The neutralizing antibodies against HPV-16, -18, -58, -45, -6 and -11 in serum samples were tested by using pseudovirus-based neutralization assays and HPV DNAs in cervical swabs were also tested by using a typing kit that can detect 21 types of HPV. The results revealed that 36% (18/50) of sera were positive for type-specific neutralizing antibodies with a titer range of 160-2560, of which 22%(11/50), 12%(6/50), 10%(5/50), 4%(2/50), 4%(2/50) and 2%(1/50) were against HPVs -6, -16, -18, -58, -45 and -1 l, respectively. Additionally, 60% (30/50) of samples were HPV DNA-positive, in which the most common types detected were HPV-68(18%), HPV-16(14%), HPV-58(12%), HPV-33(8%) and HPV-6, HPV-11, HPV-18 and HPV-52 (6% each). The concordance between HPV DNA and corresponding neutralizing antibodies was 56% (28/50) with a significant difference (P〈0.05). The full-length sequences of five HPV types (HPV -42, -52, -53, -58 and -68) were determined and exhibited 98%-100% identities with their reported genomes. The present data may have utility for investigating the natural history of HPV infection and promote the development of HPV vaccines.展开更多

The nonuniqueness can be largely reduced using Lame imp...Seismic attributes, such as P- and S-wave velocity, Poisson's ratio, and acoustic impedances, all generally can be used for distinguishing different rock types. The nonuniqueness can be largely reduced using Lame impedances instead of acoustic impedances as additional constraints. We have followed this method to constitute a petrologic composition model of the upper crust in the Bohai Bay basin, China. We briefly review the seismic parameters used for discrimination of rock types and focus our attention on the sensitivity of different combinations of parameters to determine the composition of materials. Corrections for pressure and temperature are performed in order to compare elastic wave velocities and densities measured at room temperature and surface pressure in laboratory with those for representative rock parameters. In a second step, we find the rock classes in the tested area by contrasting known data to laboratory measurements on a variety of rock samples extracted in the area. The basic field data are P-wave velocity values collected along a seismic profile conducted in the Bozhong Depression. The different rock types belonging to a particular rock class are finally constrained by the seismic velocities, Poisson's ratio, density, acoustic impedance, and Lame imoedance related to the topmost 10 km of the Bohai Bav crust.展开更多

METHODS: Twenty two 6-8 wk old HBV TGM in the third generation were d...AIM: To study the antiviral effect of Chinese medicine jiaweisinisan (JWSNS) on hepatitis B virus (HBV) infection in transgenic mice (TGM). METHODS: Twenty two 6-8 wk old HBV TGM in the third generation were divided into TGM control group and TGM treated group randomly. The normal control group included ten normal BC 57L/6 mice at the same age. The mice in treated group were administrated with JWSNS at the concentration of 4 g/mL and the dosage of 50 g/kg per d for 30 d, while the mice in TGM control group and normal control group were administrated with normal saline at the same dosage and the same time. Polymerase chain reaction (PCR) was used to assess the contents of HBV DNA in serum of HBV TGM before and after treatments, whereas blot hybridization was utilized to measure the contents of HBV DNA in the liver of both HBV TGM and normal BC 57L/6 mice. RESULTS: The levels of serum HBV DNA in TGM treated group were remarkably decreased after the treatment of JWSNS (7.662±0.78 vs 5.22±3.14, P〈0.05), while there was no obvious change after administration of normal saline in TGM control group (7.125±4.26 vs 8.932 ± 5.12, P〉 0.05). The OD values of HBV DNA in the livers of the mice in TGM treated group were significantly lower than those of TGM control group (0.274±0.096 vs 0.432 ± 0.119, P 〈 0.01). CONCLUSION: JWSNS exerts suppressive effects on HBV DNA in the serum and liver of TGM.展开更多

Objective To find neutralizing antibody candidates against hepatitis C virus (HCV) infection.Methods We constructed two eukaryotic expression vectors which contained the E1 and E2 gene of HCV, and detected their exp...Objective To find neutralizing antibody candidates against hepatitis C virus (HCV) infection.Methods We constructed two eukaryotic expression vectors which contained the E1 and E2 gene of HCV, and detected their expression in mammalian cells with transient expression. BALB/c mice were given subculaneous injections of constructed vectors combined with the IL-2 gene intraepidermally and evaluated for induced humoral immune responses by enzyme-linked immunosorbent assay (ELISA). We used an antibody-virus interaction assay to analyze the interaction of the antisera and HCV viral particles in vitro.Results Anti E1 and anti-E2 antisera were obtained from immunized mice. The serum of mice immunized with the E2 gene immunoprecipitated the HCV isolate in source serum and reacted with the isolates unrelated to the original one.Conclusions Anti-E2 antibody in induced mice can cross-reactively capture HCV particles, highlighting the possibility of generating broadly reactive anti-E2 antibodies.展开更多

Several in vitro and in vivo studies have proved that antioxidant could act as neuroprotective agent through intervening with flee-radical mediated apoptosis in the ischemic penumbra. In particular, natural products which contain antioxidant properties have undoubtedly potential for stroke treatment. In the present study, therapeutic effects of Braintone on Wistar rats undergone middle cerebral artery occlusion (MCAO) was evaluated. Gene expression levels of pro-apoptotic genes (AT2 receptor, FAS, BAX and BCL-XS) were showed to be significantly reduced in Braintone treated groups (0.4-, 0.72-, 0.76-, 0.32-fold, P〈0.05) as compared to vehicle group. Significant reduction ofimmunoreactivity of protein production of these genes, together with least nuclear green fluorescence observed in TUNEL, Braintone as an antioxidant drug, is concluded to have promising therapeutic effect for stroke treatment.展开更多
