油茶根系与内生细菌枯草芽孢杆菌互作早期的转录组分析 被引量:2

【目的】研究油茶-内生细菌互作早期的油茶根系转录组变化规律,为阐明油茶与内生细菌互作的分子机制提供依据。【方法】构建内生细菌枯草芽孢杆菌1-L-29gfpr与油茶根系互作体系,以不同时间(0、6、12、24 h)的互作体系为研究材料,利用RNA... 【目的】研究油茶-内生细菌互作早期的油茶根系转录组变化规律,为阐明油茶与内生细菌互作的分子机制提供依据。【方法】构建内生细菌枯草芽孢杆菌1-L-29gfpr与油茶根系互作体系,以不同时间(0、6、12、24 h)的互作体系为研究材料,利用RNA-Seq技术对油茶根部进行转录组测序及分析。【结果】1)转录组测序共产生52958922条序列,约46.1 Gb,差异表达基因10314条(FDR<0.05且|log_(2)FC|>1)。随着互作时间的延长差异表达基因数量呈高-低-高的趋势,接种6 h后差异表达基因数量为6306个,其中显著上调3434个,显著下调2572个;接种12 h后显著上调903个,显著下调526个;接种24 h后显著上调1195个,显著下调1384个。2)GO富集分析表明:接种6 h油茶的生物学过程、分子功能方面变化最为明显,生物学过程中磷酸代谢、含磷化合物代谢、响应激素途径、茉莉酸代谢、响应细菌途径等;分子功能中萜烯合酶活性、双加氧酶活性等;细胞组分中膜固有成分等得到富集。3)KEGG通路分析中差异表达基因主要被富集到碳水化合物代谢、氨基酸代谢、信号转导、环境适应性等通路。接种6 h后富集的代谢通路与12、24 h差异较大,其中差异明显的主要有植物激素合成信号通路、植物病原互作通路、苯丙素生物合成通路。4)基因表达量变化较大的有生长素诱导蛋白编码基因(SAUR)、木葡聚糖内糖基转移酶/水解蛋白酶编码基因(XTH)、茉莉酮酸酯ZIM结构域蛋白编码基因(TIFY)、钙调蛋白编码基因(CAM2)、转录因子MYB编码基因、抗病蛋白编码基因(RPM、RPS)、呼吸爆发氧化酶编码基因(RBOHC)、过氧化物酶编码基因(POD)。【结论】利用活体互作体系对不同时间段内生细菌-油茶互作转录组进行分析表明,内生细菌可刺激植物根系生长,提高抗性,诱导免疫反应,但随后免疫反应逐渐减弱。 展开更多

【目的】分离授粉早期玉米花丝的总蛋白质,为从蛋白质组角度揭示玉米花粉与花丝早期的相互作用奠定基础。【方法】以玉米(ZeamaysL.)自交系Ga25为试材,采用三氯乙酸-丙酮法提取总蛋白质,运用双向电泳、质谱分析与检索技术,比较自交授粉... 【目的】分离授粉早期玉米花丝的总蛋白质,为从蛋白质组角度揭示玉米花粉与花丝早期的相互作用奠定基础。【方法】以玉米(ZeamaysL.)自交系Ga25为试材,采用三氯乙酸-丙酮法提取总蛋白质,运用双向电泳、质谱分析与检索技术,比较自交授粉后1h和2h的花丝蛋白质组的差异。【结果】与授粉1h的蛋白质组相比,在授粉2h后的蛋白质组中出现28个差异蛋白点,其中,6个蛋白质点特异表达,19个蛋白质点上调表达,3个蛋白质点下调表达;通过MALDI-TOF-MS质谱测序和MASCOT序列分析,注释了23个蛋白质点,其余5个为未知蛋白;功能分类表明,差异蛋白质分别参与细胞壁合成(21.4%)、防御/抗胁迫(17.9%)、细胞组织、蛋白命运、蛋白合成、转录等细胞过程。【结论】在玉米花粉与花丝早期的相互作用过程中蛋白质组有显著的变化,与授粉后1h相比,授粉后2h的蛋白质组中大部分差异蛋白呈上调趋势,并有特异蛋白质出现;分泌型过氧化物酶、膨胀素、果胶甲基化酶抑制蛋白、谷胱甘肽转移酶与未知蛋白4可能与玉米花粉和花丝的早期互作密切相关。 展开更多

Early Stage above Ground Interaction in Wheat-Jatropha Agrisilviculture System

Experiments were conducted to analyze the initial tree crop interaction between Jatropha curcas and wheat by planting the perennial component in East-West orientation with 8-2 m distance and wheat as intercrop at National Research Centre for Agroforestry (NRCAF), Jhansi during July, 2007 to April, 2009. During planting of Jatropha the interaction to wheat was non-significant and influence of orientation is also nil. Eighteen months after plating Jatropha had attained the mean height of 124.56 cm, collar diameter of 6.37 cm, number of branches of 11.25, canopy diameter of 98.5 cm and yielded 18.5 g seeds per plant. During 18 MAP also observations revealed the non-significant influence of Jatropha on wheat. Observations also indicated the progress in wheat growth and yield attributes with the increase of distances from tree base and it follow the order of 0.5 m〈1.0 m〈2.0 m〈3.0, 4.0 m and open. Nearly, 62% and 14% yield loss in wheat grain was observed at 0.5 m and lm distance from the tree base respectively compare with open. Comparison ofwheat-Jatropha agrisilviculture system and sole wheat cropping indicated the net grain yield reduction up to 13.5% by agrisilviculture system. The comparison of wheat and straw yields between the two years clearly indicating that the system and sole cropping have not much differed while the influence of climatic factor is witnessed by the variation of yields over years. It revealed that the initial above ground interaction between Jatropha and wheat was due to the space that can be managed by the pruning of branches which is a recommended cultural practice of Jatropha. 展开更多

While a large number of studies have described animal social networks, we have a poor understanding of how these networks vary with ecological and social conditions. For example, reproductive periods are an important life-history stage that may involve changes in dominance relationships among individuals, yet no study to date has compared social networks of do- minance interactions (i.e. dominance networks) across reproductive contexts. We first analyzed a long-term dataset on captive so- cial groups of the cooperatively breeding cichlid Neolamprologuspulcher, and found that eviction events were significantly more common around reproduction than expected by chance. Next, we compared the structure of dominance networks during early pa- rental care and non-reproductive periods, using one of the first applications of exponential random graph models in behavioral biology. Contrary to our predictions, we found that dominance networks showed few changes between early parental care and non-reproductive periods. We found no evidence that dominance interactions became more skewed towards larger individuals, became more frequent between similar-sized individuals, or became more biased towards a particular sex during parental care. However, we did find that there were relatively more dominance interactions between opposite-sex dyads in the early parental care period, which may be a by-product of increased sexual interactions during this time. This is the first study in behavioral ecology to compare social networks using exponential random graph modeling, and demonstrates a powerful analytical framework for future studies in the field [Current Zoology 61 (1): 45-54, 2015]. 展开更多

The early phases of the shock interaction process on two-dimensional interfaces with different shapes are numerically investigated in this study,which are closely related to the shock refraction and reflection,vorticity production and transport.The numerical method employs an adaptive unstructured quadrilateral mesh,which can capture the wave pattern and interface evolution very well.Simulations are carried out under the conditions of an incident shock Mach number of 1.2 and the light/heavy (air/SF 6) interface.Five different shapes are considered in the simulations:rectangle,ellipse,diamond and two kinds of triangle.The results show that the interfacial shapes can influence the wave patterns particularly on the shape and evolution of refracted shock waves.The generation and the distribution of vorticity on the interfaces with five different shapes also have dissimilarities.The circulation deposition on five interfaces is quantitatively investigated and compared with theoretical model.A good agreement is found between the numerical results and the predictions by the theoretical model.Some characteristic scales of the interface are tracked.Under the influence of nonlinear-acoustic effect and vorticity effect,the interfaces present different evolution modes. 展开更多
