The increase of carbon emissions induced by human economic activi- ties has become a major driving force behind global climate change. Thus, as a matter of social responsibility, reasonable carbon con- straints should be implemented to ensure environmental security and sustainable development for every country. Based on a summary of studies that examined the relationship between carbon emissions and regional development, this paper shows that human activity-led carbon emission is caused by the combination of several influencing factors, including population size, income level, and technical pro- gress. Thus, a quantitative model derived from IPAT-ImPACT-Kaya series and STIRPAT models was established. Empirical analysis using multivariate nonlinear regression demonstrated that the origins of growing global carbon emission included the increasing influ- encing elasticity of the population size and the declining negative effect of technical progress. Meanwhile, in context of classification of country groups at different income levels, according to the comparison of fluctuating patterns of the influencing elasticity, technical progress was found as the main factor influencing carbon emission levels in high-income countries, and population size might he the controlling factor in middle-income countries. However, for low-income countries, the nonlinear relationship between carbon emission and its influencing factors was not significant, whereas population growth was identified as an important potential driving force in future carbon emissions. This study can therefore provide a reference for the formulation of policies on carbon constraints, especially to de- velop more efficient carbon mitigating policies for countries at different income levels.展开更多

A human aFGF gene was cloned into the intracellular expression vecto...Pichia pastoris expression system is similar to that of the mammal cell in modification of expressed protein, including refolding and glycosylation. A human aFGF gene was cloned into the intracellular expression vector pPIC9K. The Pichia pastoris KM71 strain was transformed with the recombined expression plasmid. Transgenic expression was observed after screening the transformants with G418. The expression and secretion of recombinant human aFGF (rhaFGF) into the culture medium were testified by ELISA assay. The yield peaked after two days of induction and was approximately 10 mgL-1 in shake-flask fermentation medium. The recombinant proteins were purified by the combination of heparin-Sepharose affinity chromatography and gel filtration chromatography. Two proteins with relative molecular masses (Mr) of 17 000 and 35 000 were purified as a single band in SDS-PAGE, whose biological activities were determined by MTT assay. It is found that the protein with Mr of 17 000 is nonglycosylated haFGF, and that with Mr of 35 000 is glycosylated haFGF; and the latter has a lower biological activity than the former.展开更多

Methods: In the Zhongshan City where there is highly incidence rate of NPC, we chose the members of the NPC clustering families as objects, and the patients of nasopharyngitis and NPC as the control group. We isolated the RNA from the nasopharyngeal tissue, and synthesized its cRNA, the genome stability and DNA repair genes chip technique, chemiluminescent detection and real-time fluorescence quantita- tive technique were used to examine the genome stability and DNA repair genes in the nasopharyngeal tissue. Results: More genome stability and DNA repair genes were up-regulated in the members of the NPC clustering families than the NPC patients, and the range of up-regulated was high, with the over up-regulated 100 times genes including TEP1, MSH4, PMS2LI. Fewer genome stability and DNA repair genes were down-regulated in the members of the NPC clustering families than the NPC patients, the ubiquitin genes almost were down-regulated, the results also could be confirmed by real-time fluorescence quantitative PCR. Conclusion: There are specially expression character of genome stability and DNA repair genes in the members of NPC clustering families.展开更多
To obtain the recombinant soluble protein of the extracellular fragment of human TRAIL gene and to identify its function preliminarily, this gene fragment was amplified from peripheral blood mononuclear cells (PBMC)...To obtain the recombinant soluble protein of the extracellular fragment of human TRAIL gene and to identify its function preliminarily, this gene fragment was amplified from peripheral blood mononuclear cells (PBMC) by RT-PCR and cloned into vector pGEM-T-Easy for sequence analysis. The expression vector pET-30a/TRAIL was then constructed by DNA recombination method with a His-tag gene at the front of the TRAIL fragment, and the recombinant protein was expressed in E. coli BL21 (DE3). Meanwhile, the expressed target protein was purified with Ni-NTA chromatography column and identified by SDS-PAGE and Western blotting. The proliferation inhibition activity of TRAIL-His was detected by MTF assay. PI staining and Wright-Giemsa staining were used to detect the presence of the TRAIL-induced cell apoptosis. It was demonstrated that the target protein expressed in E. coli BL21 showed the same relative molecular mass as that the protein expected and could be recognized by both the anti-TRAIL polyclonal antibody and anti-His monoclonal antibody. In addition, this protein could also inhibit proliferation of human lymphoma cell line Jurkat cells or induce apoptosis of this cell line. It is apparent that a recombinant soluble TRAIL protein with biological activity is obtained and this prospective study can lay solid foundation for further research on the biological activity and application in the anti-tumor therapy.展开更多

The purpose of this study, to determine the contribution of the tourism sector in the country's economy and Russian tourist profile, was examined and investigated. Survey data collection techniques were used in the study. National and international researches were supported by literature. In this study, it was aimed to determine the role and the importance of Marmaris in the development of tourism in economy. It was also aimed to determine the cause of Russian tourists preferring Marmaris. The 12 questions in the questionnaire are to determine the demographic and travel characteristics, four questions are to determine the resources of accommodation and information of Russian tourists, and the questionnaire was completed with a question which was asked to determine the reason of preferring Marmaris, It was graded with the five-point Likert scale. The results of the study show the profile of Russian tourists who prefer Marmaris. It was examined that they have come up with all-inclusive system, they have two-week holiday term, they gather information through travel agencies, and Marmaris has an intensive revisit frequency. Marmaris is expressed as a great holiday destination by Russian tourists. Accordingly, Marmaris is popular among Russian tourists as a tourist destination and is considered to be quite suitable for the holidays.展开更多

Research suggests that genes are not in independent existence and the expression of a gene will promote or inhibit the expression of another gene; if the expression of a gene makes the biochemical environment of ceils changed, the expression of a series of genes will be affected. In order to get a better understanding of the relationship between genes, all sorts of gene regulatory network models have been established by scientists. In this paper, a variety of gene regulatory networks are first introduced according to the process of this subject research, and then the most basic network (i.e. Boolean network) is emphatically analyzed, and then a new method (i.e. Boolean network based on the theory of circuit) to describe Boolean network is drawn forth. After the shortcomings of the Boolean network proposed in the past are analyzed, a simulation circuit Boolean model is established using EDA technology in order to improve the Boolean network.展开更多

The values and changing roles of the indices are the foundations of coal and gas outbursts prediction. But now, only the data of ere key monitoring station is used in the coal and gas outbursts prediction practice, and the other data are ignored. In order to overcome the human factor and make full use of the monitoring information, the technique of multi-sensor target tracking is proposed to deal with the microseismic informatiion. With the results of microseismic events, the activities of geological structure, fracure-depth of roof and floor, and the location of gas channel are obtained. These studies indicate that it is considerably possible to predict the coal and gas outbursts using microseismic monitoring with its inherent ability to remotely monitor the progressive failure caused by mining.展开更多
