It was proved benign disease by surgen hand check and puncture cytology, then divided them into two groups randomly. One group was Ru Zeng Nin capsule, another one was comparing. It was observed the condition about pain? tenderness? swelling piece after taking capsule half a year. The results showed that effective rate was 97 8%, cure rate was 80%. From this, we Rnow that Ru Zeng Nin capsule has better treatment effect to treast pain? mammary gland hypprplasia and fiber hyperplasia symptom. This capsule has safe and taking eary characteristic, so it’s easy to spread and opply in the clinical.展开更多
目的:基于网络药理学探讨乳增宁胶囊治疗乳腺增生的作用机制并对核心靶蛋白进行实验验证。方法:通过中药系统药理学数据库平台(TCMSP)检索乳增宁胶囊所有药物的化学成分和相关作用靶点;采用GeneCard数据库检索乳腺增生的相关靶基因;后采...目的:基于网络药理学探讨乳增宁胶囊治疗乳腺增生的作用机制并对核心靶蛋白进行实验验证。方法:通过中药系统药理学数据库平台(TCMSP)检索乳增宁胶囊所有药物的化学成分和相关作用靶点;采用GeneCard数据库检索乳腺增生的相关靶基因;后采用Perl语言软件筛选药物与疾病共同的靶点及药物的有效成分,并将药物与疾病共同靶蛋白导入STRING与Cytoscape 3.7.2软件构建蛋白互作关系网络(PPI),采用Cytoscape 3.7.2软件构建药物有效成分与疾病靶基因互作关系网络;采用R语言软件进行基因本体论(GO)及京都基因与基因组百科全书(KEGG)集分析,并用Cytoscape 3.7.2软件构建KEGG富集通路与富集蛋白互作网络关系,最后对富集的核心靶蛋白进行动物实验验证。结果:共检索到乳增宁胶囊有效成分76个、相关作用靶点242个;共检索到乳腺增生靶基因665个;GO分析显示乳腺增生的生物学过程主要涉及对金属离子的反应(response to metal ion)、对类固醇激素的反应(response to steroid hormone)、对氧化应激的反应(response to oxidative stress)、对细胞氧化应激的反应(cellular response to oxidative stress)、对辐射的反应(response to radiation)等20个;KEGG富集分析发现主要通路有PI3K-Akt信号通路(PI3K/AKT)、卡波西氏肉瘤相关疱疹病毒感染(KSHV)、乙型肝炎(HB)、人巨细胞病毒感染(HCMV)等20个;KEGG富集通路与富集蛋白互作网络结果发现主要核心靶蛋白有丝裂原激活的蛋白激酶1(MAPK1),PKB蛋白激酶1(AKT1)和细胞周期蛋白(CCND1)等;动物实验结果发现乳增宁胶囊对乳腺组织过度增生有显著的抑制作用。结论:乳增宁胶囊治疗乳腺增生具有多途径、多靶点的特点,并且能够显著抑制乳腺组织的过度增生。展开更多
