隐睾发生过程中DNA甲基化状态和甲基化转移酶表达的实验研究

目的研究环境内分泌干扰物邻苯二甲酸二(2-乙基已基)酯[Di—(2-ethylhexy)phthalate,DEHP]作用于孕鼠后,胎鼠睾丸组织基因组DNA甲基化水平的变化及3种DNA甲基化转移酶的表达情况;探讨DEHP所致表观遗传修饰改变在睾丸下降过程中... 目的研究环境内分泌干扰物邻苯二甲酸二(2-乙基已基)酯[Di—(2-ethylhexy)phthalate,DEHP]作用于孕鼠后,胎鼠睾丸组织基因组DNA甲基化水平的变化及3种DNA甲基化转移酶的表达情况;探讨DEHP所致表观遗传修饰改变在睾丸下降过程中的作用。方法妊娠昆明小鼠(KMmice)随机分为3组,即正常对照组、玉米油对照组和DEHP实验组,每组15只。玉米油对照组和DEHP实验组自妊娠第12.5天到妊娠第18.5天分别持续经口予以玉米油和DEHP,用量按每日500mg/kg,正常对照组不予灌药。采用高效液相色谱仪检测基因组DNA甲基化水平,实时荧光定量PCR检测3种DNA甲基化转移酶的表达量。结果正常对照组和玉米油对照组DNA甲基化水平和3种甲基化转移酶的表达均无统计学意义(P〉0.05):DEHP实验组DNA甲基化水平比对照组增高约10%。与对照组相比,差异有显著统计学意义(P〈0.05);实验组3种甲基化转移酶的表达量均比对照组高,与对照组相比,差异有显著统计学意义(Dnmt1,P〈0.05;Dnmt3a,P〈0.01;Dnmt3b,P〈0.05)。结论环境内分泌干扰物DEHP通过影响3种甲基化转移酶的表达而导致基因组DNA甲基化水平升高,从而抑制睾丸下降过程中多种基因的表达,发生隐睾。DNA甲基化等表观遗传学修饰的改变可能是DEHP诱发隐睾的分子机制之一。 展开更多

A series of extraction equilibrium experiments for aminobenzoic acid with di(2-ethylhexyl)phosphoric acid (D2EHPA) dissolved in n-octane or 1-octanol was carried out. The effects of aminobenzoic acid concentration,D2EHPA concentration and pH on the distribution ratio were discussed in detail. The infrared spectra of the organic phase loaded with solute illustrated that pH had little effect on the structure of the complex formed. There proceed ion association and cation-exchange.reaction in the extraction. An expression of the equilibrium distribution was proposed. 展开更多

The dynamic mechanical properties and miscibility of an organic hybrid made from chlorinated polyethylene (CPE) and 2, 2'-methyIene-bis-( 4-methyl-6-cyclohexylphenol) ( ZKF ) are mainly discussed in this paper. It is found that ZKF acts as an antiplasticizer in CPE matrix and has good miscibility even with large ratio in CPE /ZKF hybrids. The glass transition temperature of various CPE /ZKF hybrids determined by DSC give a good fit to Wood's equation. Bifunctional ZKF is supposed to improve the intermolecular force of CPE, and the improvement is developed when the ZKF content increases. On the other hand, the viscoelastic properties are highly improved with the addition of ZKF. TA and tanδ peak values increase when the ZKF content increase in the CPE /ZKF hybrids, the damping capacity has been improved during the glass transition of CPE /ZKF hybrids. In addition, the glass transition temperature shifts to room temperature from the low temperature with the continuous addition of ZKF to CPE. 展开更多

The extraction of cadmium and zinc with di-(2-ethylhexyl)-dithiophosphoric acid(D2EHDTPA)-toluene and D2EHDTPA-trioctyl amine(TOA)-toluene has been investigated. Infrared spectrum analysis, the nuclear magnetic resonance spectrum(NMR) analysis and conductivity measurements confirm that the probable structures of the extracted complexes are CdA2 and ZnA2. Cadmium can be extracted by D2EHDTPA very easily. Almost all the cadmium can be extracted, but stripping of cadmium from the organic phase is very difficult. When a mixture of D2EHDTPA and TOA is used, selective extraction of cadmium from zinc sulfate solution can be achieved. Cadmium can be easily stripped from the organic phase. 展开更多
