口蹄疫乳鼠毒在蚯蚓和苍蝇中实验带毒的研究 被引量:2

O型口蹄疫(FMD)乳鼠毒注射到蚯蚓(Pheretima sp.)体内后的第8天仍可检测到口蹄疫病毒(FMDV),但第14天以后则未查出。将此病毒洒在蚯蚓生活的土壤中,检测蚯蚓体内FMDV的结果亦同。本试验还间接地表明,FMDV不能在蚯蚓体内复制、增殖,蚯蚓... O型口蹄疫(FMD)乳鼠毒注射到蚯蚓(Pheretima sp.)体内后的第8天仍可检测到口蹄疫病毒(FMDV),但第14天以后则未查出。将此病毒洒在蚯蚓生活的土壤中,检测蚯蚓体内FMDV的结果亦同。本试验还间接地表明,FMDV不能在蚯蚓体内复制、增殖,蚯蚓只起携带FMDV的作用。蚯蚓属于低等动物,免疫系统尚未健全,但对非已物质有一定的免疫应答。蚯蚓与FMDV接触后,用间接血凝测得其体腔液中具有“抗体”反应。FMDV与舍蝇属(Muscasp.)和金蝇属(Chrysomyia sp.)的苍蝇接触24小时后,对其带毒检测结果表明:苍蝇只能短时在体表携带FMDV,且苍蝇体表携带FMDV的量很少。故认为在FMDV传播中苍蝇只起短时机械扩散作用。 展开更多

目的:研究肝豆扶木汤(GDFMT)对Wilson病肾纤维化小鼠的保护作用及其可能作用机制。方法:将60只成年雄性毒乳鼠(TX)随机分成模型组、GDFMT高、中、低剂量组、青霉胺组,另12只作为正常组的野生小鼠共6组。GDFMT高、中、低剂量组分别予以13... 目的:研究肝豆扶木汤(GDFMT)对Wilson病肾纤维化小鼠的保护作用及其可能作用机制。方法:将60只成年雄性毒乳鼠(TX)随机分成模型组、GDFMT高、中、低剂量组、青霉胺组,另12只作为正常组的野生小鼠共6组。GDFMT高、中、低剂量组分别予以13.92、6.96、3.48 g·kg^(-1),青霉胺组0.1 g·kg^(-1),模型组及正常组使用等量0.9%氯化钠溶液灌胃,每天1次,连续给药4周。酶联免疫吸附测定法(ELISA)检测血清中血尿素氮(BUN),肌酐(CRE),Ⅲ型前胶原(PC-Ⅲ),Ⅳ型胶原(C-Ⅳ)的含量。苏木素-伊红(HE)和马松(Masson)染色的小鼠肾的病理形态学改变。免疫荧光法测定肾细胞内瘦素(leptin)、Janus激酶2(JAK2)及信号传导及转录激活蛋白(STAT)蛋白表达情况;实时荧光定量聚合酶链式反应(Real-time PCR)测定leptin、瘦素受体(OB-R)、JAK2及STAT mRNA的情况。蛋白免疫印迹法(Western blot)检测转化生长因子-β(1 TGF-β_(1))、单核细胞趋化蛋白-1(MCP-1)表达情况。结果:与正常组比较,模型小鼠BUN、CRE、PC-Ⅲ、C-Ⅳ水平显著增加(P<0.01),与模型组比较,GDFMT高、中剂量组和青霉胺组含量明显降低(P<0.05,P<0.01),GDFMT高剂量组显著减少(P<0.01)。肾组织病理形态结果表明,模型组肾纤维组织增生,用药干预后肾组织病理损害不同程度减轻,纤维化改善。免疫荧光结果显示,模型组leptin、JAK2及STAT3蛋白肾纤维化高表达,GDFMT干预后,荧光强度降低,GDFMT高剂量组荧光强度最低。Real-time PCR结果显示,与正常组比较,模型leptin、OB-R、JAK2、STAT3 mRNA表达水平明显升高,与模型组比较,GDFMT中、高剂量组及青霉胺组表达水平明显降低(P<0.05,P<0.01)。Western blot结果显示,与正常组比较,模型组TGF-β_(1)、MCP-1表达量显著上升(P<0.01),与模型组比较,GDFMT中、高剂量组表达水平显著降低(P<0.01)。结论:GDFMT可以减轻TX小鼠肾纤维化损伤,其机制可能是通过调控leptin及JAK/STAT信号通路表达有关。 展开更多

以10000 ID50剂量的乳鼠肌肉毒通过颈部肌肉注射途径攻击牛,取代用牛舌皮强毒在牛舌面涂抹和上唇划痕各1ml的方法,进行了牛O型口蹄疫灭活疫苗效力检验试验。结果对照组牛3/3发病,免疫组牛均100%保护。

AIM:To study the effects of live and dead Lactobacillus rhamnosus GG(GG) on rotavirus infection in a neonatal rat model.METHODS:At the age of 2 d,suckling Lewis rat pups were supplemented with either live or dead GG and the treatment was continued daily throughout the experi-ment.At the age of 5 and 6 d the pups received oral rotavirus(RV) SA-11 strain.The pups were sacrificed at the age of 7 or 8 d by decapitation.The gastrointestinal tract was removed and macroscopic observations were done.The consistency of feces in the colon was classified using a four-tier system.RV was detected from the plasma,small intestine,colon and feces by real-time quantitative polymerase chain reaction(PCR).RESULTS:In this neonatal rat model,RV induced a mild-to-moderate diarrhea in all except one pup of the RV-inoculated rats.RV moderately reduced body weight development from day 6 onwards.On day 7,after 2 d of RV infection,live and dead GG groups gained significantly more weight than the RV group without probiotics [36%(P = 0.001) and 28%(P = 0.031),respectively].In addition,when compared with the RV control group,both live and dead GG reduced the weight ratio of colon/animal body weight to the same level as in the healthy control group,with reductions of 22%(P = 0.002) and 28%(P < 0.001),respectively.Diarrhea increased moderately in both GG groups.However,the diarrhea incidence and severity in the GG groups were not statistically significantly different as compared with the RV control group.Moreover,observed diarrhea did not provoke weight loss or death.The RV control group had the largest amount of RV PCR-positive samples among the RV-infected groups,and the live GG group had the smallest amount.Rats receiving live GG had significantly less RV in the colon(P = 0.027) when compared with the RV control group.Live GG was also more effective over dead GG in reducing the quantity of RV from plasma(P = 0.047).CONCLUSION:Both live and dead GG have beneficial effects in RV infection.GG may increase RV clearance from the body and reduce colon swelling. 展开更多

An optimized recombinant HPV16 E6E7 fusion gene (HPV16 ofE6E7) was constructed according to codon usage for mammalian cell expression, and a mutant of HPV16 ofE6E7 fusion gene (HPV16 omfE6E7) was generated by site-directed mutagenesis at L57G, C113R for the E6 protein and C24G, E26G for the E7 protein for HPV16 ofE6E7 [patent pending (CN 101100672)]. The HPV16 omfE6E7 gene constructed in this work not only lost the transformation capability to NIH 3T3 cells and tumorigenicity in SCID mice, but also maintained very good stability and antigenicity. These results suggests that the HPV16 omfE6E7 gene should undergo further study for application as a safe antigen-specific therapeutic vaccine for HPV16-associated tumors. 展开更多
