In order to improve the yield of MIL-C, statistically-based e...Cytosine-substituted mildiomycin analogue (MIL-C) was produced effectively by supplementing cytosine into the culture of Streptoverticillium rimofaciens. In order to improve the yield of MIL-C, statistically-based experimental designs were applied to optimize the fermentation medium for S. rimofaciens ZJU 5119. Fifteen culture conditions were examined for their significances on MIL-C production using Plackett-Burman design. The Plackett-Burrnan design and one-variable-at-a-time design indicated that glucose and rice meal as the complex carbon sources, and peanut cake meal and NH4NO3 as the complex nitrogen sources were beneficial for MIL-C production in S. rimofaciens ZJU 5119. The results of further central composition design (CCD) showed that the optimal concentration of glucose, rice meal and peanut cake meal were 18.7 g/L, 64.8 g/L and 65.1 g/L, respectively. By using this optimal fermentation medium, the MIL-C concentration was increased up to 1336.5 mg/L, an approximate 3.8-fold improvement over the previous concentration (350.0 mg/L) with un-optimized medium. This work will be very helpful to the large-scale production of MIL-C in the future.展开更多

[Method] 50 male mice were divided into 5 groups equally and randomly and among them,the three gr...[Objective] The antioxidation effect of Carya cathayensis sarg (CCS) Oil was qualitatively and quantitatively detected. [Method] 50 male mice were divided into 5 groups equally and randomly and among them,the three groups were either negative,or model control,or positive control (ginseng saponins),respectively. Two remaining groups were orally fed with low and high dosages of CCS oil ((9 ml/(kg·d) and (33 ml/(kg·d)),respectively. All groups were injected D-galactose through hypodermic except the negative control ones which were only treated with equals amount of distilled water. The superoxide dismutase (SOD),maleic dialdehyde(MDA) and monoamine oxidase(MAO) were analyzed with blood sample and brain tissue 42 days post treatment. [Result] It was showed that the activity of SOD enzyme was increased in the CCS oil groups,while MDA and MAO content were decreased in the CCS oil groups compared with that of other groups with significance observed in the group fed with high dose of CCS oil. [Conclusion] The result indicated that the CCS oil has the ability of antioxidation and is useful to human health.展开更多
The main purpose of this study was to detect an association of cytoplasmic signal transducers and activators of transcription-1 (STAT1) with milk production traits in 472 Holstein and 283 Jersey cattle breeds of Tur...The main purpose of this study was to detect an association of cytoplasmic signal transducers and activators of transcription-1 (STAT1) with milk production traits in 472 Holstein and 283 Jersey cattle breeds of Turkey. This gene, located on chromosome 2, was chosen due to its role on development of mammary gland. A polymorphism of selected 314 bp allele fragment was detected by the restriction fragment length polymorphism analysis of polymerase chain reaction-amplified fragments (PCR-RFLP) method and also confirmed by DNA sequencing. The association tests were conducted between STAT1 genotypes and some economically important dairy traits. The genotypes for C/T as a single nucleotide polymorphism (SNP) were identified at interval 60 cM to 63 cM. The effects of STAT1 gene on milk production traits were not significant in Holstein cows, although animals with CT genotypes showed fairly close to significant value for the corrected 305 d milk yield. However, Jersey cows with/7" genotype were 2.07 kg higher for test-day milk yield (P 〈 0.05), 0.13 kg for fat yield (P 〈 0.01) and 0.07 kg for protein yield (P 〈 0.05) compared with animals having CC and CT genotypes. Definitely, the further research should be conducted to search this gene intensively with larger samples to identify polymorphism and any association between the economically important traits and genotypic class in Holstein cows. Finally, based on the findings, it was concluded that STATI gene might be used as a potential candidate gene to improve milk yield and milk fat and protein contents in dairy cows breeding programs.展开更多
