Methods: We took count of HeLa cells in different i...Objective: To investigate the expressions of p53, RB1, Fas, c-fos, Ras, EGFR mRNA in human cervical cancer (HeLa) cell in response to the trichostatin A (TSA). Methods: We took count of HeLa cells in different incubation times with TSA (0.2μm/L). The result indicated that HeLa cells changed evidently when HeLa cells were incubated for 36 h. Then, we investigated the genes expression (mRNA levels) of HeLa cells after treatment for 36 h using SYBR green real-time PCR. Results: We demonstrated that trichostatin A (TSA) could make human cervical cancer (HeLa) cell morphological change and induce HeLa cell apoptosis. Furthermore, the data suggest that TSA-induced down-regulation of p53, RB1, Fas, but upregulated c-fos gene expression after treatment for 36 h, and Ras, EGFR did not show obvious response to TSA treatments. Conclusion: TSA has different effects on gene expression.展开更多

These technologies are not intended to substitute the human expert but to facilitate his task. The objective of this work is to develop a method for diagnosing cancer cervical smears using cervical-vaginal segmented to build the authors' database and a human supervisor and as an automatic tool manage and monitor the execution of the operation of diagnostic and proposing corrective actions if necessary. The Supervisor Smart is manufactured by the technique of neural networks with a success rate of 43.3% followed by the technique of fuzzy logic with a success rate equal to 56.7% and finally to improve this rate we used neuro-fuzzy approach which has a rate which reaches 94%.展开更多

The purpose of this study was to investigate theputative role of the Notch1 receptor in carcinogenesis and in theprogression of the cervical cancer. Since human papillomavirus(HPV) is a causative agent in cervical carcinoma, the interactionbetween Notch1 and HPV infection was examined.METHODS Forty cervical cancer samples and 30 normalcervical tissue specimens were examined using Western blot andRT-PCR to detect Notch1 protein and mRNA levels. HPV16 DNAwas examined in all samples using PCR.RESULTS The level of Notch1 protein expression wassignificantly lower in cervical cancer tissue than in normal tissue.Levels of Notch1 mRNA were found to be substantially downregulatedin cancer tissue. Notch1 protein expression levelswere significantly higher in carcinomas without HPV DNAthan that in carcinomas with HPV infection (55.5% vs. 3.3%, P <0.05). Down-modulation of Notch1 mRNA levels in carcinomawas demonstrated to be associated with HPVE6 transcription.Moreover, levels of Notch1 expression were shown to besignificantly higher in early stage disease than in advanced stagedisease (P = 0.001).CONCLUSION Down-modulation of Notch1 expressionprobably plays an important role in the late stages of HPVinducedcervical cancer.展开更多
