目的 建立能够同时作用于炎症相关细胞因子的药物筛选模型 ,探讨多指标综合筛选方法 ,为发现新的抗炎先导化合物提供快速有效的方法。方法 采用人血白细胞为研究对象 ,以脂多糖 (LPS)刺激诱发炎性因子生成 ,探讨最佳刺激条件 ,建立多... 目的 建立能够同时作用于炎症相关细胞因子的药物筛选模型 ,探讨多指标综合筛选方法 ,为发现新的抗炎先导化合物提供快速有效的方法。方法 采用人血白细胞为研究对象 ,以脂多糖 (LPS)刺激诱发炎性因子生成 ,探讨最佳刺激条件 ,建立多指标检测方法和综合评价药物作用的分析方法。观察不同浓度脂多糖 (0 5~ 5 0mg·L-1)及不同作用时间 (0~ 4h)炎性因子IL 1、IL 8、TNF α生成水平的变化。同时观察 5 LO抑制剂NDGA (nordihydroguaiareticacid)和COX 2抑制剂Diclofenac在多种细胞因子筛选模型上的应用。结果 脂多糖可浓度依赖性 (0 5~ 5 0mg·L-1)和时间依赖性 (0~ 4h)地刺激人血白细胞释放IL 1、IL 8和TNF α。脂多糖 (终浓度 5mg·L-1)刺激人血白细胞 4h有较高释放 ,与对照组比差异明显。NDGA和Diclofenac在脂多糖刺激 4h对炎性因子释放有显著抑制作用。结论 这种脂多糖体外刺激人血白细胞释放多种炎性因子的筛选模型可用于高通量大规模筛选具有抗炎作用的先导化合物 。 展开更多

目的 探讨食管癌术后乳糜胸的原因及治疗方法。方法 分析 8例食管癌术后发生乳糜胸者的手术过程及术后引流量等相关指标。结果 本组 8例中 7例行主动脉弓上吻合 ,1例主动脉弓下吻合。术后病理均为鳞状细胞癌 ;其中T11例 ,T2 2例 ,T3 ... 目的 探讨食管癌术后乳糜胸的原因及治疗方法。方法 分析 8例食管癌术后发生乳糜胸者的手术过程及术后引流量等相关指标。结果 本组 8例中 7例行主动脉弓上吻合 ,1例主动脉弓下吻合。术后病理均为鳞状细胞癌 ;其中T11例 ,T2 2例 ,T3 5例 ;N13例 ,N0 5例 ,M0 8例。术后引流量 2 4 1~ 16 4 1ml/d ,平均 5 82ml/d ,白蛋白总用量 0~ 5 2 0 g ,血浆总用量 4 0 0~ 10 0 0 0ml;5例行二次开胸胸导管结扎术治疗 ,两次手术间隔时间 12~34d。 3例保守治疗 ,胸腔注射重组人血白介素Ⅱ。全部病例均治愈。结论 肿瘤外侵处、主动脉弓后和弓上部位胸导管易受损伤 ,此为产生乳糜胸的主要原因 ,手术部位的良好显露在一定程度上可避免胸导管损伤。胸腔引流量平均在 30 0ml/d以上 ,保守治疗 1周无好转者 。 展开更多

The cDNA fragment of human TRAIL (TNF-related apoptosis inducing ligand) was cloned into RevTet-On, a Tetregulated and high-level gene expression system. The gene expression system was constructed in a human leukemic cell line: Jurkat. By using RevTet-On TRAIL gene expression system in Jurkat as a cell model, we studied the influence of TRAIL gene on the changes of cellular apoptosis before and after the TRAIL gene expression, which was induced by adding tetracycline derivative doxycycline (Dox). The results indicated that the cellular apoptosis ratio was largely dependent on the TRAIL gene expression level. Moreover, it was found that the apoptosis-inducing TRAIL could cause significant changes in the biophysical properties of Jurkat cells. The cell surface charge density decreased, the membrane fluidity declined, the elastic coefficients K_I increased, and the proportion of α-helix in membrane protein secondary structure decreased. Thus, the apoptosis-inducing TRAIL gene caused significant changes on the biomechanic properties of Jurkat cells. 展开更多

HEL cells, a human erythroleukemia cell line, mainly express the fetal (r)globin gene and trace amount of the embryonic (E)globin gene, but not adult (B) globin gene. Here we show that hydroxyurea (HU) can induce HEL cells to express adult (B) globin gene and lead these cells to terminal differentiation. Results showed in Gel mobility shift assays that GATA factors could specifically bind to the regulatory elements of human B- globin gene, including the proximal regulatory element (the B- promoter) and the distal regulatory elements (the DNase I hypersensitive sites in the LCR, HS2-HS4 core sequences). However, the DNA binding patterns of GATA factors were quite different between HU-induced and uninduced HEL cells. Western-blot analysis of nuclear extracts from both the uninduced and HU- induced HEL cells revealed that the level of GATA-2 transcription factor decreased, whereas the level of GATA-1 transcription factor increased following the time of hydroxyurea induction. Furthermore, using RT-PCR analysis the expression of human B-globin gene in HU-induced HEL cells could be blocked again when HEL cells were incubated in the presence of antisense oligonucleotides for hGATA-1, suggesting that the upregulation of hGATA-1 transcription factor might be critical for the expression of human β- globin gene in HU-induced HEL cells. 展开更多

Multiple lymphomatous polyposis (MLP) is an unusual form of non-Hodgkin's lymphoma characterized by polyps throughout the gastrointestinal tract. It has been reported that most MLP are observed in cases with mantle cell lymphoma of B-cell type. We herein present a case of a 66-year-old man with adult T-cell leukemia/lymphoma (ATLL). Colonoscopy revealed MLP throughout the colon and histopathological findings of ATLL cell infiltration. The patient died despite combination of chemotherapy. The literature of manifestations of colonic involvement of ATLL isreviewed and the importance of endoscopic evaluation to differentiate ATLL intestinal lesions from opportunistic infectious enterocolitis is discussed. 展开更多

AIM: To determine whether alteration of the mito- chondria DNA (mtDNA) copy number and its oxidative damage index (mtDNA△CT) can be detected by analysis of peripheral blood cells in hepatitis C virus (HCV)- infected patients. METHODS: This study enrolled two groups of pa- tients aged 40-60 years: a control group and an HCV- infected group in Department of Gastroenterology and Hepatology in Changhua Christian Hospital. Patients with co-infection with hepatitis B virus or human im- munodeficiency virus, autoimmune disease, malignant neoplasia, pregnancy, thyroid disease, or alcohol con- sumption 〉 40 g/d were excluded. HCV-infected pa- tients who met the following criteria were included: (1) positive HCV antibodies for 〉 6 mo; (2) alanine aminotransferase (ALT) levels more than twice the upper lim- it of normal on at least two occasions during the past 6 mo; and (3) histological fibrosis stage higher than F1. The mtDNA copy number and oxidative damage index of HCV mtDNA (mtDNA△CT) were measured in periph- eral blood leukocytes. The association between mtDNA copy number and mtDNA△CT was further analyzed using clinical data. RESULTS: Forty-seven normal controls (male/female: 26/21, mean age 50.51 ± 6.15 years) and 132 HCV- infected patients (male/female: 76/61, mean age 51.65 ± 5.50 years) were included in the study. The geno- types of HCV-infected patients include type 1a (n = 3), type 1b (n = 83), type 2a (n = 32), and type 2b (n = 14). Liver fibrosis stages were distributed as follows: F1/F2/F3/F4 = 1/61/45/25 and activity scores were A0/ A1/A2/A3 = 7/45/55/25. There were no age or gender differences between the two groups. HCV-infected pa- tients had higher hepatitis activity (aspartate transami- nase levels 108.77 ± 60.73 vs 23.19 ± 5.47, P 〈 0.01; ALT levels 168.69 ± 93.12 vs 23.15 ± 9.45, P 〈 0.01) and lower platelet count (170.40±58.00 vs 251.24 ± 63.42, P 〈 0.01) than controls. The mtDNA copy num- ber was lower in HCV-infected patients than in controls (173.49 vs 247.93, P 〈 0.05). The mtDNA△CT was higher in HCV-infected patients than in controls (2.92 vs 0.64, P 〈 0.05). To clarify the clinical significance of these results in HCV-infected patients, their association with different clinical parameters among HCV-infected pa- tients was analyzed. A negative association was found between mtDNA copy number and elevated aspartate transaminase levels (r = -0.17, P 〈 0.05). Changes in mtDNA copy number were not associated with HCV RNA levels, HCV genotypes, liver fibrosis severity, or inflammatory activity in the liver biopsy specimen. How- ever, a correlation was observed between mtDNA△CT and platelet count (r = -0.22, P 〈 0.01), HCV RNA level (r = 0.36, P 〈 0.01), and hepatitis activity (r = 0.20, P = 0.02). However, no difference in the change in mtDNA△CT was observed between different fibrosis stages or HCV CONCLUSION: Oxidative stress and mtDNA dam- age are detectable in patient's peripheral leukocytes. Increased leukocyte mtDNA△CT correlates with higher HCV viremia, increased hepatitis activity, and lower platelet count. 展开更多

In present study we studied the effect of all-trans retinoic acid(ATRA) and dimethylsulfoxide (DMSO) on the induction of apoptosis in HL-60 cell line. Based on morphological changes by Hochest 33342 staining and identification of internucleosomal DNA cleavage by gel electrophoresis, we observed aberrant nuclear chromatin eondensation and ladder-like pattern of DNA degradation. Using Flow Cytometric method, we found sub-G1 peak in RA-treated HL-60 cells starting 5 to 6 d after the initiation of the treatment. However, such an obvious apoptotic peak was not identified in DMSO-differentiated cells. Combining the research accomplished before, our study approves further that apoptosis could be a common mode of death of terminally differentiated HL-60 cells. 展开更多

This growth factor isoform contains exon 7, which is essential for binding to extracellular domain of VEGF receptor 2, located on endothelial cells lining the surface of blood vessels. This binding stimulates the cascade of downstream signalling events leading to process known as angiogenesis, hVEGF165 overexpressed with His-tag in BL21 E. coli cells forms inclusion bodies (insoluble protein), so the research found the procedure for its solubilization and purification on a Nickel based affinity chromatography. Although this eukaryotic signal protein needs posttranslational processing for its full function as a homodimer, author verified the biological activity of our hVEGF165 protein, obtained as monomer, by wound healing test. 展开更多
