生地注射液对中风模型神经元保护的实验研究 被引量:6

目的运用凝血酶复制中风后神经元损伤模型,观察生地注射液对该损伤神经元的保护作用。方法将培养7 d后的胎鼠大脑皮层神经元分为3组,即正常对照组、模型组与中药组,继续培养24 h后检测培养液中乳酸脱氢酶(lactic dehydrogenase,LDH)活... 目的运用凝血酶复制中风后神经元损伤模型,观察生地注射液对该损伤神经元的保护作用。方法将培养7 d后的胎鼠大脑皮层神经元分为3组,即正常对照组、模型组与中药组,继续培养24 h后检测培养液中乳酸脱氢酶(lactic dehydrogenase,LDH)活性与存活率指标(methyl thiazdyl tetrazolium,MTT)值。结果正常组与中药组LDH值较模型组低,与后者比较具统计学意义(P<0.01);正常组与中药组MTT值较模型组高,与后者比较亦具统计学意义(P<0.01)。结论生地注射液能够减轻凝血酶导致的神经元损伤,提高神经元损伤后的细胞存活率,从而显示了较好的神经元保护作用。 展开更多

中风具有高致死率、致残率及复发率的特点,一直是人类未能攻克的疾病。中风主要分为出血性中风和缺血性中风,后者占所有中风事件的百分之八十。科学家们一直致力于开发有效的预防治疗中风的药物,方便实用的模型是病理机制研究和药物的... 中风具有高致死率、致残率及复发率的特点,一直是人类未能攻克的疾病。中风主要分为出血性中风和缺血性中风,后者占所有中风事件的百分之八十。科学家们一直致力于开发有效的预防治疗中风的药物,方便实用的模型是病理机制研究和药物的筛选过程中非常重要的支撑和保障。在众多缺血性中风模型中,光照血栓模型以操作简便且创伤性小、不需要开颅手术、死亡率低。等独特的优势得到了越来越多的认可。该模型可通过调整激光照射大脑区域内的位置来控制血栓形成的部位,通过调整造模染料的剂量及激光照射的强度和时间来控制缺血灶形成的大小,适合于缺血性中风过程中神经退行性病变,神经保护和神经再生的细胞和分子机制研究及抗中风药物的开发。本文对光照血栓性中风模型的原理、优缺点以及在药物开发中的应用进行了叙述。 展开更多

目的从下调脑区的激活,研究复康片治疗缺血性中风后遗症的作用机制。方法电凝法制备MCAO大鼠模型。将180只大鼠随机分为模型组、药物组和假手术组。于造模成功后5周,药物组按4.5g生药/kg灌胃给予复康片,余两组分别灌胃给予同等量生理盐... 目的从下调脑区的激活,研究复康片治疗缺血性中风后遗症的作用机制。方法电凝法制备MCAO大鼠模型。将180只大鼠随机分为模型组、药物组和假手术组。于造模成功后5周,药物组按4.5g生药/kg灌胃给予复康片,余两组分别灌胃给予同等量生理盐水,1日1次,共2周。以BWT评分法进行神经行为学评价,结合局部脑血流量(rCBF)测定、免疫组化和原位杂交法分别观察大鼠脑组织生长相关蛋白(GAP43)、突触素P38(SYN)的表达和大鼠脑微血管内皮细胞ET1、ecNOS、P gpmRNA的变化。结果模型组大鼠病灶远隔区存在低灌注现象,突触活性降低,脑微血管内皮细胞对脑微循环的调节机能下降;药物组可明显改善这种低活动状态,并能显著提高大鼠运动功能。结论复康片对缺血性中风后遗症模型神经功能有显著改善作用,激活下调脑区是其重要机制。 展开更多

结合光学相干层析技术的三维成像能力和动态散射技术的运动识别能力,可以实现无标记的三维光学微血管造影,在不牺牲线扫描速度的前提下,通过帧间分析的方法提高血流造影的灵敏度,实现毛细血管水平的探测.提出小波域分量复合的方法降低... 结合光学相干层析技术的三维成像能力和动态散射技术的运动识别能力,可以实现无标记的三维光学微血管造影,在不牺牲线扫描速度的前提下,通过帧间分析的方法提高血流造影的灵敏度,实现毛细血管水平的探测.提出小波域分量复合的方法降低静态组织信号和动态血流信号之间的分割误差,实现高对比度的血管造影分别利用组织血流模拟样品和活体大鼠脑组织进行实验验证,结果发现,采用小波域分量复合之后,血管分割误差分别减小了83%和71%,造影图对比度增强,并且具有更好的血管连接性.进而,利用研制的系统对大鼠脑血管局部缺血性中风模型进行了初步的成像研究,清晰地呈现了中风模型形成,血管受损和血管恢复的整个过程,有助于对局部缺血性中风模型机理的研究. 展开更多

The output of 25 models used in the Coupled Model Intercomparison Project phase 3 (CMIP3) were evaluated,with a focus on summer precipitation in eastern China for the last 40 years of the 20th century.Most mod-els failed to reproduce rainfall associated with the East Asian summer monsoon (EASM),and hence the seasonal cycle in eastern China,but provided reasonable results in Southwest (SW) and Northeast China (NE).The simula-tions produced reasonable results for the Yangtze-Huai (YH) Basin area,although the Meiyu phenomenon was underestimated in general.One typical regional phe-nomenon,a seasonal northward shift in the rain belt from early to late summer,was completely missed by most models.The long-term climate trends in rainfall over eastern China were largely underestimated,and the ob-served geographical pattern of rainfall changes was not reproduced by most models.Precipitation extremes were evaluated via parameters of fitted GEV (Generalized Ex-treme Values) distributions.The annual extremes were grossly underestimated in the monsoon-dominated YH and SW regions,but reasonable values were calculated for the North China (NC) and NE regions.These results suggest a general failure to capture the dynamics of the EASM in current coupled climate models.Nonetheless,models with higher resolution tend to reproduce larger decadal trends and annual extremes of precipitation in the regions studied. 展开更多

AIM: To explore the association between single nucleotide polymorphisms (SNPs) at 8q24 and gastric cancer risk. METHODS: A case-control investigation including 212 gastric cancer patients and 377 healthy controls was conducted. The genotypes of SNPs (rs6983267, rs7008482 and rs10808555) were examined and established through polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Multivariate logistic regression models were used to evaluate the association between SNPs and gastric cancer. RESULTS: The genotype frequencies of rs6983267 in gastric cancer patients were obviously different from those in the control (P = 0.005). GT genotype of rs6983267 was associated with an increased risk of gastric cancer compared with GG genotype (adjusted odds ratio = 2.01, 95% confidence interval: 1.28-3.14). Further stratified analysis indicated that rs6983267 GT genotype facilitated the risk of gastric cancer of non-cardiac and intestinal type (OR: 2.638, 95% CI: 1.464-4.753; OR: 1.916, 95% CI: 1.166-3.150, respectively). CONCLUSION: This study demonstrates for the first time that rs6983267 is involved in susceptibility to gastric cancer, although further large-sample investigations are still needed. 展开更多

The healthy development of SMEs is very important, whereas, SMEs’ financing is a worldwide problem, and the result of financing game between banks and enterprises depends on a variety of decision-making factors. Free radical induced neuronal damage is involved in stroke. Several in vitro and in vivo studies have proved that antioxidant could act as neuroprotective agent through intervening with flee-radical mediated apoptosis in the ischemic penumbra. In particular, natural products which contain antioxidant properties have undoubtedly potential for stroke treatment. In the present study, therapeutic effects of Braintone on Wistar rats undergone middle cerebral artery occlusion (MCAO) was evaluated. Gene expression levels of pro-apoptotic genes (AT2 receptor, FAS, BAX and BCL-XS) were showed to be significantly reduced in Braintone treated groups (0.4-, 0.72-, 0.76-, 0.32-fold, P〈0.05) as compared to vehicle group. Significant reduction ofimmunoreactivity of protein production of these genes, together with least nuclear green fluorescence observed in TUNEL, Braintone as an antioxidant drug, is concluded to have promising therapeutic effect for stroke treatment. 展开更多

OBJECTIVE: To observe the effect of tonifying liver and kidney-essence herbs on expression of a nerve regeneration inhibitor, Nogo for neuron A (No- go-A), and its associated signaling molecule, low-af- finity neurotrophin receptor p75 (p75TR), in rats with cerebral ischemic stroke (CIS), with the aim of exploring the possible mechanism of tonifying liver and kidney-essence herbs in recovery following in- jury to the central nervous system. METHODS: A cerebral ischemic stroke model in SD rats was established with the suture-occlusion method. Successful model rats were divided into placebo and herb groups at random; sham-operat- ed and control groups were set up simultaneously.Each of these groups was divided into six sub- groups at random. Expression of Nogo-A and p75TR was evaluated with immunofluorescence microsco- py at days 3, and weeks1, 2, 3, 4 and 8 after adminis- tration. RESULTS: Tonifying liver and kidney-essence herbs suppressed the expression of Nogo-A and p75T (P〈0.05 and P〈0.01, respectively). CONCLUSION: Suppressing the expression of No- go-A and p75NTR is possibly one of the mechanisms underlying the ability of tonifying liver and kid- ney-essence herbs to promote recovery of the in- jured central nervous system. 展开更多
