymphocytes (P 〈 0.05). It is concluded that photoperiod maintained at 6L: 18D for Piracanjubas permits a greater gonadal development and an amount of immune cells over the photoperiod maintained in 18L: 6D. 展开更多

Abstract:Objective To map the epitopes on Wegener's granulomatosis autoantigen proteinase 3.Methods Antigenicity of proteinase 3 was studied with Western blot analysis in which proteinase 3 was prepared under reducing and non-reducing conditions. Two anti-proteinase 3 monoclonal antibodies, HZ1F12 and HZ1H3, were used to inhibit each other and to inhibit 22 anti-proteinase 3 positive sera from patients with Wegener's granulomatosis in competitive inhibition enzyme-linked immunosorbent assays (ELISA) and Western blot analysis. Results All monoclonal antibodies and patient sera recognized proteinase 3 under non-reducing conditions in Western blot analysis. HZ1F12 was inhibited 74% by HZ1H3. 10/22 (46%) sera were completely or partially inhibited by HZ1F12; 9/22 (41%) sera were partially inhibited by HZ1H3; and 6/22 (27%) were inhibited by both monoclonal antibodies. In inhibition Western blot analysis, the binding of patient sera to proteinase 3 could be inhibited by HZ1F12. Conclusions The epitopes of Wegener's granulomatosis autoantigen were conformational. Anti-proteinase 3 monoclonal HZ1F12 and HZ1H3 recognized similar or overlapping epitopes on the proteinase 3 molecule. Epitopes of proteinase 3 recognized by anti-proteinase 3 positive sera were not restricted. 展开更多
