Enantioselective extraction of hydrophobic clorprenaline (CPE) enantiomers from organic phase to aqueous phases with sulfobutylether-β-cyclodextrin (SBE-β-CD) as the selector was investigated with insight into a number of important process variables, such as the type of organic solvent, concentration of selector, pH, and temperature. Equilibrium of the extraction system was modeled using a reactive extraction modcl with a homogeneous aqueous phase reaction. The important parameters of this model were determined experimentally. The physical distribution coefficients for molecular and ionic CPE were determined as 0.3 and 8.93, respectively. The equilibrium constants of the complexation reaction with SBE-β-CD were determined as 152 and 110 L/mol for R- and S-CPE, respectively. Results show that the experimental data agree with the model predictions perfectly. Comprehensively considering the experiment and model, the extraction conditions are optimized and the best extraction conditions are: pH of 6.0, SBE-β-CD concentration of 0.04 tool/L, and temperature of 5 ℃, providing the enantioselectivity (a) of 1.25, the fraction of R-CPE (φR) in aqueous phase of 0.71 and performance factor (pf) of 0.025. 展开更多

Potentiometric experiments were carried out on the proton binding equilibria of FA extracted from a weathered coal and HA and Fa extracted from a dark loessial soil.The affinity spectrum model was employed to treat the experimental data.The affinity spectrum model technique could“magnify” the heterogeneity of the proton binding equilibria.so it was useful for comparing and studying the characteristics of humic substances with similar properties.According to the affinity spectra,we also found that the direction of the titration could affect the properties of the equilibria of FA from the weathered coal,and the acidic functional groups contained in FA from the weathered coal were larger in quantity than those contained in HA and FA from the dark loessial soil. 展开更多

The inheritance mode of seven microsatellite markers was investigated in Patinopecten yessoensis larvae from four con-trolled crosses,and the feasibility of using these markers for kinship estimation was also examined. All the seven microsatellite loci were compatible with Mendelian inheritance. Neither sex-linked barriers to transmission nor major barriers to fertilization between gametes from the parents were evident. Two of the seven loci showed the presence of null alleles in two families,suggesting the need to conduct comprehensive species-specific inheritance studies for microsatellite loci used in population genetic studies. However,even if the null allele heterozygotes were considered as homozygotes in the calculation of genetic distance,offspring from four families were all unambiguously discriminated in the neighbor-joining dendrogram. This result indicates that the microsatellite markers used may be capable of discriminating between related and unrelated scallop larvae in the absence of pedigree information,and of investigating the effective number of parents contributing to the hatchery population of the Japanese scallop. 展开更多

A non-equilibrium chromatographic rate model was employed to simulate the affinity chromatography of urokinase. The chromatography process was developed to a yield of high purity product of urokinase from crude materials. The affinity gel used in the process was prepared by an epichlorohydrin-activation method using epichlorohydrin activated Sepharose 4B as a matrix and p-aminobenzamidine as a ligand. The chromatographic process were numerically simulated and analyzed with the aid of VERSE-LC computer simulator. Considering the basic principles, rate model with the back mixing in column inlet was utilized in simulating and studying the effect of the column inlet pattern on other parameters. Comparison of the simulation results with the experimental data showed that the rate model can be used to describe the affinity chromatography of urokinase in a fixed bed column with satisfactory accuracy. 展开更多

Thymidylate synthase (TS, E.C.2.1.1.45) catalyzes a critical reaction in the only pathway of de novo synthesis of thymidylate (dTMP) in human cells, and is an important target of chemotherapy. To evaluate the inhibitory activities of novel compounds to TS, a convenient method of activity assay using 6x His-tagged recombinant human TS (rhTS) was established and 49 novel synthetic folate analogues were screened to discover potential TS inhibitors. During the process, 4 novel compounds were found to effectively inhibit TS, while the IC 50 of a positive control raltitrexed was 3.4 μM in this assay. 展开更多
