采用基因工程方法将HPV16E6、E7基因融合后插入痘苗病毒载体,通过同源重组构建表达人乳头瘤病毒16型E6/E7融合蛋白的非复制型重组痘苗病毒疫苗,用C57BL/6小鼠观察其免疫原性和抗肿瘤移植情况。测序结果表明融合的HPV16E6、E7基因序列与... 采用基因工程方法将HPV16E6、E7基因融合后插入痘苗病毒载体,通过同源重组构建表达人乳头瘤病毒16型E6/E7融合蛋白的非复制型重组痘苗病毒疫苗,用C57BL/6小鼠观察其免疫原性和抗肿瘤移植情况。测序结果表明融合的HPV16E6、E7基因序列与设计相符;构建的非复制型重组痘苗病毒经Dot blot鉴定,显示有E6、E7融合基因的插入;Western blot检测表明该重组病毒在鸡胚成纤维细胞中能表达HPV16型E6/E7融合蛋白。动物免疫试验表明,该重组病毒在小鼠体内可诱发E6、E7特异性抗体;被免疫小鼠能抵抗TC-1肿瘤细胞的攻击。此结果为将来进一步研制HPV16、18型联合疫苗打下了基础。 展开更多

宫颈环形电切除术在治疗宫颈高度鳞状上皮内病变中的价值

目的探讨宫颈环形电切除术(LEEP)治疗宫颈高度鳞状上皮内病变(≥CIN2)临床价值及术后随访途径。方法随机选取我院宫颈门诊,阴道镜下宫颈活检病理诊断≥CIN2患者113例,均行宫颈LEEP治疗,切除术后标本均送病理学检查,并利用HPV及阴道镜进... 目的探讨宫颈环形电切除术(LEEP)治疗宫颈高度鳞状上皮内病变(≥CIN2)临床价值及术后随访途径。方法随机选取我院宫颈门诊,阴道镜下宫颈活检病理诊断≥CIN2患者113例,均行宫颈LEEP治疗,切除术后标本均送病理学检查,并利用HPV及阴道镜进行术后随访。结果 LEEP术前术后病理结果一致者54例,级别下降者34例,级别上升者25例。其中级别上升者有8例为宫颈早期浸润癌,术前高危型HPV感染102例,其中单独感染68例,复合感染34例。术后6个月HPV转阴率85.29%、12月转阴率97.06%,差异有统计学意义(P<0.05)。持续性高危型HPV感染者,阴道镜下宫颈活检术+宫颈管搔刮发现病变残留占40%。结论 LEEP治疗宫颈高度鳞状上皮内病变安全经济有效,但由于宫颈病变的多灶性,术后可采用高危型HPV监测联合阴道镜下活检+宫颈管搔刮术,以尽早发现病变残留和(或)复发。 展开更多

OBJECTIVE To investigate the detection rate of humanpapilloma virus (HPV) DNA in the Kazakh esophageal carcinoma(EC) patients of Xinjiang.METHODS We detected the prevalence of a HPV gene in tumortissues from 318 esophageal squamous cell carcinoma (ESCC).Tumor tissues were kept in formalin and embedded in paraffin.One hundred seventeen samples used crude cell suspension, whilethe other 201 used the method of DNA extraction with phenol-Tris/chloroform. We analyzed the relevance to EC of Kazakh's inXinjiang.RESULTS In the ESCC samples of Kazakh's in Xinjiang, totaldetection rate for HPV DNA was 64.5% (205/318). The positiverate of HPV in group of crude cell suspensions was 82.9% (97/117)compared with the rate of 53.7% (108/201) in the group of DNAextraction. The results in the two groups showed significantdiffference (x^2 = 5.711, P < 0.05).CONCLUSION HPV DNA infection may be one of the mostimportant factors related to EC of Kazakh's in Xinjiang. 展开更多

Objective To detect HPV16 E6, E7 genes and cell-type specific enhancer (CTSE) of long control region (LCR) in breast carcinoma (BC). Methods HPV16 E6 ,E7 genes and CTSE were detected in 40 BCs and 20 normal breast tissue (NBT) using polymerase chain reaction (PCR). Results The positive rates of HPV16 E6 , E7genes and CTSE were 60% ( 24/40 ) , 55% ( 22/40 ) and 67. 5% ( 27/40 ) respectively in BCs, whereas only 5% (1/20), 5% (1/20) and 15% (3/20) in NBTs ( P 〈 O. 05). There exited significant correlation between E6 gene and CTSE in BCs ( P 〈0. 05) , as well as E7 gene and CTSE. The infection of HPV16 E6, E7 and CTSE had no statistic relationship with pathological features. Conclusion There were HPV16 E6 , E7 genes and CTSE together in BCs and CTSE may play an important role in pathogenesis of BC. 展开更多
