去整合素kistrin对人晶状体上皮细胞株凋亡的影响

目的探讨去整合素kistrin对人晶状体上皮细胞株SRA01/04的细胞凋亡的影响。方法取对数生长期的人晶状体上皮细胞株SRA01/04用于实验。分别用含有不同浓度(80μg.L-1、160μg.L-1、320μg.L-1)kistrin的完全培养基孵育8h、16h、32h,然后用流式细胞仪检测细胞凋亡情况,并与阴性对照组(kistrin浓度为0μg.L-1)比较。用含kistrin的终浓度为160μg.L-1的培养基孵育细胞,于24h、32h后进行吖啶橙染色,倒置相差荧光显微镜观察细胞凋亡形态学变化,并与对照组(kistrin浓度为0μg.L-1)比较。结果kistrin作用8h时,阴性对照组与80μg.L-1、160μg.L-1、320μg.L-1kistrin孵育细胞组细胞凋亡率分别为(1.29±0.43)%、(5.48±0.66)%、(11.23±1.16)%、(20.77±1.46)%,kistrin作用16h时不同组别细胞凋亡率分别为(3.04±0.12)%、(7.68±0.23)%、(19.76±2.22)%、(32.36±3.29)%,kistrin作用32h时不同组别细胞凋亡率分别为(4.75±0.08)%、(72.15±7.04)%、(54.09±3.77)%、(43.27±3.32)%;在每个时间点各浓度的细胞凋亡率与阴性对照组相比,差异均有统计学意义(P均<0.01)。与对照组相比,浓度为160μg.L-1的kistrin作用24h后,细胞形态可见改变,kistrin作用32h后细胞出现明显的各种凋亡形态学改变。结论去整合素kistrin对人晶状体上皮细胞株SRA01/04有诱导凋亡的作用,为药物防治后发性白内障提供了新思路。
