The results showed that cry1A ha...We investigated the inheritance and expression of cry1A gene in transgenic maize ( Zea mays L.) by Southern blotting analysis and enzyme_linked immunosorbent assays (ELISA). The results showed that cry1A had been transmitted to progeny of transgenic maize as a single gene. Contents of cry1A insecticidal protein were significantly different among transgenic maize lines and various tissues of the same transgenic lines. High expression of cry1A protein occurred in green tissues, such as leaf and husk leaf, and low expression occurred in pith, tassel, ear pith, pollen and silk. The results also showed that the contents of cry1A insecticidal protein in leaves of transgenic maize increased with the advance of development and there was no significant difference in cry1A expression level among various generations of transgenic maize.展开更多
Electron transport system (ETS ) / dehydrogenase activity in a paddy field soil was measured under a variety of incubation conditions using the reduction of 2- (p-iodophenyl- 3- (p-nitrophenyl ) -5- phellyl tetrazoliu...Electron transport system (ETS ) / dehydrogenase activity in a paddy field soil was measured under a variety of incubation conditions using the reduction of 2- (p-iodophenyl- 3- (p-nitrophenyl ) -5- phellyl tetrazolium chloride (INT) to iodonitrotetrazolium formazan (INTF). The results exhibited a high positive correlation between the ETS activity and the incubation temperature and soil moisture. Dehydrogenase/ETS activity displayed a negative correlation with insecticide concentrations, and the activity affected adversely as the concentration of the insecticide increased. The higher doses, 5 and 10 field rates (1 field rate ~ 1500 mL ha-1), of insecticide significantly inhibited ETS activity, while lower rates failed to produce any significant reducing effect. Inorganic N (as urea) of concentrations from 0 to 100 ug N g-1 soil showed a positive response to ETS activity. However, at concentrations of 200 and 400 ug N g-1, the activity was reduced significantly.展开更多

The cytotoxicity of carbaryl to FG cells was ...In this study, we determined the cytotoxicity and genotoxicity of carbamate insecticide carbaryl to flounder gill (FG) cells and its teratogenicity to zebrafish embryos. The cytotoxicity of carbaryl to FG cells was determined with methods including MTT and neutral red uptaking (NRU), lactate dehydrogenase (LDH) releasing and Hoechst 33342 and propidiurn idodide (PI) double staining. Moderate cytotoxicity in a concentration-dependent manner was observed. The 24h-IC50 value of 53.48 ± 1.21, 59.13 ± 1.19 and 46.21 ± 1.24 mgL 1 carbaryl was obtained through MTT, NRU and LDH assays, respectively. Double fluorescence staining dem- onstrated that carbaryl induced the death of FG cells mainly through necrosis. There was no significant genotoxicity found in the FG cells exposed to the highest testing concentration of carbaryl (20mgL-1, P〉0.05) as was demonstrated by Comet assay. Zebrafish embryos exposed to carbaryl at concentrations 〉10 mg L-1 displayed moderate toxic effects on the survival, spontaneous movement, hatching, heart rates of the embryos and their development, which were evidenced by yolk and pericardial sac edemas, body length reduction and tail flexure in time- and concentration-dependent manners at specific stages. The 24h-, 48h- and 96h-LC50 values of carbaryl to zebra fish embryos were 41.80 ± 1.10, 17.80 ± 1.04 and 14.46 ± 1.05 mg L-1, respectively. These results suggested that car- baryl is moderately toxic to FG ceils cultured in vitro and zebrafish embryos, and the FG cells were similar to zebrafish embryos in their sensitivity to carbaryl as 24h-IC50 and LC50 indicated.展开更多

Therefore, it is important to investigate the ...Leafhopper Cicadulina bipunctata is represented the main insect as a pathogen for phytoplasma disease occurring by insect-transmitted plant viruses in date palm orchards. Therefore, it is important to investigate the potential effect of some insecticides against such insect. The adults of leafhopper C. bipunctata were collected from date palm orchards in Alhasa, Eastern province, Saudi Arabia. Three insecticides from different classes--beta-cyfluthrin (pyrethroids), imidacloprid (neonicotinoids) and abamectin (natural compounds)--have been evaluated in vivo against adults C. bipunctata. This stage was exposed to residual film of various concentrations of each insecticide on transparent plastic cups using a Potter precision laboratory spray tower. Bioassay test showed that both beta-cyfluthrin and imidacloprid caused 100% mortality by 500 ppm at 24 h after treatment, whereas abamectin gave the same mortality by 50 ppm at the same time. Toxicity values revealed that abameetin was the most potent insecticide compared with beta-cyfluthrin and imidacloprid, where the lethal concentrations LC50 and LC95 were 24.58 ppm and 116.73 ppm at 3 h after treatment, respectively. Therefore, abamectin can be a possible candidate to be applied on date palm or ground grass by the Ministry of Agriculture after successful field experiments.展开更多
In order to determine the combined effects of migration and gene flow on evolution of insecticide resistance in the mosquito Culex pipiens, four samples were collected in China, among which two were collected along th...In order to determine the combined effects of migration and gene flow on evolution of insecticide resistance in the mosquito Culex pipiens, four samples were collected in China, among which two were collected along the railway from Beijing to Guangzhou. Bioassay data showed that the resistance levels of the four populations to dichlorvos were high and to parathion moderate as compared with the susceptible strain and there was no significant difference among the four populations to the same organophosphate (OP) insecticide. Starch electrophoresis was done to identify the frequency of known overproduced esterases and to analyze genetic diversity among various populations by electrophoretic polymorphism of five presumably neutral loci. The results indicated that the gene flow between populations existed and the number of effective migrants (Nm) was related to collection geography (Nm from 1.67 to 40.07). In contrast with lower genetic differentiation between two nearby populations (between GZ and ZS, ZZ and SQ) and higher genetic differentiation between two distant populations (between GZ and ZZ), there was a significant and inconsistent difference in the distribution of resistance alleles, A2-B2 when explained only with active migration. This divergent situation could be straightened out when considering passive migration (such as railway transport) which increased the spread of A2-B2 along the railway, i.e., in GZ and ZZ. The resistance alleles, A2-B2, dispersing to around areas by active migration suffered from the limitation of gene flow and the speed of invasion.展开更多

Fully inactivating SARS-Co V-2, the virus causing coronavirus disease 2019, is of key importance for interrupting virus transmission but is currently performed by using biologically or environmentally hazardous disinf...Fully inactivating SARS-Co V-2, the virus causing coronavirus disease 2019, is of key importance for interrupting virus transmission but is currently performed by using biologically or environmentally hazardous disinfectants. Herein, we report an eco-friendly and efficient electrochemical strategy for inactivating the SARS-Co V-2 using in-situ formed nickel oxide hydroxide as anode catalyst and sodium carbonate as electrolyte. At a voltage of 5 V, the SARS-Co V-2 viruses can be rapidly inactivated with disinfection efficiency reaching 95% in only 30 s and 99.99% in 5 min. Mass spectrometry analysis and theoretical calculations indicate that the reactive oxygen species generated on the anode can oxidize the peptide chains and induce cleavage of the peptide backbone of the receptor binding domain of the SARS-Co V-2 spike glycoprotein, and thereby disables the virus. This strategy provides a sustainable and highly efficient approach for the disinfection of the SARS-CoV-2 viruliferous aerosols and wastewater.展开更多
